    Developing a Method for Rapid and Accurate Identification of Bacillus Species in Clinical Isolates Using Polymerase Chain Reaction

    Bacillus species have caused a variety of diseases in humans throughout history. These include foodborne illness, wound infections, and anthrax poisoning. With over 266 identified species within the Bacillus genus, isolates previously described as independent species (e.g. B. anthracis, B. cereus, B. thuringiensis) have been discovered to be so genetically and phenotypically similar that they are often very difficult to discriminate between. Therefore, there have been instances of misidentification reported in scientific literature. Therefore, the majority of clinical tests done may not be precise enough to distinguish between some species of Bacillus, as they are so genetically similar. A new software, called MALDI-TOF, has been very successful in species discrimination. However, this technology is relatively new and not widely available for all clinical settings. The misidentification of clinical Bacillus isolates from infected patients could have medical significance and negatively affect treatment. By finding improved approaches for correctly identifying and distinguishing between Bacillus species, the scientific community can increase its understanding of the pathogenic potential of certain Bacillus species and positively impact patient diagnosis and treatment. This study was intended to develop a technique for better distinguishing between Bacillus species. Recent research has suggested that the pycA gene may be the key to separating Bacillus species. A series of specially designed primers will be used to target the pycA gene in Bacillus thuringiensis and Bacillus cereus strains grown from isolates obtained from the NRRL repository. With the use of these primers, PCR testing will be performed on the various Bacillus strains. This will allow for identification of the isolate strains based on the PCR products and comparison to the initial repository identification. If successful, this approach could prove to be a more rapid and precise way to distinguish between Bacillus species in clinical settings, possibly improving the speed of diagnosis and treatment.     Resonance in Human Walking Economy: How Natural Is It?

    Locomotion and movement economy are cornerstone topics in movement science. Modeling the leg as a hybrid mass-spring pendulum shows walking economy should be optimized when stride frequency matches the resonant frequency of the limb. Human walking is described as self-optimizing because mean preferred (PSF) and modeled resonant (RSF) stride frequencies usually are statistically equivalent, but this depiction may not be fully justified. Purpose: To more thoroughly examine the self-optimization characterization and the consequences of obligating use of the RSF. Methods: Forty-seven individuals of diverse statures completed 3 consecutive days of preferred walking trials on a treadmill where stride rate, stride length, walking speed, heart rate and walking economy measures were made under steady state heart rate conditions. Anthropometric measures were taken to build a hybrid model of the leg and model the RSF. Reliability across days was evaluated via repeated measures analysis of variance (ANOVA) and intra-class correlation (á=.05) and correlations were calculated for PSF and RSF. A separate sample of 20 participants walked under 3 conditions, (1) completely preferred; (2) at the original preferred speed using the RSF; and (3) with the option to establish a new preferred speed while using the RSF. Results: Gait characteristics were fundamentally reliable across days and the correlation between PSF and RSF was weak (8% explained variance). Walking economy improved 14% when using the RSF and allowed to self-select the speed / stride length used at that cadence.     Anion Effects on Gas Solubility in Ionic Liquids

    This work presents the results of solubility measurements for a series of gases in 1-n-butyl-3-methyl imidazolium tetrafluoroborate and 1-n-butyl-3-methyl imidazolium bis(trifluoromethylsulfonyl) imide. The gases considered include benzene, carbon dioxide, nitrous oxide, ethylene, ethane, oxygen, and carbon monoxide. Carbon dioxide and oxygen solubilities are also reported in methyl-tributylammonium bis(trifluoromethylsulfonyl) imide, butyl-methyl pyrrolidinium bis(trifluoromethylsulfonyl) imide, and tri-isobutyl-methyl phosphonium p-toluenesulfonate. We report the associated Henry's constants and enthalpies and entropies of absorption. In general, benzene, followed by carbon dioxide and nitrous oxide, have the highest solubilities and strongest interactions with the ionic liquids, followed by ethylene and ethane. Oxygen had very low solubilities and weak interactions. Carbon monoxide had a solubility below the detection limit of our apparatus. Ionic liquids with the bis(trifluoromethylsulfonyl) imide anion had the largest affinity for CO_2, regardless of whether the cation was imidazolium, pyrrolidinium, or tetraalkylammonium. These results suggest that the nature of the anion has the most significant influence on the gas solubilities

