    Photoelectrochemical Behavior of n-Type GaAs(100) Electrodes Coated by a Single Layer of Graphene

    Methods for cell-selective analysis of proteome dynamics will facilitate studies of biological processes in multicellular organisms. Here we describe a mutant murine methionyl-tRNA synthetase (designated L274GMmMetRS) that charges the noncanonical amino acid azidonorleucine (Anl) to elongator tRNA^(Met) in hamster (CHO), monkey (COS7), and human (HeLa) cell lines. Proteins made in cells that express the synthetase can be labeled with Anl, tagged with dyes or affinity reagents, and enriched on affinity resin to facilitate identification by mass spectrometry. The method does not require expression of orthogonal tRNAs or depletion of canonical amino acids. Successful labeling of proteins with Anl in several mammalian cell lines demonstrates the utility of L274GMmMetRS as a tool for cell-selective analysis of mammalian protein synthesis

    The New Community Policing: Developing a Partnership-Based Theoretical Foundation

    This paper presents a Partnership Model of Community Policing based on Partnership concepts developed by Riane Eisler and undergirded by Cultural Transformation Theory as a guiding principle (1987, 2010, 2013). This model is more reflective of the daily lived experiences of community police officers. It is culturally relevant and based on the whole of the police officer’s relationship with the community within the context in which the interactions occur. This "New Community Policing" is an extension of Riane Eisler’s Cultural Transformation Theory and is an attempt to answer her call for a movement towards a partnership model of social organization. Ultimately, "8 Pillars of the New Community Policing" are developed to aid in defining and implementing community policing

    Reference evapotranspiration from coarse-scale and dynamically downscaled data in complex terrain: Sensitivity to interpolation and resolution

    The main objective of this study was to investigate whether dynamically downscaled high resolution (4-km) climate data from the Weather Research and Forecasting (WRF) model provide physically meaningful additional information for reference evapotranspiration (E) calculation compared to the recently published GridET framework that uses interpolation from coarser-scale simulations run at 32-km resolution. The analysis focuses on complex terrain of Utah in the western United States for years 1985–2010, and comparisons were made statewide with supplemental analyses specifically for regions with irrigated agriculture. E was calculated using the standardized equation and procedures proposed by the American Society of Civil Engineers from hourly data, and climate inputs from WRF and GridET were debiased relative to the same set of observations. For annual mean values, E from WRF (EW) and E from GridET (EG) both agreed well with E derived from observations (r2 = 0.95, bias \u3c 2 mm). Domain-wide, EW and EG were well correlated spatially (r2 = 0.89), however local differences ΔE=EW-EG were as large as +439 mm year−1 (+26%) in some locations, and ΔE averaged +36 mm year−1. After linearly removing the effects of contrasts in solar radiation and wind speed, which are characteristically less reliable under downscaling in complex terrain, approximately half the residual variance was accounted for by contrasts in temperature and humidity between GridET and WRF. These contrasts stemmed from GridET interpolating using an assumed lapse rate of Γ = 6.5K km−1, whereas WRF produced a thermodynamically-driven lapse rate closer to 5K km−1 as observed in mountainous terrain. The primary conclusions are that observed lapse rates in complex terrain differ markedly from the commonly assumed Γ = 6.5K km−1, these lapse rates can be realistically resolved via dynamical downscaling, and use of constant Γ produces differences in E of order as large as 102 mm year−1

    The Sloan Lens ACS Survey. VII. Elliptical Galaxy Scaling Laws from Direct Observational Mass Measurements

    We use a sample of 53 massive early-type strong gravitational lens galaxies with well-measured redshifts (ranging from z=0.06 to 0.36) and stellar velocity dispersions (between 175 and 400 km/s) from the Sloan Lens ACS (SLACS) Survey to derive numerous empirical scaling relations. The ratio between central stellar velocity dispersion and isothermal lens-model velocity dispersion is nearly unity within errors. The SLACS lenses define a fundamental plane (FP) that is consistent with the FP of the general population of early-type galaxies. We measure the relationship between strong-lensing mass M_lens within one-half effective radius (R_e/2) and the dimensional mass variable M_dim = G^-1 sigma_e2^2 R_e/2 to be log_10 [M_lens/10^11 M_Sun] = (1.03 +/- 0.04) log_10 [M_dim/10^11 M_Sun] + (0.54 +/- 0.02) (where sigma_e2 is the projected stellar velocity dispersion within R_e/2). The near-unity slope indicates that the mass-dynamical structure of massive elliptical galaxies is independent of mass, and that the "tilt" of the SLACS FP is due entirely to variation in total (luminous plus dark) mass-to-light ratio with mass. Our results imply that dynamical masses serve as a good proxies for true masses in massive elliptical galaxies. Regarding the SLACS lenses as a homologous population, we find that the average enclosed 2D mass profile goes as log_10 [M(<R)/M_dim] = (1.10 +/- 0.09) log_10 [R/R_e] + (0.85 +/- 0.03), consistent with an isothermal (flat rotation curve) model when de-projected into 3D. This measurement is inconsistent with the slope of the average projected aperture luminosity profile at a confidence level greater than 99.9%, implying a minimum dark-matter fraction of f_DM = 0.38 +/- 0.07 within one effective radius.     Metabolomic and Functional Genomic Analyses Reveal Varietal Differences in Bioactive Compounds of Cooked Rice

