    Exploring the scope of [Pt2(4-FC6H4)4(μ-SEt2)2] as a precursor for new organometallic platinum(II) and platinum(IV) antitumor agents

    The new compound [Pt2(4-FC6H4)4(μ-SEt2)2] (A) was prepared and fully characterized. The reactions of compound A with ligands ArCH=NCH2CH2NMe2 (Ar = 2-BrC6H4, 1a; 2,6-Cl2C6H3, 1b; 4-ClC6H4, 1c; 2-Cl,6-FC6H3, 1d) were studied under different conditions and produced platinum(II) compounds [Pt(4-FC6H4)2(ArCH=NCH2CH2NMe2)] (2b−2d), containing a bidentate [N,N′] ligand, as well as cyclometalated platinum(IV) or platinum(II) compounds such as [PtBr(4-FC6H4)2(C6H4CH=NCH2CH2NMe2)] (4a) or [PtCl{(3-FC6H3)(2-XC6H3)CH=NCH2CH2NMe2}] (5b: X = Cl; 5d: X = F), containing a tridentate [C,N,N′] ligand and either a five (4a) or a seven (5b, 5d) membered metallacycle. These compounds exhibit a great antiproliferative activity against non-small lung cancer cells (A549), and the best result was obtained for compound 2c (IC50 = 0.3 ± 0.1 μM). While compounds 5 alter the mobility of plasmid DNA in a similar way to cisplatin, compound 4 was less efficient in removing the supercoils from DNA. In spite of the very low IC50 value obtained for compound 2c, this compound does not interact with DNA, and it is neither an intercalator nor a topoisomerase I inhibitor

    A Systems Biology Approach Identifies Molecular Networks Defining Skeletal Muscle Abnormalities in Chronic Obstructive Pulmonary Disease

    Chronic Obstructive Pulmonary Disease (COPD) is an inflammatory process of the lung inducing persistent airflow limitation. Extensive systemic effects, such as skeletal muscle dysfunction, often characterize these patients and severely limit life expectancy. Despite considerable research efforts, the molecular basis of muscle degeneration in COPD is still a matter of intense debate. In this study, we have applied a network biology approach to model the relationship between muscle molecular and physiological response to training and systemic inflammatory mediators. Our model shows that failure to co-ordinately activate expression of several tissue remodelling and bioenergetics pathways is a specific landmark of COPD diseased muscles. Our findings also suggest that this phenomenon may be linked to an abnormal expression of a number of histone modifiers, which we discovered correlate with oxygen utilization. These observations raised the interesting possibility that cell hypoxia may be a key factor driving skeletal muscle degeneration in COPD patients

    Estudi del paper de la JMJD3 en la progressió tumoral induïda per TGFB

    La demetilasa d’histones JMJD3 (Jumonji domain containing protein 3), és un enzim capaç de de demetilar específicament la lisina 27 a la histona 3 (H3K27), eliminant així una marca epigenètica relacionada amb la repressió transcripcional. Recentment s’ha descrit que està implicada en el manteniment de la pluripotència de les cèl•lules mare embrionàries (ESCs). A més, també s’ha demostrat el seu paper en la regulació de processos fisiològics d’inflamació, de reprogramació epigenètica i de diferenciació, així com en la progressió del càncer de colon. En aquesta línia, resultats previs del grup han demostrat que l’expressió de JMJD3 està regulada per TGFB, en línies cel•lulars derivades de glioma. Tenint en compte aquests antecedents, l’objectiu principal d’aquest projecte ha estat estudiar el paper principal de la JMJD3 en la regulació epigenètica de la progressió tumoral induïda per TGFB. Els nostres resultats demostren que l’expressió de JMD3 en cèl•lules A549, derivades d’un adenocarcinoma de pulmó, es veu fortament induïda després d’un tractament amb TGFB. Aquest augment es produeix ràpidament i es manté almenys 48 hores, temps en el que té lloc la transició epitelio-mesenquimal (EMT). Per tal d’estudiar el paper de la JMD3 en aquest procés de transdiferenciació, vam generar línies cel•lulars estables mitjançant la infecció amb vectors lentivirals que expressaven shRNAs específics contra la seva seqüència. El knockdown de JMJD3 va bloquejar significativament l’expressió de marcadors mesenquimals, tant a nivell RNA com de proteïna en presència de TGFB. Aquests resultats suggereixen que la demetilasa d’histones JMJD3 té un paper clau en la regulació de la EMT induïda per TGFB

    Laser vibrometer condition assessment of cemented materials using wavelet synchro-squeezed transform

    Current non-destructive ultrasonic techniques (NDT) are based mostly on wave velocity analysis. While current techniques can identify severe damage, they fail to detect early deterioration. Therefore, the proposed method, based on the propagation of surface waves, takes into account not only changes in wave velocity but also changes in wave attenuation. In practical/field applications, access to a structure is often limited to one side only (i.e. concrete slabs, vacuum building walls). Thus, surface wave analysis is a natural solution. To improve the reliability of wave attenuation measurements, responses of ultrasonic transducers are measured using a high-frequency Doppler laser vibrometer. Firstly, the ultrasonic transducer is characterized using the laser vibrometer. Then, based on the sensitive frequency ranges, response signals measured for the cemented sand specimen are analyzed, and the relative attenuation index based on spectral energy is proposed. The method is further improved using the wavelet synchro-squeezed transform

    Role of Survivin in Bladder Cancer: Issues to Be Overcome When Designing an Efficient Dual Nano-Therapy

    Bladder cancer is the 10th most diagnosed cancer, with almost 10 M cancer deaths last year worldwide. Currently, chemotherapy is widely used as adjuvant therapy after surgical transurethral resection. Paclitaxel (PTX) is one of the most promising drugs, but cancer cells acquire resistance, causing failure of this treatment and increasing the recurrence of the disease. This poor chemotherapeutic response has been associated with the overexpression of the protein survivin. In this work, we present a novel dual nano-treatment for bladder cancer based on the hypothesis that the inhibition of survivin in cancer cells, using a siRNA gene therapy strategy, could decrease their resistance to PTX. For this purpose, two different polymeric nanoparticles were developed to encapsulate PTX and survivin siRNA independently. PTX nanoparticles showed sizes around 150 nm, with a paclitaxel loading of around 1.5%, that produced sustained tumor cell death. In parallel, siRNA nanoparticles, with similar sizes and loading efficiency of around 100%, achieved the oligonucleotide transfection and knocking down of survivin expression that also resulted in tumor cell death. However, dual treatment did not show the synergistic effect expected. The root cause of this issue was found to be the cell cycle arrest produced by nuclear survivin silencing, which is incompatible with PTX action. Therefore, we concluded that although the vastly reported role of survivin in bladder cancer, its silencing does not sensitize cells to currently applied chemotherapies
