ttl mutants are impaired in cellulose biosynthesis under osmotic stress

As sessile organisms, plants require mechanisms to sense and respond to the challenging environment, that encompass both biotic and abiotic factors that results in differential development. In these conditions is essential to balance growth and stress responses. As cell walls shape plant growth, this differential growth response cause alterations to the plant cell wall and cellulose is a major component. Therefore, understanding the mechanisms that regulate cellulose biosynthesis is essential to develop strategies to improve plant production. Previous studies have shown that the GSK3 kinase BIN2 modulate cellulose biosynthesis through phosphorylating cellulose synthases and that the expression of cellulose synthases are regulated by Brassinosteroids. Our previous work reveals that the tetratricopeptide-repeat thioreoxin-like (TTL) TTL1, TTL3, and TTL4 genes, in addition to their reported role in abiotic stress tolerance, are positive regulators of BR signaling. We observe association of TTL3 with most core components in traducing BR signalling, such as LRR-RLK BRI1, BIN2 and the transcription factor BES1 that positively regulate cellulose biosynthesis. We show that ttl mutants are affected in cellulose biosynthesis, particularly in osmotic stress conditions. Furthermore, TTL3 associates with LRR-RLKs that have been shown to be important for cellulose biosynthesis such as FEI1 in the FEI1/FEI2/SOS5 pathway. We aim to investigate the mechanisms by which TTL proteins regulate cellulose biosynthesis using a combination of genetics, biochemical, and molecular and cell biology approaches. Joint water quantity/quality management analysis in a biofuel production area: Using an integrated economic-hydrologic model

    Get PDF
"Water management in the Pirapama River Basin in northeastern Brazil is affected by both water quantity and water quality constraints. The region is known for significant sugarcane-based ethanol production—which is key to the Brazilian economy and expected to grow dramatically under recent global changes in energy policy. Sugarcane production in the region goes hand in hand with controlled fertirrigation practices with potentially significant adverse impacts on the environment. To assess sustainable water allocation in the basin, an integrated hydrologic-economic basin model is adapted to study both water quantity and water quality aspects. The model results show that incorporating water quality aspects into water allocation decisions leads to a substantial reduction in application of vinasse to sugarcane fields. To enforce water quality restrictions, the shadow price for maintaining water in the reservoir could be used as a pollution tax for fertirrigated areas, which are currently not subject to pollution charges." from authors' abstractWater quality, River basin model, Integrated economic-hydrologic modeling, Nonlinear optimization, Biofuels, Water resources, Environmental impacts,

Automatic Recognition of Prosodic Patterns in Semantic Verbal Fluency Tests - an Animal Naming Task for Edutainment Applications

    Get PDF
This paper automatically detects prosodic patterns in the domain of semantic fluency tests. Verbal fluency tests aim at evaluating the spontaneous production of words under constrained conditions. Mostly used for assessing cognitive impairment, they can be used in a plethora of domains, as edutainment applications or games with educational purposes. This work discriminates between list effects, disfluencies, and other linguistic events in an animal naming task. Recordings from 42 Portuguese speakers were automatically recognized and AuToBI was applied in order to detect prosodic patterns, using both European Portuguese and English models. Both models allowed to differentiate list effects from the other events, mostly represented by the tunes: L* H/L(-%) (English models) or L*+H H/L(-%) (Portuguese models). However, English models proved to be more suitable because they rely in substantial more training material.info:eu-repo/semantics/publishedVersio

4SpecID: reference DNA libraries auditing and annotation system for forensic applications

    Get PDF
Forensic genetics is a fast-growing field that frequently requires DNA-based taxonomy, namely, when evidence are parts of specimens, often highly processed in food, potions, or ointments. Reference DNA-sequences libraries, such as BOLD or GenBank, are imperative tools for taxonomic assignment, particularly when morphology is inadequate for classification. The auditing and curation of these datasets require reliable mechanisms, preferably with automated data preprocessing. Software tools were developed to grade these datasets considering as primary criterion the number of records, which is not compliant with forensic standards, where the priority is validation from independent sources. Moreover, 4SpecID is an efficient and freely available software tool developed to audit and annotate reference libraries, specifically designed for forensic applications. Its intuitive user-friendly interface virtually accesses any database and includes specific data mining functions tuned for the widespread BOLD repositories. The built tool was evaluated in laptop MacBook and a dual-Xeon server with a large BOLD dataset (Culicidae, 36,115 records), and the best execution time to grade the dataset on the laptop was 0.28 s. Datasets of Bovidae and Felidae families were used to evaluate the quality of the tool and the relevance of independent sources validation.Portuguese funds are supporting this work through FCT-Fundação para a Ciência e a Tecnologia/Ministério da Ciência, Tecnologia e Inovação in the framework of the project “Institute for Research and Innovation in Health Sciences” (POCI-01-0145-FEDER-007274) and “Center of Mathematics of the University of Porto” (UID/MAT/00144/2019).
