    Telomeres are nucleoprotein structures that caps the ends of chromosomes. Human telomeres are repeats of TTAGGG sequences that measures between 5 and 15 kb. Telomeres protect chromosome ends from DNA damage and are associated with the nuclear membrane to maintain nuclear organization. During the process of DNA synthesis and cell division, telomere length decreases as a result of the incomplete replication of chromosome ends. Telomerase is a ribonuclear protein-enzyme complex that synthesizes telomeric repeats by adding hexameric (TTAGGG) repeats onto the telomeric ends of chromosomes. Telomerase is active in germ line cells and stem cells but not in most adult tissue. Telomere shortening has been proposed to play an important role in the process of cellular senescence. Cellular senescence may represent therefore, the manifestation of the aging process at the cellular level. In this work, telomere length was analized in the Sardinian population in relation to age and sex. The age of subjects enrolled in the research project Akea (A Kent'Annos ) was comprised between 20 and 105 years. Telomere length was determined using a method based on Cawthon’s quantitative real time PCR. Results have shown a negative correlation between the average length of telomeres and age. In addition, telomere length in women was lower than in men, a result in opposition with data observed in other scientific works. Finally, some centenarian samples showed a LTL unusually high

    Behavior of Ternary Mixtures of Hydrogen Bond Acceptors and Donors in Terms of Band Gap Energies

    Three ternary mixtures composed by choline chloride (ChCl), ethylene glycol (EG), and a second hydrogen bond donor (HBD) as ethanol (A), 2-propanol (B), and glycerol (C) were studied in terms of composition related to the band gap energy (BGE). A Design of Experiments (DoE) approach, and in particular a Simple Lattice three-components design, was employed for determining the variation of the BGE upon the composition of each system. UV-VIS analysis and subsequent Tauc plot methodology provided the data requested from the DoE, and multivariate statistical analysis revealed a drop of the BGE in correspondence to specific binary compositions for systems A and B. In particular, a BGE of 3.85 eV was registered for the mixtures ChCl/EtOH (1:1) and ChCl/2-propanol (1:1), which represents one of the lowest values ever observed for these systems

    Evaluation of different Bacillus thuringiensis sv kurstaki formulations against Lymantria dispar and Malacosoma neustria larvae infesting Quercus suber trees

    Lepidopteran defoliators such as Lymantria dispar and Malacosoma neustria represent a major concern for cork oak forest, especially during population outbreaks. To contain their infestations over large areas, the use of entomopathogenic microorganisms such as Bacillus thuringiensis serovar kurstaki (Btk) is one of the available option. However, the features of the microbial control agent formulations represent a key factor for the success of application programs. The results of two years efficacy trials with different formulations of Btk, conducted in 2012 and 2013 in a cork oak forest in North-Western Sardinia, are reported. In the first year, trials were carried out on a M. neustria population, while in the second year a mixed population of L. dispar and M. neustria was involved. Trials included two formulations of Btk strain EG 2348 (RapaxÂź and Rapax Experimental) in comparison with two other commercial formulations (Foray 48BÂź and DelfinÂź). Both formulations of Btk strain EG 2348 proved to be effective in controlling the two pest species, showing a forest protection potential comparable to that of the reference products, Foray 48BÂź and DelfinÂź, containing spores and insecticidal Cry proteins of strains HD-1 and SA-11, respectively. Also the defoliation levels were significantly higher in untreated control trees than in treated ones

    Abstract Cerambyx cerdo is a large saproxylic species included in the Annex IV of the European Habitats Directive as a 'priority species'. Although C. cerdo populations have shown a significant decline in size and distribution in central and northern European forests, in the Mediterranean basin it is often considered as a pest in urban and peri‐urban areas. Based on European legislations currently in use, we propose a simplified decision‐making flowchart that should be followed regarding the adoption of different control measures against severe C. cerdo infestations

    Optimization of Nazarov Cyclization of 2,4-Dimethyl-1,5-Diphenylpenta-1,4-Dien-3-One in Deep Eutectic Solvents by a Design of Experiments Approach

