    Pregnancy weight gain limitation by a supervised nutritional program influences placental NF-κB/IKK complex expression and oxidative stress

    Objective: Nuclear factor kappa B (NF-κB) pathway and oxidative stress participate in endothelial dysfunction, which is one of the causes of preeclampsia. Among the human antioxidant mechanisms there are the enzymes catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD). Our aim was to measure NF-κB, its inhibitor (IKK) and oxidative stress in placenta and umbilical cord of pregnant women submitted to a supervised nutritional program. Methods: Two groups were conformed: A) 14 pregnant women with individualized nutritional counseling and B) 12 pregnant women without nutritional guidance. NF-κB and IKK were assessed by real time PCR (RT-PCR). Enzymatic activity of CAT, GPx, lipoperoxidation (LPO) and SOD were also evaluated. Results: Pregnant women that followed-up a supervised nutritional program had lower level of systolic (p=0.03) and diastolic pressure (p=0.043) although they were heavier than a control group (p=0.048). Among all women, the Spearman correlation was positive between weight gain and placental NF-κB expression (1, p≤0.01). In placenta, women with nutritional advice had lower enzymatic activity of GPx (p≤0.038) and showed a tendency of IKK to be higher than in women without a nutritional supervised program. Conclusion: A supervised nutritional program in pregnancy offers a proven option to control weight gain, hypertension, NF-κB/IKK complex expression and oxidative stress reactions in placenta

    Molecular Epidemiology Surveillance of SARS-CoV-2: Mutations and Genetic Diversity One Year after Emerging

    In December 2019, the first cases of the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) were identified in the city of Wuhan, China. Since then, it has spread worldwide with new mutations being reported. The aim of the present study was to monitor the changes in genetic diversity and track non-synonymous substitutions (dN) that could be implicated in the fitness of SARS-CoV-2 and its spread in different regions between December 2019 and November 2020. We analyzed 2213 complete genomes from six geographical regions worldwide, which were downloaded from GenBank and GISAID databases. Although SARS-CoV-2 presented low genetic diversity, there has been an increase over time, with the presence of several hotspot mutations throughout its genome. We identified seven frequent mutations that resulted in dN substitutions. Two of them, C14408T>P323L and A23403G>D614G, located in the nsp12 and Spike protein, respectively, emerged early in the pandemic and showed a considerable increase in frequency over time. Two other mutations, A1163T>I120F in nsp2 and G22992A>S477N in the Spike protein, emerged recently and have spread in Oceania and Europe. There were associations of P323L, D614G, R203K and G204R substitutions with disease severity. Continuous molecular surveillance of SARS-CoV-2 will be necessary to detect and describe the transmission dynamics of new variants of the virus with clinical relevance. This information is important to improve programs to control the virus

    Spatiotemporal Changes in Plasmodium vivax msp142 Haplotypes in Southern Mexico: From the Control to the Pre-Elimination Phase

    For 20 years, Plasmodium vivax has been the only prevalent malaria species in Mexico, and cases have declined significantly and continuously. Spatiotemporal genetic studies can be helpful for understanding parasite dynamics and developing strategies to weaken malaria transmission, thus facilitating the elimination of the parasite. The aim of the current contribution was to analyze P. vivax-infected blood samples from patients in southern Mexico during the control (1993–2007) and pre-elimination phases (2008–2011). Nucleotide and haplotype changes in the pvmsp142 fragment were evaluated over time. The majority of multiple genotype infections occurred in the 1990s, when the 198 single nucleotide sequences exhibited 57 segregating sites, 64 mutations, and 17 haplotypes. Nucleotide and genetic diversity parameters showed subtle fluctuations from across time, in contrast to the reduced haplotype diversity and the increase in the R2 index and Tajima’s D value from 2008 to 2011. The haplotype network consisted of four haplogroups, the geographical distribution of which varied slightly over time. Haplogroup-specific B-cell epitopes were predicted. Since only high-frequency and divergent haplotypes persisted, there was a contraction of the parasite population. Given that 84% of haplotypes were exclusive to Mesoamerica, P. vivax flow is likely circumscribed to this region, representing important information for parasite surveillance

    Yeasts are microscopic organisms that are distributed in the biomes of the whole Earth, so some yeasts species exhibit diverse metabolic adaptations that allow them to proliferate in extreme environments. Yeasts that inhabit the Antarctica represent a relatively unexplored group of cold-adapted fungi. This review describes some of the metabolic adaptations that allow yeasts to inhabit extreme environments such as those in the Antarctica. This review also addresses the relevant considerations to know whether a yeast is extremophilic, as well as the criteria used to classify by its growth and temperature. The role of carotenoid and lipid biosynthesis pathways to mitigate reactive oxygen species generated by oxidative stress in pigmented and oleaginous yeasts including the Rhodotorula genus is described. Further, this review also considers the importance of basic research in oleaginous yeast from Antarctica and the development of biotechnological applications.Las levaduras son organismos microscópicos que están distribuidos en toda la Tierra, de modo que algunas han adaptado su metabolismo para proliferar en ambientes extremos. Las levaduras que habitan en la Antártica son un grupo de microorganismos adaptados al frío que han sido poco estudiadas. En esta revisión se describen algunas de las adaptaciones metabólicas que les permiten habitar en ambientes extremos, por ejemplo, el de la Antártica. También se abordan las consideraciones relevantes para saber si una levadura es extremófila, así como los criterios utilizados para clasificar a las levaduras por crecimiento y temperatura. Además, se explica el papel de las vías de biosíntesis de carotenoides y lípidos que están involucradas en contrarrestar a las especies reactivas de oxígeno generadas por estrés oxidante en levaduras pigmentadas y oleaginosas del género Rhodotorula. La revisión también considera aspectos de investigación básica y la importancia de las levaduras oleaginosas de la Antártica para el desarrollo de algunas aplicaciones biotecnológicas

    Clinical Characteristics of Coronavirus Disease (COVID-19) in Mexican Children and Adolescents

    Background: We analyzed the demographic, clinical, and diagnostic data of children and adolescents in Mexico, from the first case of coronavirus disease (COVID-19) to 28 February 2022. Methods: Using the open databases of the Ministry of Health and a tertiary pediatric hospital, we obtained demographic and clinical data from the beginning of the COVID-19 pandemic until 28 February 2022. In addition, quantitative reverse-transcription polymerase chain reaction outputs were used to determine the viral load, and structural protein-based serology was performed to evaluate IgG antibody levels. Results: Of the total 437,832 children and adolescents with COVID-19, 1187 died. Of these patients, 1349 were admitted to the Hospital Infantil de Mexico Federico Gómez, and 11 died. Obesity, asthma, and immunosuppression were the main comorbidities, and fever, cough, and headache were the main symptoms. In this population, many patients have a low viral load and IgG antibody levels. Conclusion: During the first 2 years of the COVID-19 pandemic in Mexico, children and adolescents had low incidence and mortality. They are a heterogeneous population, but many patients had comorbidities such as obesity, asthma, and immunosuppression; symptoms such as fever, cough, and headache; and low viral load and IgG antibodies
