    Peer review for the evaluation of the academic research: the Italian experience

    Peer review, that is the evaluation process based on judgments formulated by independent experts, is generally used for different goals: the allocation of research funding, the review of the research results submitted for publication in scientific journals, and the assessment of the quality of research conducted by Universities and university-related Institutes. The paper deals with the latter type of peer review. The aim is to understand how the characteristics of the Italian experience provide useful lessons for improving peer review effectiveness for evaluating the academic research. More specifically, the paper investigates the peer review process developed within the Three-Year Research Assessment Exercise (VTR) in Italy. Our analysis covers four disciplinary sectors: chemistry, biology, humanities and economics. Thus, the choice includes two “hard science” sectors, which have similar type of research output submitted for the three-year evaluation process, and two sectors with different types of output. The results provide evidences, which highlight the important role played by peer review for judging the quality of the academic research in different fields of science, and for comparing different institutions’ performance. Moreover, some basic features of the evaluation process are discussed, in order to understand their usefulness for reinforcing the effectiveness of the peers’ final outcome.Scientific research, Evaluation, Peer review, University, Academic institutions

    Does investor attention influence stock market activity? The case of spin-off deals

    This paper investigates empirically the nature of the interactions between mass media, investor attention and the stock market using data from a sample of 16 spin-off deals traded on NYSE and published between 2004 and 2010 in “Wall Street Journal”, the US’s second-largest newspaper by circulation. The results show that: i) the impact of media sentiment on the stock market reactions is enhanced / moderated by the level of attention of investors; ii) individual investors’ attention is grabbed by stocks experiencing high trading volumes on the previous day; iii) high attention could result in downward pressure on stock market returns.Media Sentiment, Investor Attention, Internet Search, Spin-off

    Counteracting aged DNA methylation states to combat ageing and age-related diseases

    DNA methylation (DNAm) overwrites information about multiple extrinsic factors on the genome. Age is one of these factors. Age causes characteristic DNAm changes that are thought to be not only major drivers of normal ageing but also precursors to diseases, cancer being one of these. Although there is still much to learn about the relationship between ageing, age-related diseases and DNAm, we now know how to interpret some of the effects caused by age in the form of changes in methylation marks at specific loci. In fact, these changes form the basis of the so called "epigenetic clocks", which translate the genomic methylation profile into an "epigenetic age". Epigenetic age does not only estimate chronological age but can also predict the risk of chronic diseases and mortality. Epigenetic age is believed to be one of the most accurate metrics of biological age. Initial evidence has recently been gathered pointing to the possibility that the rate of epigenetic ageing can be slowed down or even reversed. In this review, we discuss some of the most relevant advances in this field. Expected outcome is that this approach can provide insights into how to preserve health and reduce the impact of ageing diseases in humans

    L’influenza del diritto comunitario sull’ ordinamento nazionale, ha fatto sì che il processo amministrativo perdesse le caratteristiche proprie dei modelli processuali a carattere soggettivo, per assumere le caratteristiche di un modello ibrido, per certi aspetti molto vicino ai modelli processuali a carattere oggettivo, caratterizzati da amplissimi poteri del g.a. e da nuovi equilibri tra le parti.     Context-Aware Kubernetes Scheduler for Edge-native Applications on 5G

    This paper is an extension of work originally presented in SoftCOM 2019 [1]. The novelty of this work reside in its focused improvement of our scheduling algorithm towards its usage on a real 5G infrastructure. Industrial IoT applications are often designed to run in a distributed way on the devices and controller computers with strict service requirements for the nodes and the links between them. 5G, especially in concomitance with Edge Computing, will provide the desired level of connectivity for these setups and it will permit to host application run-time components in edge clouds. However, allocation of the edge cloud resources for Industrial IoT (IIoT) applications, is still commonly solved by rudimentary scheduling techniques (i.e. simple strategies based on CPU usage and device readiness, employing very few dynamic information). Orchestrators inherited from the cloud computing, like Kubernetes, are not satisfying to the requirements of the aforementioned applications and are not optimized for the diversity of devices which are often also limited in capacity. This design is especially slow in reacting to the environmental changes. In such circumstances, in order to provide a proper solution using these tools, we propose to take the physical, operational and network parameters (thus the full context of the IIoT application) into consideration, along with the software states and orchestrate the applications dynamically

    Assessment of Poly(ADP-ribose) Polymerase1 (PARP1) expression and activity in cells purified from blood and milk of dairy cattle

