    SAFETY ASSESSMENT OF LINUM USITATISSIMUM (LINN.) INGESTION IN NEW ZEALAND RABBITS

    Background: The therapeutic safety of herbal medicine is a major concern for consumers and users. After studying the effects of linseed on hair growth in rabbits, the turn is to evaluate its safety by the observation of some clinical, biological and anatomo-pathological aspects. Materials and Methods: A study was conducted during a period of three months on two groups of rabbits (control and test). Test group daily received feed supplemented with 3g of ground linseed while the control animals received the same feed without any additives. Weekly, rabbits were weighed and monthly blood samples were taken. By the end of the trial, liver and kidneys biopsies were analyzed for histological and cytological lesions. Results: There was no significant improvement in weight gain in the test group rabbits, in which biochemical parameters had differentially evolved with a decrease in their Glycemia and cholesterolemia. There were also no modifications in their serum hepatic and renal marker enzymes and their liver and kidneys exhibited noticeably normal histology without any anatomically detectable anomalies. Conclusion: These findings confirm that prolonged linseed ingestion in rabbits is safe

    First serological and molecular investigation of hepatitis E virus infection in dromedary camels in Algeria

    Hepatitis E is an acute self-limited or fulminant infection in humans, caused by the hepatitis E virus (HEV). This member of the Hepeviridae family has been identified in a wide range of domestic and wild animals all over the world, with a possible transmission to humans through fecal oral route, direct contact and ingestion of contaminated meat products, making it one of the global zoonotic and public health major concerns. Since there is no monitoring program and a lack of data on HEV in animals in Algeria, the current preliminary survey has been undertaken to elucidate the exposure to the virus in camels at abattoirs of six southern provinces of Algeria. Two-hundred and eight sera/plasma were collected and analyzed (by double antigen sandwich ELISA) for the presence of total anti-HEV antibodies, among which 35.1% were positive, but no HEV RNA could be isolated from them (by two pan-HEV nested RT-PCR and broad range real-time reverse transcription RT-PCR). The univariate analysis showed significant associations (p < 0.05) between HEV seroprevalence and province of origin, age, and sex of camels, whereas the multivariable logistic regression analysis revealed a negative impact of camels’ age on it. The obtained results confirm that HEV infection is widespread established in the camelid population of Algeria

    Preliminary investigation of the antimicrobial and mechanisms of resistance of Enterobacteria isolated from minced meat in the Northeast of Algeria: The case of butchers from Constantine

    peer-reviewedFood products of animal origin such as fresh meat are easily contaminated by microorganisms if handling, processing and storage conditions are not fully respected. The present study aimed first to evaluate the bacterial load and microbial contamination rates of ground raw beef to identify the main pathogenic flora that dominate and second, to determine the resistance patterns and extended-spectrum beta-lactamase (ESBL) of isolated Gram-negative strains against certain families of antibiotics. Therefore, 39 samples have been collected from 5 butcher shops located in Constantine province in the North-East of Algeria. The samples were analysed for total bacterial count, presence of total and faecal coliforms, Staphylococci and Salmonella. Furthermore, 23 antibiotics were tested using the diffusion method on Mueller-Hinton agar, towards 22 strains isolates. Bacterial analyses showed a high contamination by total aerobic bacteria, total and faecal coliforms. Escherichia coli, Citrobacter spp., Enterobacter spp., Hafnia alvei, Salmonella pullorum and Staphylococcus spp (except Staphylococcus aureus) were further revealed in some samples. The results of the antibiogram test exhibit multi-resistance to more than eight antibiotics with varied effects. From the whole tested strains isolates, the fully susceptibility effect was for spectinomycin (SPT). This study reveals that the analysed minced meat was found to be highly contaminated with antibiotic resistant bacteria. This study allows concluding that appropriate use of antibiotics in compliance with good hygiene practices is essential to reduce the antibiotic resistance identified in this preliminary study

    Evaluation of crude flaxseed (Linum usitatissimum L.) oil in burn wound healing in New Zealand rabbits

    Background: Burns are among the most prevalent injuries in humans with high cost in health care and heavy prolonged or permanent physical, psychological and social consequences. Commercial antimicrobial creams and dressing agents are unsuccessful in healing deep burn wounds.Materials and Methods: A study was conducted to assess the impact of crude linseed oil (LSO) topical application on burn wounds healing in rabbits in comparison with untreated wounds (NAT) and those treated with Vaseline gel(VAG) and Cicatryl-Bio ointment (CBO). By the 28th day post burning, skin biopsies were analyzed for histological and cytological lesions. The presence of various bioactive phytochemical groups in linseed was also screened.Results: Phytochemical screening has resulted in high concentrations of flavonoids and terpenoids, low amounts of catechic tannins and total absence of alkaloids and saponosides. All along the trial, the rate of wounds contraction was found to be significantly higher in burns treated with LSO which had also a significant shorter healing period (26±5.89 days) as compared to the other treatments. LSO healed wounds included less inflammatory cells, complete epithelium regeneration with a reduced thickness of the new formed dermis, discreet fibrosis, enhanced neo-vascularization, increased number of collagen fibers, fibroblasts and many myofibroblasts. Additionally, no adverse effects of LSO on cicatrization process were recorded.Conclusion: These findings prove the safety and efficaciousness of linseed oil topical application in the therapy of burn wounds.Keywords: Linseed oil, phytochemical screening, topical application, burn wounds, healing, rabbit

