    The Mysteries of the White Truffle: Its Biology, Ecology and Cultivation

    Tuber magnatum Picco is the most expensive of the truffles and a great deal of research has been carried out in an attempt to solve the mysteries of its ecology and biology. However, considerable work remains to be done particularly on those secrets of its life cycle that remain a mystery. It is known that T. magnatum is heterothallic, but it has yet to be determined how fertilization occurs between the two strains of different mating types. It is also known that the white truffle is an ectomycorrhizal fungus, and its mycorrhizas can be produced in greenhouses, but then they seem to disappear in the field. The role of other soil microorganisms, fungi and bacteria, on its soil mycelial development and fructification is intriguing but is far from being completely understood. All these uncertainties have made the cultivation of T. magnatum extremely difficult and only recently have we had the scientific proofs that it is possible. Even so, many questions remain unanswered and the management practices of T. magnatum plantations are still to be better defined to also enable the taming of this truffle

    In vitro interactions between Bradyrhizobium spp. and Tuber magnatum mycelium

    Tuber magnatum is the most expensive truffle, but its large-scale cultivation is still a challenge compared to other valuable Tuber species. T. magnatum mycelium has never been grown profitably until now, which has led to difficulties to studying it in vitro. This study describes beneficial interactions between T. magnatum mycelium and never before described bradyrhizobia, which allows the in vitro growth of T. magnatum mycelium. Three T. magnatum strains were co-isolated on modified Woody Plant Medium (mWPM) with aerobic bacteria and characterised through microscopic observations. The difficulties of growing alone both partners, bacteria and T. magnatum mycelium, on mWPM demonstrated the reciprocal dependency. Three bacterial isolates for each T. magnatum strain were obtained and molecularly characterised by sequencing the 16S rRNA, glnII, recA and nifH genes. Phylogenetic analyses showed that all nine bacterial strains were distributed among five subclades included in a new monophyletic lineage belonging to the Bradyrhizobium genus within the Bradyrhizobium jicamae supergroup. The nifH genes were detected in all bacterial isolates, suggesting nitrogen-fixing capacities. This is the first report of consistent T. magnatum mycelium growth in vitro conditions. It has important implications for the development of new technologies in white truffle cultivation and for further studies on T. magnatum biology and genetics

    Map of suitability for the spontaneous growth of Tuber magnatum in Emilia-Romagna (Italy)

    In this work we used an inductive and deductive approach to produce the map of Suitability for the Spontaneous Growth of Tuber magnatum (white truffle) (SSGT) in Emilia-Romagna region (northern Italy). This map was produced in order to identify the environments where appropriate actions should be applied to protect this threatened truffle species.The steps used to define the map were: 1) surveying and mapping the actual productive areas (APAs) in two provinces of Emilia-Romagna (Bologna and Modena); 2) identification of some morphological, climatic, vegetational and pedological properties related to T. magnatum development; 3) production of digital maps representing the value that each property assumes in discrete land portions; 4) overlapping the map of APAs with each of the thematic maps and selection of the properties showing the strongest relationships with the presence of T. magnatum; 5) creating the map of SSGT, covering the whole hilly area of Emilia Romagna region; 6) assessing the reliability of the SSGT map, by overlapping the APAs map and the areas of the provinces of Parma and Piacenza where the mycelium of T. magnatum was found using specific PCR.The relationships found by means of the inductive approach (comparison between selected properties and observed frequency of truffle) demonstrated its effectiveness in predicting deductively the areas with distinct suitability for truffle

    The role of wild boars in spore dispersal of hypogeous fungi

    Wild boars (Sus scrofa L.) are well-known for soil disturbance in natural and cultivated truffières but their role in spore dispersal is poorly investigated. In the present work we studied the occurrence of hypogeous fungal spores in faecal contents of 14 wild boars randomly hunted in “Parco dei Gessi e Calanchi dell’Abbadessa” Regional Park (North of Italy) where truffle production has been previously investigated for three years. Six methods for spore analysis in faeces were compared and the suspension of faeces in ZnSO4 (70%) solution resulted to be the most reliable. Hypogeous fungal spores, including Tuber magnatum and Tuber aestivum spores, were detected in 9 animals. This result suggests that the detection of fungal spores in faeces of wild boars may provide information on the presence of hypogeous fungi in an area. However, the poor abundance of spores suggests that the wild boar can be considered an opportunistic mycophagist, ingesting truffles only occasionally, as a seasonal source of food. Considering the magnitude of wild boar movements during seasonal migrations, it is possible to speculate that they play a key role in truffle long distance dispersal

    Comparison of two schizophyllum commune strains in production of acetylcholinesterase inhibitors and antioxidants from submerged cultivation

    In recent years, fungi have been recognized as producers of acetylcholinesterase (AChE) inhibitors, agents important for the prevention of Alzheimer’s disease (AD). This study aimed to examine the AChE inhibitory, the antioxidative and antibacterial activity of two different Schizophyllum commune strains that originated from Serbia (SRB) and Italy (IT). Submerged cultivation of grown mycelia (M) and fermentation broth (F) of ethanol (EtOH) and polysaccharide (PSH) extracts lasted for 7, 14, 21 and 28 days. For AChE activity Ellman method was performed, while for antioxidative activity, sevendifferent assays were conducted: DPPH, ABTS, FRAP, SOA, OH, NO together with total phenolic content. Antimicrobial screen, LC–MS/MS technique and FTIR measurements were performed. Different isolates exhibited different AChE activity, with PSH being the strongest (SRB, M, 28 days IC90 79.73 ± 26.34 µg/mL), while in EtOH extracts, IT stood out (F, 14 days, IC50 0.8 ± 0.6 µg/mL). PSH extracts (7 days) exhibit significant antioxidative activity (AO), opposite to EtOH extracts where 14 and 21days periods stood out. Only tw extracts showed antibacterial activity. Following LC–MS/MS analysis p-hydroxybenzoic and gallic acids were the most abundant phenolics. PSH extracts demonstrated remarkable results, making this study debut and introducing S. commune as a valuable resource of AChE inhibitors

    Software engineering for self-adaptive systems:research challenges in the provision of assurances

    The important concern for modern software systems is to become more cost-effective, while being versatile, flexible, resilient, dependable, energy-efficient, customisable, configurable and self-optimising when reacting to run-time changes that may occur within the system itself, its environment or requirements. One of the most promising approaches to achieving such properties is to equip software systems with self-managing capabilities using self-adaptation mechanisms. Despite recent advances in this area, one key aspect of self-adaptive systems that remains to be tackled in depth is the provision of assurances, i.e., the collection, analysis and synthesis of evidence that the system satisfies its stated functional and non-functional requirements during its operation in the presence of self-adaptation. The provision of assurances for self-adaptive systems is challenging since run-time changes introduce a high degree of uncertainty. This paper on research challenges complements previous roadmap papers on software engineering for self-adaptive systems covering a different set of topics, which are related to assurances, namely, perpetual assurances, composition and decomposition of assurances, and assurances obtained from control theory. This research challenges paper is one of the many results of the Dagstuhl Seminar 13511 on Software Engineering for Self-Adaptive Systems: Assurances which took place in December 2013
