    Prostate Stereotactic Ablative Radiation Therapy Using Volumetric Modulated Arc Therapy to Dominant Intraprostatic Lesions

    PurposeTo investigate boosting dominant intraprostatic lesions (DILs) in the context of stereotactic ablative radiation therapy (SABR) and to examine the impact on tumor control probability (TCP) and normal tissue complication probability (NTCP).Methods and MaterialsTen prostate datasets were selected. DILs were defined using T2-weighted, dynamic contrast-enhanced and diffusion-weighted magnetic resonance imaging. Four plans were produced for each dataset: (1) no boost to DILs; (2) boost to DILs, no seminal vesicles in prescription; (3) boost to DILs, proximal seminal vesicles (proxSV) prescribed intermediate dose; and (4) boost to DILs, proxSV prescribed higher dose. The prostate planning target volume (PTV) prescription was 42.7 Gy in 7 fractions. DILs were initially prescribed 115% of the PTVProstate prescription, and PTVDIL prescriptions were increased in 5% increments until organ-at-risk constraints were reached. TCP and NTCP calculations used the LQ-Poisson Marsden, and Lyman-Kutcher-Burman models respectively.ResultsWhen treating the prostate alone, the median PTVDIL prescription was 125% (range: 110%-140%) of the PTVProstate prescription. Median PTVDIL D50% was 55.1 Gy (range: 49.6-62.6 Gy). The same PTVDIL prescriptions and similar PTVDIL median doses were possible when including the proxSV within the prescription. TCP depended on prostate α/β ratio and was highest with an α/β ratio = 1.5 Gy, where the additional TCP benefit of DIL boosting was least. Rectal NTCP increased with DIL boosting and was considered unacceptably high in 5 cases, which, when replanned with an emphasis on reducing maximum dose to 0.5 cm3 of rectum (Dmax0.5cc), as well as meeting existing constraints, resulted in considerable rectal NTCP reductions.ConclusionsBoosting DILs in the context of SABR is technically feasible but should be approached with caution. If this therapy is adopted, strict rectal constraints are required including Dmax0.5cc. If the α/β ratio of prostate cancer is 1.5 Gy or less, then high TCP and low NTCP can be achieved by prescribing SABR to the whole prostate, without the need for DIL boosting

    Automatic Testing and Improvement of Machine Translation

    This paper presents TransRepair, a fully automatic approach for testing and repairing the consistency of machine translation systems. TransRepair combines mutation with metamorphic testing to detect inconsistency bugs (without access to human oracles). It then adopts probability-reference or cross-reference to post-process the translations, in a grey-box or black-box manner, to repair the inconsistencies. Our evaluation on two state-of-the-art translators, Google Translate and Transformer, indicates that TransRepair has a high precision (99%) on generating input pairs with consistent translations. With these tests, using automatic consistency metrics and manual assessment, we find that Google Translate and Transformer have approximately 36% and 40% inconsistency bugs. Black-box repair fixes 28% and 19% bugs on average for Google Translate and Transformer. Grey-box repair fixes 30% bugs on average for Transformer. Manual inspection indicates that the translations repaired by our approach improve consistency in 87% of cases (degrading it in 2%), and that our repairs have better translation acceptability in 27% of the cases (worse in 8%)

    Adaptations in a hierarchical food web of southeastern Lake Michigan

    Two issues in ecological network theory are: (1) how to construct an ecological network model and (2) how do entire networks (as opposed to individual species) adapt to changing conditions? We present a novel method for constructing an ecological network model for the food web of southeastern Lake Michigan (USA) and we identify changes in key system properties that are large relative to their uncertainty as this ecological network adapts fromone time point to a second time point in response to multiple perturbations. To construct our foodweb for southeastern Lake Michigan,we followed the list of seven recommendations outlined in Cohen et al. [Cohen, J.E., et al., 1993.Improving foodwebs. Ecology 74, 252–258] for improving food webs. We explored two inter-related extensions of hierarchical system theory with our food web; the first one was that subsystems react to perturbations independently in the short-term and the second onewas that a system’s properties change at a slower rate than its subsystems’ properties. We used Shannon’s equations to provide quantitative versions of the basic food web properties: number of prey, number of predators, number of feeding links, and connectance (or density).We then compared these properties between the two time-periods by developing distributions of each property for each time period that took uncertainty about the property into account.We compared these distributions, and concluded that non-overlapping distributions indicated changes in these properties that were large relative to their uncertainty. Two subsystems were identified within our food web system structure (p \u3c 0.001). One subsystem had more non-overlapping distributions in food web properties between Time 1 and Time 2 than the other subsystem. The overall system had all overlapping distributions in food web properties between Time 1 and Time 2. These results supported both extensions of hierarchical systems theory. Interestingly, the subsystemwithmore non-overlapping distributions in foodweb propertieswas the subsystemthat contained primarily benthic taxa, contrary to expectations that the identifiedmajor perturbations (lower phosphorous inputs and invasive species) would more greatly affect the subsystem containing primarily pelagic taxa. Future food-web research shouldemploy rigorous statistical analysis and incorporate uncertainty in food web properties for a better understanding of how ecological networks adapt

