Sarconesin: Sarconesiopsis magellanica Blowfly Larval Excretions and Secretions With Antibacterial Properties

Larval therapy (LT) is an alternative treatment for healing chronic wounds; its action is based on debridement, the removal of bacteria, and stimulating granulation tissue. The most important mechanism when using LT for combating infection depends on larval excretions and secretions (ES). Larvae are protected against infection by a spectrum of antimicrobial peptides (AMPs); special interest in AMPs has also risen regarding understanding their role in wound healing since they degrade necrotic tissue and kill different bacteria during LT. Sarconesiopsis magellanica (Diptera: Calliphoridae) is a promising medically-important necrophagous fly. This article reports a small AMP being isolated from S. magellanica ES products for the first time; these products were obtained from third-instar larvae taken from a previously-established colony. ES were fractionated by RP-HPLC using C18 columns for the first analysis; the products were then lyophilised and their antimicrobial activity was characterized by incubation with different bacterial strains. These fractions’ primary sequences were determined by mass spectrometry and de novo sequencing; five AMPs were obtained, the Sarconesin fraction was characterized and antibacterial activity was tested in different concentrations with minimum inhibitory concentrations starting at 1.2 μM. Potent inhibitory activity was shown against Gram-negative (Escherichia coli D31, E. coli DH5α, Salmonella enterica ATCC 13314, Pseudomonas aeruginosa 27853) and Gram-positive (Staphylococcus aureus ATCC 29213, S. epidermidis ATCC 12228, Micrococcus luteus A270) bacteria. Sarconesin has a significant similarity with Rho-family GTPases which are important in organelle development, cytoskeletal dynamics, cell movement, and wound repair. The data reported here indicated that Sarconesin could be an alternative candidate for use in therapeutics against Gram-negative and Gram-positive bacterial infections. Our study describes one peptide responsible for antibacterial activity when LT is being used. The results shown here support carrying out further experiments aimed at validating S. magellanica AMPs as novel resources for combating antibacterial resistance

Nursing Student´s Knowledge Assessment about Chagas Disease

Objective: To determine the level of knowledge about Chagas disease that nursing students possess in a higher education institution in the department of Santander. Materials and methods: Cross-sectional study, with students of VIII and X nursing level. For the collection of information, a questionnaire based on nursing outcome indicators was applied: knowledge of the disease process related to Chagas. A univariate and bivariate descriptive analysis of the information was carried out presenting qualitative variables by absolute and relative frequencies, quantitative variables through median and range, test hypothesis by Mann Whitney U. Results: Statistically significant evidence is present in the process indicators of the disease and complications of the disease, value p <0.02. Regarding the indicator process of the disease, 48.7% of the students obtained substantial classified knowledge and 29% moderate knowledge. 55.3% have knowledge of the complications of the disease, being substantial; Similar degree of knowledge was obtained in indicators: measures to minimize the progression of the disease (60.5%) and causes or contributing factors (52.6%); in the indicator of signs and symptoms of the disease, 52.6% of the students obtained extensive knowledge, with a NOC score of 5. Conclusion: The indicator that showed a greater degree of knowledge of Chagas disease by students was signs and symptoms with extensive knowledge. This level obtained is crucial for the diagnosis, early treatment and control of the disease by the health team

Aplicación de estrategias educativas del plan de uso eficiente y ahorro de agua para disminuir el consumo de agua en el socorro

Problema Central: En el Socorro, en época seca, el suministro de agua potable es muy deficiente ya que los embalses de La Honda y Aguilitas se encuentran en su mínimo nivel. Objetivo: Implementar las estrategias educativas del Plan de Uso Eficiente y Ahorro de Agua en la comunidad de la facultad de Ingeniería Ambiental. Metodología: Se recopiló la información para aplicar estrategias educativas, se determinó la muestra por utilizar. De acuerdo con las necesidades municipales se diseñó un volante con las 8 principales estrategias de ahorro y uso eficiente de agua. Se aplicó una entrevista diseñada especialmente para establecer generalidades del proyecto. Se dieron a conocer las estrategias en cada vivienda donde se recopilaron los recibos de agua de cada una para hacer las respectivas comparaciones. Se sistematizaron y se analizaron los resultados obtenidos. Resultados: El consumo de agua registrado después de la aplicación de las estrategias en las viviendas se redujo o se mantuvo en su mayoría y se establece que esta disminución estadísticamente no es considerable y no se logró el 20 % de ahorro esperado. Conclusión: Aplicando el Plan de Ahorro y Uso Eficiente del Agua en la comunidad universitaria no se obtuvieron los resultados esperados, ya que el tiempo y el interés por parte de la ciudadanía son factores indispensables para lograr el objetivo

