    Availability Of Dental Anomaly Phenotype In Individuals With Familial Adenomatous Polyposis

    Background: Mutations in the APC gene cause familial adenomatous polyposis (FAP), an autosomal dominant colorectal cancer predisposition associated with the development of hundreds to thousands of adenomatous colorectal polyps beginning in childhood or adolescence. Both malignant and non-malignant extracolonic manifestations are associated with APC gene mutations, including approximately 17% of individuals with various dental anomalies. The availability of dental anomaly information in the medical record remains to be evaluated. Methods: Medical records were reviewed for documentation of dental anomalies. Dental questionnaires were mailed to 271 individuals with FAP at The University of Texas M. D. Anderson Cancer Center (UTMDACC) to assess self-reported dental phenotype. Demographic data was obtained from chart review and included current age or age at death, age at diagnosis of FAP, sex, surgical procedure for polyposis, available dental phenotype information, date of last contact at UTMDACC, and APC gene mutation and codon. Results: The response rate to the dental questionnaire was 21%. The majority of individuals (82%) were did not have dental anomaly information available in the medical record. Forty-four (16%) had self-reported dental anomalies in either the medical record or on the dental questionnaire. The most frequently reported anomalies were dental crowding and supernumerary teeth. Conclusion: Our findings are consistent with previous reports of the prevalence of dental anomalies in individuals with FAP. The results of this study indicate that dental anomalies in individuals with FAP are not consistently recorded in the medical record. Ultimately, consistent documentation of these anomalies in the medical record can aid in detection of FAP in individuals for whom genetic testing is not available. This highlights the importance of interdisciplinary approaches between clinicians, genetic counselors, and dentists to provide the best and most accurate clinical phenotype description in individuals with FAP

    The receptor for hepatocyte growth factor, also known as scatter factor (HGF/SF), has recently been identified as the 190-kDa heterodimeric tyrosine kinase encoded by the MET proto-oncogene (p190MET). The signaling pathway(s) triggered by HGF/SF are unknown. In A549 cells, a lung epithelial cell line, nanomolar concentrations of HGF/SF induced tyrosine phosphorylation of the p190MET receptor. The autophosphorylated receptor coprecipitated with phosphatidylinositol 3-kinase (PI 3-kinase) activity. In GTL16 cells, a cell line derived from a gastric carcinoma, the p190MET receptor, overexpressed and constitutively phosphorylated on tyrosine, coprecipitated with PI 3-kinase activity and with the 85-kDa PI 3-kinase subunit. In these cells activation of protein kinase C or the increase of intracellular [Ca2+] inhibits tyrosine phosphorylation of the p190MET receptor as well as the association with both PI 3-kinase activity and the 85-kDa subunit of the enzyme. In an in vitro assay, tyrosine phosphorylation of the immobilized p190MET receptor was required for binding of PI 3-kinase from cell lysates. These data strongly suggest that the signaling pathway activated by the HGF/SF receptor includes generation of D-3-phosphorylated inositol phospholipids

    Dynamics and Lax-Phillips scattering for generalized Lamb models

    This paper treats the dynamics and scattering of a model of coupled oscillating systems, a finite dimensional one and a wave field on the half line. The coupling is realized producing the family of selfadjoint extensions of the suitably restricted self-adjoint operator describing the uncoupled dynamics. The spectral theory of the family is studied and the associated quadratic forms constructed. The dynamics turns out to be Hamiltonian and the Hamiltonian is described, including the case in which the finite dimensional systems comprises nonlinear oscillators; in this case the dynamics is shown to exist as well. In the linear case the system is equivalent, on a dense subspace, to a wave equation on the half line with higher order boundary conditions, described by a differential polynomial p(x)p(\partial_x) explicitely related to the model parameters. In terms of such structure the Lax-Phillips scattering of the system is studied. In particular we determine the incoming and outgoing translation representations, the scattering operator, which turns out to be unitarily equivalent to the multiplication operator given by the rational function p(iκ)/p(iκ)-p(i\kappa)^*/p(i\kappa), and the Lax-Phillips semigroup, which describes the evolution of the states which are neither incoming in the past nor outgoing in the future

    Changes in Cytokines of the Bone Microenvironment during Breast Cancer Metastasis

