    Inventory and world geographical distribution of the olive tree (Olea europaea L.) diseases caused by viruses, bacteria and phytoplasma

    The objective of this work is to prepare an Inventory about the viral and bacterial diseases of the olive tree (Olea europaea L.) and to study their Geographical distribution in the world. Fifteen viruses were considered as pathogens on the olive tree, these viruses were reported in 22 countries in five continents showing different symptoms on different olive tree varieties. All viruses were reported in Italy, some of these viruses were found only in the Eurepean continent, such as Olive vein yellows associated virus (OVYaV), Olive yellow mottling and decline associated virus (OYMDaV), Olive mild mosaic virus (OMMV) and Olive semilatent virus (OSLV). Two bacteria were reported as pathogen on the olive tree; Pseudomonas savastanoi. pv. savastanoi responsible bacteria of thr olive knot; a serious disease of olive (Olea europaea L.) in the Mediterranean region. This bacteria was detected in 22 countries in five continents, the majority of these countries exist in the Mediterranean Basin. Another bacteria was Xylella fastidiosa responsible of the quick decline syndrome of olive (OQDS), was deteted in Europe in four countries (Italy, Spain, Portugal and Slovenia) and in America in three countries (USA, Argentina and Brazil). Phtoplasmas was reported in 5 countries in three continents (Europe, Asia and Australia)

    Combined concomitant boost radiotherapy and chemotherapy in stage III-IV head and neck carcinomas: A comparison of toxicity and treatment results with those observed after radiotherapy alone

    Background Alteration of radiation therapy (RT) fractionation and the combination of chemotherapy (CT) with RT represent two predominant fields of current research in the treatment of head and neck carcinomas. To assess the potential integration of these two fields, a retrospective comparison of toxicity and treatment outcome was carried out in stage III-IV patients treated with a concomitant boost RT schedule with or without CT. Patients and methods Fifty-two patients were treated by RT alone and 35 by RT and CT. In the RT group, there were significantly fewer T3-4 tumors (56% vs. 88%, P=0.002) and higher proportion of planned neck dissections (35% vs. 14%, P=0.047). The planned total dose was 69.9 Gy delivered over 5.5 weeks. In 10 cases CT was given before RT and in 25 concomitantly with RT, either alone or with neoadjuvant and/or adjuvant CT. All patients but two had cisplatin-based (CDDP, 100 mg/m2) CT, associated in 28 patients with 5-fluorouracil (5-FU, 1000 mg/m2/24 h Ă— 5). The median follow-up for the surviving patients was 21 and 31 months for the RT and RT-CT groups respectively. Results Grade 3-4 acute toxicity (RTOG) was observed in 73% and 86% of patients, and grade 3 dysphagia in 31% and 57% (P=0.02) respectively in the RT and RT-CT groups. The rates of grade 3-4 late complications were similar in the two groups (5% vs. 12%). At three years, actuarial loco-regional control (LRC) was 57% and 66% (P%0.66) and overall survival was 56% and 47% (P=0.99) in the RT and RT-CT groups respectively. Conclusion While acute toxicity was higher compared with RT alone, this accelerated RT schedule was feasible in association with 5-FU/CDDP, even administered concomitantly. Despite the significant proportion of more advanced disease in the RT-CT group, LRC was similar to that obtained by RT alone. Combinations of concomitant boost RT and chemotherapy merit further investigation in prospective trial

    Multiple myeloma presenting with external ear canal mass

    The manifestations of multiple myeloma are protean and related to bony osteolytic lesions, and to medullar and renal insufficiency. We report a patient who presented with otalgia as the inaugural symptom of multiple myeloma. Local irradiation combined with systemic chemotherapy led to the disappearance of the temporal bone mass and the accompanying symptoms. To date, 24 months after the diagnosis, the patient is still in remission. The literature on otological involvement in multiple myeloma is reviewed.     Neoadjuvant radiochemotherapy for locally advanced gastric cancer: a phase I-II study

