    Calcification depth of deep-dwelling planktonic foraminifera from the eastern North Atlantic constrained by stable oxygen isotope ratios of shells from stratified plankton tows

    Stable oxygen isotopes (delta O-18) of planktonic foraminifera are one of the most used tools to reconstruct environmental conditions of the water column. Since different species live and calcify at different depths in the water column, the delta O-18 of sedimentary foraminifera reflects to a large degree the vertical habitat and interspecies delta O-18 differences and can thus potentially provide information on the vertical structure of the water column. However, to fully unlock the potential of foraminifera as recorders of past surface water properties, it is necessary to understand how and under what conditions the environmental signal is incorporated into the calcite shells of individual species. Deep-dwelling species play a particularly important role in this context since their calcification depth reaches below the surface mixed layer. Here we report delta O-18 measurements made on four deep-dwelling Globorotalia species collected with stratified plankton tows in the eastern North Atlantic. Size and crust effects on the delta O-18 signal were evaluated showing that a larger size increases the delta O-18 of G. inflata and G. hirsuta, and a crust effect is reflected in a higher delta O-18 signal in G. truncatulinoides. The great majority of the delta O-18 values can be explained without invoking disequilibrium calcification. When interpreted in this way the data imply depth-integrated calcification with progressive addition of calcite with depth to about 300m for G. inflata and to about 500m for G. hirsuta. In G. scitula, despite a strong subsurface maximum in abundance, the vertical delta O-18 profile is flat and appears dominated by a surface layer signal. In G. truncatulinoides, the delta O-18 profile follows equilibrium for each depth, implying a constant habitat during growth at each depth layer. The delta O-18 values are more consistent with the predictions of the Shackleton (1974) palaeotemperature equation, except in G. scitula which shows values more consistent with the Kim and O'Neil (1997) prediction.     Strategies for PS processing of large sentinel-1 datasets

    Several advanced DInSAR techniques have been used to map surface deformations due to volcanism, active tectonics, landslides, subsidence, and uplift as well as to monitor the deformation of critical infrastructure such as bridges and dams. Recently, studies have explored the potential of these techniques to be integrated into a permanently operating monitoring system. ESA’s Sentinel-1 satellites have been providing SAR images for such a purpose since 2014. Nowadays, it is easy to access more than 230 SAR images of any area of interest, and update this dataset every six days with a new image. Due to the high frequency of the data acquisition, the question arises on how to best handle such a dataset. Is it suitable to always consider the whole available dataset or would a partial processing of the dataset and combining the results at a later point be more appropriate? To answer these questions, three different processing strategies are investigated in this paper. The first is a continuously growing dataset and for the second and third strategy, the dataset was divided into sub-stacks with and without overlap. In this study, the key parameters of each strategy are analyzed. In addition, the size of the sub-stacks is varied and the results are compared

    Volume-Function Analysis (LiMAx Test) in Patients with HCC and Cirrhosis Undergoing TACE-A Feasibility Study

    Background Transarterial chemoembolization (TACE) is an important therapy for hepatocellular carcinoma (HCC) in cirrhosis. In particular in advanced cirrhosis, post-TACE hepatic failure liver (PTHF) failure may develop. Currently, there is no standardization for the periinterventional risk assessment. The liver maximum capacity (LiMAx) test assesses the functional liver capacity, but has not been investigated in this setting. Aims The aim of this study was to prospectively evaluate periinterventional LiMAx and CT volumetry measurements in patients with cirrhosis and HCC undergoing repetitive TACE. Methods From 06/2016 to 11/2017, eleven patients with HCC and cirrhosis undergoing TACE were included. LiMAx measurements (n=42) were conducted before and after each TACE. Laboratory parameters were correlated with the volume–function data. Results The median LiMAx levels before (276±166 µg/kg/h) were slightly reduced after TACE (251 ±122 µg/kg/h; p=0.08). This corresponded to a median drop of 7.1%. Notably, there was a signifcant correlation between LiMAx levels before TACE and bilirubin (but not albumin nor albumin–bilirubin [ALBI] score) increase after TACE (p=0.02, k=0.56). Furthermore, a signifcantly higher increase in bilirubin in patients with LiMAx≤150 µg/kg/h was observed (p=0.011). LiMAx levels at diferent time points in single patients were similar (p=0.2). Conclusion In our prospective pilot study in patients with HCC and cirrhosis undergoing multiple TACE, robust and reliable LiMAx measurements were demonstrated. Lower LiMAx levels before TACE were associated with surrogate markers (bilirubin) of liver failure after TACE. Specifc subgroups at high risk of PTHF should be investigated. This might facilitate the future development of strategies to prevent occurrence of PTHF