    : The unprecedented Nazarov cyclization of a model divinyl ketone using phosphonium-based Deep Eutectic Solvents as sustainable non-innocent reaction media is described. A two-level full factorial Design of Experiments was conducted for elucidating the effect of the components of the eutectic mixture and optimizing the reaction conditions in terms of temperature, time, and substrate concentration. In the presence of the Deep Eutectic Solvent (DES) triphenylmethylphosphonium bromide/ethylene glycol, it was possible to convert more than 80% of the 2,4-dimethyl-1,5-diphenylpenta-1,4-dien-3-one, with a specific conversion, into the cyclopentenone Nazarov derivative of 62% (16 h, 60 °C). For the reactions conducted in the DES triphenylmethylphosphonium bromide/acetic acid, quantitative conversions were obtained with percentages of the Nazarov product above 95% even at 25 °C. Surface Responding Analysis of the optimized data furnished a useful tool to determine the best operating conditions leading to quantitative conversion of the starting material, with complete suppression of undesired side-reactions, high yields and selectivity. After optimization, it was possible to convert more than 90% of the model substrate into the desired cyclopentenone with cis percentages up to 77%. Experimental validation of the implemented model confirmed the robustness and the suitability of the procedure, leading to possible further extension to this specific combination of experimental designs to other substrates or even to other synthetic processes of industrial interest

    Long-term monitoring and microbiological control programs against lepidopteran defoliators in Sardinian cork oak forests (Italy)

    The gypsy moth, Lymantria dispar (L.), and the tent caterpillar, Malacosoma neustrium (L.), are the main cork oak, Quercus suber L., pests in the Mediterranean area and cause complete defoliation in large forest districts. In order to control infestations, large scale aerial applications of insecticides based on Bacillus thuringiensis subsp. kurstaki (Btk) have been carried out in Sardinia (Italy) since 2001. This paper evaluated the frequency of outbreaks in forest districts with varying homogeneity of land use, forest areas annually exposed to defoliation and the effectiveness of control programs based on Btk insecticide applications.The volume of areas annually exposed to defoliation depends on forest homogeneity, as infestations are more frequent in cork oak areas with a lower than 25% canopy cover rate. The microbiological control programme efficiently protected cork oaks from lepidopteran defoliators and caused an overall annual mean mortality of over 60%, with maximum rates of 89.9 and 98.0% for L. dispar and M. neustrium, respectively. To date, approximately 180,000 hectares of cork oak forests have been protected by spraying Btk-based insecticides

    Plasma clusterin and lipid profile: a link with aging and cardiovascular diseases in a population with a consistent number of centenarians

    The role of Clusterin in attenuation of inflammation and reverse cholesterol transfer makes this molecule a potential candidate as a marker for cancer, cardiovascular disease, diabetes mellitus, and metabolic syndrome. In elderly subjects cardiovascular diseases represent the primary cause of death and different clinical studies have shown a positive correlation of these diseases with changes in the lipid pattern. This work aimed at evaluating the relationship between circulating clusterin and the biochemical parameters that characterize the lipid profile of a Sardinian population divided into five age groups including centenarians; the high frequency in Sardinia of these long-lived individuals gave us the opportunity to extend the range of the age groups to be analyzed to older ages and to better evaluate the changes in the lipid balance during ageing and its relationship with clusterin concentration in plasma. Our results showed that Clusterin concentration values of the youngest group were more similar with the centenarian’s group compared to the other age groups, and a positive correlation arises with LDL. Furthermore given the high prevalence of cardiovascular diseases in the population examined and the association of Clusterin with these pathologies we evaluated Clusterin concentration variation in two groups with or without cardiovascular diseases. In presence of cardiovascular disease, Clusterin is significantly related to the most atherogenic components of lipid profile (total cholesterol and LDL), especially in women, suggesting its potential role in modulating cardiovascular metabolic risk factors

    Climate and landscape composition explain agronomic practices, pesticide use and grape yield in vineyards across Italy

