    The effects of equine-assisted activities on the social functioning in children and adolescents with autism spectrum disorder

    Equine-assisted activities and therapies are increasing in popularity for treatment of ASD symptoms. This research evaluated effects of a 5-week programme of therapeutic riding on social functioning of children/adolescents (N = 15) with ASD. The effectiveness of the programme was evaluated using the autism spectrum quotient, the Vineland Adaptive Behaviour Scale and the empathising and systemising quotient. Results established that the TR intervention increased empathising and reduced maladaptive behaviours. The findings also indicated that specific adaptive behaviours like socialization and communication were not affected by the intervention. Thus, a complex picture of the effects of this intervention emerges: while TR does not change all of the child’s behaviour, it can improve specific aspects of social functioning and also reduce maladaptive ASD traits

    In vitro evolution of an HIV integrase binding protein from a library of C-terminal domain γS-crystallin variants

    A protein without natural binding functions was engineered to bind HIV-1 integrase. Phage display selections applied a library of variants based on the C-terminal domain of the eye lens protein human γS-crystallin. Multiple loop regions were altered to encode libraries with ≈3.6×10(11) different variants. A crystallin variant, termed Integrase-Binding Protein-10 (IBP-10), inhibits integrase catalysis with nanomolar K(i) values. IBP-10 interacts with the integrase C-terminal domain and inhibits integrase substrate affinity. This allosteric mechanism allows IBP-10 to inhibit drug resistant integrase variants. The results demonstrate the applicability of the crystallin scaffold for the discovery of binding partners and enzyme inhibitors

    Mentalizing and Emotional Labor Facilitate Equine-Assisted Social Work with Self-harming Adolescents

    This article explores equine-assisted social work (EASW). Horses' capacities to mirror human emotions create possibilities for authentic relationships between clients and staff. This study examines what eases or counteracts the horse's capacity to facilitate relationships perceived by humans to be authentic. Video recordings of the human-horse interactions of three staff members and four female self-harming clients aged 15–21 years in a residential treatment facility were analyzed. The findings show that if the staff gave instructions and advice similar to traditional equestrian sports in combination with viewing the horse as an object, EASW is not facilitated. EASW seems to be facilitated when the horse is perceived as a subject by both staff and clients, provided that the staff gave meaning to the horse's behavior. The staff needed to highlight empathy for the horse when the horse is not able to fulfill its task without adding depth to the client's performance, to avoid raising defense mechanisms. The essence of EASW were perceived as eased by staff members when they focus on the client's emotions and help the client understand that the horse is acting in response to the client's and the staff's behavior through mentalizing and enacting emotional labor in regarding the horse as a subject. The results indicate the need for higher demands on staff members in order to facilitate EASW. Depending on whether the staff and the clients focus on performance or on emotions, different positive or negative outcomes on communication, self-confidence, self-esteem, and self-image will be likely to emerge.Publ online Jan 2015</p

    Genomic landscape of carcinogen-induced and genetically induced mouse skin squamous cell carcinoma

    Mouse models of cancers are routinely used to study cancer biology. However, it remains unclear whether carcinogenesis in mice is driven by the same spectrum of genomic alterations found in humans. Here we conducted a comprehensive genomic analysis of 9,10-dimethyl-1,2-benzanthracene (DMBA)-induced skin cancer, the most commonly used skin cancer model, which appears as benign papillomas that progress into squamous cell carcinomas (SCCs). We also studied genetically induced SCCs that expressed G12D mutant Kras (Kras G12D) but were deficient for p53. Using whole-exome sequencing, we uncovered a characteristic mutational signature of DMBA-induced SCCs. We found that the vast majority of DMBA-induced SCCs presented recurrent mutations in Hras, Kras or Rras2 and mutations in several additional putative oncogenes and tumor-suppressor genes. Similar genes were recurrently mutated in mouse and human SCCs that were from different organs or had been exposed to different carcinogens. Invasive SCCs, but not papillomas, presented substantial chromosomal aberrations, especially in DMBA-induced and genetically induced Trp53-mutated SCCs. Metastasis occurred through sequential spreading, with relatively few additional genetic events.
