Inhaled Nitric Oxide Reverses Vascular and Respiratory Effects of ET-1 and PAF in Pigs
In anaesthetized paralysed, mechanically ventilated pigs, the
vascular and respiratory effects of 80 ppm nitric oxide (NO) inhaled
for 6 min were evaluated. To evoke different levels of smooth
muscle contraction ET-1 or PAF, mediators involved in pulmonary
disorders, were used. In control conditions, inhaled NO caused
selective pulmonary vasodilatation without affecting respiratory
resistances. This pulmonary vascular activity influenced the
distensibility of the respiratory system and decreased inspiratory
work. ET-1 administration significantly increased pulmonary arterial
pressure and modestly changed mechanical properties of the
respiratory system, while PAF caused potent vasoconstriction and
bronchoconstriction associated with a marked change in
volume-pressure relationship. In both cases, the changes in vascular
and mechanical properties of the respiratory system increased
inspiratory work. The vascular and respiratory activities of inhaled
NO were correlated with preconstriction levels. The data show that
the combination of vascular and respiratory effects improves
pulmonary function, suggesting that inhalation of NO is a possible
therapeutic approach for obstructive and inflammatory pulmonary
diseases
Factors influencing interactions in zoos: animal-keeper relationship, animal-public interactions and solitary animals groups
Interactions that animals experience can have a significant influence on their health and welfare. These interactions can occur between animals themselves, but also between animals and keepers, and animals and the public. Human and non-human animals come into contact with each other in a variety of settings, and wherever there is contact there is the opportunity for interaction to take place. Interaction with companion animals are well known, but humanâanimal interaction (HAR) (Hosey, 2008) also occurs in the context of farms (Hemsworth and Gonyou, 1997; Hemsworth, 2003), laboratories (Chang and Hart, 2002), zoos (Kreger and Mench, 1995) and even the wild (e.g. Cassini, 2001). This project proposes a permanent monitoring scheme to record animal-human interactions and animal-animal interactions in zoos. This will be accompanied by a survey of animal personality for welfare, husbandry, breeding programs and reintroduction purposes. The pilot project is currently based on direct monitoring of animal behaviour, use of time lapse cameras and animal personality questionnaires completed by experienced keepers. The goal of this project is to create a network between zoos to explore the aforementioned interactions to produce husbandry protocols and explore personality and behavioural traits in multiple species. We present provisional data regarding polar bear (Fasano Zoosafari, Italy), Sumatran tigers, Amur tigers and Asiatic lion (ZSL London and Whipsnade zoo) interactions with humans and conspecifics. This data is collected across a broad range of environmental conditions and outlines the monitoring protocols developed to collect this data. The first year data show the great adaptability of these species to ex situ environments, low or absent negative impact of visitorsâ presence and the relevance of individual personality in these interactions
Mechanical ventilation and volutrauma: study in vivo of a healthy pig model
Mechanical ventilation is essential in intensive care units. However, it may itself induce lung injury. Current studies are based on rodents, using exceptionally large tidal volumes for very short periods, often after a "priming" pulmonary insult. Our study deepens a clinically relevant large animal model, closely resembling human physiology and the ventilator setting used in clinic settings. Our aim was to evaluate the pathophysiological mechanisms involved in alveolo/capillary barrier damage due to mechanical stress in healthy subjects. We randomly divided 18 pigs (sedated with medetomidine/tiletamine-zolazepam and anesthetised with thiopental sodium) into three groups (n=6): two were mechanically ventilated (tidal volume of 8 or 20 ml/kg), the third breathed spontaneously for 4 hours, then animals were sacrifi ced (thiopental overdose). We analyzed every 30' hemogasanalysis and the main circulatory and respiratory parameters. Matrix gelatinase expression was evaluated on bronchoalveolar lavage fl uid after surgery and before euthanasia. On autoptic samples we performed zymographic analysis of lung, kidney and liver tissues and histological examination of lung. Results evidenced that high V T evoked profound alterations of lung mechanics and structure, although low V T strategy was not devoid of side effects, too. Unexpectedly, also animals that were spontaneously breathing showed a worsening of the respiratory functions
Domestic dogs (Canis familiaris) grieve over the loss of a conspecific.
Behavioural reactions towards a dead conspecific have been observed rarely in wild canids and there is no documented scientific evidence of grief in pet dogs. A quantitative analysis of grief-related responses in both dogs and owners was conducted, using the validated online Mourning Dog Questionnaire. The survey was completed by 426 Italian adults who had owned at least two dogs, one of whom died while the other was still alive. This research aims to explore whether, how and what a dog may experience over the loss of a companion dog. Multiple logistic regression indicates that both a friendly or parental relationship between two dogs but also the fact that dogs used to share food and the owner's grief and anger are principal predictors of negative behavioural changes. According to dog owners' answers, the surviving dog after the death of the companion dog changed both in terms of activities ("playing", "sleeping", and "eating") and emotions (fearfulness), which occurred as a function of the quality of the relationship between the two animals. By contrast, the time the two dogs had spent together had no effect on the behaviours of surviving dog. Owner perceptions about their dog's reactions and emotions were not related to the memory or suffering of the event that tended to diminish over time. These findings indicate that a dog may show grief-related behavioural and emotional patterns when a close conspecific dies, with aspects of the latter possibly related to the owner's emotional status
Comparative Personality Traits Assessment of Three Species of Communally Housed Captive Penguins.
Understanding animal personalities has notable implications in the ecology and evolution of animal behavior, but personality studies can also be useful in optimizing animal management, with the aim of improving health and well-being, and optimizing reproductive success, a fundamental factor in the species threatened with extinction. Modern zoos are increasingly being structured with enclosures that host different species, which permanently share spaces. This condition has undeniable positive aspects, but, in some species, it could determine the appearance of collective or synchronized behaviors. The aim of this study was to verify, in a colony of three species of communally housed penguins (Pygoscelis papua, Aptenodytes patagonicus and Eudyptes moseleyi), through a trait-rating assessment, if interspecific group life impacts on the expression of personality traits, and if it is possible to highlight specie-specific expression of personality traits, despite the influence of forced cohabitation. For many of the personality traits we analyzed, we have observed that it was possible to detect an expression that differed, according to the species. From a practical point of view, these data could ameliorate the management of the animals, allowing to design animal life routines, according to the different behavioral characteristics of the cohabiting species

Familiarity and Interest in Working with Livestock Decreases the Odds of Having Positive Attitudes towards Non-Human Animals and Their Welfare among Veterinary Students in Italy
We investigated the attitudes of veterinary students towards animals and their welfare in Italy. Regression analyses revealed predictors that are significant in differentiating students’ scoring tendency based on their gender, familiarity, and intention to work with a specific animal species, type of diet, and membership in an animal rights association. Female students, who were mostly familiar with pets and aspired to work with species other than livestock, following an animal-free diet and being a member of an animal rights association, had a significantly greater odds of having a high Animal Attitude Scale score (AAS), i.e., very positive attitude towards animals, versus a less positive attitude. Conversely, the familiarity with livestock and preference for working with livestock significantly increased the odds of a low AAS. Overall, students considered all of the Brambell Report’s Five Freedoms important for animal welfare protection. However, students scored higher for companion animals than for livestock, particularly regarding the freedom to express normal behaviour and the absence of fear and distress. This study suggests that veterinary students place less importance on the psychological aspects of welfare for livestock, and there is a tendency for students who are mostly familiar, or aspire to work, with livestock to have a less positive attitude towards non-human animals and their welfare. These findings should be considered within the veterinary educational curriculum due to their potential impact on animal welfare
