    Non-destructive structural analysis of the heat conducting path in power electronics and solid state lighting by thermal transient testing

    Thermal transient testing is a tool for examining structural details of a heat conducting path composed of heat conduction and heat convection sections. Using the change of inherent heat in electronics, temperature transients provide a characterisation technique where using x-ray or acoustic microscopy would be troublesome and time-consuming. A complete toolkit for the accurate dynamic characterisation of subassemblies and cooling mounts is described in the paper. After giving the theoretical background, it is shown that the methodology can directly generate a valid and detailed structural equivalent from a single transient measurement. The descriptive thermal functions such as time constants and structure functions are demonstrated in practical examples, the capability of the structure functions for validating cooling concepts and detecting assembly problems is shown.     Photoreceptor Cell Death, Proliferation and Formation of Hybrid Rod/S-Cone Photoreceptors in the Degenerating STK38L Mutant Retina

    A homozygous mutation in STK38L in dogs impairs the late phase of photoreceptor development, and is followed by photoreceptor cell death (TUNEL) and proliferation (PCNA, PHH3) events that occur independently in different cells between 7–14 weeks of age. During this period, the outer nuclear layer (ONL) cell number is unchanged. The dividing cells are of photoreceptor origin, have rod opsin labeling, and do not label with markers specific for macrophages/microglia (CD18) or MĂŒller cells (glutamine synthetase, PAX6). Nestin labeling is absent from the ONL although it labels the peripheral retina and ciliary marginal zone equally in normals and mutants. Cell proliferation is associated with increased cyclin A1 and LATS1 mRNA expression, but CRX protein expression is unchanged. Coincident with photoreceptor proliferation is a change in the photoreceptor population. Prior to cell death the photoreceptor mosaic is composed of L/M- and S-cones, and rods. After proliferation, both cone types remain, but the majority of rods are now hybrid photoreceptors that express rod opsin and, to a lesser extent, cone S-opsin, and lack NR2E3 expression. The hybrid photoreceptors renew their outer segments diffusely, a characteristic of cones.     Environmental drivers of forest biodiversity in temperate mixed forests – A multi-taxon approach

    Harmonization of timber production and forest conservation is a major challenge of modern silviculture. For the establishment of ecologically sustainable forest management, the management-related environmental drivers of multi-taxon biodiversity should be explored. Our study reveals those environmental variables related to tree species diversity and composition, stand structure, litter and soil conditions, microclimate, landscape, and land-use history that determine species richness and composition of 11 forest-dwelling organism groups. Herbs, woody regeneration, ground-floor and epiphytic bryophytes, epiphytic lichens, terricolous saprotrophic, ectomycorrhizal, and wood-inhabiting macrofungi, spiders, carabid beetles, and birds were sampled in West Hungarian mature mixed forests. The correlations among the diversities and compositions of different organism groups were also evaluated. Drivers of organism groups were principally related to stand structure, tree species diversity and composition, and microclimate, while litter, soil, landscape, and land-use historical variables were less influential. The complex roles of the shrub layer, deadwood, and the size of the trees in determining the diversity and composition of various taxa were revealed. Stands with more tree species sustained higher stand-level species richness of several taxa. Besides, stands with different dominant tree species harbored various species communities of organism groups. Therefore, landscape-scale diversity of dominant tree species may enhance the diversity of forest-dwelling communities at landscape level. The effects of the overstory layer on forest biodiversity manifested in many cases via microclimate conditions. Diversity of organism groups showed weaker relationship with the diversity of other taxa than with environmental variables. According to our results, the most influential drivers of forest biodiversity are under the direct control of the actual silvicultural management. Heterogeneous stand structure and tree species composition promote the different organism groups in various ways. Therefore, the long-term maintenance of the structural and compositional heterogeneity both at stand and landscape scale is an important aspect of ecologically sustainable forest management

    Recommendations of the International Society of Intraoperative Neurophysiology for intraoperative somatosensory evoked potentials

