    É crescente a pressão para a publicação de artigos em periódicos de renome. Ressalta-se que os artigos publicados nesses periódicos são cada vez mais empíricos e com utilização de técnicas estatísticas progressivamente mais sofisticadas. Para isso, é importante que existam pesquisadores e programas capacitados para a análise quantitativa de dados. Neste trabalho, apresentam-se os resultados de um survey realizado com pesquisadores nacionais para verificar seu grau de proficiência em métodos estatísticos. Constatou-se que pesquisadores e programas brasileiros estão menos preparados para o uso desses métodos em relação a pesquisadores norte-americanos, conforme pesquisa usada como referência. Sugere-se não só a preparação de professores e alunos em métodos quantitativos, mas, sobretudo, a existência de estruturas de apoio que possam dar suporte para a melhoria de trabalhos desse tipo.En el ámbito académico hay una presión creciente para la publicación de artículos en revistas de reconocido prestigio. Se resalta que los artículos publicados en estos periódicos son cada vez más empíricos y se utilizan técnicas estadísticas progresivamente más sofisticadas. Por ello, es importante que haya investigadores y programas que sean capaces de analizar datos cuantitativos. En este trabajo se presentan los resultados de un estudio survey aplicado a investigadores brasileños con el fin de comprobar su nivel de competencia en métodos estadísticos. Se encontró que los investigadores y los programas brasileños están menos preparados para usar estos métodos que los investigadores estadounidenses, de acuerdo con un estudio de referencia. Se sugiere en este estudio que se realice la preparación adecuada de los docentes y estudiantes en métodos cuantitativos y que, sobre todo, se desarrollen estructuras de apoyo que puedan ayudar a mejorar dichos trabajos.There is a growing pressure for the publishing of articles in renowned periodicals. We reinforce that articles published in this kind of journals are becoming more empirical and are progressively using more sophisticated statistical techniques. This demands better works and more statistic data analysis papers. For this it is important that there are skilled researchers and programs for the quantitative data analysis. This work presents the results of a survey on Brazilian researchers to evaluate their proficiency in statistic research methods. We verified that our researchers and programs are less prepared to the use of these methods in relation to American researchers, according to a survey used as a reference. We suggest not only researchers and students training in quantitative methods, but also the availability of support structures to enable the improvement of the quantitative research

    Simulation of the scalar transport above and within the Amazon forest canopy

    The parallelized large-eddy simulation model (PALM) was used to understand better the turbulent exchanges of a passive scalar above and within a forested region located in the central Amazon. Weak (2 ms−1) and strong (6 ms−1) wind conditions were simulated. A passive scalar source was introduced to the forest floor for both simulations. The simulations reproduced the main characteristics of the turbulent flow and of the passive scalar transport between the forest and the atmosphere. Noteworthily, strong and weak wind conditions presented different turbulence structures that drove different patterns of scalar exchange both within and above the forest. These results show how passive scalar concentration is influenced by the wind speed at the canopy top. Additionally, higher wind speeds are related to stronger sweep and ejection regimes, generating more intense plumes that are able to reduce the passive scalar concentration inside the forest canopy.     The effect of the matrix system in the delivery and in-vitro bioactivity of microencapsulated oregano essential oil

    Microencapsulation allows bioactive compounds protection from external factors. Innovation in food industry often requires adding functional ingredients, to tailor flavour and texture, to improve preservation, to control bioactive compounds stability and controlled release during processing/storage. Oregano, besides richness in aroma compounds, is also known by potential antioxidant and antimicrobial activities. These sensitive compounds need protection in order to allow their use in a wider range of processes. In this study, oregano essential oil (EO) was microencapsulated by spray/freeze drying in: rice starch (with/without bonding agents), gelatine/sucrose and inulin, dried at different temperatures. Microencapsulates were analysed for morphology and structure (SEM, CLSM, X-ray diffraction and FTIR). Releasing ability of entrapped EO (UV–VIS spectroscopy) was evaluated by diffusion coefficient (D). Antioxidant activity (AA) - ORAC and HORAC- and antimicrobial activities against pathogens were evaluated. Rice starch spherules, presenting interconnecting cavities, were formed. Spray-dried inulin and gelatine/sucrose systems formed continuous walled and smooth surface spherical capsules (3-4.5 and 0.9-10m, respectively). EO was uniformly distributed inside the structures (CLSM) and its presence confirmed by FTIR. Depending on the system, D varied among 10-13 (starch), 10-13-10-15, (gelatine/sucrose) and 10-16 m2/s (inulin). In starch system, D was mainly influenced by the gelatin concentration, increasing with it. X- ray diffraction and FTIR results suggest some kind of linkage between gelatine and starch. Spray-dried gelatine/sucrose system, revealed to be unsuitable for EO encapsulation due to capsules disintegration but freeze-drying was effective. The D of EO from inulin capsules decreases when these are produced above 140 ºC. The impact of encapsulation method on EO bioactivity and product stability during 6 months, was verified through the determination of microcapsules AA, using free EO value as reference. The results obtained provide information on the release/stability of oregano EO from different matrices, relevant for functional ingredients microencapsulation

