    Validation of a capillary zone electrophoresis method for the determination of ciprofloxacin, gatifloxacin, moxifloxacin and ofloxacin in pharmaceutical formulations

    An alternative capillary zone electrophoresis (CZE) method for the determination of ciprofloxacin (CPFLX), gatifloxacin (GTFLX), moxifloxacin (MFLX) and ofloxacin (OFLX) through a simple aqueous electrolyte system consisting of 25 mmol L-1 of TRIS/ hydrochloride and 15 mmol L-1 of sodium tetraborate buffer mixture (pH 8.87) using direct UV detection at 282 nm within 3 min was validated. The analytical parameters of validation evaluated were: linearity (r > 0.998), selectivity (comparison between slope of the calibration curve of external standard and calibration curve of standard addition), repeatability in area for sample (RSD%: < 3.94% for CPFLX, < 3.87% for GTFLX, 1.30% for MFLX and < 1.88% for OFLX), intermediate precision in area for sample (RSD%: < 3.59% for CPFLX, < 3.09% for GTFLX, 2.67% for MFLX and < 2.25% for OFLX), accuracy (mean of recovery range: 101.2% for CPFLX, 101.0% for GTFLX, 101.3% for MFLX and 99.9% for OFLX), limit of detection (mg L-1: 2.72 for CPFLX, 1.92 for GTFLX, 0.795 for MFLX and 1.05 for OFLX), limit of quantification (mg L-1: 9.06 for CPFLX, 6.40 for GTFLX, 2.65 for MFLX and 3.50 for OFLX) and robustness. Due to its simplicity, selectivity, precision, accuracy and rapidity, the methodology can be an interesting alternative for quality assurance in the pharmaceutical industry of these drugs

    Effects of consumption of galactooligosaccharides obtained through whey enzymatically modified on the faecal flora and nutritional parameters of hamsters

    The aim of this research was to evaluate the influence of wheyenzymatically modified rich in galactooligosaccharides in thenutritional characteristics and effects in the microflora of cecumcontents by the study with Golden Syrian hamsters (Mesocricetusauratus) for 28 days (controlled conditions). Three isoproteic dietswere prepared (20% w/w): C (casein), W (whey) and G (wheymodified). The groups studied differed positively from the C regardingfeed and protein efficiency ratio. The relationships (w/w) oforgan/body were found proportional in all diets. The counts ofprobiotics from the cecum contents the groups showed no difference.The pHs of studied groups were lower than C, this acidity can atimpairs the ability of pathogens to grow in the intestine. Resultssuggest that using whey enzymatically modified rich ingalactooligosaccharides could replace the standard diet withnutritional efficiency and possible inhibit the microorganismspathogenic without induce damage in health.Fil: Dos Santos Da Fonseca, Renata Aline.     Assessment of Physical-Chemical Characteristics of Water and Sediments from a Brazilian Tropical Estuary: Status and Environmental Implications

    The environmental quality of the Jacuípe River's estuary (very important in northeastern Brazil) was assessed during 2007 and 2008. In water, concentrations (mg L−1) of NO2− (<0.004 to 0.016), NO3− (0.01 to 0.33), soluble PO43− (<0.02 to 0.22), dissolved oxygen (3.9 to 9.6), total contents (mg L−1) of Cd (<0.001), Cu (<0.01), Pb (<0.01), and Zn (<0.1), pH (5.60 to 8.00), and electrical conductivity (0.12 to 48.60 mS cm−1) agreed with environmental standards. In sediments, clay and total organic matter (%, m/m) varied, respectively, from 8.8 to 12.0 and from 1.1 to 8.8, while infrared, thermogravimetric profile, electronic micrograph, as well as X-Ray analyses showed desirable adsorptive characteristics. However, maximum exchangeable levels (mg kg−1) of Cd (1.3), Cu (44.6), Pb (35.7), and Zn (43.7) and their respective maximum pseudototal concentrations (mg kg−1): 19.4, 95.1, 68.2, and 30.3 were below the recommended limits. In this sense, it was possible to demonstrate good environmental preservation even with the growing number of industries and touristic activities in the evaluated estuarine area

    Atividades biológicas de espécies amazônicas de Rubiaceae

    Rubiaceae is a plant family with great chemical and biological potential. In this work a phytochemical prospection was performed and it was assessed the cytotoxic, antioxidant and antifungal activities of dichloromethanic and methanolic extracts of the leaves of Duroia saccifera, Ferdinandusa goudotiana, F. hirsuta, F. paraensis, Ferdinandusa sp., Palicourea corymbifera and P. guianensis. The cytotoxicity evaluation was carried out using the test toxicity on Artemia salina. The antifungal activity was determined by agar diffusion method using fungi: Epidermophyton floccosum, Microsporum canis 32905, M. gipseum 29/00, Trichophyton mentagrophytes ATCC 9533/03, T. rubrum ATCC 28189, T. tonsurans 21/97, Cladosporium cladosporioides, C. sphaerospermum, Fusarium U. 662/06, Scytalidium U. 661/06, Candida albicans ATCC 3632 and C. albicans U. 5/99. The antioxidant activity was determined by testing the amount of free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the discoloration of the radical cation 6-sulfonic acid-2,2-azinobis-3-ethylbenzothiazoline (ABTS). For the toxicity test on A. salina greater toxicity in the methanol extract of F. goudotiana was observed, which proved to be toxic up to a concentration of 5 μg mL-1. The presence of antifungal principles was observed in the F. hirsuta and F. paraensis against four tested fungi. In the evaluation of the antioxidant activity, the methanol extracts showed greater activity than the dichloromethanic ones, being similar the obtained result through both methods (reduction of DDPH and ABTS decolorization). The results create possibilities for future researches related to the structure and activity of the components of each active extract. © 2015, Instituto de Biociencias. All rights reserved
