Sperm physiology and quality in two marine teleosts: Anguilla anguilla & Takifugu niphobles
The conservation status of the species studied in this thesis
(European eel and pufferfish) is currently frail, thus the main goal of
this research was to develop, improve and apply several techniques
and protocols with the aim of increasing the knowledge about their
sperm biology, improving their reproductive performance and even
helping future breeding in captivity.
The reproductive performance of the males is often assessed through
the sperm motion parameters analysed by the CASA system, so first
we focused on how to standardize this technique in terms of
procedural and biological settings. In this respect, we laid the
foundations for applying a standard method to assess sperm quality
in fish, making it possible for sperm studies to be compared both
intra- and inter-laboratories using the proper CASA settings.
Secondly, with the aim of improving the reproductive performance of
European eel males, 3 thermal regimes (two of them variable: T10
and T15; and one of them constant: T20) and 3 hormonal treatments
(hCG, hCGrec and PSMG) were assessed based on different sperm
quality parameters. In the case of the thermal regimes, our results
demonstrated that the onset and progression of spermiation are
strongly influenced by water temperature, with treatment T20
showing the best results in all the sperm quality parameters. In the
case of hormonal treatments, hCGrec produced the best results in all
the sperm quality parameters, becoming an economically
profitable/viable treatment and an effective alternative to the
standard hCG treatment often used to induce spermiation in eel
species.
A preliminary physiological study regarding the changes to the main
ions involved in the fish sperm activation process was carried out.
Our results showed that intracellular concentrations of Ca2+
and K+
increased upon eel sperm activation, while pH gradually decreased over time, thus it is likely that all of them play an important role in
the initiation of sperm motility in the European eel, as with other
marine and freshwater teleosts.
In the second part of this thesis, which focuses on the pufferfish, an
in-depth study into the sperm of this species was carried out for
future application in aquacultural matters. A short-term storage
method for pufferfish sperm was developed, enabling us to preserve
the sperm quality parameters for a relatively long time period (7
days) compared to fresh sperm samples. Moreover, the effects of
both the osmolality and the ion composition of the activation media
on the sperm motion parameters were evaluated, concluding that
both factors play an essential role in the initiation of sperm motility
of pufferfish sperm and probably, in marine fish sperm.
Finally, in vitro fertilization trials were developed to assertain how
the quantity and quality of male gametes affects the fertilization and
hatching rates. We demonstrated that sperm/egg ratio and sperm
quality are strongly related factors, suggesting that both should be
taken into account as unique interrelated elements. In addition,
coefficients of correlation among all the spermatozoa motion
parameters provided by a CASA system and fertilization and hatching
rates were estimated for the first time in a marine species.
Spermatozoa velocities showed the highest coefficients (¿0.80),
suggesting that the kinetics of the spermatozoa are a key factor in
the fertilization process.Gallego Albiach, V. (2013). Sperm physiology and quality in two marine teleosts: Anguilla anguilla & Takifugu niphobles [Tesis doctoral no publicada].
Microbial fermentation in an in vitro model of colonic fermentation with different foods
[EN] The interaction between diet and gut microbiota, and ultimately their link to health, has become
the focus of huge research showing that diet and lifestyle have a strong influence on the gut
microbiota. That is why there is a growing interest in resolving questions about the relation
between the gut microbiome and host metabolism.
The human intestine is densely populated by trillions of microbial symbionts. The symbiotic gut
microbiota helps nutrient absorption through the fermentation of dietary fibre, provides
protection from invading pathogens and helps to develop and regulate the immune system.
However, the mechanisms underlying interactions between diet, gut microbiome and host
metabolism are still poorly understood. Here, we discuss how meta-omics datasets can be
obtained through an in vitro model model of colonic fermentation in order to study how the food
that is consumed can shape the diversity and composition of the gut microbiota. In order to
analyze microbial community structure, tools and approaches such as next generation
sequencing (NGS) of 16S rRNA amplicons and bioinformatic analysis are used. The study will
be performed with bread that will be digested and then submitted to an in vitro fermentation
process with fecal inocula from healthy Spanish adults from Granada.
In the end, the influence of bread on the structure of the gut microbiota from different types of
fecal samples will be evaluated, as well as the variability of the microbioal taxonomy,
composition and alpha and beta diversity when fecal samples are analyzed before and after
fermentation and before and after a freezing treatment at -80 Cº. Our results suggest that despite
a loss of bacterial groups that alters the microbial composition, freezing the samples before
fermentation is not a serious problem because the effect of fermentation on the samples remains
significant in terms of richness, diversity and abundance of the main bacterial groups. This will
be studied in line with the main objectives of a larger project (Stance4Health) of developing a
Nacher Albiach, P. (2019). Microbial fermentation in an in vitro model of colonic fermentation with different foods. http://hdl.handle.net/10251/125065TFG
Titelles i transgressió
ABSTRACT: The puppet theater has traditionally been a neglected form of popular literature in Europe, considered a minor form and left out of any cultural considerations. However, it is an art form that has often characterized by its ability to satire, mockery, transgression and contestation of political or religious authority. The recovery of puppet theater for adults and its presence in the audiovisual world are two examples of the present and the future of this theatrical form. Learning Diversity: A Collaborative Work Experience of Literary Education in a Spanish Teacher Training Classroom
The aim of this paper is to present an experience carried out in a group of 50 university students of the subject Literary Education in the Childhood Classroom in the 2014/2015 academic year. The activity is based on the need to educate on diversity and includes literature, drama and puppets as powerful tools for the awareness, reflection and dialogue about the topic of diversity. Based on research about children's literature focused on diversity, the work groups started a debate on the (in)visibility of most of these works. Once a text was chosen, students produced and dramatized a puppet play. The process included every aspect of production, from the initial conception to the final performance, focusing on an element of reflection and multiplicity of roles during the entire process. We believe the experience was very positive and beneficial for the literary training of the teacher. It provided a framework for the programming and development of activities about all kinds of diversity in the early childhood education classroom
On Puppets and Literary Education in Diverse Schools: A Review from Spain
Intercultural education and the attention to diversity have become two of the most important aspects of education in recent decades. There are many areas of diversity that can be addressed in the classroom, always based on tolerance and the acceptance of difference. However, intercultural education is destined to go one step further: from an inalienable foundation of respect and tolerance for everyone, we aim to develop diversity as a positive value that must be understood and accepted. There is no doubt that we are faced with one of the great challenges of education. Literature and theatre, on the other hand, is always a meeting point, first and foremost between the author and the recipient, often separated by temporal, spatial, linguistic, and cultural factors. In this sense, as Josep Ballester (2015) reminds us that the different sets of values and worldviews presented by the different human communities, as well as certain patterns of conduct and folk wisdom developed over generations, are stored and alive in language and literature. We will focus our educational approach precisely in this capacity of literature and drama -puppets specifically- to confront different human cultures and bring them to dialogue
