    Concurrent environmental stressors and jellyfish stings impair caged European sea bass (Dicentrarchus labrax) physiological performances

    none8siThe increasing frequency of jellyfish outbreaks in coastal areas has led to multiple ecological and socio-economic issues, including mass mortalities of farmed fish. We investigated the sensitivity of the European sea bass (Dicentrarchus labrax), a widely cultured fish in the Mediterranean Sea, to the combined stressors of temperature, hypoxia and stings from the jellyfish Pelagia noctiluca, through measurement of oxygen consumption rates (MO2), critical oxygen levels (PO2crit), and histological analysis of tissue damage. Higher levels of MO2, PO2crit and gill damage in treated fish demonstrated that the synergy of environmental and biotic stressors dramatically impair farmed fish metabolic performances and increase their health vulnerability.     Population trends of the fan mussel Pinna nobilis from Portofino MPA (Ligurian Sea, Western Mediterranean Sea) before and after a mass mortality event and a catastrophic storm

    Two Pinna nobilis populations thriving inside the borders of the Portofino Marine Protected Area (MPA) (Ligurian Sea, western Mediterranean Sea) were monitored before (2012) and after (September 2018) a dire mass mortality event that, since September 2016, spread through the whole Mediterranean Sea. In Portofino MPA, recorded mortality rates reached values of 91.29% and 43.94% in the two populations. The presence of a Haplosporidium protozoan parasite, considered to be the main cause of the mortality episodes, was confirmed from histological evidence: sporocysts and plasmodia were observed in all the tubules of the digestive glands of the collected specimens. Moreover, a catastrophic storm hit the Ligurian coasts at the end of October 2018, causing considerable damages both below and above the surface; a new survey conducted in November 2018 showed the complete annihilation of the two studied populations, as a probable combination of the continued parasite infections and the mechanical impacts caused by the storm. Finally, in June 2020 the sites were monitored again looking for traces of recovery, but no new specimens were recorded, indicating that P. nobilis became virtually absent from the MPA

    Seamounts as hot-spots of large pelagic aggregations

    The distribution of four top predators in the Tyrrhenian Sea, a sub-basin of the Mediterranean Sea, was investigated by means of random forest regression considering depth, distance from the coast, seafloor slope, and distance from seamounts as habitat descriptors on a 2x2–nautical mile regular grid. RF results are processed to estimate variable importance and model performance. Random forest architecture reached optimal sensitivity and specificity, thus providing a consistent support tool for identifying suitable habitats. The considered species are characterized as having patched suitable habitats with a number of hot-spot areas where the different species' habitats overlap. These hot-spot areas' locations correspond to those of specific seamounts identifying the attraction effect of these topographic structures. The mean features typifying the most attractive seamounts are investigated and found to be shallow peak and base depths but wide base area and high relative elevation

    Effects of Dietary Plant Protein Replacement with Insect and Poultry By-Product Meals on the Liver Health and Serum Metabolites of Sea Bream (Sparus aurata) and Sea Bass (Dicentrarchus labrax)

    The liver health of Gilthead sea bream and European sea bass, fed with fish meal-free diets, including various proportions of plant proteins, as well as insect and poultry by-product meals, was investigated through biochemical and histological analyses using a new liver index (LI) formula. Four isoproteic (45% Dry Matter, DM) and isolipidic (20% DM) diets were compared, including a plant-based control diet (CV) and three other test diets, in which 40% of a plant protein-rich ingredient mixture was replaced with meals from Hermetia illucens (H40) or poultry by-product (P40) alone, or in combination (H10P30). The trials lasted 12 and 18 weeks for sea bream and sea bass, respectively. The results obtained thus far highlighted species-specific differences in the physiological response to dietary changes. In sea bream, the biochemical and histological responses suggest favorable physiological and liver health statuses, with higher serum cholesterol (CHO) and triglyceride (TAG) levels, as well as moderate hepatocyte lipid accumulation, with the H10P30 diet compared to the CV (p < 0.05). In sea bass, all diets resulted in elevated serum TAG levels and lipid accumulation in the liver, particularly in fish fed the P40 one (p < 0.05), which resulted in the highest LI, coupled with a higher frequency of severe lipid accumulation, hypertrophy, cord loss, peripheral nuclei displacement, and pyknosis. In conclusion, sea bream adapted well to the test diets, whereas sea bass exhibited altered hepatic lipid metabolism leading to incipient liver steatosis, likely due to the high lipid contents of the diets, including the insect and poultry meals. The LI formula developed in this study proved to be a reliable tool for assessing the effects of dietary changes on the liver health of sea bream and sea bass, consistent with biochemical and histological findings

