    GIS, GPS, and Remote Sensing Technologies in Extension Services: Where to Start, What to Know

    The rapid development and integration of spatial technologies such as Geographic Information Systems, the Global Positioning System, and remote sensing, have created many new tools for Extension professionals, but have also widened the digital divide, leaving many with little understanding of the technology and potential applications. This article presents examples of Extension-related applications of GIS-GPS-RS technologies and discusses how to go about learning more and determining if these technologies would be useful. Examples and recommendations are taken from the literature and from the authors\u27 own experience

    Using a Participatory Stakeholder Process to Plan Water Development in Koraro, Ethiopia

    This article reports the results of a one-day participatory workshop in Koraro, Ethiopia conducted prior to major development interventions in the region. The methodology of the workshop, structured to generate data useful for understanding the physical and social systems integral to water resources planning, provides a framework for future water need explorations in similar settings in Ethiopia and elsewhere. The use of only improved water sources as a metric for access to water under-represents the situation in Koraro, as many rely on streambeds for water due to the perceived cleanliness and low salinity of this unimproved water source. The reliance on metrics common in the Millennium Development Goals and Sustainable Development Goals, such as a minimum distance to a water source and the categorization of potable water based on type of water source, using varying figures (from as many as 30 to as few as four) can lead to assessments regarding the amount of additional sources necessary to allow access to specific locales, that are not consistent with actual need. Since the workshop, the Millennium Village Project has constructed over 30 wells in the region, following the most commonly used distance and source type metrics with less than desirable results. The water access evaluations alone do not address the needs of Koraro residents

    This paper describes a case of colic that ended in death due to gastric impaction occurred to a mare of 12 years of age. This case was triggered by the accidental and voracious consumption of concentrate feed intended for alpacas during a failure of supervision by staff. The clinical picture was characterized by severe depression, severe abdominal pain, lateral decubitus and inability to rise, with a rapid course and poor prognosis which turned fatal. Gross lesions included severe stomach distension, which was impacted with plenty content partially digested, fibrous and dry, with mucosal ulcerations and presence of serous exudate in the abdominal cavity.El presente reporte describe un caso clínico de cólico que terminó en muerte, por impacción gástrica de origen alimenticio ocurrido una yegua de 12 años. Este caso se desencadenó por el consumo accidental y voraz de alimento concentrado para alpacas durante un descuido del personal. El cuadro clínico se caracterizó por depresión severa, dolor abdominal intenso, decúbito lateral e incapacidad para levantarse, siendo el curso rápido, de pronóstico grave, que devino en muerte. Las lesiones macroscópicas incluyeron distensión severa del estómago, el cual estaba impactado con abundante contenido parcialmente digerido, fibroso y seco, con úlceras en la mucosa y presencia de exudado seroso en la cavidad abdominal

    Protective Antigens Against Glanders Identified by Expression Library Immunization

    Burkholderia are highly evolved Gram-negative bacteria that primarily infect solipeds but are transmitted to humans by ingestion and cutaneous or aerosol exposures. Heightened concern over human infections of Burkholderia mallei and the very closely related species B. pseudomallei is due to the pathogens’ proven effectiveness as bioweapons, and to the increased potential for natural opportunistic infections in the growing diabetic and immuno-compromised populations. These Burkholderia species are nearly impervious to antibiotic treatments and no vaccine exists. In this study, the genome of the highly virulent B. mallei ATCC23344 strain was examined by expression library immunization for gene-encoded protective antigens. This protocol for genomic-scale functional screening was customized to accommodate the unusually large complexity of Burkholderia, and yielded 12 new putative vaccine candidates. Five of the candidates were individually tested as protein immunogens and three were found to confer significant partial protection against a lethal pulmonary infection in a murine model of disease. Determinations of peripheral blood cytokine and chemokine profiles following individual protein immunizations show that interleukin-2 (IL-2) and IL-4 are elicited by the three confirmed candidates, but unexpectedly interferon-γ and tumor necrosis factor-α are not. We suggest that these pathogen components, discovered using genetic immunization and confirmed in a conventional protein format, will be useful toward the development of a safe and effective glanders vaccine

