    Digestibility and rumen fermentation of a high forage diet pre-treated with a mixture of cellulase and xylanase enzymes

Unlocking the nutritional potential of poor-quality tropical forages with fibrolytic enzymes would improve forage digestibility and utilization. Using in vitro and in vivo methods this study investigated the effect of pre-treating Smutsfinger hay for 24 hours with a mixture of fibrolytic enzyme (100% cellulase; 75% cellulase: 25% xylanase; 50% cellulase: 50% xylanase; 25% cellulase: 75% xylanase; 100% xylanase and a control with no enzyme) on ruminal fermentation and digestibility of nutrients by sheep. For in vitro fermentation, dry matter, neutral detergent fibre (NDF) degradability and volatile fatty acids (VFA) were determined with standard procedures. The same treatments were used for an in vivo digestibility trial using Merino sheep in a 6 x 6 Latin square design. Feed intake and total tract digestibility were recorded. Rumen fluid samples were collected daily, preserved, and analysed for VFA. The addition of 100% cellulase enzyme to Smutsfinger hay in vitro increased (P <0.05) NDF degradability and gas production compared with the control and inclusion of 100% xylanase enzyme. Both 100% cellulase and xylanase enzymes significantly reduced in vitro end time fermentation pH. A 50:50 mixture of cellulase and xylanase plus enzyme in vivo, increased acetate, total VFA concentration, and higher NDF and ADF digestibility of the test feed compared with the control. Inclusion of a 50-75% mixture of cellulase and 50-25% xylanase enzymes treatment led to higher gas production and butyrate concentration, decreased ruminal pH and improved nutrient digestibility

    Activation of glucocorticoid receptors is associated with the suppression of antioxidant responses in the liver of goats fed a high-concentrate diet

    This study investigated changes in oxidative stress and the relevant mechanisms in the liver of goats fed a high-concentrate (HC) diet for 5 weeks. Twelve goats were randomly assigned to a low-concentrate (concentrate-to-forage = 55:45, LC, n = 6) or HC diet (concentrate-to-forage = 90:10, n = 6), with dry matter as the base. Enzyme activity assays, real-time polymerase chain reaction and western blotting were used to evaluate antioxidant parameters and gene expression in the liver. Superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), total nitric oxide synthase activity and total antioxidant capacity (T-AOC) in the liver declined (p < .01) in HC-fed goats compared to those in LC-fed goats. The mRNA levels of GPX1, CAT and SOD1 were down-regulated (3.69, 47.37 and 27.61%, respectively) in HC-fed goats compared to those in LC-fed goats. Furthermore, glutathione S-transferase M1 (GSTM1) was upregulated (466.35%, p < .01) in the liver of HC-fed goats. The mRNA and protein levels of the nuclear factor E2-related factor 2 (NRF2) and total glucocorticoid receptor (GR) declined (p < .05) in HC-fed goats (by 28.57, 33.1, 30.85 and 34%, respectively). However, the nuclear translocation of GR increased (p < .05; by 44.75%) in HC-fed goats. Negative correlations were detected for hepatic nuclear GR protein expression with hepatic CAT activity and GPx activity. In conclusion, feeding an HC diet to goats for 5 weeks suppressed NRF2-dependent antioxidant responses and enhanced GR nuclear translocation in the liver

    Anti-staphylococcal properties of four plant extracts against sensitive and multi-resistant bacterial strains isolated from cattle and rabbits

    The aim of this study is to investigate the biopotency of methanolic extracts of Vitex mollis, Psidium guajava, Dalbergia retusa, and Crescential alata leaves against various staphylococcal strains isolated from cattle and rabbits. Methicillin-resistant S. aureus strains were isolated from cattle, while other strains were isolated from rabbits using standard methodology. The total phytochemical phenolic and saponins contents were obtained being the main groups of the antinutritional factors. The antimicrobial activity of the extracts against the standard culture of S. aureus (control) and S. aureus isolated from cattle and rabbits were investigated comparatively relative to that of oxacillin. It was found that both the control S. aureus and the isolated S. aureus are susceptible to all the four plant extracts, and sensitive to oxacillin. Of all the S. aureus including the control, MRSA2 is the most susceptible to all the extracts at 1000 μg/mL, except that of V. mollis where it is the least susceptible. Among all the plant extracts, P. guajava is the most active against MRSA2 and SOSA2. Therefore, the isolates from cattle (MRSA1 and MRSA2) are more susceptible to all the plant extracts than the isolates from rabbits. Among all the rabbit isolates, CoNS3 is the least susceptible to the extracts. Since all the plant extracts exhibit remarkable inhibitory activities against all the S. aureus strains, they are promising towards the production of therapeutic drugs

