    Evaluación fotogramétrica de los estándares de tolerancia geométrica: un estudio de caso de modelo de Darpa

    A DARPA Suboff scale model was used for the development of an analysis methodology for geometric deviations of the hull, aiming the quality control of model manufacturing process. The surface three-dimensional coordinates were obtained through a photogrammetric survey and analyzed based on statistical tools. The results were classified according to the ITTC and DIN ISO geometric tolerance standards, allowing the investigation of the compliance of the model to such criteria and the study of local geometric deviations.Se utilizó un modelo a escala DARPA Suboff para el desarrollo de una metodología de análisis para las desviaciones geométricas del casco, con el objetivo de controlar la calidad del modelo del proceso de fabricación. Las coordenadas tridimensionales de la superficie se obtuvieron a través de un estudio fotogramétrico y se analizaron en base a herramientas estadísticas. Los resultados se clasificaron de acuerdo con los estándares de tolerancia geométrica del ITTC y DIN ISO, lo que permitió investigar el cumplimiento del modelo con dichos criterios y estudiar las desviaciones geométricas locales

    Spatio-temporal study and population structure of Daptonema Oxycerca (Nematoda: Xyalidae) in Coroa Grande, Rio de Janeiro, Brazil

    Daptonema oxycerca foi descrita originalmente para o Mar do Norte, mas até o momento nada se conhece sobre essa espécie em regiões tropicais. A distribuição espaço-temporal e a estrutura populacional de D. oxycerca foram estudadas durante o período de um ano (de Maio 1998 a Abril 1999) em Coroa Grande, Rio de Janeiro, Brasil. Para a amostragem, dois transectos fixos e perpendiculares à linha d'água foram estabelecidos, e, em cada transecto, quatro níveis foram marcados para representar as zonas superior e inferior do médiolitoral. As menores temperaturas foram observadas no final do inverno e início da primavera e as maiores temperaturas no verão. A densidade de D. oxycerca variou durante o ano, com um pico em agosto. Essa tendência foi observada para os machos, as fêmeas grávidas e as fêmeas não-grávidas. A densidade da espécie correlacionou-se negativamente com a temperatura. A variação da razão sexual não foi significativa. D. oxycerca foi mais abundante no nível superior do médiolitoral em ambos os transectos. Esse trabalho representa o primeiro registro de D. oxycerca no Brasil, assim como, confirma a influência da temperatura na regulação de sua densidade populacional.Daptonema oxycerca was originally described from the North Sea and, up to now, nothing is known of this species in tropical regions. The spatio-temporal distribution and the population structure of this species was studied during one-year period (from May 1998 through April 1999) in Coroa Grande, Rio de Janeiro, Brazil. For sampling, two fixed, parallel transects were established perpendicularly to the shoreline, and at each transect, four, equally spaced levels were marked to represent the upper and the low intertidal zones. The lowest temperatures occurred in the end of the austral winter and first month of the spring and the highest temperatures occurred in the austral summer. The density of D. oxycerca varied during the year, with a peak in August. This tendency was observed for males, ovigerous females, and non-ovigerous females. The species density was negative correlated with temperature. The sex ratio was not statistically significant. D. oxycerca was most abundant at the upper intertidal level in both transects. This work provides the first record of D. oxycerca in Brazil, and confirms the influence of temperature in regulating its population density

    Study of the electrochemical reduction of amoebicide Teclozan and its amperometric determination in pharmaceutical formulations