    It is commonly accepted that cancer cells interact with host cells to create a microenvironment favoring malignant colonization. The complex bone microenvironment produces an ever changing array of cytokines and growth factors. In this study, we examined levels of MCP-1, IL-6, KC, MIP-2, VEGF, MIG, and eotaxin in femurs of athymic nude mice inoculated via intracardiac injection with MDA-MB-231GFP human metastatic breast cancer cells, MDA-MB-231BRMS1GFP, a metastasis suppressed variant, or PBS. Animals were euthanized (day 3, 11, 19, 27 after injection) to examine femoral cytokine levels at various stages of cancer cell colonization. The epiphysis contained significantly more cytokines than the diaphysis except for MIG which was similar throughout the bone. Variation among femurs was evident within all groups. By day 27, MCP-1, MIG, VEGF and eotaxin levels were significantly greater in femurs of cancer cell-inoculated mice. These pro-osteoclastic and angiogenic cytokines may manipulate the bone microenvironment to enhance cancer cell colonization

    Improved Boundary Layer Depth Retrievals from MPLNET

    Continuous lidar observations of the planetary boundary layer (PBL) depth have been made at the Micropulse Lidar Network (MPLNET) site in Greenbelt, MD since April 2001. However, because of issues with the operational PBL depth algorithm, the data is not reliable for determining seasonal and diurnal trends. Therefore, an improved PBL depth algorithm has been developed which uses a combination of the wavelet technique and image processing. The new algorithm is less susceptible to contamination by clouds and residual layers, and in general, produces lower PBL depths. A 2010 comparison shows the operational algorithm overestimates the daily mean PBL depth when compared to the improved algorithm (1.85 and 1.07 km, respectively). The improved MPLNET PBL depths are validated using radiosonde comparisons which suggests the algorithm performs well to determine the depth of a fully developed PBL. A comparison with the Goddard Earth Observing System-version 5 (GEOS-5) model suggests that the model may underestimate the maximum daytime PBL depth by 410 m during the spring and summer. The best agreement between MPLNET and GEOS-5 occurred during the fall and they diered the most in the winter

    Molecular changes in detrained & retrained adult jaw muscle

    A hypofunctional masticatory system was developed in 21-day-old male rats by feeding them a soft diet for 27 weeks. Retraining of a parallel group for 6 weeks was achieved by switching back to a hard diet after 21 weeks. A control group was fed a hard diet for 27 weeks. At the end of the experimental period, the expression levels of the myosin heavy chain isoform genes MYH 1 and 2 (fast), 3 (embryonic) and 7 (slow) in the deep masseter were compared using qRT-PCR analysis. The gene expressions of MYH 3 and MYH 7 were significantly higher in the rehabilitation group compared with the normal and hypofunctional group, but no significant differences were found in regards to the gene expression of MYH 1 and 2. Retraining made it possible for the slow (MYH 7) isoform levels to adapt to the increased mechanical load. The increased level of embryonic (MYH 3) isoform could be due to the need of creation of new MYH isoform

    Incidence of emergency department visits for electric rental scooters using detailed ridership data

    INTRODUCTION: Electric scooter (e-scooter) rental usage has increased exponentially around the country, expanding to more than 120 cities by the end of 2018. Early attempts to capture the safety effects of widespread adoption of this technology have been hampered by lack of accurate ridership data. Here we describe a 17-month evolution of ridership characteristics in St. Louis, Missouri, and the frequency of e-scooter rental-related injuries serious enough to require an emergency department (ED) visit over this time frame; we also provide estimates of incidence rates of injuries based on company ridership data. METHODS: We performed a combination retrospective chart review and prospective questionnaire-based analysis of adult e-scooter rental-related ED visits in both downtown St. Louis Level 1 trauma centers during the first 17 months of e-scooter rental usage (August 2018-December 2019). The retrospective portion focused on demographics, alcohol use, helmet use, disposition, operative repair, and temporal and severity markers. The prospective portion focused on more detailed crash and rider data. Finally, we used ridership data from both e-scooter rental companies in St. Louis to estimate incidence and temporal trends. RESULTS: A total of 221 patients had e-scooter rental-related ED visits. The median age of our population was 31 years with 58.8% male and 53.8% White. There were no deaths. Ninety-two patients were found to have fractures with 38% requiring surgery. Of the 21 patients diagnosed with head injury, five had an intracranial bleed. Overall incidence of ED visits related to e-scooters was 2.1 per 10,000 trips and 2.2 per 10,000 miles with the number of ED visits by month closely correlated with the number of rides per month (Pearson correlation coefficient = 0.95). CONCLUSION: The number of e-scooter rental-related injuries seen in St. Louis trauma centers was relatively low and correlated closely with overall number of rides. The number of injuries decreased and were less severe from 2018 to 2019 with infrequent intracranial injuries and a large percentage of fractures requiring operative repair
