    Muscle pathology in myotonic dystrophy: light and electron microscopic investigation in eighteen patients

    Myotonic dystrophy (DM) is the most common muscular dystrophy in adults. Two known genetic subtypes include DM1 (myotonic dystrophy type 1) and DM2 (myotonic dystrophy type 2). Genetic testing is considered as the only reliable diagnostic criterion in myotonic dystrophies. Relatively little is known about DM1 and DM2 myopathology. Thus, the aim of our study was to characterise light and electron microscopic features of DM1 and DM2 in patients with genetically proven types of the disease. We studied 3 DM1 cases and 15 DM2 cases from which muscle biopsies were taken for diagnostic purposes during the period from 1973 to 2006, before genetic testing became available at our hospital. The DM1 group included 3 males (age at biopsy 15–19). The DM2 group included 15 patients (5 men and 10 women, age at biopsy 26–60). The preferential type 1 fibre atrophy was seen in all three DM1 cases in light microscopy, and substantial central nucleation was present in two biopsies. Electron microscopy revealed central nuclei in all three examined muscle biopsies. No other structural or degenerative changes were detected, probably due to the young age of our patients. Central nucleation, prevalence of type 2 muscle fibres, and the presence of pyknotic nuclear clumps were observed in DM2 patients in light microscopy. Among the ultrastructural abnormalities observed in our DM2 group, the presence of internal nuclei, severely atrophied muscle fibres, and lipofuscin accumulation were consistent findings. In addition, a variety of ultrastructural abnormalities were identified by us in DM2. It appears that no single ultrastructural abnormality is characteristic for the DM2 muscle pathology. It seems, however, that certain constellations of morphological changes might be indicative of certain types of myotonic dystrophy. (Folia Morphol 2011; 70, 2: 121–129

    Physical activity levels as a quantifier in police officers and cadets

    Objectives: The aim of the present study was to determine the physical activity levels of active duty police officers and police academy cadets in different life domains and intensities. These parameters were treated as potential quantifiers that could be used when assessing individuals preparing for work as future police officers. Material and Methods: The study recruited 153 active police officers and 176 cadets attending a police academy and administered a diagnostic survey, the long-form version of the International Physical Activity Questionnaire, while in the statistical analysis the Student's t-test for independent groups was applied. Results: It was determined that police officers present high physical activity levels within the work domain, which are developed from initial training at a police academy and then throughout their police career. Conclusions: Such data are important in the light of the role police officers play in public safety as well as the prominence of physical activity within a particular profession and how it can be targeted and tailored to their needs

    Objectives: The aim of the study was to assess physical activity (PA) level in a representative sample of Polish adults.Materials and Methods: A cross-sectional analysis was performed among 2413 randomly selected individuals (51.5% women) aged 18-79 years, who participated in the Nationwide Study of Occurrence of Risk Factors of Cardiovascular Diseases NATPOL 2011 (March-July 2011). The study procedures consisted of a questionnaire as well as of anthropometric, blood pressure and biochemical measurements. Leisure-time, occupational and commuting PA were assessed by the use of a questionnaire interview. Results: About 48.2% of adults do exercise for at least 30 minutes on most days of a week. About 11% of the respondents declare a sedentary lifestyle. About 26.5% of working population report hard physical work, while sedentary work is reported by 47.6% of the employed participants. Active commuting is declared by 27.3% of working/studying population. About 47.2%, 36.6%, and 15.3% spend 30 min per day, respectively, on this kind of PA. Conclusions: PA level of more than half of Polish adults is still not satisfactory. Promotion of an active lifestyle should concern mainly leisure-time and commuting PA with paying special attention to substantial differences in various socio-demographic groups

    Incidence, risk factors, and outcomes of second neoplasms in patients with acute promyelocytic leukemia:the PETHEMA-PALG experience

    The most important challenges in acute promyelocytic leukemia (APL) is preventing early death and reducing long-term events, such as second neoplasms (s-NPLs). We performed a retrospective analysis of 2670 unselected APL patients, treated with PETHEMA “chemotherapy based” and “chemotherapy free” protocols. Only de novo APL patients who achieved complete remission (CR) and completed the three consolidation cycles were enrolled into the analysis. Out of 2670 APL patients, there were 118 (4.4%) who developed s-NPLs with the median latency period (between first CR and diagnosis of s-NPL) of 48.0 months (range 2.8–231.1): 43.3 (range: 2.8–113.9) for s-MDS/AML and 61.7 (range: 7.1–231.1) for solid tumour. The 5-year CI of all s-NPLs was of 4.43% and 10 years of 7.92%. Among s-NPLs, there were 58 cases of s-MDS/AML, 3 cases of other hematological neoplasms, 57 solid tumours and 1 non-identified neoplasm. The most frequent solid tumour was colorectal, lung and breast cancer. Overall, the 2-year OS from diagnosis of s-NPLs was 40.6%, with a median OS of 11.1 months. Multivariate analysis identified age of 35 years (hazard ratio = 0.2584; p &lt; 0.0001) as an independent prognostic factor for s-NPLs. There were no significant differences in CI of s-NPLs at 5 years between chemotherapy-based vs chemotherapy-free regimens (hazard ratio = 1.09; p = 0.932). Larger series with longer follow-up are required to confirm the potential impact of ATO+ATRA regimens to reduce the incidence of s-NPLs after front-line therapy for APL.</p