    Neste trabalho, a redução eletroquímica do amebicida Teclozan (TEC) foi estudada sobre um eletrodo de carbono vítreo em meio de acetonitrila. Eletrólises com potencial controlado foram realizadas visando, tanto a determinação do número de elétrons envolvidos na redução do fármaco, quanto a identificação dos produtos eletrogerados, os quais foram isolados por extração líquido-líquido e caracterizados por 1H RMN. Foi verificado que o TEC apresenta dois picos voltamétricos, cada um associado à quebra redutiva de duas ligações C-Cl. Em presença de um doador de prótons, foi observado que o primeiro pico voltamétrico em −1,8 V corresponde principalmente à redução dos grupamentos -CHCl2 a -CH2Cl; enquanto o segundo pico em −2,2 V é responsável pela redução dos grupos -CH2Cl a CH3, fornecendo como único produto o derivado totalmente desalogenado do TEC, com rendimentos entre 82 e 97%. Este trabalho descreve também o desenvolvimento de um método eletroanalítico baseado na detecção amperométrica do TEC em condições hidrodinâmicas, o qual forneceu um limite de detecção de 8,9 × 10-6 mol L-1.The electrochemical reduction of amoebicide Teclozan (TEC) was studied on a glassy carbon electrode in acetonitrile. Controlled-potential electrolyses were performed for coulometric and preparative purposes. The electrogenerated products were isolated by liquid-liquid extraction and characterized by 1H NMR. It was observed that TEC presents two voltammetric peaks, each one associated with the cleavage of two C-Cl bonds. In presence of a proton donor it was observed that the first peak at −1.8 V promotes mainly the reduction of the groups CHCl2 to CH2Cl and the second one at −2.2 V promotes the reduction of the groups CH2Cl to CH3 giving as the sole product the completely dechlorinated TEC derivative with yields between 82 and 97%. In addition, a comparative study between the analytical performance of voltammetric techniques and amperometric detection of TEC in hydrodynamic conditions was performed. The amperometric detection was more sensitive than all evaluated voltammetric techniques, providing a detection limit of 8.9 × 10-6 mol L-1.FAPESP; CAPES; FC

    Fatty acid binding proteins have the potential to channel dietary fatty acids into enterocyte nuclei

    Intracellular lipid binding proteins, including fatty acid binding proteins (FABPs) 1 and 2, are highly expressed in tissues involved in the active lipid metabolism. A zebrafish model was used to demonstrate differential expression levels of fabp1b.1, fabp1b.2, and fabp2 transcripts in liver, anterior intestine, and brain. Transcription levels of fabp1b.1 and fabp2 in the anterior intestine were upregulated after feeding and modulated according to diet formulation. Immunofluorescence and electron microscopy immunodetection with gold particles localized these FABPs in the microvilli, cytosol, and nuclei of most enterocytes in the anterior intestinal mucosa. Nuclear localization was mostly in the interchromatin space outside the condensed chromatin clusters. Native PAGE binding assay of BODIPY-FL-labeled FAs demonstrated binding of BODIPY-FLC12 but not BODIPY-FLC5 to recombinant Fabp1b.1 and Fabp2. The binding of BODIPY-FLC12 to Fabp1b.1 was fully displaced by oleic acid. In vivo experiments demonstrated, for the first time, that intestinal absorption of dietary BODIPY-FLC12 was followed by colocalization of the labeled FA with Fabp1b and Fabp2 in the nuclei. These data suggest that dietary FAs complexed with FABPs are able to reach the enterocyte nucleus with the potential to modulate nuclear activity

    A Potential Link Between Prolonged Cork Exposure and Intestinal-Type Sinonasal Adenocarcinoma – Special Findings of a Retrospective Cohort Analysis

    Introduction: intestinal-type sinonasal adenocarcinoma (ITAC) is a rare epithelium tumor of the nasal cavities and paranasal sinuses. Exposure to wood and leather dusts is a strong etiological factor related to its development. Prolonged cork exposure has rarely been associated. Materials and methods: thirty-seven-year (1981-2018) retrospective cohort analysis of all consecutive patients with sinonasal cancer (SNC) followed at our institution. Medical records were reviewed to determine patient demographics, occupational/environmental exposure, location and extent of the tumor, stage, histopathology findings, treatment strategies, and oncologic outcomes. Survival analysis was done using Kaplan-Meier method. Results: we evaluated 379 patients with SNC, including 39 (10.3%) ITAC. Patient median age was 73 years (range 49-87), 56% male and 69% with identified professional occupational exposure (54% for cork; 69.2% considering only those for which an agent has been identified). Seventy-two percent had locally advanced disease (stage III or IVA-B). The initial treatment was surgery in 77%, and 54% received adjuvant radiotherapy. The median time to progression, progression-free survival, and overall-survival was 2.36 years (95% CI 1.54-8.70), 1.96 years (95% CI 1.43-3.74), and 3.51 years (95% CI 2.33-10.02), respectively. Conclusion: ITAC is an uncommon malignancy that grows silently, which contributes to delayed diagnosis, advanced stage and low survival rates. In our cohort, we observed a high prevalence of cork occupational exposure. This finding may lead to the implementation of protection measures and suggest a potential link to be further studied.info:eu-repo/semantics/publishedVersio