    T helper 17 cells (Th17) are critical for fighting infections at mucosal surfaces; however, they have also been found to contribute to the pathogenesis of multiple autoimmune diseases and have been targeted therapeutically. Due to the role of Th17 cells in autoimmune pathogenesis, it is important to understand the factors that control Th17 development. Here we identify the activating transcription factor 7 interacting protein (ATF7ip) as a critical regulator of Th17 differentiation. Mice with T cell-specific deletion of Atf7ip have impaired Th17 differentiation secondary to the aberrant overproduction of IL-2 with T cell receptor (TCR) stimulation and are resistant to colitis in vivo. ChIP-seq studies identified ATF7ip as an inhibitor of Il2 gene expression through the deposition of the repressive histone mark H3K9me3 in the Il2-Il21 intergenic region. These results demonstrate a new epigenetic pathway by which IL-2 production is constrained, and this may open up new avenues for modulating its production

    Toward Automation of the Supine Pressor Test for Preeclampsia

    Preeclampsia leads to increased risk of morbidity and mortality for both mother and fetus. Most previous studies have largely neglected mechanical compression of the left renal vein by the gravid uterus as a potential mechanism. In this study, we first used a murine model to investigate the pathophysiology of left renal vein constriction. The results indicate that prolonged renal vein stenosis after 14 days can cause renal necrosis and an increase in blood pressure (BP) of roughly 30 mmHg. The second part of this study aimed to automate a diagnostic tool, known as the supine pressor test (SPT), to enable pregnant women to assess their preeclampsia development risk. A positive SPT has been previously defined as an increase of at least 20 mmHg in diastolic BP when switching between left lateral recumbent and supine positions. The results from this study established a baseline BP increase between the two body positions in nonpregnant women and demonstrated the feasibility of an autonomous SPT in pregnant women. Our results demonstrate that there is a baseline increase in BP of roughly 10-14 mmHg and that pregnant women can autonomously perform the SPT. Overall, this work in both rodents and humans suggests that (1) stenosis of the left renal vein in mice leads to elevation in BP and acute renal failure, (2) nonpregnant women experience a baseline increase in BP when they shift from left lateral recumbent to supine position, and (3) the SPT can be automated and used autonomously

    Evaluating quality of life tools in North American patients with erythropoietic protoporphyria and X-linked protoporphyria.

    BackgroundErythropoietic protoporphyria (EPP) and X-linked Protoporphyria (XLP) are rare photodermatoses presenting with severe phototoxicity. Although anecdotally, providers who treat EPP patients acknowledge their life-altering effects, tools that fully capture their impact on quality of life (QoL) are lacking.MethodsAdult patients with EPP/XLP were given four validated QoL tools: the Patient Reported Outcomes Measurement Information System 57 (PROMIS-57), the Hospital Anxiety and Depression Scale (HADS), the Illness Perception Questionnaire Revised (IPQR), and an EPP-Specific tool. All patients received the PROMIS-57 while the HADS, IPQR, and EPP-Specific tools were introduced at a later date. Associations between responses and clinical phenotypes were explored.ResultsTwo hundred and two patients were included; 193 completed PROMIS-57, 104 completed IPQR, 103 completed HADS, and 107 completed the EPP-Specific tool. The IPQR showed that patients strongly believed EPP/XLP had a negative impact on their lives. Mean scores in anxiety and depression domains of both HADS and PROMIS-57 were normal; however, anxiety scores from HADS were borderline/abnormal in 20% of patients. The EPP-Specific tool revealed a decreased QoL in most patients. The PROMIS-57 showed that 21.8% of patients have clinically significant pain interference. Several tool domains correlated with measures of disease severity, most being from the PROMIS-57.ConclusionsImpaired QoL is an important consequence of EPP/XLP. PROMIS-57 was most sensitive in evaluating impaired QoL in EPP/XLP. Further research is needed to compare the effectiveness of it for assessing response to treatment

    Predictors of Achievement When Virtual Manipulatives are Used for Mathematics Instruction

    The purpose of this study was to determine variables that predict performance when virtual manipulatives are used for mathematics instruction. This study included the following quasi-experimental design features: 1) a large number of students (N=350); 2) within-class random-assignment to treatment groups; 3) retention effects measured by post-test and delayed post-test; 4) fidelity of instructional treatments documented through observations; and, 5) instrument development for the unit of study. This design was used to determine variables that predict student performance on tests of fraction knowledge for third- and fourth-grade students in two treatment groups: classroom instruction using texts and physical manipulatives (CI), and computer lab instruction using virtual fraction applets (VM). The Pre-test, Post-test 1, and Post-test 2 measured learning and retention of fraction concepts. Observation ethograms documented representation use. The results revealed that fewer demographic predictors of student performance (e.g., socio-economic status, English language learner status, and gender) exist during fraction instruction when virtual manipulatives were used. When instructors used virtual manipulatives, there was an equalizing effect on achievement in third and fourth grade classrooms, in that fewer demographic factors were influential for VM groups compared to CI groups