    Influence of dietary supplementation with sunflower oil and quebracho tannins on growth performance and meat fatty acid profile of Awassi lambs

    The influence of dietary supplementation with quebracho tannins (QT) or/and sunflower oil (SF) on growth performance and fatty acid profile in meat of Awassi lambs was studied. Seventy-two lambs were assigned randomly to nine diets (8 per group) in the experiment following a 32 factorial design with all possible combinations of two factors, SF and QT each at three levels i.e. 0 (control), 20 or 40 g/kg diet DM. A significant SF ×QT interaction was observed on the conjugated linoleic acid content of intramuscular fat (P =0.007), but not on average daily weight gain (P=0.11). Addition of QT at 40 g/ kg DM significantly increased average daily gain comparing to control diet. The inclusion of QT in the diet increased the content of the conjugated linoleic acid in the lamb meat above that achieved with SF supplementation only. Addition of the combination of QT and SF to the diet enhanced the content of conjugated linoleic acid (C18:2 c9t11) of the lamb meat. This could be due to the reduction in the bio-hydrogenation of unsaturated fatty acids in the diet.     The Dietary Components and Feeding Management as Options to Offset Digestive Disturbances in Horses

    Equines are faced with digestive disorders derived from abrupt feed changes from mainly forages to high-starch cereal grains. This diet change aimed at meeting the nutritional requirements to optimize the horse's performance causes microbial imbalances in the gut, which results in unintended but inevitable health problems such as colic, diarrhea, gastric ulcer, and laminitis. Several strategies are recommended to control these problems. This review is meant to offer plausible and adoptable solutions to digestive disturbances in equines. High-forage diets seem to be the primary solution. However, there are other options such as feeding horses with alfalfa hay for gut pH modulation and gastric ulcer management, avoidance of the off-feed for long periods within a day especially between 01:00 AM and 09:00 AM, yeast supplementation, feeding equines about 2–3 times a day without increasing the quantity of the daily ration, and dietary fatty acid supplementation such as eicosapentaenoic acid and docosahexaenoic acid to increase insulin sensitivity. Protozoa and faunation seem to be a good modulator of gut pH because they help to control the rate of starch degradation without generating methane in horses because acetogenesis is the main hydrogen sink in equines. An appropriate grain mixture of barley–corn or barley–oat avoids gut pH depression. Furthermore, the use of crop by-products such as soybean hull and pectin are good alternative energy sources for equines. Therefore, the use of some sources of fiber in the diet of equines can replace high-starch cereal grains in equine diets because they are capable of providing the required energy to support the energy demands during work

    Effects of addition of exogenous fibrolytic enzymes on digestibility and milk and meat production: A systematic review