    Background: To study in a phase I-II trial the maximum tolerated dose, the toxicity, and the tolerance of adding radiotherapy to systemic chemotherapy administered preoperatively in patients with locoregionally advanced gastric adenocarcinoma. Patients and methods: Patients with adenocarcinoma of the stomach (T3-4Nany or TanyN+), performance status ≤1, normal hematological, hepatic and renal functions received two cycles of cisplatin 100 mg/m2 on day 1, 5-FU 800 mg/m2 on days 1 to 4 and leucovorin 60 mg b.i.d. on days 1 to 4 q3w, concomitantly with radiation therapy escalated in three dose tiers (31.2, 38.4 and 45.6 Gy). Results: Nineteen patients were accrued and 18 completed neoadjuvant therapy. Major toxicity consisted of grade 3/4 leucopenia and mucositis in 89% and 36% of the patients, respectively. Only one episode of febrile neutropenia was recorded. Dose level number 2 (38.4 Gy) with the chemotherapy given q4w is the recommended dose level. All patients were subsequently operated and no fatalities occurred. Pathological assessment showed one complete and eight partial responses. Two- and 3-year relapse-free survival rates were 57% and 50%, respectively. Only one patient relapsed locally. The peritoneum was the most frequent site of relapse. Conclusions: This neoadjuvant therapeutic program is relatively well tolerated, does not seem to increase the operative risk, and might increase the locoregional control of the disease.     The purpose of this study was to assess the potential prognostic and/or predictive value of the expression of cyclin D1, cyclin E, and p21 protein in a series of 98 anal carcinomas (T1-4, N0-3) treated by radiotherapy with (51) or without (47) chemotherapy in one institution. Correlation with Mib1 index and p53 expression was also investigated. Median follow-up for surviving patients was 124 months (range: 30-266). Immunohistochemical staining was performed on pretreatment biopsies, applying a standard ABC technique for cyclin D1 (clone DSC6, DAKO, 1 : 300), cyclin E (clone 13A3, Novocastra, 1 : 100), p21(WAF/CIP1) (clone SX118, DAKO, 1 : 50), p53 (clone DO7, DAKO, 1 : 200), and Mib1 (Ki-67, Dianova, 1 : 20). Tumours were classified into low- or high-expression groups according to the expression level of the protein considered. High expression was found in 51% of tumours for cyclin E, in 33.7% for cyclin D1, and in 65% for p21. None of those factors were significantly associated with clinical variables such as advanced T or N categories. In a monovariate analysis, advanced T and N categories and longer overall treatment time were the only variables that correlated significantly with low rate of local control (LC) and disease-free survival. However, in a subgroup analysis, high p21 expression correlated with a trend for significantly higher 5-year LC (87 vs 68%, P=0.07) in the N0 patients. The results of this study suggest that the cell-cycle proteins investigated are unlikely to be clinically useful in predicting treatment response or prognosis in patients with anal carcinomas

    Oxaliplatin combined with irinotecan and 5-fluorouracil/leucovorin (OCFL) in metastatic colorectal cancer: a phase I-II study