    Factors controlling the depth habitat of planktonic foraminifera in the subtropical eastern North Atlantic

    Planktonic foraminifera preserved in marine sediments archive the physical and chemical conditions under which they built their shells. To interpret the paleoceano-graphic information contained in fossil foraminifera, the recorded proxy signals have to be attributed to the habitat and life cycle characteristics of individual species. Much of our knowledge on habitat depth is based on indirect methods, which reconstruct the depth at which the largest portion of the shell has been calcified. However, habitat depth can be best studied by direct observations in stratified plankton nets. Here we present a synthesis of living planktonic foraminifera abundance data in vertically resolved plankton net hauls taken in the eastern North Atlantic during 12 oceanographic campaigns between 1995 and 2012. Live (cytoplasm-bearing) specimens were counted for each depth interval and the vertical habitat at each station was expressed as average living depth (ALD). This allows us to differentiate species showing an ALD consistently in the upper 100m (e.g., Globigerinoides ruber white and pink), indicating a shallow habitat; species occurring from the surface to the subsurface (e.g., Globigerina bulloides, Globorotalia inflata, Globorotalia truncatulinoides); and species inhabiting the subsurface (e.g., Globorotalia scitula and Globorotalia hirsuta). For 17 species with variable ALD, we assessed whether their depth habitat at a given station could be predicted by mixed layer (ML) depth, temperature in the ML and chlorophyll a concentration in the ML. The influence of seasonal and lunar cycle on the depth habitat was also tested using periodic regression. In 11 out of the 17 tested species, ALD variation appears to have a predictable component. All of the tested parameters were significant in at least one case, with both seasonal and lunar cyclicity as well as the environmental parameters explaining up to >50% of the variance. Thus, G. truncatulinoides, G. hirsuta and G. scitula appear to descend in the water column towards the summer, whereas populations of Trilobatus sacculifer appear to descend in the water column towards the new moon. In all other species, properties of the mixed layer explained more of the observed variance than the periodic models. Chlorophyll a concentration seems least important for ALD, whilst shoaling of the habitat with deepening of the ML is observed most frequently. We observe both shoaling and deepening of species habitat with increasing temperature. Further, we observe that temperature and seawater density at the depth of the ALD were not equally variable among the studied species, and their variability showed no consistent relationship with depth habitat.     Heteroarylketones inhibit astroglial interleukin-6 expression via a STAT3/NF-κB signaling pathway

    <p>Abstract</p> <p>Background</p> <p>Elevated brain levels of the pleiotropic cytokine interleukin-6, which is mainly secreted from activated local astrocytes, contribute to pathological events including neuroinflammation and neurodegeneration. Thus, inhibition of pathological IL-6 expression provides a rationale strategy for targeting the onset or further progression of neurological disorders including Alzheimer's disease, multiple sclerosis, Parkinson's disease and traumatic brain injury. The purpose of this study was to identify and to characterize new potent inhibitors of astrocytic IL-6 expression for further therapeutic development of novel anti-inflammatory and neuroprotective drugs.</p> <p>Methods</p> <p>Oncostatin M (OSM)-treated human glioma U343 cells were used as model for induction of astrocytic IL-6 expression. This model was characterized by immunoblotting, siRNA technique, ELISA and qRT-PCR and used to screen low molecular weight compound libraries for IL-6-lowering effects. To validate bioactive compounds identified from library screens, bacterial lipopolysaccharide was used to induce IL-6 expression in cultivated primary astrocytes and in mice <it>in vivo</it>. To dissect underlying molecular mechanisms, protein extracts from OSM-treated U343 cells were analyzed by phospho-specific immunoblotting and immunocytochemistry as well as by co-immunoprecipitation.</p> <p>Results</p> <p>OSM-treatment (100 ng/ml; 24 h) led to 30-fold increase of IL-6 secretion from U343 cells. The temporal profile of IL-6 mRNA induction displayed a biphasic induction pattern with peak synthesis at 1 h (6.5-fold) and 16 h (5.5-fold) post stimulation. IL-6 protein release did not show that biphasic pattern and was detected as early as 3 h post stimulation reaching a maximum at 24 h. The screen of compound libraries identified a set of heteroarylketones (HAKs) as potent inhibitors of IL-6 secretion. HAK compounds affected the second peak in IL-6 mRNA synthesis, whereas the first peak was insensitive to HAK treatment. HAK compounds also suppressed lipopolysaccharide-induced IL-6 expression in primary murine astrocytes as well as in brain and plasma samples from lipopolysaccharide-treated mice. Finally, HAK compounds were demonstrated to specifically suppress the OSM-induced phosphorylation of STAT3 at serine 727 and the physical interaction of pSTAT3<sup>S727 </sup>with p65.</p> <p>Conclusion</p> <p>Heteroarylketone compounds are potent inhibitors of IL-6 expression <it>in vitro </it>and <it>in vivo </it>and may represent a new class of potent anti-inflammatory and neuroprotective drugs.</p