    Establishing induced pluripotent stem cell lines from two dominant optic atrophy patients with distinct OPA1 mutations and clinical pathologies

    Dominant optic atrophy (DOA) is an inherited disease that leads to the loss of retinal ganglion cells (RGCs), the projection neurons that relay visual information from the retina to the brain through the optic nerve. The majority of DOA cases can be attributed to mutations in optic atrophy 1 (OPA1), a nuclear gene encoding a mitochondrial-targeted protein that plays important roles in maintaining mitochondrial structure, dynamics, and bioenergetics. Although OPA1 is ubiquitously expressed in all human tissues, RGCs appear to be the primary cell type affected by OPA1 mutations. DOA has not been extensively studied in human RGCs due to the general unavailability of retinal tissues. However, recent advances in stem cell biology have made it possible to produce human RGCs from pluripotent stem cells (PSCs). To aid in establishing DOA disease models based on human PSC-derived RGCs, we have generated iPSC lines from two DOA patients who carry distinct OPA1 mutations and present very different disease symptoms. Studies using these OPA1 mutant RGCs can be correlated with clinical features in the patients to provide insights into DOA disease mechanisms

    Reversal of cell, circuit and seizure phenotypes in a mouse model of DNM1epileptic encephalopathy

    Dynamin-1 is a large GTPase with an obligatory role in synaptic vesicle endocytosis at mammalian nerve terminals. Heterozygous missense mutations in the dynamin-1 gene (DNM1) cause a novel form of epileptic encephalopathy, with pathogenic mutations clustering within regions required for its essential GTPase activity. We reveal the most prevalent pathogenic DNM1 mutation, R237W, disrupts dynamin-1 enzyme activity and endocytosis when overexpressed in central neurons. To determine how this mutation impacted cell, circuit and behavioural function, we generated a mouse carrying the R237W mutation. Neurons from heterozygous mice display dysfunctional endocytosis, in addition to altered excitatory neurotransmission and seizure-like phenotypes. Importantly, these phenotypes are corrected at the cell, circuit and in vivo level by the drug, BMS-204352, which accelerates endocytosis. Here, we demonstrate a credible link between dysfunctional endocytosis and epileptic encephalopathy, and importantly reveal that synaptic vesicle recycling may be a viable therapeutic target for monogenic intractable epilepsies.<br/

    Capturing health and eating status through a nutritional perception screening questionnaire (NPSQ9) in a randomised internet-based personalised nutrition intervention : the Food4Me study

    BACKGROUND: National guidelines emphasize healthy eating to promote wellbeing and prevention of non-communicable diseases. The perceived healthiness of food is determined by many factors affecting food intake. A positive perception of healthy eating has been shown to be associated with greater diet quality. Internet-based methodologies allow contact with large populations. Our present study aims to design and evaluate a short nutritional perception questionnaire, to be used as a screening tool for assessing nutritional status, and to predict an optimal level of personalisation in nutritional advice delivered via the Internet. METHODS: Data from all participants who were screened and then enrolled into the Food4Me proof-of-principle study (n = 2369) were used to determine the optimal items for inclusion in a novel screening tool, the Nutritional Perception Screening Questionnaire-9 (NPSQ9). Exploratory and confirmatory factor analyses were performed on anthropometric and biochemical data and on dietary indices acquired from participants who had completed the Food4Me dietary intervention (n = 1153). Baseline and intervention data were analysed using linear regression and linear mixed regression, respectively. RESULTS: A final model with 9 NPSQ items was validated against the dietary intervention data. NPSQ9 scores were inversely associated with BMI (β = -0.181, p < 0.001) and waist circumference (Β = -0.155, p < 0.001), and positively associated with total carotenoids (β = 0.198, p < 0.001), omega-3 fatty acid index (β = 0.155, p < 0.001), Healthy Eating Index (HEI) (β = 0.299, p < 0.001) and Mediterranean Diet Score (MDS) (β = 0. 279, p < 0.001). Findings from the longitudinal intervention study showed a greater reduction in BMI and improved dietary indices among participants with lower NPSQ9 scores. CONCLUSIONS: Healthy eating perceptions and dietary habits captured by the NPSQ9 score, based on nine questionnaire items, were associated with reduced body weight and improved diet quality. Likewise, participants with a lower score achieved greater health improvements than those with higher scores, in response to personalised advice, suggesting that NPSQ9 may be used for early evaluation of nutritional status and to tailor nutritional advice.     Brote de Alta Mortalidad en Terneros Lecheros por Diarrea Neonatal Producida por Cryptosporidium sp Asociado a Bacteriemia en un Establo Lechero de Lima

