    Biomarkers of postoperative delirium and cognitive dysfunction

    Elderly surgical patients frequently experience postoperative delirium (POD) and the subsequent development of postoperative cognitive dysfunction (POCD). Clinical features include deterioration in cognition, disturbance in attention and reduced awareness of the environment and result in higher morbidity, mortality and greater utilization of social financial assistance. The aging Western societies can expect an increase in the incidence of POD and POCD. The underlying pathophysiological mechanisms have been studied on the molecular level albeit with unsatisfying small research efforts given their societal burden. Here, we review the known physiological and immunological changes and genetic risk factors, identify candidates for further studies and integrate the information into a draft network for exploration on a systems level. The pathogenesis of these postoperative cognitive impairments is multifactorial; application of integrated systems biology has the potential to reconstruct the underlying network of molecular mechanisms and help in the identification of prognostic and diagnostic biomarkers

    Digitalization as a Catalyst to Overcome the Negative Consequences of the Pandemic and Martial Law

    The article is aimed at studying the peculiarities of the manifestation of the digital economy during the COVID-19 pandemic and martial law, identifying the impact of digitalization on the prospects for the development of Ukrainian business. The digital economy, the functioning of which is based on the creation, distribution and use of digital technologies and related products and services, is constantly evolving in accordance with new challenges of economic development, which is reflected in the expansion of its architecture. In recent years, the transformation of economic activity models caused by the use of end-to-end digital technologies (artificial intelligence, robotics, the Internet of Things, wireless technologies, etc.) has been actualized, which has turned digitalization into the most important factor of economic growth. Digital technologies were used in various spheres of economic activity even before the COVID-19 pandemic, but the latter significantly intensified the digital transformation of business processes. The use of online applications, the provision of online services, the creation of hybrid business models that combine the features of offline and online services, has allowed many economic actors to switch to a remote work format, reducing losses from social distancing and other preventive measures, and some companies have even managed to increase the efficiency of their activities. Improving the efficiency of management and production activities based on digital solutions has turned digitalization from an anti-crisis tool into a source of profitability for companies in the long term. A certain similarity of the challenges caused by the coronavirus crisis and the disruption of the domestic economy as a result of Russian aggression has led to the need to intensify the use of digital technologies by business and the State.     Aerosol and its radiative effects during the aeroradcity 2018 Moscow experiment

    During the AeroRadCity-2018 spring aerosol experiment at the Moscow State University Meteorological Observatory the aerosol properties of the atmosphere and radiative aerosol effects were analyzed using a wide complex of measurements and model COSMO-ART simulations over Moscow domain. The program of measurements consisted of columnar aerosol AERONET retrievals, surface PM10, black carbon (BC) and aerosol gas precursors mass concentrations, as well as radiative measurements under various meteorological conditions. We obtained a positive statistically significant dependence of total and fine aerosol optical depth (AOD) mode (R2 ~0.4) with PM concentrations. This dependence has revealed a pronounced bifurcation point around PM10=0.04 mgm-3. The modelled BC concentration is in agreement with the observations and has a pronounced correlation with PM, but not with the AODs. The analysis of radiative effects of aerosol has revealed up to 30% loss for UV irradiance and 15% - for shortwave irradiance at high AOD in Moscow. Much intensive radiation attenuation is observed in the afternoon when remote pollution sources may affect solar fluxes at elevated boundary layer conditions. Negative (cooling) radiative forcing effect at the top of the atmosphere from -18 Wm-2 to -4 Wm-2 has been evaluated. Mean difference in visible AOD between urban and background conditions in Moscow and Zvenigorod was about 0.01 according to measurements and model simulations, while in some days the difference may increase up to 0.05. The generation of urban aerosol was shown to be more favorable in conditions with low intensity of pollutant dispersion, when mean deltaAOD550 was doubled from 0.01 to 0.02

    Fruit and seed plumpness, germination and viability of rare plants in Yakutia

    Background. With the increasing anthropogenic impact on nature, conservation and renewal of rare and endangered plants in natural cenoses is an urgent problem. Quality assessment, identification of full-fledged plump seeds, and determination of their viability is the way to develop reproduction technologies and provide a possibility of repatriating these species to their natural habitats. Collecting and studying natural flora species in botanical gardens helps to save biological resources of useful species and recommend them for urban landscaping.Materials and methods. X-ray techniques were used to analyze seed quality and viability of rare species in the flora of Yakutia: Adonis sibirica Patrin, Phlojodicarpus sibiricus (Steph. ex Spreng.) Koso-Pol., Iris laevigata Fisch. et C.A. Mey., Iris sanguinea Donn, and Paeonia anomala L. Methods developed for the application of microfocus radiography to fruits and seeds were used in the X-ray analysis.Results. Seed quality was assessed according to guidelines. It was shown that fruits and seeds of the studied species developed under the conditions of Yakutia were full-fledged (plump), but due to the presence of a morphophysiological type of dormancy in them, they could germinate only after a cold or multistage stratification.Conclusion. Assessment of reproductive diaspore quality and viability in rare and endangered plant species of Yakutia will make it possible to develop technologies for increasing their seed germination rates and restoring the natural populations of these species