    Emergence of Methicillin-Resistant Staphylococcus aureus ST239/241 SCCmec-III Mercury in Eastern Algeria

    International audienceIn this paper, we investigate the epidemiology of infections-associated Staphylococcusaureus (S. aureus) from the Medical Intensive Care Unit (MICU) at University Hospital Center of Constantine (UHCC) in Algeria, with a special emphasis on methicillin-resistant strains (MRSA) revealed by cefoxitin disks (30 μg), then confirmed by penicillin-binding protein (PBP2a) agglutination and real-time polymerase chain reaction (RT-PCR) targeting mecA and mecC genes. Staphylococcal Cassette Chromosome mec (SCCmec type), staphylococcal protein A (spa-type), multilocus sequence type (MLST), Panton–Valentine Leucocidin (PVL), and toxic shock syndrome toxin-1 (TSST-1) were further investigated in all isolates, and whole genome sequencing was performed for a selected subset of three hospital-acquired MRSA (HA-MRSA) isolates. A measurement of 80% out of the 50 S. aureus isolates were identified as HA-MRSA harbouring the mecA gene, and 72.5% of them were multidrug resistant (MDR). Twelve STs, four different SCCmec cassettes, fourteen spa types, ten isolates Panton–Valentine Leukocidin (PVL)-positive, and three isolates TSST-1 were identified. Interestingly, there was a high prevalence (n = 29; 72.5%) of a worrisome emerging clone: the HA-MRSA ST239/241 SCCmec-III mercury with PVL negative, resistant to β-lactams, aminoglycosides, quinolones, and tetracyclines. Other clones of HA-MRSA isolates were also identified, including PVL-positive ST80 SCCmec-IV/SCCmec-unknown (22.5%), ST34 SCCmec-V with TSST-1 positive (2.5%), and PVL-negative ST72 SCCmec-II (2.5%). Genome analysis enables us to describe the first detection of both PVL-negative HA-MRSA ST239/241 SCCmec-III mercury carrying ccrC, as well as SCCmec-V cassette, which dramatically changes the epidemiology of S. aureus infections in one of the hospitals in eastern Algeria

    International audienceBackground: The aims of this study were to investigate Salmonella contamination in broiler chicken farms and slaughterhouses, to assess the antibiotic resistance profile in avian and human Salmonella isolates, and to evaluate the relationship between avian and human Extended Spectrum beta-Lactamase (ESBL)-producing isolates. Salmonella was screened in different sample matrices collected at thirty- two chicken farms and five slaughterhouses. The human isolates were recovered from clinical specimens at the University Teaching Hospital of Constantine (UTH). All suspected colonies were confirmed by MALDI-TOF (Matrix Assisted Laser Desorption Ionization Time OF light) and serotyped. Susceptibility testing against 13 antibiotics including, amoxicillin/clavulanic acid, ticarcillin, cefoxitin, cefotaxime, aztreonam, imipenem, ertapenem, gentamicin, amikacin, ciprofloxacin, colistin, trimethoprim/sulfamethoxazole and fosfomycin, was performed using the disk diffusion method on Mueller-Hinton agar. ESBL-production was screened by the double-disk synergy test and confirmed by molecular characterization using PCR (polymerase chain reaction) amplification and sequencing of ESBL encoding genes. Clonality of the avian and human strains was performed using the Multi Locus Sequencing Typing method (MLST). Results: Forty-five isolated avian Salmonella strains and 37 human collected ones were studied. Five S. enterica serotypes were found in avian isolates (mainly Kentucky) and 9 from human ones (essentially Infantis). 51.11% and 26.6% of the avian isolates were resistant to ciprofloxacin and cefotaxime, respectively, whereas human isolates were less resistant to these antibiotics (13.5% to ciprofloxacin and 16.2% to cefotaxime). Eighteen (12 avian and 6 human) strains were found to produce ESBLs, which were identified as bla(CTX-M-1) (n = 12), bla(CTX-M-15) (n = 5) and bla(TEM) group (n = 8). Interestingly, seven of the ESBL-producing strains (5 avian and 2 human) were of the same ST (ST15) and clustered together, suggesting a common origin. Conclusion: The results of the combined phenotypic and genotypic analysis found in this study suggest a close relationship between human and avian strains and support the hypothesis that poultry production may play a role in the spread of multidrug-resistant Salmonella in the human community within the study region