    This paper presents and discusses the clinical and pathological findings of an outbreak of neonatal diarrhea that caused a high mortality in a dairy farm in Lima, Peru. The diarrheas did not respond to antibiotics and occurred in calves less than one month old. The animals showed signs of dehydration, fluid feces, fever and marked depression of sensory. They were negative for coronavirus and rotavirus but were positive for Criptosporidium sp. In the post mortem study, a severe diffuse acute mucohemorrhagic enterocolitis was found and the histopathological diagnosis corroborated that it was a severe acute diffuse catarrhal enteritis associated with Cryptosporidium sp and bacterial nests. In the barn was found deficiencies in the management of colostrum and hygiene of the containers in which the milk was offered to the calves. The problem was solved introducing a series of good practices in calf handling.En el presente trabajo se presenta y discute los hallazgos clínicos y patológicos de un brote de diarrea neonatal que causó una alta mortalidad en un establo lechero de Lima, Perú. Los cuadros de diarrea no respondían a la antibioterapia y se presentaron en terneros menores de un mes de edad. Los animales presentaban signos de deshidratación, heces líquidas, fiebre y marcada depresión de sensorio. Fueron negativos a coronavirus y rotavirus, pero fueron positivos a Criptosporidium sp. Al estudio posmortem se encontró una severa enterocolitis mucohemorrágica difusa aguda y al diagnóstico histopatológico se corroboró que se trataba de una severa enteritis catarral difusa aguda asociada a Cryptosporidium sp y nidos bacterianos. En el establo se encontró deficiencias en el manejo del calostro e higiene de los recipientes en los que se ofrecía la leche a los terneros. El problema fue solucionado introduciendo una serie de buenas prácticas de manejo del terneraje

    Conservation of the endemic dwarf carnivores of Cozumel Island, Mexico.

    Cozumel Island, Mexico, harbours two endemic species of dwarf procyonids: the Pygmy Raccoon Procyon pygmaeus and the Dwarf Coati Nasua nelsoni. Both species are Critically Endangered, and are among the world&rsquo;s most threatened Carnivora. Here we summarise the research we have been conducting on their ecology, evolution, genetics, and conservation. We also summarise the conservation initiatives we have been undertaking and promoting in order to advance the conservation of these unique species and their habitats. This effort illustrates the importance of an interdisciplinary approach in conservation science and action in maximising effectiveness. Nevertheless, the precarious status of the species make it imperative to continue and expand the work we have carried out in Cozumel to prevent two imminent global extinctions.<br /

Las actividades se han realizado con el apoyo del Programa de Cooperación Universitaria PCI-AECID IBEROAMÉRICA, de la Dirección de Cooperación para el Desarrollo de la Universidad Politécnica de Madrid y de los fondos propios de las Universidades latinoamericanas, con especial mención a la Universidad de Costa Rica, coordinadora de los trabajos en Centroamérica.