    The effect of encapsulating matrix on retention, protection and delivery of Oregano essential oil (EO) was studied. EO was encapsulated in rice starch porous spheres, inulin and gelatine/sucrose capsules by spray drying. Gelatine/sucrose matrix was also dried by freeze drying. Experimental designs were applied to test the effect of bonding agents and solids content for rice starch and drying temperature and solids content for inulin and gelatine/sucrose systems. The ratio of gelatine/sucrose was also tested. EO was identified (confocal laser scanning microscopy and FT-IR) in all tested matrices and the release profiles,antioxidant activity and antimicrobial activity of encapsulates evaluated. Results showed that the three tested materials are able to encapsulate Oregano EO. Higher diffusion coefficients were obtained for starch microcapsules (about 1013 m2/s) followed by spray-dried gelatine/sucrose systems (about 1015 m2/s) and inulin microcapsules (about 1016 m2/s).     Tuning the bioactivity of tensioactive deoxy glycosides to structure: antibacterial activity versus selective cholinesterase inhibition rationalized by molecular docking

    New octyl/dodecyl 2,6-dideoxy-D-arabino-hexopyranosides have been synthesized by a simple but efficient methodology based on the reaction of glycals with alcohols catalysed by triphenylphosphane hydrobromide, deprotection, regioselective tosylation and reduction. Their surface-active properties were evaluated in terms of adsorption and aggregation parameters and compared with those of 2-deoxy-D-glycosides and 2,6-dideoxy-L-glycosides. Deoxygenation at the 6-position led to a decrease in the critical micelle concentration, and an increase in the adsorption efficiency (pC20) promoting aggregation more efficiently than adsorption. With regard to the antibacterial activity, dodecyl 2,6-dideoxy-α-L-arabino-hexopyranoside was the most active compound towards Bacillus anthracis (MIC 25 μM), whereas its enantiomer exhibited a MIC value of 50 μM. Both 2,6-dideoxy glycosides were active towards Bacillus cereus, Bacillus subtilis, Enterococcus faecalis and Listeria monocytogenes. In contrast, none of the 2-deoxy glycosides was significantly active. These results and the data on surface activity suggest that aggregation is a key issue for antimicrobial activity. Beyond infection, Alzheimer’s disease also threatens elderly populations. In the search for butyrylcholinesterase (BChE) selective inhibition, 2- deoxy glycosides were screened in vitro by using Ellman’s assay. Octyl 2-deoxy-α-D-glycoside was found to be a BChE selective inhibitor promoting competitive inhibition. Docking studies supported these results as they pinpoint the importance of the primary OH group in stabilizing the BChE inhibitor complex. A size-exclusion mechanism for inhibition has been proposed based on the fact that acetylcholinesterase (AChE) exhibits several bulky residues that hinder access to the active-site cavity. This work shows how the deoxygenation pattern, configuration and functionality of the anomeric centre can tune physical and surface properties as well as the bioactivity of these multifunctional and stereochemically rich molecules.FEDER e FCTinfo:eu-repo/semantics/publishedVersio

    Influence of passage number on the impact of the secretome of adipose tissue stem cells on neural survival, neurodifferentiation and axonal growth

    Mesenchymal stem cells (MSCs), and within them adipose tissue derived stem cells (ASCs), have been shown to have therapeutic effects on central nervous system (CNS) cell populations. Such effects have been mostly attributed to soluble factors, as well as vesicles, present in their secretome. Yet, little is known about the impact that MSC passaging might have in the secretion therapeutic profile. Our aim was to show how human ASCs (hASCs) passage number influences the effect of their secretome in neuronal survival, differentiation and axonal growth. For this purpose, post-natal rat hippocampal primary cultures, human neural progenitor cell (hNPCs) cultures and dorsal root ganglia (DRGs) explants were incubated with secretome, collected as conditioned media (CM), obtained from hASCs in P3, P6, P9 and P12. Results showed no differences when comparing percentages of MAP-2 positive cells (a mature neuronal marker) in neuronal cultures or hNPCs, after incubation with hASCs secretome from different passages. The same was observed regarding DRG neurite outgrowth. In order to characterize the secretomes obtained from different passages, a proteomic analysis was performed, revealing that its composition did not vary significantly with passage number P3 to P12. Results allowed us to identify several key proteins, such as pigment epithelium derived factor (PEDF), DJ-1, interleucin-6 (IL-6) and galectin, all of which have already proven to play neuroprotective and neurodifferentiating roles. Proteins that promote neurite outgrowth were also found present, such as semaphorin 7A and glypican-1.     Evolution of electronic and ionic structure of Mg-clusters with the growth cluster size

    The optimized structure and electronic properties of neutral and singly charged magnesium clusters have been investigated using ab initio theoretical methods based on density-functional theory and systematic post-Hartree-Fock many-body perturbation theory accounting for all electrons in the system. We have systematically calculated the optimized geometries of neutral and singly charged magnesium clusters consisting of up to 21 atoms, electronic shell closures, binding energies per atom, ionization potentials and the gap between the highest occupied and the lowest unoccupied molecular orbitals. We have investigated the transition to the hcp structure and metallic evolution of the magnesium clusters, as well as the stability of linear chains and rings of magnesium atoms. The results obtained are compared with the available experimental data and the results of other theoretical works.Comment: 30 pages, 10 figures, 3 table
