    Herança transgeracional: a circularidade e a concentração do trauma

    The trans-generational kind of trauma may be diffused in two basic ways. Through the circulation of the trauma, its plot is transmitted to all member of the family. Through the traumatic confluence of a specific member, in which some differences are implied. This kind constitutes the trauma of the absolute, in which there is a over-investment of hate and horror in the representations: of being abandoned, repelled, unloved, among others. Both forms may be used by the families; they are not impervious categories. Two clinical cases are presented in order to reflect upon their specificities. Concepts developed by some authors allowed the author of the present study to comprehend these questions.El trauma de carácter transgeracional puede ser propagado de dos formas básicas. En la circularidad del trauma, su trama es transmitida para todos los miembros de la familia. En la confluencia traumática sobre un miembro específico, algunos diferenciales están presentes. Este tipo constituye el trauma de lo absoluto, en el qual hay el sobre-investimiento de ódio y horror en las representaciones: ser abandonado, rechazado, no-amado, entre otras. Las dos formas pueden ser utilizadas por las familias, no siendo categorias estanques. Dos historiales clínicos son presentados de modo a pensar se sus especificidades. Conceptos de algunos pensadores han permitido a la autora pensar esas cuestiones.Les traumatismes de caractère transgénérationnelle peuvent se propager en deux formes de base. Circularité dans le traumatisme, sa trame est transmise à tous les membres de la famille. Dans le confluent traumatique d’un membre spécifique, certaines différences sont présentes. Ce type constitue le traumatisme de l’absolu, pour lequel il existe un surinvestissement de la haine et l’horreur des représentations : être abandonné, être rejeté, pas aimé, entre autres. Ces deux les formes peuvent être utilisées par les familles,ne pas être des catégories étanches. Deux historiques cliniques sont présentés pour penser à leurs spécificités. Les concepts de quelques penseurs ont permis à l’auteur de penser à ces questions.     Efficacy, Stability, and Safety Evaluation of New Polyphenolic Xanthones Towards Identification of Bioactive Compounds to Fight Skin Photoaging

    Antioxidants have long been used in the cosmetic industry to prevent skin photoaging, which is mediated by oxidative stress, making the search for new antioxidant compounds highly desirable in this field. Naturally occurring xanthones are polyphenolic compounds that can be found in microorganisms, fungi, lichens, and some higher plants. This class of polyphenols has a privileged scaffold that grants them several biological activities. We have previously identified simple oxygenated xanthones as promising antioxidants and disclosed as hit, 1,2-dihydroxyxanthone (1). Herein, we synthesized and studied the potential of xanthones with different polyoxygenated patterns as skin antiphotoaging ingredients. In the DPPH antioxidant assay, two newly synthesized derivatives showed IC₅₀ values in the same range as ascorbic acid. The synthesized xanthones were discovered to be excellent tyrosinase inhibitors and weak to moderate collagenase and elastase inhibitors but no activity was revealed against hyaluronidase. Their metal-chelating effect (FeCl₃ and CuCl₂) as well as their stability at different pH values were characterized to understand their potential to be used as future cosmetic active agents. Among the synthesized polyoxygenated xanthones, 1,2-dihydroxyxanthone (1) was reinforced as the most promising, exhibiting a dual ability to protect the skin against UV damage by combining antioxidant/metal-chelating properties with UV-filter capacity and revealed to be more stable in the pH range that is close to the pH of the skin.     Pharmaceutical services in primary health care : are pharmacists and users on the same page?

    This study investigated structural and organizational characteristics of the Pharmaceutical Services based on Primary Health Care (PHCPS) from the viewpoints of users and pharmacists. A mixed method design was applied, combining one-to-one semi-structured interviews with four pharmacists in charge of five public dispensing facilities and 69 users, with a secondary database analysis. Data were collected from February to August 2014 in Divinopolis, a municipality in Minas Gerais State. PHCPS were similar in terms of general activities performed and staff profile and background. While users were concerned about medicines' availability and improvements related to the PHCPS' conveniences and personnel, pharmacists pointed out problems regarding infrastructure to storage. Despite most users had low information on how to use their medicines, no pharmacists declared to participate in medicines dispensing activities. There was a low match between users and pharmacists viewpoints and advantages for concentrate medicines dispensing in a smaller number of facilities were not clear

ReisParacoccidioides brasiliensis is the causative agent of paracoccidioidomycosis, a disease that affects 10 million individuals in Latin America. This report depicts the results of the analysis of 6,022 assembled groups from mycelium and yeast phase expressed sequence tags, covering about 80% of the estimated genome of this dimorphic, thermo-regulated fungus. The data provide a comprehensive view of the fungal metabolism, including overexpressed transcripts, stage-specific genes, and also those that are up- or down-regulated as assessed by in silico electronic subtraction and cDNA microarrays. Also, a significant differential expression pattern in mycelium and yeast cells was detected, which was confirmed by Northern blot analysis, providing insights into differential metabolic adaptations. The overall transcriptome analysis provided information about sequences related to the cell cycle, stress response, drug resistance, and signal transduction pathways of the pathogen. Novel P. brasiliensis genes have been identified, probably corresponding to proteins that should be addressed as virulence factor candidates and potential new drug targets

    Eficácia clínica de novos estudos para o tratamento da Dermatite Atópica: uma revisão integrativa

    A dermatite atópica (DA) é a doença de pele mais comum dentre as doenças inflamatórias crônicas da pele, acometendo de 10 a 20% das crianças e até 10% dos adultos em países desenvolvidos. O presente estudo de revisão buscou avaliar a eficácia clínica de novos estudos para o tratamento da dermatite atópica, documentados por meio de estudos clínicos e randomizados. Trata-se de uma pesquisa de revisão integrativa realizada por meio da base de dados PubMed, que levou em consideração os seguintes critérios de inclusão: ensaios clínicos e testes controlados e randomizados; artigos publicados no último ano; que possuíam texto completo disponível e que abordassem acerca da terapêutica da dermatite atópica (DA). Ficou constatado que o brepocitinibe tópico se mostrou bem tolerado nos pacientes que possuem DA de grau leve e moderado, da mesma forma que o upadacitinibe, o qual apresenta um perfil de risco-benefício favorável para o  tratamento da DA moderada e grave em adolescentes. Além disso, o abrocitinibe também demonstrou resultados eficazes, com melhora da qualidade de vida e da coceira noturna em pacientes com DA. Verificou-se, ainda, que os anticorpos neutralizantes de IL-13 apresentam eficácia clínica comprovada em pacientes com DA, em especial quando associados aos costicosteroides tópicos. Essa associação com o traloquinumabe trouxe melhorias de sintomas, com redução de interferência no sono relacionado ao eczema, resultados semelhantes de eficácia quando os corticosteroides são associados ao lebrikizumabe
