    An Integrated Solution among Social, Personal and Formal Learning for Lifelong Competences

    In the Knowledge Society the worker should benefit from learning experiences which intersect formal learning moments with both individual and collaborative informal learning, according to a vision of authentic lifelong edu-cation” The paper suggests an innovative answer to the “lifelong competence” management approach. In this context a Lifelong Learning Model (LLM) finds its realization through an integrated solution among the personal, social and formal learning. The vision is sustained by a conceptual architecture, which represents a distinctive and enabling factor for the management of competence allowing to customize training paths on the worker profile. The idea is related to the integration between a Personal Learning Environment (PLE), a learning community and the solution of the Polo di Eccellenza L&K, the learning plat-form IWT (Intelligent Web Teacher)

    The kinesin Eg5 inhibitor K858 induces apoptosis but also survivin-related chemoresistance in breast cancer cells

    Inhibitors of kinesin spindle protein Eg5 are characterized by pronounced antitumor activity. Our group has recently synthesized and screened a library of 1,3,4-thiadiazoline analogues with the pharmacophoric structure of K858, an Eg5 inhibitor. We herein report the effects of K858 on four different breast cancer cell lines: MCF7 (luminal A), BT474 (luminal B), SKBR3 (HER2 like) and MDA-MB231 (basal like). We demonstrated that K858 displayed anti-proliferative activity on every analyzed breast cancer cell line by inducing apoptosis. However, at the same time, we showed that K858 up-regulated survivin, an anti-apoptotic molecule. We then performed a negative regulation of survivin expression, with the utilization of wortmannin, an AKT inhibitor, and obtained a significant increase of K858-dependent apoptosis. These data demonstrate that K858 is a potent inhibitor of replication and induces apoptosis in breast tumor cells, independently from the tumor phenotype. This anti-proliferative response of tumor cells to K858 can be limited by the contemporaneous over-expression of survivin; consequently, the reduction of survivin levels, obtained with AKT inhibitors, can sensitize tumor cells to K858-induced apoptosis

    Personalization and Contextualization of Learning Experiences based on Semantics

    Context-aware e-learning is an educational model that foresees the selection of learning resources to make the e-learning content more relevant and suitable for the learner in his/her situation. The purpose of this paper is to demonstrate that an ontological approach can be used to define leaning contexts and to allow contextualizing learning experiences finding out relevant topics for each context. To do that, we defined a context model able to formally describe a learning context, an ontology-based model enabling the representation of a teaching domain (including context information) and a methodology to generate personalized and context-aware learning experiences starting from them. Based on these theoretical components we improved an existing system for personalized e-learning with contextualisation features and experimented it with real users in two University courses. The results obtained from this experimentation have been compared with those achieved by similar systems

    It is estimated that in Italy at least 3% of babies are born with some congenital malformation. Scientific evidence demonstrates that the intake of folic acid by a woman prior the conception and during the early stages of pregnancy plays a role in preventing neural tube defects and severe birth anomalies due to the lack of neural tube closure at either the upper or lower end during the third and fourth week of pregnancy. The intake of folic acid during the periconception period can reduce the risk of other neonatal malformations such as congenital heart defects, urinary tract anomalies, oral facial clefts, limb defects. The Italian Network for Folic Acid Promotion, established in 2004 and coordinated by the National Centre for Rare Diseases of the Istituto Suepriore di Sanit? has elaborated and diffused a recommendation for the periconceptional supplementation of folic acid.     Color segmentation and neural networks for automatic graphic relief of the state of conservation of artworks

    none5noThis paper proposes a semi-automated methodology based on a sequence of analysis processes performed on multispectral images of artworks and aimed at the extraction of vector maps regarding their state of conservation. The graphic relief of the artwork represents the main instrument of communication and synthesis of information and data acquired on cultural heritage during restoration. Despite the widespread use of informatics tools, currently, these operations are still extremely subjective and require high execution times and costs. In some cases, manual execution is particularly complicated and almost impossible to carry out. The methodology proposed here allows supervised, partial automation of these procedures avoids approximations and drastically reduces the work times, as it makes a vector drawing by extracting the areas directly from the raster images.     Functional interaction of reverse gyrase with single-strand binding protein of the archaeon Sulfolobus

    Reverse gyrase is a unique hyperthermophile-specific DNA topoisomerase that induces positive supercoiling. It is a modular enzyme composed of a topoisomerase IA and a helicase domain, which cooperate in the ATP-dependent positive supercoiling reaction. Although its physiological function has not been determined, it can be hypothesized that, like the topoisomerase–helicase complexes found in every organism, reverse gyrase might participate in different DNA transactions mediated by multiprotein complexes. Here, we show that reverse gyrase activity is stimulated by the single-strand binding protein (SSB) from the archaeon Sulfolobus solfataricus. Using a combination of in vitro assays we analysed each step of the complex reverse gyrase reaction. SSB stimulates all the steps of the reaction: binding to DNA, DNA cleavage, strand passage and ligation. By co-immunoprecipitation of cell extracts we show that reverse gyrase and SSB assemble a complex in the presence of DNA, but do not make stable protein–protein interactions. In addition, SSB stimulates reverse gyrase positive supercoiling activity on DNA templates associated with the chromatin protein Sul7d. Furthermore, SSB enhances binding and cleavage of UV-irradiated substrates by reverse gyrase. The results shown here suggest that these functional interactions may have biological relevance and that the interplay of different DNA binding proteins might modulate reverse gyrase activity in DNA metabolic pathways