Sarconesin II, a New Antimicrobial Peptide Isolated from Sarconesiopsis magellanica Excretions and Secretions

Antibiotic resistance is at dangerous levels and increasing worldwide. The search for new antimicrobial drugs to counteract this problem is a priority for health institutions and organizations, both globally and in individual countries. Sarconesiopsis magellanica blowfly larval excretions and secretions (ES) are an important source for isolating antimicrobial peptides (AMPs). This study aims to identify and characterize a new S. magellanica AMP. RP-HPLC was used to fractionate ES, using C18 columns, and their antimicrobial activity was evaluated. The peptide sequence of the fraction collected at 43.7 min was determined by mass spectrometry (MS). Fluorescence and electronic microscopy were used to evaluate the mechanism of action. Toxicity was tested on HeLa cells and human erythrocytes; physicochemical properties were evaluated. The molecule in the ES was characterized as sarconesin II and it showed activity against Gram-negative (Escherichia coli MG1655, Pseudomonas aeruginosa ATCC 27853, P. aeruginosa PA14) and Gram-positive (Staphylococcus aureus ATCC 29213, Micrococcus luteus A270) bacteria. The lowest minimum inhibitory concentration obtained was 1.9 µM for M. luteus A270; the AMP had no toxicity in any cells tested here and its action in bacterial membrane and DNA was confirmed. Sarconesin II was documented as a conserved domain of the ATP synthase protein belonging to the Fli-1 superfamily. The data reported here indicated that peptides could be alternative therapeutic candidates for use in infections against Gram-negative and Gram-positive bacteria and eventually as a new resource of compounds for combating multidrug-resistant bacteria. © 2019 by the authors

El poder extraordinario que tiene la literatura, en la vida de un ser humano, es incalculable; no sólo es abrir un libro y pasar los ojos por las páginas; ¡No! Es dejarse atrapar, dejarse llevar por mundos y contextos distintos, diversos, pletóricos de vivencias humanas que nos hacen únicos e irrepetibles. Cada vez que abrimos un libro y nos damos el permiso de leerlo, nos volvemos mejores seres humanos, porque comprendemos más al otro, o por lo menos, tratamos de reconocer cómo vivían, cómo pensaban, cómo actuaban, en otras épocas, incluyendo la nuestra. Con la lectura, podemos entender por qué los seres humanos son como son, potenciamos la empatía y fortalecemos la conciencia moral, entre algunas ventajas que tiene este proceso que nos acompaña a lo largo de toda la vida. Experiencias en el aula: tercer encuentro de prácticas pedagógicas innovadoras.

Experiencias de profesores en su quehacer en los distintos ambientes de aprendizaje presenciales y a distancia.Para el Centro de Excelencia Docente aeiou constituye un honor presentar la publicación del Tercer Encuentro de Prácticas Pedagógicas Innovadoras en el que se destacan cuarenta trabajos de profesores de UNIMINUTO provenientes de diferentes sedes. Con este encuentro son ya tres que bajo la dirección de aeiou los profesores han compartido su quehacer en los distintos ambientes de aprendizaje presenciales y a distancia. Cada año el Centro de Excelencia Docente invita a los profesores a participar en este evento, para el 2108 además de la inscripción voluntaria por parte de cada profesor, se invitó al estudiantado a que postularan a sus profesores que consideraban eran innovadores y creativos en el cumplimiento de su función docente y se obtuvo una respuesta importante por parte de los estudiantes, que para algunos profesores resultó sorpresiva porque quizás no habían considerado que el trabajo que hacían en su ambiente de aprendizaje era diferente, fuera de lo común. Luego de una evaluación de jurados nacionales e internacionales de las prácticas presentadas y de la realización del evento, que tuvo como novedad hacerlo de forma simultánea en cuatro sedes donde UNIMINUTO tiene presencia: Buga, Ibagué, Pereira y Bogotá, se comparte la presente publicación para tener como referencia y evidencia el trabajo que los profesores hacen a diario