    Population wide testing pooling strategy for SARS-CoV-2 detection using saliva

    SARS-CoV-2 pandemic has forced frequent testing of populations. It is necessary to identify the most cost-effective strategies for the detection of COVID-19 outbreaks. Nasopharyngeal samples have been used for SARS-CoV-2 detection but require a healthcare professional to collect the sample and cause discomfort and pain to the individual. Saliva has been suggested as an appropriate fluid for the diagnosis of COVID-19. We have investigated the possibility of using pools of saliva samples to detect SARS-CoV-2 in symptomatic and asymptomatic patients. Two hundred and seventy-nine saliva samples were analyzed through RT-PCR of Envelope, Nucleocapsid and Open Reading Frame 1ab genes. Reproducibility assays showed an almost perfect agreement as well as high sensitivity (96.6%), specificity (96.8%), positive predicted value (96.6%), and negative predicted value (96.8%). The average Cycle Threshold of the genes detected was 29.7. No significant differences (p > 0.05) were detected when comparing the cycle threshold average of two consecutive reactions on the same positive saliva samples. Saliva samples have a higher median viral load (32.6) than in nasopharyngeal samples (28.9), although no significant differences were detected (p > 0.05). Saliva-pool samples allowed effective SARS-CoV-2 screening, with a higher sensibility (96.9%) on 10-sample pools than in 20-sample pools (87.5%). Regardless of pools size specificity was high (99.9%) and an almost perfect agreement was observed. Our strategy was successfully applied in population wide testing of more than 2000 individuals, showing that it is possible to use pooled saliva as diagnostic fluid for SARS-CoV-2 infection.info:eu-repo/semantics/publishedVersio

    The Transcriptome of the Salivary Glands of Amblyomma aureolatum Reveals the Antimicrobial Peptide Microplusin as an Important Factor for the Tick Protection Against Rickettsia rickettsii Infection

    The salivary glands (SG) of ixodid ticks play a pivotal role in blood feeding, producing both the cement and the saliva. The cement is an adhesive substance that helps the attachment of the tick to the host skin, while the saliva contains a rich mixture of antihemostatic, anti-inflammatory, and immunomodulatory substances that allow ticks to properly acquire the blood meal. The tick saliva is also a vehicle used by several pathogens to be transmitted to the vertebrate host, including various bacterial species from the genus Rickettsia. Rickettsia rickettsii is a tick-borne obligate intracellular bacterium that causes the severe Rocky Mountain spotted fever. In Brazil, the dog yellow tick Amblyomma aureolatum is a vector of R. rickettsii. In the current study, the effects of an experimental infection with R. rickettsii on the global gene expression profile of A. aureolatum SG was determined by next-generation RNA sequencing. A total of 260 coding sequences (CDSs) were modulated by infection, among which 161 were upregulated and 99 were downregulated. Regarding CDSs in the immunity category, we highlight one sequence encoding one microplusin-like antimicrobial peptide (AMP) (Ambaur-69859). AMPs are important effectors of the arthropod immune system, which lack the adaptive response of the immune system of vertebrates. The expression of microplusin was confirmed to be significantly upregulated in the SG as well as in the midgut (MG) of infected A. aureolatum by a quantitative polymerase chain reaction preceded by reverse transcription. The knockdown of the microplusin expression by RNA interference caused a significant increase in the prevalence of infected ticks in relation to the control. In addition, a higher rickettsial load of one order of magnitude was recorded in both the MG and SG of ticks that received microplusin-specific dsRNA. No effect of microplusin knockdown was observed on the R. rickettsii transmission to rabbits. Moreover, no significant differences in tick engorgement and oviposition were recorded in ticks that received dsMicroplusin, demonstrating that microplusin knockdown has no effect on tick fitness. Further studies must be performed to determine the mechanism of action of this AMP against R. rickettsii
