Analysis of TSG101 tumour susceptibility gene transcripts in cervical and endometrial cancers
Carcinoma of the uterine cervix is a common malignancy among women that has been found to show loss of heterozygosity in the chromosome 11p. Recent studies have localized the TSG101 gene in this region, and also demonstrated a high frequency of abnormalities of this gene in human breast cancer. To determine the role of the TSG101 gene in the carcinogenesis of cervical and uterine carcinoma, 19 cases of cervical carcinoma and five cases of endometrial carcinoma, as well as nearby non-cancerous tissue from the same patients, and 16 blood samples from healthy persons as normal control were analysed by Southern blot analysis of genomic DNA, reverse transcription of the TSG101 mRNA followed by PCR amplification and sequencing of the products. We found that abnormal transcripts of the TSG101 gene were common both in cancerous or non-cancerous tissues of the uterus and cervix and in normal peripheral mononuclear cells. There was no genomic deletion or rearrangement in spite of the presence of abnormal transcripts, and no definite relationship between the abnormal transcripts and HPV infection was found. Although the frequency of abnormal transcripts was higher in cancerous than in non-cancerous tissue, normal peripheral mononuclear cells also had abnormal transcripts. Given these findings, the role of the TSG101 gene as a tumour-suppressor gene should be re-evaluated. Because some aberrant transcripts could be found at the first PCR reaction, we suggest that the aberrant transcripts might be the result of imperfect minor splicesome products. © 1999 Cancer Research Campaig
Revised Lithostratigraphy of the Sonsela Member (Chinle Formation, Upper Triassic) in the Southern Part of Petrified Forest National Park, Arizona
BACKGROUND: Recent revisions to the Sonsela Member of the Chinle Formation in Petrified Forest National Park have presented a three-part lithostratigraphic model based on unconventional correlations of sandstone beds. As a vertebrate faunal transition is recorded within this stratigraphic interval, these correlations, and the purported existence of a depositional hiatus (the Tr-4 unconformity) at about the same level, must be carefully re-examined. METHODOLOGY/PRINCIPAL FINDINGS: Our investigations demonstrate the neglected necessity of walking out contacts and mapping when constructing lithostratigraphic models, and providing UTM coordinates and labeled photographs for all measured sections. We correct correlation errors within the Sonsela Member, demonstrate that there are multiple Flattops One sandstones, all of which are higher than the traditional Sonsela sandstone bed, that the Sonsela sandstone bed and Rainbow Forest Bed are equivalent, that the Rainbow Forest Bed is higher than the sandstones at the base of Blue Mesa and Agate Mesa, that strata formerly assigned to the Jim Camp Wash beds occur at two stratigraphic levels, and that there are multiple persistent silcrete horizons within the Sonsela Member. CONCLUSIONS/SIGNIFICANCE: We present a revised five-part model for the Sonsela Member. The units from lowest to highest are: the Camp Butte beds, Lot's Wife beds, Jasper Forest bed (the Sonsela sandstone)/Rainbow Forest Bed, Jim Camp Wash beds, and Martha's Butte beds (including the Flattops One sandstones). Although there are numerous degradational/aggradational cycles within the Chinle Formation, a single unconformable horizon within or at the base of the Sonsela Member that can be traced across the entire western United States (the "Tr-4 unconformity") probably does not exist. A study of a couple with type 2 diabetes: dyadic adjustment and psychological morbidity
Objective: this study assessed dyadic adjustment and psychological morbidity
in type 2 diabetic patients and their partners, focusing on the role of gender.
Methods: 214 diabetic patients and their partners participated in the cross-sectional
study and were assessed on psychological morbidity (HADS) and marital
adjustment (RDAS). Data was analyzed using dyadic analysis, a statistical process
that studies the patient/partner dyads simultaneously.
Results: results revealed that the negative relationship between dyadic adjustment
and psychological morbidity in female patients was stronger than in male
diabetic patients or in partners of male diabetic patients. On the other hand, the
relationship between dyadic adjustment and psychological morbidity in partners
of diabetic men was stronger than the same relationship in partners of diabetic
women.
Conclusion: since gender is a moderator, it is important to attend to the different
needs of female and male patients and the education of diabetic patients

Dyadic adjustment, family coping, body image, quality of life and psychological morbidity in patients with psoriasis and their partners
Background Psoriasis is an incurable and chronic disease
that includes unpredictable periods of remission and relapse
requiring long-term therapy.
Purpose This paper focuses on the relationship among
family coping, psychological morbidity, body image,
dyadic adjustment and quality of life in psoriatic patients
and their partners.
Method One hundred and one patients with psoriasis and
78 partners comprised the sample. They were regular users
of the Dermatology Service of a Central Northern hospital
in Portugal and a private dermatology clinic. Patients with
psoriasis were assessed on anxiety, depression, body image,
quality of life, dyadic adjustment and family coping.
Partners were assessed on the same measures except body
image and quality of life.
Results A positive relationship among dyadic adjustment,
psychological morbidity and family coping in patients and
their partners was found. Also, patients with lower levels of
quality of life had partners with higher levels of depressive
and anxious symptoms. Better dyadic adjustment predicted
family coping in the psoriatic patient. High levels of dyadic
adjustment in patients and low partners’ trait anxiety
predicted better dyadic adjustment in partners.