    Uncovering the molecular mechanisms of lignocellulose digestion in shipworms

    Abstract Lignocellulose forms the structural framework of woody plant biomass and represents the most abundant carbon source in the biosphere. Turnover of woody biomass is a critical component of the global carbon cycle, and the enzymes involved are of increasing industrial importance as industry moves away from fossil fuels to renewable carbon resources. Shipworms are marine bivalve molluscs that digest wood and play a key role in global carbon cycling by processing plant biomass in the oceans. Previous studies suggest that wood digestion in shipworms is dominated by enzymes produced by endosymbiotic bacteria found in the animal’s gills, while little is known about the identity and function of endogenous enzymes produced by shipworms. Using a combination of meta-transcriptomic, proteomic, imaging and biochemical analyses, we reveal a complex digestive system dominated by uncharacterized enzymes that are secreted by a specialized digestive gland and that accumulate in the cecum, where wood digestion occurs. Using a combination of transcriptomics, proteomics, and microscopy, we show that the digestive proteome of the shipworm Lyrodus pedicellatus is mostly composed of enzymes produced by the animal itself, with a small but significant contribution from symbiotic bacteria. The digestive proteome is dominated by a novel 300 kDa multi-domain glycoside hydrolase that functions in the hydrolysis of β-1,4-glucans, the most abundant polymers in wood. These studies allow an unprecedented level of insight into an unusual and ecologically important process for wood recycling in the marine environment, and open up new biotechnological opportunities in the mobilization of sugars from lignocellulosic biomass

    Feasibility of a mHealth Approach to Nutrition Counseling in an Appalachian State

    Abstract: West Virginia is a rural state with an aging population that may experience barriers to accessing nutritional and lifestyle counseling. This study examined feasibility of an online personalized nutrition tracking application, Good Measures (GM), with patients at seven health care clinics throughout the state. Fourteen healthcare providers and 64 patients 18 years or older with a Body Mass Index (BMI) greater than or equal to 30 and access to the Internet were recruited for this 12-week feasibility study. Patient participants logged meals and exercise into the GM application via smart phone, tablet, or computer and virtually engaged with a Registered Dietitian Nutritionist (RDN) in one-on-one sessions. The primary endpoint was to examine feasibility of the program by usage of the application and feedback questions regarding the benefits and challenges of the application. Participants were predominately white (92%) and female (76%). Minimal improvements in weight and systolic blood pressure were found. Participant attitude survey data declined from 4-weeks to 12-weeks of the intervention. Interestingly though, patients in a rural clinic had lesser declines in attitudes than peri-urban participants. Qualitative feedback data identified participants predominately had a positive overall feeling toward the approach. Participants expressed favorability of RDN access, the variety of foods, but did give suggestions for in-person meetings and more updating of the application. Implementing a technology approach to nutrition in rural areas of West Virginia using a mobile application with RDN access may be one strategy to address public health issues such as obesity

    The Spacecraft INterface and control Board for NomAD (SINBAD) is an electronic interface designed by the Instituto de Astroffisica de Andalucfia (IAA-CSIC). It is part of the Nadir and Occultation for MArs Discovery instrument (NOMAD) on board in the ESAs ExoMars Trace Gas Orbiter mission. This mission was launched in March 2016. The SINBAD Flight Software (SFS) is the software embedded in SINBAD. It is in charge of managing the interfaces, devices, data, observing sequences, patching and contingencies of NOMAD. It is presented in this paper the most remarkable aspects of the SFS design, likewise the main problems and lessons learned during the software development process