    WaterPower: studying physical and socio-political factors in water security

    This briefing paper presents the initial findings as well as the integrative approach of the project WaterPower. WaterPower is a four year research project that is currently in its second year. The project combines multiple scientific perspectives to study how social and biophysical factors are interacting to shape the various uses and flows of water in Accra. The city of Accra has been chosen as a case study due to its status as a coastal capital in the global south where major global trends such as climate change and rapid urbanization intersect. Water as a natural resource, we content, is not only of great importance for the sustainability of ecosystems and socioeconomic systems. Flows and uses of water are also bound to questions of politics, security and accessibility. However, in spite of these interrelated social and biophysical challenges we maintain that Water Studies as a field of research is still surprisingly disciplinary in its approach and scientific outlook. This briefing paper therefore pursues a twofold objective. On the one hand, we seek to show how the WaterPower research approach can contribute to a more integrated understanding of Water Studies in an urban context. On the other hand, we present our initial findings on how individual and collective capacities for responding to the multiple water-related pressures at hand can be strengthened. In sum, we emphasize that an ongoing and intensive exchange with stakeholders, policy-makers and interested parties in Ghana is essential for WaterPower’s integrative approach

    German general practitioners' self-reported management of patients with chronic depression

    Background: Patients with chronic depression (persisting symptoms for >= 2 years) are a clinically relevant group with extensive (co) morbidity, high functional impairment and associated costs in primary care. The General Practitioner (GP) is the main health professional attending to these patients. The aim of this study was to examine the GPs' perception on managing patients with chronic depression. Methods: We performed an explorative cross-sectional study with a systematic sample of GPs in central Germany. Source of data was a written questionnaire (46 items). Descriptive analysis was carried out. Results: Two hundred twenty (out of 1000;22%) GPs participated. 93% of the GPs distinguish between care for patients with chronic depression and acute depressive episode. 92% would recommend psychotherapeutic co-treatment to the chronically depressed patient. 52% of GPs would favour a general restraint on antidepressants (ADs) in older chronically depressed patients (>= 75 years) whereas 40% suggest long-term pharmacotherapy. If severe physical comorbidity is present GPs would be restrictive in prescribing ADs (65%) or would urgently refer to specialist psychiatric services (40%). In case of a comorbid anxiety disorder 66% of the GPs would suggest a combined psycho- und pharmacotherapy. If a substance use disorder coexists 84% would prefer urgent referrals to specialist services. Conclusions: Participating GPs report awareness towards chronic depression in their patients. Physical and mental comorbidity seem to play an important role in GPs' treatment decisions

    Two New Loci for Body-Weight Regulation Identified in a Joint Analysis of Genome-Wide Association Studies for Early-Onset Extreme Obesity in French and German Study Groups