    Intraoperative somatosensory evoked potentials (SEPs) provide dorsal somatosensory system functional and localizing information, and complement motor evoked potentials. Correct application and interpretation require in-depth knowledge of relevant anatomy, electrophysiology, and techniques. It is advisable to facilitate cortical SEPs with total intravenous propofol-opioid or similarly favorable anesthesia. Moreover, SEP optimization is recommended to enhance surgical feedback speed and accuracy by maximizing signal-to-noise ratio (SNR); it consists of selecting highest-SNR peripheral and cortical derivations while omitting low-SNR channels. Confounding factors causing non-surgical SEP reduction should be excluded before issuing a warning. It is advisable to facilitate their identification with peripheral SEP controls and cortical SEP systemic controls whenever possible. Warning criteria should adjust for baseline drift and reproducibility. The recommended adaptive warning criterion is visually obvious amplitude reduction from recent pre-change values and clearly exceeding trial-to-trial variability, particularly when abrupt and focal. Acquisition and interpretation should be done by qualified technical and professional level personnel. Indications for SEP monitoring include intracranial, posterior fossa, and spinal neurosurgery, as well as orthopedic spine, cerebrovascular, and descending aortic surgery. Indications for SEP mapping include sensorimotor cortex and dorsal column midline identification. Future advances could modify current recommendations

    Isolation of chick retina cones and study of their diversity based on oil droplet colour and nucleus position

    The chick retina has four morphological cone types that differ not only in shape, but also in the visual pigment in the outer segment, in the colour of the oil droplet in the inner segment and in synaptic connectivity. Neither the type of droplet nor the visual pigment has been definitively established for the four cone types. The main aim of the present work has been the isolation of entire live photoreceptors in order to study the oil droplet colour in each cone type and to quantify each type. We have improved an earlier retinal cell isolation method and obtained large numbers of entire cones. Principal cones (27% of the cones) possess a yellow or colourless droplet. Accessory cones (27% of the cones) all contain a small pale green droplet. Straight cones (44% of the cones) have a red, orange, yellow, or colourless droplet. Oblique cones (1.66% of the cones) all have a colourless droplet. We have found that straight cones with a red, orange, or yellow droplet differ in terms of the position of the nucleus and their percentage and conclude that they are distributed in three rows in the outer nuclear layer (ONL) of the central retina. Our study of 4,6-diamidino-2-phenylindole-stained retinal sections has revealed three rows of nuclei instead of the two currently thought to form the ONL. Together, our results show a larger cone diversity than previously known, suggest a larger functional diversity and provide an efficient method for isolating entire chick photoreceptors

    Cone pigments in a North American marsupial, the opossum (Didelphis virginiana)

    Only two of the four cone opsin gene families found in vertebrates are represented in contemporary eutherian and marsupial species. Recent genetic studies of two species of South American marsupial detected the presence of representatives from two of the classes of cone opsin genes and the structures of these genes predicted cone pigments with respective peaks in the ultraviolet and long-wavelength portions of the spectrum. The Virginia opossum (Didelphis virginiana), a profoundly nocturnal animal, is the only marsupial species found in North America. The prospects for cone-based vision in this species were examined through recordings of the electroretinogram (ERG), a commonly examined retinal response to photic stimulation. Recorded under flickering-light conditions that elicit signals from cone photoreceptors, the spectral sensitivity of the opossum eye is well accounted for by contributions from the presence of a single cone pigment having peak absorption at 561–562 nm. A series of additional experiments that employed various chromatic adaptation paradigms were conducted in a search for possible contributions from a second (short-wavelength sensitive) cone pigment. We found no evidence that such a mechanism contributes to the ERG in this marsupial

    Pan-retinal characterisation of Light Responses from Ganglion Cells in the Developing Mouse Retina

    International audienceWe have investigated the ontogeny of light-driven responses in mouse retinal ganglion cells (RGCs). Using a large-scale, high-density multielectrode array, we recorded from hundreds to thousands of RGCs simultaneously at pan-retinal level, including dorsal and ventral locations. Responses to di erent contrasts not only revealed a complex developmental pro le for ON, OFF and ON-OFF responses, but also unveiled di erences between dorsal and ventral RGC responses. At eye-opening, dorsal RGCs of all types were more responsive to light, perhaps indicating an environmental priority to nest viewing for pre-weaning pups. The developmental pro le of ON and OFF responses exhibited antagonistic behaviour, with the strongest ON responses shortly after eye-opening, followed by an increase in the strength of OFF responses later on. Further, we found that with maturation receptive eld (RF) center sizes decrease, spike-triggered averaged responses to white noise become stronger, and centers become more circular while maintaining di erences between RGC types. We conclude that the maturation of retinal functionality is not spatially homogeneous, likely re ecting ecological requirements that favour earlier maturation of the dorsal retina