    El articulo esta publicado en una revista de Open accessExogenous fibrolytic enzymes (EFE) added to the ruminant diet can increase fiber digestibility and production efficiency. A systematic review was conducted to understand the interactions between EFE and diet on digestibility and animal performance. The database included variables from 384 experiments with EFE and 264 controls from 85 papers published since 2000 (classification criteria: 1) type of study (in vitro, in situ, in vivo), 2) type of ruminants (sheep, Buffaloes, goats, beef and dairy cattle), 3) primary EFE activity (Cellulases (Cel) or Xylanases (Xyl)), 4) forage proportion (FP), 5) type of plant (TP: legumes or grasses), 6) number of ingredients in diets, and 7) application time (AT)). In over of 52.85% of cases, EFE improved the degradability of dry matter (DMD), neutral and acid detergent fiber (NDFD and ADFD), in vitro gas production (GP), volatile fatty acids (VFA), the acetate: propionate ratio (A:P ratio), protein and fat milk, milk yield and average daily gain (ADG) (by 7.78–21.85%). Cel improved organic matter degradability (OMD), GP, VFA, milk yield, and milk protein and fat content. EFE in FP≥40% diets enhanced the ADG, and in grass-based diets increased the dry matter intake (DMI). The AT of EFE affected the DMD, NDFD, and ADFD. Significant correlations were found between the improvements of NDFD or ADFD with DMD (r>0.59), milk yield (r=0.64), and ADG (r=0.59). In conclusion, many factors interact with EFE supplementation effects, but EFE consistently enhanced the DMD, NDFD, and ADFD of ruminant diets, which are related to improvements in productive performance

    Optimal Nitrogen Fertilization to Reach the Maximum Grain and Stover Yields of Maize (Zea mays L.): Tendency Modeling

    Utilization of maize stover to the production of meat and milk and saving the grains for human consumption would be one strategy for the optimal usage of resources. Variance and tendency analyses were applied to find the optimal nitrogen (N) fertilization dose (0, 100, 145, 190, 240, and 290 kg/ha) for forage (F), stover (S), cob (C), and grain (G) yields, as well as the optimal grain-to-forage, cob-to-forage, and cob-to-stover ratios (G:F, C:F, and C:S, respectively). The study was performed in central Mexico (20.691389° N and −101.259722° W, 1740 m a.m.s.l.; Cwa (Köppen), 699 mm annual precipitation; alluvial soils). N-190 and N-240 improved the individual yields and ratios the most. Linear and quadratic models for CDM, GDM, and G:F ratio had coefficients of determination (R2) of 0.20–0.46 (p < 0.03). Cubic showed R2 = 0.30–0.72 (p < 0.02), and the best models were for CDM, GDM, and the G:F, C:F, and C:S DM ratios (R2 = 0.60–0.72; p < 0.0002). Neither SHB nor SDM negatively correlated with CDM or GDM (r = 0.23–0.48; p < 0.0001). Excess of N had negative effects on forage, stover, cobs, and grains yields, but optimal N fertilization increased the proportion of the G:F, C:F, and C:S ratios, as well as the SHB and SDM yields, without negative effects on grain production

    Antimicrobial and bactericidal impacts of Bacillus amyloliquefaciens CECT 5940 on fecal shedding of pathogenic bacteria in dairy calves and adult dogs

    Two experiments were carried out to evaluate the bactericidal impacts of Bacillus amyloliquefaciens CECT 5940 on the shedding of faecal pathogenic bacteria in dairy calves (Experiment 1) and in adults dogs (experiment 2). In the calves experiment, a completely randomized design was used to investigate the faecal bacteria profile of Holstein dairy calves fed with either pasteurized waste milk (PWM; n = 9) or a formulated non-medicated milk replacer (NMR; n = 9) for 60 d. The NMR containing sodium-butyrate and the active probiotic B. amyloliquefaciens CECT 5940. In the dogs experiment, addition of same probiotic (i.e., B. amyloliquefaciens CECT 5940) was carried out in two stages. The first stage started from day 7–37, and the second from day 44–71. The assessment of faecal score measured on day 22, 37, 42, 57, 71 and 77 to determine the texture of the stools. Calves received PWM consumed (P < 0.05) more starter feed between day 16 and day 45. The calves fed NMR had more moisture faeces and less cough reflux than the PWM-calves. Feeding NMR to calves increased faecal Klebsiella oxytoca and Proteus vulgaris counts in comparison to PWM-calves. The administration of B. amyloliquefaciens CECT 5940 to the dog diet has no significant effect on the hardness of the stool. Meanwhile, the bacillus count increases while the coliforms count decreases upon B. amyloliquefaciens CECT 5940 administration. This reveals that B. amyloliquefaciens CECT 5940 survived the gastrointestinal passage and rapidly colonized the dog intestine, which could positively affect the metabolism and composition of the intestinal microflora. These results show that B. amyloliquefaciens are a promising probiotic with an antimicrobial and bactericidal activities against the intestinal pathogenic bacteria for dairy calves and adult dogs