    Growth and Welfare of Rainbow Trout (Oncorhynchus mykiss) in Response to Graded Levels of Insect and Poultry By-Product Meals in Fishmeal-Free Diets

    This study compared the nutrient-energy retention, digestive function, growth performance, and welfare of rainbow trout (ibw 54 g) fed isoproteic (42%), isolipidic (24%), fishmeal-free diets (CV) over 13 weeks. The diets consisted of plant-protein replacement with graded levels (10, 30, 60%) of protein from poultry by-product (PBM) and black soldier fly H. illucens pupae (BSFM) meals, either singly or in combination. A fishmeal-based diet was also tested (CF). Nitrogen retention improved with moderate or high levels of dietary PBM and BSFM relative to CV (p &lt; 0.05). Gut brush border enzyme activity was poorly affected by the diets. Gastric chitinase was up-regulated after high BSFM feeding (p &lt; 0.05). The gut peptide and amino acid transport genes were differently regulated by protein source and level. Serum cortisol was unaffected, and the changes in metabolites stayed within the physiological range. High PBM and high BSFM lowered the leukocyte respiratory burst activity and increased the lysozyme activity compared to CV (p &lt; 0.05). The BSFM and PBM both significantly changed the relative percentage of lymphocytes and monocytes (p &lt; 0.05). In conclusion, moderate to high PBM and BSFM inclusions in fishmeal-free diets, either singly or in combination, improved gut function and nutrient retention, resulting in better growth performance and the good welfare of the rainbow trout

    The Seabed Makes the Dolphins: Physiographic Features Shape the Size and Structure of the Bottlenose Dolphin Geographical Units

    The common bottlenose dolphin (Tursiops truncatus) is a cosmopolitan delphinid, regularly present in the Mediterranean Sea. According to previous studies, this dolphin tends to form resident geographical units scattered on the continental shelf. We investigated how the physiographic characteristics of the area of residence, with special reference to the size and shape of the continental shelf, affect the home range and the group size of the local units. We analysed and compared data collected between 2004-2016 by 15 research groups operating in different study areas of the Mediterranean Sea: the Alboran Sea, in the South-Western Mediterranean, the Gulf of Lion and the Pelagos Sanctuary for the marine mammals, in the North-Western Mediterranean, and the Gulf of Ambracia, in the North-Central Mediterranean Sea. We have found that in areas characterised by a wide continental platform, dolphins have wider home ranges and aggregate into larger groups. In areas characterized by a narrow continental platform, dolphins show much smaller home ranges and aggregate into smaller groups. The results obtained from this collective research effort highlight the importance of data sharing to improve our scientific knowledge in the field of cetaceans and beyond

    Adult Xenopus laevis were exposed in vivo to ethinylestradiol, tamoxifen, methyldihydrotestosterone and flutamide as (anti)estrogenic and (anti)androgenic compounds, respectively, for four weeks at a concentration of 10(-8) M and to Lambro river water, a polluted river from Italy. Effects of the treatments were analysed by mRNA expression of retinol-binding protein (RBP), transferrin (TF), transthyretin (TTR) and vitellogenin (VTG) in the liver of male and female X. laevis, to analyse the potential of these genes to detect endocrine disrupting compounds (EDC) with different modes of action. In addition, plasma VTG and sex steroid levels, estradiol-17beta (E(2)) and testosterone (T), were analysed. Sex steroids were depressed by ethinylestradiol in both sexes whereas tamoxifen increased E(2) in females. The induction of VTG protein plasma levels was more pronounced at the protein level compared to hepatic VTG mRNA expression in response to estrogenic treatment but VTG mRNA expression detected both, estrogenic and antiestrogenic EDC. The mRNA expression of TF was decreased by estrogenic and increased by antiestrogenic treatment while TTR mRNA expression was down-regulated and RBP mRNA up-regulated by estrogenic exposure. The other treatments did not affect the mRNA expression of the examined genes