    Background:: A phase I-II multicenter trial was conducted to define the maximal tolerated dose and describe the activity of an OCFL combination using oxaliplatin (OHP), irinotecan (CPT-11) and 5-fluorouracil (FU)/leucovorin (LV) in metastatic colorectal cancer (CRC). Patients and methods:: CRC patients not pretreated with palliative chemotherapy, with performance status ≤1 and adequate haematological, kidney and liver function, were eligible. Treatment consisted in weekly 24-h infusion 5-FU (2300 mg/m2)/LV (30 mg) and alternating OHP (70-85 mg/m2, days 1 and 15) and CPT-11 (80-140 mg/m2, days 8 and 22) repeated every 5 weeks. OHP and CPT-11 were escalated in cohorts of three to six patients. Results:: Thirty patients received a median of five cycles. Dose-limiting toxicity occurred at dose level 3, and the recommended dose was OHP 70 mg/m2, CPT-11 100 mg/m2, LV 30 mg and 5-FU 2300 mg/m2/24 h. Grade ≥3 toxicities were diarrhea 23%, neutropenia 20%, fatigue 7%, and neurologic 7%. Two febrile neutropenia episodes (one fatal) were recorded. Among 28 patients with measurable disease (90%), we observed two complete and 20 partial responses; overall RR was 78% (95% CI, 59% to 92%). Median time to progression and overall survival were 9.5 and 25.4 months, respectively. Seven patients underwent liver metastases resection.     In this study, four species of Entolomataceae family, were studied namely Entoloma papillatum, Entoloma rhodopolium, Nolanea hirtipes and N. cucullata are harvested for the first time in Morocco. The genus Entoloma' species were encountered in Quercus suber in the forest of Mamora (Northwestern Morocco) and those of the Nolanea' genus under Quercus rotundifolia in the forest El Harcha (Northeast of the Central Plateau)

    Genotype x nutrition interactions in European sea bass (Dicentrarchus labrax): effects on gut health and intestinal microbiota

    One of the main objectives of the present study was an effective replacement of dietary fishmeal/fish oil (FM/FO) by new raw materials without negatively affecting European sea bass (Dicentrarchus labrax) performance and health status within a selection breeding context. The genomic selection of this species is still in its infancy if compared to salmonids and in particular, the role of D.labrax genome in shaping the gut microbiome, has been scarcely investigated. Wildtype (WT) and selected (GS) sea bass were fed with two diets: a control (20% FM/ 5–9% FO), and a “future” (F) diet, in which FM was decreased to 10% being replaced by poultry meal, whereas FO was completely replaced by a blend of rapeseed, poultry, and algae oils. The morphological evaluation of the intestine revealed a well-organized folding pattern and a conserved gut epithelial barrier for all fish groups. Despite a basal level of inflammation in the proximal intestine of WT fish, no differences were observed neither in the morphometric characteristics of goblet cells nor in the expression of GALT-related genes in response to fish genotype or diet. At distal intestine, WT fish showed a higher inflammatory status and larger goblet cells than GS fish and within the same genotype, fish fed the F diet had in general larger goblet cells. In distal intestine, a significant effect was found on the expression of 3 out of 7 target GALT genes. In particular, the expression of cytokines il-1β, tnf-α, and il-10 was different, showing an interaction effect diet x genotype. Diet had a lower influence upon gut bacterial composition than genotype. Indeed, regardless of the diet, WT fish showed higher species richness than GS genotype and this could be a direct consequence of selective breeding for multiple traits selection including growth, external morphology for lower abdominal fat deposition and adaptation to multiple and successive feed sources and composition across generations of selection. Furthermore, the gut microbiota of GS fish shared a reduced individual variability, indicating an enhanced capacity to cope with changes in diet composition. The less changes of GS sea bass at the level of gut bacterial composition in cumulating data collected with the feeds, demonstrate a capacity to reshape their microbiota thus better adapting to the diet, but with no negative impact on their growth performances, and even a better growth. A significant genotype effect was found for specific bacterial taxa, such as Paracoccus genus and other genera belonging to Moraxellaceae family, which were enriched in WT fish, regardless of the diet.     Pathogenicity of Helminthosporium rostrata on rice varieties widely grown in Morocco

    The plants of rice varieties (Arco, Thaibonnet and Elio) were inoculated with three isolates of Helminthosporium rostrata (HR1 HR2 and HR3), isolated for the first time in Morocco from the rice seed of Taibonnet variety at the end of the growing season. The results obtained showed that all the isolates are able to induce the disease on rice plants and sporulate on the foliar lesions. HR1, HR2 and HR3 was respectively the most pathogenic on Elio (I.C = 113), Arco (I.C = 212.5), and Taibonnet (130.48). The symptoms induced by the isolates are similar to those induced by Helminthosporium sativum on rice