    Meta-analyses of population-based genome-wide association studies (GWAS) in adults have recently led to the detection of new genetic loci for obesity. Here we aimed to discover additional obesity loci in extremely obese children and adolescents. We also investigated if these results generalize by estimating the effects of these obesity loci in adults and in population-based samples including both children and adults. We jointly analysed two GWAS of 2,258 individuals and followed-up the best, according to lowest p-values, 44 single nucleotide polymorphisms (SNP) from 21 genomic regions in 3,141 individuals. After this DISCOVERY step, we explored if the findings derived from the extremely obese children and adolescents (10 SNPs from 5 genomic regions) generalized to (i) the population level and (ii) to adults by genotyping another 31,182 individuals (GENERALIZATION step). Apart from previously identified FTO, MC4R, and TMEM18, we detected two new loci for obesity: one in SDCCAG8 (serologically defined colon cancer antigen 8 gene; p = 1.85610 x 10(-8) in the DISCOVERY step) and one between TNKS (tankyrase, TRF1-interacting ankyrin-related ADP-ribose polymerase gene) and MSRA (methionine sulfoxide reductase A gene; p = 4.84 x 10(-7)), the latter finding being limited to children and adolescents as demonstrated in the GENERALIZATION step. The odds ratios for early-onset obesity were estimated at similar to 1.10 per risk allele for both loci. Interestingly, the TNKS/MSRA locus has recently been found to be associated with adult waist circumference. In summary, we have completed a meta-analysis of two GWAS which both focus on extremely obese children and adolescents and replicated our findings in a large followed-up data set. We observed that genetic variants in or near FTO, MC4R, TMEM18, SDCCAG8, and TNKS/MSRA were robustly associated with early-onset obesity. We conclude that the currently known major common variants related to obesity overlap to a substantial degree between children and adults

    Ausweitung des Sojaanbaus in Deutschland durch züchterische Anpassung sowie pflanzenbauliche und verarbeitungstechnische Optimierung

    Die Arbeiten im Sojaforschungsprojekt waren erfolgreich und konnten wichtige Impulse für die Ausweitung des Sojaanbaus in Deutschland geben. So sind die entwickelten Stämme und Kreuzungsnachkommen eine Basis für den Aufbau einer eigenständigen deutschen Sojazüchtung. Die Sorten Korus und Protibus erwiesen sich als besonders geeignet für die Tofuherstellung. Die im Projekt entwickelte Labortofurei ist ein Züchtungsinstrument zur Identifikation vielversprechender Genotypen, mit dem auch die weitere Entwicklung frühreifer Tofusojasorten unterstützt werden kann. In Gefäßversuchen konnte gezeigt werden, dass die Reaktion auf Kühlestress während der Hülsenansatzphase zwischen den Sorten variiert und es tolerante, kompensierende und sensitive Sorten gibt. Die praktische Selektion auf Kältetoleranz war erfolgreich und für die Selektion auf Unkrauttoleranz konnte ein System etabliert werden. Bis auf das Präparat Radicin können die vorhandenen kommerziellen Bradyrhizobienpräparate für den Praxiseinsatz empfohlen werden. Die Hypothese, dass die Selektion des Symbiosepartners auf Kühletoleranz lohnenswert ist, wurde bestätigt. Bei der Sortenprüfung in ganz Deutschland zeigte sich, dass die Anbauwürdigkeit von Soja gut und nur an wenigen der geprüften Standorte nicht gegeben war. Die 00-Sorte ES-Mentor lieferte insgesamt die höchsten Relativerträge sowie den höchsten Rohproteinertrag, bei den 000-Sorten schnitt Sultana besonders gut ab. Eine Variation der Saatzeit sowie verschiedene Verfrühungstechniken erweisen sich nicht als ertragsrelevant. Beim Erfolg der Unkrautregulierung mit Torsionshacke, Fingerhacke und Flachhäufler gab es keine Unterschiede. Im Dammanbau lassen sich Sojabohnen mit gutem Unkrautregulierungserfolg kultivieren. Bei der Sojaaufbereitung sollte eine unnötig hohe Erhitzung der Bohnen bei der Aufbereitung vermieden werden, da durch die Erhitzung neben der Trypsininhibitoraktivität auch Eiweißverdaulichkeit reduziert werden. Mit ausschließlich indirekter, länger einwirkender, trockener Wärme (z. B. Biogasabwärme), ist es schwierig, gute Aufbereitungsqualitäten zu erzielen. Der Wissenstransfer mit Feldtagen und Website www.sojainfo.de war wichtig und erfolgreich zur Steigerung des Interesses am heimischen Sojaanbau