    Bottlenose Dolphins and Seabirds Distribution Analysis for the Identification of a Marine Biodiversity Hotspot in Agrigento Waters

    The aim of this study is to evaluate the presence of biodiversity hotspots in Agrigento waters (Mediterranean Sea) to define the conservation area for bottlenose dolphins (Tursiops truncatus) and seabirds (Calonectris diomedea, Puffinus yelkouan, and Hydrobates pelagicus), according to European directives. With this purpose, the maximum entropy algorithm (MaxEnt) was applied to the sighting points of the focal species. They co-occur in the study area and have been documented to forage behind trawlers. In this study, a fishing rate was designed and used as an explanatory variable of the species distribution, together with physiographic variables. Data were collected during 68 surveys in the waters off Agrigento province. MaxEnt models showed a strong predictive power, with distance from the coast being the greatest predicting variable, followed by slope, depth, and fishing rate. For all the species considered, the probability of presence increased as the fishing rate grew. Cartographic analysis revealed one area shared by the species, which occupies 529 km2, from the shoreline to 100 m depth. This study increases knowledge on the distribution and habitat preferences of the target species in the Sicilian waters. Evaluating the influence of fisheries is a promising method that needs further testing to apply effective management measures

    Seriola dumerilii, (Carangidae, Perciformes) is a gonochoric teleost occurring in the Mediterranean Sea. Despite the growing interest for this species, little is known on sex steroid pattern during the reproductive season. Aim of this study was to verify if 17-20ß-dihydroxy-progesterone (DHP) could be involved as maturation inducing substance (MIS) in S. dumerilii as in other species. 244 females of wild S. dumerilii were collected in South Mediterranean Sea (Pelagie Islands), during three spawning seasons and histologically staged. Blood samples were withdrawn by puncturing the gill artery and plasma aliquots were frozen at -20°C for steroid RIAs. DHP plasma values were measured in wild females at the end of vitellogenesis and during final maturation (FOM). DPH values were related both to 17ß-estradiol and testosterone values and to morphological features of oocytes. All specimens with ovaries characterised by a population of oocytes undergoing FOM, showed a significant increase (P<0.01) in DHP value. Furthermore, we analysed DHP plasma values in relation to three different stages occurring during FOM. Early FOM, including GV migration and lipid droplets coalescence, showed the first increase in DHP values (xxpg/ml±xx). Middle FOM corresponding to GVBD was characterised by the higher values of DHP (xxxpg/ml±xx). In late FOM (hydration), DHP started to decrease (xxxpg/ml±xxx). These results suggest that DHP could be considered as the MIS in S. dumerili

    Assessment of thyrrenian seamounts attractiveness on Stenella coeruleoalba.

    In the framework of the PROMETEOS project, 64 seamounts were identified in Tyrrhenian Sea and specific morphological characteristics have been calculated for each seamount. Random Forest regression technique has been applied to striped dolphin distribution aiming at assessing importance and effect of seamounts on this species. A total of 467 sightings have been considered deriving from OBISSEAMAP (Tyrrhenian aerial survey of Tethys Institute and ISPRA), and two dedicated campaigns (Tirreno 2013 and Tirreno 2014). Distance from seamounts, together with other morphological features such as depth and distance from the coast, resulted the most important variables affecting presence of striped dolphin. Moreover, results clearly showed that differences in the structures appeal are due to some specific seamounts characteristics (slope and minimum peak depth) allowing to rank mountains in terms of capacity to attract Stenella coeruleoalba and thus to possibly address management and conservation practice