Given that the imbalance in pro- and anti-inflammatory cytokines plays an important role in the pathogenesis of sepsis, estimation of its prognostic value in patients in the early stages from the disease onset or injury is of concern.Aim of study. To study changes in concentrations of biomarkers for inflammation on day 1–2 in patients with a high risk for sepsis.Materials and methods. On day 1-2 from the admission, blood tests were performed in 48 patients with a high risk of sepsis: 9 — with mediastinitis, 6 — with widespread purulent peritonitis, 24 — with severe concomitant injury, 9 — with acute destructive pancreatitis. Sepsis occurred in 28 patients of group 1. Group 2 consisted of 20 patients without sepsis. We investigated concentration of procalcitonin, C-reactive protein, interleukine-6 and -10, lipopolysaccharide-bindidng protein and soluble receptor for interleukine-2.Result. Significant differences in concentrations of groups were identified on day 1-2 in CRP (P=0.00009), IL-2R (P=0.0005) LBP (P=0.00002) and IL-6 (P=0.0192).Conclusion. Hyper LBP and CRP on the background of high IL-2R, IL-6, increase the risk of sepsis

    EFFICIENCY OF THE FORMATION OF ANTITUMOR IMMUNE RESPONSES IN THE SYSTEM OF PREVENTIVE VACCINATION OF MICE WITH TESTICULAR ANTIGENS

    Аppearance of a malignant tumor is associated with impaired mechanisms of proliferation, differentiation, apoptosis ability. However, these changes are not enough for immune system to recognize and destroy mutated cells. Weak immunogenicity of tumor-associated antigens (TAA) and the insufficiency of co-stimulating molecules on the surface of tumor cells is a reason for this phenomenon Since biochemical processes of tumor cells and healthy tissue cells are identical, therefore creation of effective chemotherapeutic drugs is limited not by selectivity of their action. So antitumor vaccination is the most effective specific method for both preventing recurrence of a disease and a therapeutic treatment tool in oncology. Xenogeneic proteins are highly immunogenic and effective in breaking immune tolerance to human analogs. In our work, we used sheep testicular AG as a source xenogenic TAAs. Sheep testicles contain a large set of TAAs. Experimental mice were immunized with type liposomal testicular vaccine from sheep, one month after vaccination, to induce tumor growth, cells of carcinoma LLC were implanted in mouse. The life expectancy of the experimental group of mice was 2 times higher compared to the syngenetic control and 20% of them did not develop the tumor at all. In the spleen of mice who did not have tumors after pre-vaccination sheep liposomal testicular AG, T-regulatory cells and T-memory cells were measured. We found a credible decrease in both naive Treg (CD4+CD25+), activated (CD4+CD25+FoxP3+) and both T-memory (CD4+CD44+) and central memory (CD4+CD44+CD62L+) in spleen pre-vaccination mice with compared to the contral intact spleen. Content of IFNg and IL-10 in supernatants of mouse slenocytes derived from vaccinated mice with no tumors was investigated and showed a reliable decrease in the amount of IL-10, but not IFNg. We believe that immunization with xenogenic tumor AGs can lead to the formation of a protective antitumor response

    Analysis of the dynamic co-expression network of heart regeneration in the zebrafish.

    The zebrafish has the capacity to regenerate its heart after severe injury. While the function of a few genes during this process has been studied, we are far from fully understanding how genes interact to coordinate heart regeneration. To enable systematic insights into this phenomenon, we generated and integrated a dynamic co-expression network of heart regeneration in the zebrafish and linked systems-level properties to the underlying molecular events. Across multiple post-injury time points, the network displays topological attributes of biological relevance. We show that regeneration steps are mediated by modules of transcriptionally coordinated genes, and by genes acting as network hubs. We also established direct associations between hubs and validated drivers of heart regeneration with murine and human orthologs. The resulting models and interactive analysis tools are available at http://infused.vital-it.ch. Using a worked example, we demonstrate the usefulness of this unique open resource for hypothesis generation and in silico screening for genes involved in heart regeneration

В 1–2-е сут с момента поступления исследована кровь 48 пациентов с высоким риском развития сепсиса: 9 — с медиастинитом, 6 — с распространенным гнойным перитонитом, 24 — с тяжелой сочетанной травмой, 9 — с острым деструктивным панкреатитом. Сепсис развился у 28 пациентов 1-й группы. 2-я группа включала 20 пациентов без сепсиса. Исследовали концентрации прокаль- цитонина, С-реактивного белка (СРБ), интерлейкинов-6 (ИЛ-6) и -10 (ИЛ-10), липополисахарид- связывающего белка (ЛСБ) и растворимого рецептора к интерлейкину-2 (ИЛ-2R).РЕЗУЛЬТАТЫ. Статистически значимые различия концентраций в группах были выявлены уже в 1–2 сут в отношении СРБ (р=0,00009), ИЛ-2R (р=0,0005 ), ЛСБ (р=0,00002) и ИЛ-6 (р=0,0192).ЗАКЛЮЧЕНИЕ. При гиперпродукции ЛСБ и СРБ на фоне высокой концентрации ИЛ-2R и ИЛ-6, риск развития сепсиса повышается