    Context Worldwide, organic farming is being promoted as one of the main alternatives to intensive conventional farming. However, the benefits of organic agriculture are still controversial and need to be tested across wide environmental gradients. Objective Here, we carried out an observational study to test how agronomic practices, pest management, environmental impact and yield of conventional and organic vineyards changed along wide climatic and landscape gradients across Italy. Methods We used a block design with 38 pairs of conventional and organic vineyards across Italy. Results and conclusions Most agronomic practices did not differ between conventional and organic vineyards. By contrast, landscape composition and climate were strong predictors of management in both systems. First, increasing semi-natural areas around the vineyards reduced pesticide pressure and related environmental impacts, but was also associated with lower yield. Second, irrespective of the farming system, a warm and dry climate was associated with reduced fungicide pressure. Conventional farming had a yield gain of 40% in cold and wet climate compared to organic but the yield gap disappeared in the warmest regions. Significance In both farming systems, we observed a large variability in management practices that was mainly explained by climate and landscape composition. This large variability should be considered when evaluating the benefits and drawbacks of different farming systems under contrasting environmental contexts

    Preliminary Evidence for Cell Membrane Amelioration in Children with Cystic Fibrosis by 5-MTHF and Vitamin B12 Supplementation: A Single Arm Trial

    Cystic fibrosis (CF) is one of the most common fatal autosomal recessive disorders in the Caucasian population caused by mutations of gene for the cystic fibrosis transmembrane conductance regulator (CFTR). New experimental therapeutic strategies for CF propose a diet supplementation to affect the plasma membrane fluidity and to modulate amplified inflammatory response. The objective of this study was to evaluate the efficacy of 5-methyltetrahydrofolate (5-MTHF) and vitamin B12 supplementation for ameliorating cell plasma membrane features in pediatric patients with cystic fibrosis.A single arm trial was conducted from April 2004 to March 2006 in an Italian CF care centre. 31 children with CF aged from 3 to 8 years old were enrolled. Exclusion criteria were diabetes, chronic infections of the airways and regular antibiotics intake. Children with CF were supplemented for 24 weeks with 5-methyltetrahydrofolate (5-MTHF, 7.5 mg /day) and vitamin B12 (0.5 mg/day). Red blood cells (RBCs) were used to investigate plasma membrane, since RBCs share lipid, protein composition and organization with other cell types. We evaluated RBCs membrane lipid composition, membrane protein oxidative damage, cation content, cation transport pathways, plasma and RBCs folate levels and plasma homocysteine levels at baseline and after 24 weeks of 5-MTHF and vitamin B12 supplementation. In CF children, 5-MTHF and vitamin B12 supplementation (i) increased plasma and RBC folate levels; (ii) decreased plasma homocysteine levels; (iii) modified RBC membrane phospholipid fatty acid composition; (iv) increased RBC K(+) content; (v) reduced RBC membrane oxidative damage and HSP70 membrane association.5-MTHF and vitamin B12 supplementation might ameliorate RBC membrane features of children with CF.ClinicalTrials.gov NCT00730509

    Gypsy Moth Management with LdMNPV Baculovirus in Cork Oak Forest

    (1) Research Highlights: Applications of a species-specific baculovirus is a promising method to control the gypsy moth and regulate its population dynamics in forest ecosystems. (2) Background and Objectives: Cork oak protection against the Lepidopteran defoliator Lymantria dispar requires an appropriate forest ecosystem management program, involving the application of eco-sustainable microbial products during population outbreaks. The species-specific multicapsid nucleopolyhedrovirus (LdMNPV), agent of natural epizootics in gypsy moth populations, represents an option that was investigated in a multi-year field study, involving viral applications either from the ground or by aerial treatment. (3) Materials and Methods: Efficacy trials against L. dispar populations were conducted in 2018 and 2019 in Sardinia, according to a randomized block design. Each year, two trials were conducted, applying a baculovirus commercial formulation with an atomizer from the ground and assessing the effects of different doses and application timing, respectively. An aerial application trial distributing LdMNPV at ultra-low volumes (2 L/ha) was also conducted in 2019 to assess the virus efficacy at a larger field scale. (4) Results: In both years, a significant increase in larval mortality was detected in plots treated with higher viral occlusion body (OB) doses and with an earlier application targeting younger larvae, in comparison with untreated controls. Due to an observed retrogradation phase of the target pest in 2019, no significant differences in larval density between areas treated from a helicopter and control were detected, but in the few weeks following application, a meaningful vitality decrease in larval samples from treated plots was observed. (5) Conclusions: Based on the results of this study, the use of LdMNPV in forest protection programs against gypsy moth can be worth consideration in multi-year integrated program strategies to modulate population dynamics
