    Seismic site response of unstable steep slope using noise measurements : the case study of Xemxija Bay area, Malta

    Landslide phenomena involve the northern coast of Malta, affecting in particular the urban area of Xemxija. Limestones overlying a clayey formation represent the shallower lithotypes that characterize the surficial geology of this area, where lateral spreading phenomena and rockfalls take place. Ambient noise records, processed through spectral ratio techniques, were analysed in order to characterize the dynamic behavior of the rock masses affected by the presence of fractures linked to the landslide body existing in the area. Experimental spectral ratios were also calculated after rotating the horizontal components of the seismic signal, and a direct estimate of the polarization angle was also performed in order to investigate the existence of directional effects in the ground motion. The results of the morphologic survey confirmed the existence of large cliff-parallel fractures that cause cliff-edge and unstable boulder collapses. Such phenomena appear connected to the presence, inside the clay formation, of a sliding surface that was identified through the interpretation of the noise measurement data. The boundaries of the landslide area appear quite well defined by the pronounced polarization effects, trending in the northeastern direction, observed in the fractured zone and in the landslide body in particular.peer-reviewe

    Rosmarinus officinalis extract inhibits human melanoma cell growth.

    Rosmarinus officinalis L. is receiving increasing attention due to its anti-inflammatory and antioxidative constituents. Our recent studies showed that R. officinalis extract, containing 31.7 % of carnosic acid, was able to counteract the deleterious effects of UV-R, by protecting plasmid DNA from hydroxyl radicals generated by UV-A. In this work, we evaluated the effects of this extract on pBR322 DNA cleavage induced by nitric oxide, and the growth inhibitory activity against two human melanoma cell lines, M14 and A375. The extract showed a protective effect on plasmid DNA damage, and at concentrations of 10-80 ÎĽg/mL was able to reduce significantly (p<0.001) the growth (MTT assay) of both melanoma cell lines. In addition, our results indicate that apoptotic cell demise is induced in M14 and A375 cells. No statistically significant increase in LDH release was observed in melanoma cells, correlated to a fragmentation of genomic DNA, determined by COMET assay

    Analysis of apyrase 5' upstream region validates improved Anopheles gambiae transformation technique

    <p>Abstract</p> <p>Background</p> <p>Genetic transformation of the malaria mosquito <it>Anopheles gambiae </it>has been successfully achieved in recent years, and represents a potentially powerful tool for researchers. Tissue-, stage- and sex-specific promoters are essential requirements to support the development of new applications for the transformation technique and potential malaria control strategies. During the <it>Plasmodium </it>lifecycle in the invertebrate host, four major mosquito cell types are involved in interactions with the parasite: hemocytes and fat body cells, which provide humoral and cellular components of the innate immune response, midgut and salivary glands representing the epithelial barriers traversed by the parasite during its lifecycle in the mosquito.</p> <p>Findings</p> <p>We have analyzed the upstream regulatory sequence of the <it>An. gambiae </it>salivary gland-specific <it>apyrase </it>(<it>AgApy</it>) gene in transgenic <it>An. gambiae </it>using a <it>piggyBac </it>transposable element vector marked by a <it>3xP3 </it>promoter:<it>DsRed </it>gene fusion. Efficient germ-line transformation in <it>An. gambiae </it>mosquitoes was obtained and several integration events in at least three different G<sub>0 </sub>families were detected. <it>LacZ </it>reporter gene expression was analyzed in three transgenic lines/groups, and in only one group was tissue-specific expression restricted to salivary glands.</p> <p>Conclusion</p> <p>Our data describe an efficient genetic transformation of <it>An. gambiae </it>embryos. However, expression from the selected region of the <it>AgApy </it>promoter is weak and position effects may mask tissue- and stage- specific activity in transgenic mosquitoes.</p

    The personal and interpersonal components of perfectionism: the Italian validation of “Multidimensional Inventory of Perfectionism in Sport”

    The present research focused on the general theme of perfectionism in the sport domain, and it provided the first empirical validation of the original 72-item “Multidimensional Inventory of Perfectionism in Sport” (MIPS) among Italian athletes. The study, specifically, also focused on the relations linking personal and interpersonal components of perfectionism to athletes’ competitive anxiety. The research overall relied on data from 644 Italian sport science students and professional athletes and included both cross-sectional and longitudinal designs. Data analyses primarily focused on structural equation modeling, and the findings overall supported the psychometric and construct validity of the Italian version of the MIPS, also highlighting the key role of the personal components of perfectionism

    A Model for QCD at High Density and Large Quark Mass

    We study the high density region of QCD within an effective model obtained in the frame of the hopping parameter expansion and choosing Polyakov type of loops as the main dynamical variables representing the fermionic matter. To get a first idea of the phase structure, the model is analyzed in strong coupling expansion and using a mean field approximation. In numerical simulations, the model still shows the so-called sign problem, a difficulty peculiar to non-zero chemical potential, but it permits the development of algorithms which ensure a good overlap of the Monte Carlo ensemble with the true one. We review the main features of the model and present calculations concerning the dependence of various observables on the chemical potential and on the temperature, in particular of the charge density and the diquark susceptibility, which may be used to characterize the various phases expected at high baryonic density. We obtain in this way information about the phase structure of the model and the corresponding phase transitions and cross over regions, which can be considered as hints for the behaviour of non-zero density QCD.Comment: 21 pages, 29 figure