    Volunteer satisfaction in sports clubs: A multilevel analysis in 10 European countries

    Regular voluntary engagement is a basic resource for sports clubs that may also promote social cohesion and active citizenship. The satisfaction of volunteers is an imperative factor in this engagement, and the purpose of this article is to explore individual and organizational determinants of volunteer satisfaction in sports clubs. Theoretically, our study builds on the actor-theory concepts where volunteer satisfaction depends on subjective evaluations of expectations and experiences in a sports club (‘logic of situation’), so that positive evaluations lead to higher satisfaction and, hopefully, retention of volunteers. This research uses a sample of 8131 volunteers from 642 sports clubs in 10 European countries, and is the first analysis to combine determinants at the level of the club and the volunteer (multilevel). Results show that the most important determinants of satisfaction are the conditions of volunteering (recognition, support, leadership and material incentives) and the workload of volunteers. Surprisingly, club characteristics, size or having paid staff are not significant determinants of volunteer satisfaction. The results of this analysis can assist more effective volunteer management in sports clubs that are facing challenges of individualization and professionalization

    Oat–buckwheat breads – technological quality, staling and sensory properties

    peer reviewedThe technological and sensory properties and the staling of breads made from oat flour (OF) and buckwheat flour (BF) were analysed. Significant differences in protein and ash content were found in the experimental breads due to significant differences in the composition of the BF and OF used. As the proportion of BF in the recipe increased, a deterioration in the technological properties of the dough and bread as well as an increase in the crumb hardness were observed. The presence of OF in the recipe increased the bread volume, significantly enhanced the lightness of the crust and crumb and improved the overall sensory quality. The OF used in the recipe decreased the starch retrogradation enthalpy value, which is strongly related to a delay in bread staling. The proposed bakery products can be attractive to consumers who are looking for new food products

    W pracy przedstawione zostały wyniki pomiarów mikrotwardości z wgłębnikiem Vickersa. Badania przeprowadzono na układzie warstwa DLC – podłoże krzemowe. Jednocześnie z pomiarem mikrotwardości realizowany był zapis generowanych sygnałów emisji akustycznej. Wyniki mikrotwardości w postaci wykresów sił obciążania-odciążania oraz charakterystyki czasowe sygnałów EA były porównywane ze zdjęciami SEM odcisków wgłębnika. W pomiarach zaobserwowano efekt "pop-in", który korelował z występowaniem impulsów EA. Zwiększenie nacisków obciążenia skutkowało zwiększeniem uszkodzenia i odzwierciedlało się większą liczbą impulsów EA. Cienka warstwa DLC zwiększyła odporność na uszkodzenia.This paper presents the results of the measurements of microindentation using Vickers’ indenter. The research was carried out with the following system: the DLC (Diamond Like Carbon) layer deposited on silicon substrate. At the same time, the measurements of micro-indentation acoustic emission signals were recorded. The results of microindentation forces in the form of load-displacement curves and registered AE-time characteristics of the AE signals were compared with the SEM images of indentation. During the measurements, "pop-in" effects were observed as discontinuities of the loading curve, which correlate with AE impulses. A rise of the maximal load resulted in the increase of damage and caused multiplication of AE pulses. Due to the fact the DLC layer was thin, most AE pulses were generated during the damaging of Si substrate. The thin layer of DLC increased mechanical resistance to damage

    W pracy przedstawione są wyniki badań tribologicznych wykonanych na próbkach z warstwami o grubości 50 nm, 500 nm i 1300 nm nałożonymi na podłoże ze stali narzędziowej 316L. Warstwy nakładane były metodą RF PACVD. Testy tribologiczne realizowano w węźle tarcia typu kula–powierzchnia płaska z postępowo-zwrotnym ruchem ślizgowym kulki. Grubsze warstwy DLC wykazały stabilny i niski współczynnik tarcia. Ślady tarcia analizowano za pomocą elektronowego mikroskopu skaningowego oraz mikroskopu interferencyjnego. Najmniejsze zużycie zaobserwowano dla próbki z warstwą DLC o grubości 500 nm. Nawet w przypadku intensywnego uszkodzenia, warstwy DLC pełniły funkcję zabezpieczającą i zmniejszały zakres i zmieniały rodzaj zniszczeń.This paper presents the results of tribological tests made on the samples with a layer thickness of 50 nm, 500 nm and 1300 nm deposited on the surface of 316L tool steel. The DLC layers were made by the RF PACVD method. Tribological tests were carried out on the ball-flat surface friction node, with the reciprocating sliding movement of the ball. The thicker DLC layers showed a stable and low friction coefficient. Friction traces were analysed by scanning electron microscopy and interference microscopy. The lowest wear was observed for the DLC layer with thickness of 500 nm. Even in the case of heavy damage, the DLC layer acted as a protection and reduced the extend and changed the type of damage