    Poly(ADP-ribosyl)ation (PAR) is a post-translational protein modification catalysed by enzyme member of the poly(ADP-ribose) polymerases (PARPs) family. The activation of several PARPs is triggered by DNA strand breakage and the main PARP enzyme involved in this process is PARP1. Besides its involvement in DNA repair, PARP1 is involved in several cellular processes including transcription, epigenetics, chromatin re-modelling as well as in the maintenance of genomic stability. Moreover, several studies in human and animal models showed PARP1 activation in various inflammatory disorders. The aims of the study were (1) to characterize PARP1 expression in bovine peripheral blood mononuclear cells (PBMC) and (2) to evaluate PAR levels as a potential inflammatory marker in cells isolated from blood and milk samples following different types of infection, including mastitis. Our results show that (i) bovine PBMC express PARP1; (ii) lymphocytes exhibit higher expression of PARP1 than monocytes; (iii) PARP1 and PAR levels were higher in circulating PBMCs of infected cows; (iv) PAR levels were higher in cells isolated from milk with higher Somatic Cell Counts (SCC > 100,000 cells/mL) than in cells from milk with low SCCs. In conclusion, these findings suggest that PARP1 is activated during mastitis, which may prove to be a useful biomarker of mastitis

    Poly(ADP-ribosyl)ation is involved in the epigenetic control of TET1 gene transcription

    TET enzymes are the epigenetic factors involved in the formation of the Sixth DNA base 5-hydroxymethylcytosine, whose deregulation has been associated with tumorigenesis. In particular, TET1 acts as tumor suppressor preventing cell proliferation and tumor metastasis and it has frequently been found down-regulated in cancer. Thus, considering the importance of a tight control of TET1 expression, the epigenetic mechanisms involved in the transcriptional regulation of TET1 gene are here investigated. The involvement of poly(ADP-ribosyl)ation in the control of DNA and histone methylation on TET1 gene was examined. PARP activity is able to positively regulate TET1 expression maintaining a permissive chromatin state characterized by DNA hypomethylation of TET1 CpG island as well as high levels of H3K4 trimethylation. These epigenetic modifications were affected by PAR depletion causing TET1 downregulation and in turn reduced recruitment of TET1 protein on HOXA9 target gene. In conclusion, this work shows that PARP activity is a transcriptional regulator of TET1 gene through the control of epigenetic events and it suggests that deregulation of these mechanisms could account for TET1 repression in cancer

    Validation of suitable internal control genes for expression studies in aging.

    Quantitative data from experiments of gene expression are often normalized through levels of housekeeping genes transcription by assuming that expression of these genes is highly uniform. This practice is being questioned as it becomes increasingly clear that the level of housekeeping genes expression may vary considerably in certain biological samples. To date, the validation of reference genes in aging has received little attention and suitable reference genes have not yet been defined. Our aim was to evaluate the expression stability of frequently used reference genes in human peripheral blood mononuclear cells with respect to aging. Using quantitative RT-PCR, we carried out an extensive evaluation of five housekeeping genes, i.e. 18s rRNA, ACTB, GAPDH, HPRT1 and GUSB, for stability of expression in samples from donors in the age range 35-74 years. The consistency in the expression stability was quantified on the basis of the coefficient of variation and two algorithms termed geNorm and NormFinder. Our results indicated GUSB be the most suitable transcript and 18s the least for accurate normalization in PBMCs. We also demonstrated that aging is a confounding factor with respect to stability of 18s, HPRT1 and ACTB expression, which were particularly prone to variability in aged donors

    A green approach to clean iron stains from marble surfaces

    AbstractIn the field of cultural heritage restoration, the removal of iron corrosion stains is a difficult problem to deal with, especially in stone materials. Many studies in recent years have been aimed at finding simple and reliable methods using non-toxic chelating compounds. The search for natural non-toxic compounds is therefore of great relevance, especially in the conservation of cultural heritage, where the use of toxic chemical compounds often involves risks for the environment and human health. Following this trend, the purpose of this preliminary work was to verify the use of two proteins, Lactotransferrin (Ltf) and Ovotransferrin (Ovt), for the removal of iron-based stains on marble surfaces. The two proteins, whose high affinity for iron "in vivo" has been widely documented, were extracted from their natural matrices. The protein extracts were then immobilized using a common cellulose pulp. The poultices obtained were spread on the surfaces of artificially stained marble specimens and, after a set time, were easily removed. The effectiveness of the removal, visually evident, was detected by spectrocolorimetry and image analysis. The surface analyses, before and after the treatment, carried out by X-ray photoelectron spectroscopy (XPS), confirmed that both proteins have a selective and effective complexing capacity for the ferric ions of rust stains