    El articulo esta publicado en una revista de Open accessThe objective was to review the potential effects of adding anthocyanin delphinidin-3-O-sambubioside (DOS) and cyanidin-3-O-sambubioside (COS) of HS in animal diets. One hundred and four scientific articles published before 2021 in clinics, pharmacology, nutrition, and animal production were included. The grains/concentrate, metabolic exigency, and caloric stress contribute to increasing the reactive oxygen species (ROS). COS and DOS have antioxidant, antibacterial, antiviral, and anthelmintic activities. In the rumen, anthocyanin might obtain interactions and/or synergisms with substrates, microorganisms, and enzymes which could affect the fiber degradability and decrease potential methane (CH4) emissions; since anthocyanin interferes with ruminal fatty acids biohydrogenation (BH), they can increase the n-3 and n-6 polyunsaturated fatty acids (PUFA), linoleic acid (LA), and conjugated linoleic acid (CLA) in milk and meat, as well as improving their quality. Anthocyanins reduce plasma oxidation and can be deposited in milk and meat, increasing antioxidant activities. Therefore, the reduction of the oxidation of fats and proteins improves shelf-life. Although studies in ruminants are required, COS and DOS act as inhibitors of the angiotensin-converting enzyme (ACEi) and rennin expression, regulating the homeostatic control and possibly the milk yield and body weight. By-products of HS contain polyphenols as calyces with positive effects on the average daily gain and fat meat quality

    Antibacterial activities of the phytochemicals-characterized extracts of Callistemon viminalis, Eucalyptus camaldulensis and Conyza dioscoridis against the growth of some phytopathogenic bacteria

    Three bacterial isolates were isolated from infected potato tubers showing soft and brown rots like symptoms as well as one isolate from infected peach tree showing crown gall symptom. The morphological, biochemical and molecular assays proved that bacterial isolates belonging to Pectobacterium carotovorum subsp. carotovorum, Ralstonia solanacearum, Dickeya spp. and Agrobacterium tumefaciens. The acetone (AcE) and n-butanol (ButE) extracts of Callistemon viminalis flowers and essential oil from aerial parts of Conyza dioscoridis as well as ButE of Eucalyptus camaldulensis bark are evaluated at different concentrations against the growth of the isolated bacteria. The diameter of inhibition zone (IZ) and the minimum inhibitory concentrations (MICs) are compared. Results indicated that the highest IZ values were 20.0 mm and 18.3 mm for E. camaldulensis bark ButE and C. viminalis flower ButE, respectively, against P. carotovorum; 16.3 mm and 16.0 mm for E. camaldulensis bark ButE and C. viminalis flower ButE, respectively, against R. solanacearum; 18.5 mm for C. viminalis flower AcE and C. dioscoridis aerial parts EO against Dickeya spp.; and 15.0 mm for C. viminalis flower AcE against A. tumefaciens. MICs ranged from4000 μg/mL for A. tumefaciens. It was proved that C. viminalis flowers AcE contains mainly 5-hydroxymethylfurfural (20.6%), palmitic acid (18.5%), and pyrogallol (16.4%); while C. viminalis flower ButE contains palmitic acid (36.3%), 2-hydroxymyristic acid (9.4%), 5-hydroxymethylfurfural (7.2%), and shikimic acid (6.6%); whereas E. camaldulensis bark ButE contains 8-nonynoic acid methyl ester (45.6), camphor (30.9%), menthol (8.8%), and 1,8-cineole (eucalyptol) (8.2%), whilst the EO of C. dioscoridis aerial parts comprises Z-(13,14-epoxy)tetradec-11-en-1-ol acetate (11.6%), γ-elemene (10.2%), tau.- muurolol (7.1%), and cadina-3,9-diene (4.7%). It can be concluded that phytochemical extracts of C. viminalis, E. camaldulensis and C. dioscoridis demonstrated strong to moderate antibacterial effects against the studied plant bacterial pathogens