    Emerging evidence supports that cooked rice (Oryza sativa L.) contains metabolites with biomedical activities, yet little is known about the genetic diversity that is responsible for metabolite variation and differences in health traits. Metabolites from ten diverse varieties of cooked rice were detected using ultra performance liquid chromatography coupled to mass spectrometry. A total of 3,097 compounds were detected, of which 25% differed among the ten varieties. Multivariate analyses of the metabolite profiles showed that the chemical diversity among the varieties cluster according to their defined subspecies classifications: indica, japonica, and aus. Metabolite-specific genetic diversity in rice was investigated by analyzing a collection of single nucleotide polymorphisms (SNPs) in genes from biochemical pathways of nutritional importance. Two classes of bioactive compounds, phenolics and vitamin E, contained nonsynonymous SNPs and SNPs in the 5′ and 3′ untranslated regions for genes in their biosynthesis pathways. Total phenolics and tocopherol concentrations were determined to examine the effect of the genetic diversity among the ten varieties. Per gram of cooked rice, total phenolics ranged from 113.7 to 392.6 µg (gallic acid equivalents), and total tocopherols ranged between 7.2 and 20.9 µg. The variation in the cooked rice metabolome and quantities of bioactive components supports that the SNP-based genetic diversity influenced nutritional components in rice, and that this approach may guide rice improvement strategies for plant and human health

    Bremelanotide for Treatment of Female Hypoactive Sexual Desire

    Hypoactive sexual desire disorder (HSDD) is a persistent deficiency or absence of sexual fantasies and desire resulting in significant distress or interpersonal difficulty. Women with this disorder may display a lack of motivation for sexual activity, reduced responsiveness to erotic cues, a loss of interest during sexual activity, and avoidance of situations that could lead to sexual activity. The pathophysiology of HSDD is thought to be centered around inhibitory and excitatory hormones, neurotransmitters, and specific brain anatomy. Due to the multifactorial nature of HSDD, treatment can be complex and must attempt to target the biological and psychosocial aspects of the disorder. Bremelanotide is a melanocortin receptor agonist and has been recently approved by the FDA to treat HSDD. Bremelanotide is administered intranasally or as a subcutaneous injection. The recommended dosage of bremelanotide is 1.75 mg injected subcutaneously in the abdomen or thigh at least 45 min before sexual activity. Studies showed improvements in desire, arousal, and orgasm scores when 1.75 mg of bremelanotide was administered before sexual activity compared to a placebo. Bremelanotide is a promising way to treat HSDD

    Quantum-enhanced metrology with the single-mode coherent states of an optical cavity inside a quantum feedback loop

    In this paper, we use the nonlinear generator of dynamics of the individual quantum trajectories of an optical cavity inside an instantaneous quantum feedback loop to measure the phase shift between two pathways of light with a precision above the standard quantum limit. The feedback laser provides a reference frame and constantly increases the dependence of the state of the resonator on the unknown phase. Since our quantum metrology scheme can be implemented with current technology and does not require highly efficient single photon detectors, it should be of practical interest until highly entangled many-photon states become more readily available

    Accurate identification of hospital admissions from care homes; development and validation of an automated algorithm

    Background: measuring the complex needs of care home residents is crucial for resource allocation. Hospital patient administration systems (PAS) may not accurately identify admissions from care homes. Objective: to develop and validate an accurate, practical method of identifying care home resident hospital admission using routinely collected PAS data. Method: admissions data between 2011 and 2012 (n = 103,105) to an acute Trust were modelled to develop an automated tool which compared the hospital PAS address details with the Care Quality Commission’s (CQC) database, producing a likelihood of care home residency. This tool and the Nuffield method (CQC postcode match only) were validated against a manual check of a random sample of admissions (n = 2,000). A dataset from a separate Trust was analysed to assess generalisability. Results:the hospital PAS was inaccurate; none of the admissions from a care home identified on manual check had a care home source of admission recorded on the PAS. Both methods performed well; the automated tool had a higher positive predictive value than the Nuffield method (100% 95% confidence interval (CI) 98.23–100% versus 87.10% 95%CI 82.28–91.00%), meaning those coded as care home residents were more likely to actually be from a care home. Our automated tool had a high level of agreement 99.2% with the second Trust’s data (Kappa 0.86 P &lt; 0.001). Conclusions: care home status is not routinely or accurately captured. Automated matching offers an accurate, repeatable, scalable method to identify care home residency and could be used as a tool to benchmark how care home residents use acute hospital resources across the National Health Service