Conclusion The results highlight the importance of incorporating
family variables in psychological interventions in
psoriasis’ care, particularly family coping and dyadic
adjustment as well as the need for psychological intervention
to focus both on patients and partners
Whole-genome sequencing for an enhanced understanding of genetic variation among South Africans
The Southern African Human Genome Programme is a national initiative that aspires to
unlock the unique genetic character of southern African populations for a better understanding
of human genetic diversity. In this pilot study the Southern African Human Genome
Programme characterizes the genomes of 24 individuals (8 Coloured and 16 black southeastern
Bantu-speakers) using deep whole-genome sequencing. A total of ~16 million unique
variants are identified. Despite the shallow time depth since divergence between the two
main southeastern Bantu-speaking groups (Nguni and Sotho-Tswana), principal component
analysis and structure analysis reveal significant (p < 10−6) differentiation, and FST analysis
identifies regions with high divergence. The Coloured individuals show evidence of varying
proportions of admixture with Khoesan, Bantu-speakers, Europeans, and populations from the
Indian sub-continent. Whole-genome sequencing data reveal extensive genomic diversity,
increasing our understanding of the complex and region-specific history of African populations
and highlighting its potential impact on biomedical research and genetic susceptibility to
disease
Expression of thymidylate synthase in human cells is an early G1 event regulated by CDK4 and p16INK4A but not E2F
Thymidylate synthase (TS) is the enzyme that catalyses the last step in de novo thymidylate synthesis. It is of interest clinically because it is an effective target for drugs such as 5-fluorouracil, often used in combination therapy. Despite a number of earlier reports indicating that TS is a cell cycle-dependent enzyme, this remains equivocal. Here, we show that in HCT116 cells synchronised by serum starvation, there is a clear dissociation between the expression of cyclin E (a well-characterised cell-cycle protein) and TS. Although both cyclin E and TS mRNA and protein increased during G1, TS upregulation was delayed. Moreover, TS levels did not decrease following S-phase completion while cyclin E decreased sharply. Similarly, clear differences were seen between cyclin E and TS as asynchronously growing HCT116 cells were growth-inhibited by low-serum treatment. In contrast to previous reports using rodent cells, adenovirus-mediated over-expression of E2F1 and cyclin E in three human cell lines had no effect on TS. Cell-cycle progression was blocked by treatment of cells with pharmacological inhibitors of CDK2 and CDK4 and by ectopic expression of p16INK4A. Whereas CDK2 inhibition had no effect on TS levels, inhibition of CDK4 was associated with decreased TS protein levels. These results provide the first evidence that drugs targeting CDK4 may be useful with anti-TS drugs as combination therapy for cancer
Determinants of recovery from post-COVID-19 dyspnoea: analysis of UK prospective cohorts of hospitalised COVID-19 patients and community-based controls
Background The risk factors for recovery from COVID-19 dyspnoea are poorly understood. We investigated determinants of recovery from dyspnoea in adults with COVID-19 and compared these to determinants of recovery from non-COVID-19 dyspnoea. Methods We used data from two prospective cohort studies: PHOSP-COVID (patients hospitalised between March 2020 and April 2021 with COVID-19) and COVIDENCE UK (community cohort studied over the same time period). PHOSP-COVID data were collected during hospitalisation and at 5-month and 1-year follow-up visits. COVIDENCE UK data were obtained through baseline and monthly online questionnaires. Dyspnoea was measured in both cohorts with the Medical Research Council Dyspnoea Scale. We used multivariable logistic regression to identify determinants associated with a reduction in dyspnoea between 5-month and 1-year follow-up. Findings We included 990 PHOSP-COVID and 3309 COVIDENCE UK participants. We observed higher odds of improvement between 5-month and 1-year follow-up among PHOSP-COVID participants who were younger (odds ratio 1.02 per year, 95% CI 1.01–1.03), male (1.54, 1.16–2.04), neither obese nor severely obese (1.82, 1.06–3.13 and 4.19, 2.14–8.19, respectively), had no pre-existing anxiety or depression (1.56, 1.09–2.22) or cardiovascular disease (1.33, 1.00–1.79), and shorter hospital admission (1.01 per day, 1.00–1.02). Similar associations were found in those recovering from non-COVID-19 dyspnoea, excluding age (and length of hospital admission). Interpretation Factors associated with dyspnoea recovery at 1-year post-discharge among patients hospitalised with COVID-19 were similar to those among community controls without COVID-19. Lawson criterion for ignition exceeded in an inertial fusion experiment
For more than half a century, researchers around the world have been engaged in attempts to achieve fusion ignition as a proof of principle of various fusion concepts. Following the Lawson criterion, an ignited plasma is one where the fusion heating power is high enough to overcome all the physical processes that cool the fusion plasma, creating a positive thermodynamic feedback loop with rapidly increasing temperature. In inertially confined fusion, ignition is a state where the fusion plasma can begin "burn propagation" into surrounding cold fuel, enabling the possibility of high energy gain. While "scientific breakeven" (i.e., unity target gain) has not yet been achieved (here target gain is 0.72, 1.37Â MJ of fusion for 1.92Â MJ of laser energy), this Letter reports the first controlled fusion experiment, using laser indirect drive, on the National Ignition Facility to produce capsule gain (here 5.8) and reach ignition by nine different formulations of the Lawson criterion