    Novel cDNAs encoding salivary proteins from the malaria vector Anopheles gambiae

    AbstractSeveral genes encoding salivary components of the mosquito Anopheles gambiae were identified using a selective trapping approach. Among these, five corresponded to genes expressed specifically in female glands and their role may possibly be linked to blood-feeding. Our collection included a fourth member of the D7 protein family and two polypeptides that showed weak similarity to anti-coagulants from distantly related species. Moreover, we identified two additional members of a novel group of proteins that we named glandins. The isolation of tissue-specific genes represents a first step toward a deeper molecular analysis of mosquito salivary secretions

    CORAL: the prediction of biodegradation of organic compounds with optimal SMILES-based descriptors

    Abstract CORAL software (http:/www.insilico.eu/coral) has been used to build up quantitative structure-biodegradation relationships (QSPR). The normalized degradation percentage has been used as the measure of biodegradation (for diverse organic compounds, n=445). Six random splits into sub-training, calibration, and test sets were examined. For each split the QSPR one-variable linear regression model based on the SMILES-based optimal descriptors has been built up. The average values of numbers of compounds and the correlation coefficients (r2) between experimental and calculated biodegradability values of these six models for the test sets are n=88.2±11.7 and r2=0.728±0.05. These six models were further tested against a set of chemicals (n=285) for which only categorical values (biodegradable or not) were available. Thus we also evaluated the use of the model as a classifier. The average values of the sensitivity, specificity, and accuracy were 0.811±0.019, 0.795±0.024, and 0.803±0.008, respectively

    Comparative study of two methods for rare earth elements analysis in human urine samples using inductively coupled plasma-mass spectrometry

    The application of rare earth elements (REEs) in several areas, including hightech technology, agriculture, medicine, and fuels, has made them an essential component of our everyday life. This extensive use of REEs in several technologies is expected to potentially impact human health. Even if several studies investigated the levels of REEs in human matrices, until now no standard method has been established for analyzing these elements in human matrices. The sample analysis should be of high quality, and the methods should be validated properly to ensure the quality of the procedure and traceability of the analytical data. In this research, we compared the validation and effectiveness of two different methods of sample preparation for human urine samples: a simple dilution of the sample (DIL) was compared with microwave assisted-acid decomposition (MIN) for tracing REE levels in human urine samples. The analysis was carried out by inductively coupled plasma mass spectrometry (ICP-MS). The working conditions have been set in high-sensitivity mode. Accuracy of the proposed method was evaluated by spiking the sample matrix with known concentrations of analyte standards. Both methods showed adequate precision of repeatability and intra-laboratory reproducibility, with the DIL method showing better precision of both repeatability and reproducibility than the MIN method. The CVr% values of repeatability range from 1.5 to 12% for the DIL and from 8.4 to 16% for the MIN method. The CVr% values of reproducibility range from 6.2–23% for the DIL and from 8.6 to 24% for the MIN method. REE recoveries for both methods were very close to 100%. Both methods proved to be effective for the determination of REE levels in human urine matrices

    Clinical, Pathological and Prognostic Features of Rare BRAF Mutations in Metastatic Colorectal Cancer (mCRC): A Bi-Institutional Retrospective Analysis (REBUS Study)

    Simple SummarySomatic BRAF mutations occur in approximately 10% of metastatic colorectal cancers (mCRCs) and, according to the involved codon, are classified as V600E and in non-V600, accounting for 80% and 20%, respectively. Being the most frequent mutation, the BRAF V600E mutation has been extensively investigated and up to now its clinical, pathological and molecular phenotype and its prognostic impact have been clearly described. On the contrary, evidence concerning BRAF non-V600 is weaker. We retrospectively evaluated 537 mCRC patients treated at two Italian Institutions. This study corroborates and strengthens available evidence concerning phenotype and prognostic performance of BRAF non-V600 compared to BRAF V600E and BRAF wild-type mCRCs. This deeper insight on rare BRAF non-V600 mutated mCRC is a primary issue in the precision oncology era, since the wider application of NGS is expected to increase the identification of those aberrations.Recently, retrospective analysis began to shed light on metastatic colorectal cancers (mCRCs) harboring rare BRAF non-V600 mutations, documenting a distinct phenotype and a favorable prognosis. This study aimed to confirm features and prognosis of rare BRAF non-V600 mCRCs compared to BRAF V600E and BRAF wild-type mCRCs treated at two Italian Institutions. Overall, 537 cases were retrospectively evaluated: 221 RAS/BRAF wild-type, 261 RAS mutated, 46 BRAF V600E and 9 BRAF non-V600. Compared to BRAF V600E mCRC, BRAF non-V600 mCRC were more frequently left-sided, had a lower tumor burden and displayed a lower grade and an MMR proficient/MSS status. In addition, non-V600 mCRC patients underwent more frequently to resection of metastases with radical intent. Median overall survival (mOS) was significantly longer in the non-V600 compared to the V600E group. At multivariate analysis, only age &lt; 65 years and ECOG PS 0 were identified as independent predictors of better OS. BRAF V600E mCRCs showed a statistically significant worse mOS when compared to BRAF wild-type mCRCs, whereas no significant difference was observed between BRAF non-V600 and BRAF wild-type mCRCs. Our study corroborates available evidence concerning incidence, clinicopathologic characteristics and prognosis of BRAF-mutated mCRCs
