    Non-invasive intestinal biomarkers: a new ELISA test for Pancreatitis Associated Protein detection in pig

    Feed additives are commonly used to improve pig performance and health, but they need to be tested so new biomarkers for intestinal health, non- or minimally invasive, are under investigations.The quantification of Myeloperoxidase (MPO) and Pancreatitis Associated Protein (PAP) in feces could prove useful to non-invasively monitor intestinal health (Niewold, 2015). MPO is an enzyme that permits to quantify the number of inflammatory cells present in tissues and feces (Prokopowicz et al., 2012) , while PAP is a protein mainly produced in the small intestine with anti-inflammatory and bactericidal activity (Cash et al., 2006; Mukherjee et al., 2014). Because of the lack of a commercial ELISA kit for porcine PAP detection, the main aim of this study was to develop and validate a new sandwich ELISA test for the quantification of PAP in pig fecal samples. Our study consisted of two phases: test development and test validation. During the development phase we used polyclonal antibodies previously immunized from rabbit serum with a pure peptide containing the N-terminus of pig PAP (Soler et al., 2015). The validation of the test was then performed with fecal extraction samples derived from animals with known high or low growth performance.Moreover, the temperature stability of PAP in feces and the optimal extraction method was tested. Even if only preliminary, our results seem to show a fair relationship between fecal consistency, used as health indicator, and PAP fecal concentrations. Furthermore, no relevant differences in PAP concentration after 24h of incubation at 37 °C, 4°C or room temperature were detected.To date, the present results suggest that PAP seems to be exceptionally stable in feces and is a very promising candidate as a non-invasive (fecal) biomarker for intestinal health and growth

    Effects of Fat Supplementation in Dairy Goats on Lipid Metabolism and Health Status

    Fat supplementation has long been used in dairy ruminant nutrition to increase the fat content of milk and supply energy during particularly challenging production phases. Throughout the years, advances have been made in the knowledge of metabolic pathways and technological treatments of dietary fatty acids (FAs), resulting in safer and more widely available lipid supplements. There is an awareness of the positive nutraceutical effects of the addition of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) to fat supplementation, which provides consumers with healthier animal products through manipulation of their characteristics. If it is true that benefits to human health can be derived from the consumption of animal products rich in bioactive fatty acids (FAs), then it is reasonable to think that the same effect can occur in the animals to which the supplements are administered. Therefore, recent advances in fat supplementation of dairy goats with reference to the effect on health status have been summarized. In vivo trials and in vitro analysis on cultured cells, as well as histological and transcriptomic analyses of hepatic and adipose tissue, have been reviewed in order to assess documented relationships between specific FAs, lipid metabolism, and immunity

    Domain size effects on the dynamics of a charge density wave in 1T-TaS2

    Recent experiments have shown that the high temperature incommensurate (I) charge density wave (CDW) phase of 1T-TaS2 can be photoinduced from the lower temperature, nearly commensurate (NC) CDW state. Here we report a time-resolved x-ray diffraction study of the growth process of the photoinduced I-CDW domains. The layered nature of the material results in a marked anisotropy in the size of the photoinduced domains of the I-phase. These are found to grow self-similarly, their shape remaining unchanged throughout the growth process. The photoinduced dynamics of the newly formed I-CDW phase was probed at various stages of the growth process using a double pump scheme, where a first pump creates I-CDW domains and a second pump excites the newly formed I-CDW state. We observe larger magnitudes of the coherently excited I-CDW amplitude mode in smaller domains, which suggests that the incommensurate lattice distortion is less stable for smaller domain sizes.Comment: 8 pages, 8 figure

    Selection of new markers for animal by-products characterization by classical microscopy

    The aim of this study was to identify possible markers to distinguish differences between land animals by using the microscopic method in association with computer image analysis. For this purpose bone fragments from poultry and mammals were obtained and analysed by microscopic method. Through a digital camera and an image analysis software 85 bone lacunae images have been processed and elaborated in order to obtain for each lacuna a monochrome mask on which several measurements were performed. Data were analysed by ANOVA and LDA. Results obtained in the present study indicated that of 32 descriptors processed by image analysis software, only 12 were significantly (P<0.001) different between mammalian and poultry. However, when morphometric measurements were analysed by LDA, 86% of lacunae were correctly classified into the animal class of origin (i.e. mammalian as mammalian and poultry as poultry). By contrast 14% of lacunae were incorrectly classified. In conclusion, data here presented indicate that some of descriptors used by image analysis appears promising not only for a reliable distinction between the different origins of animal meal at the level of vertebrate classes, but also for further characterisation and identification of processed animal proteins in animal feeds

    Green tea and pomegranate extract administered during critical moments of the production cycle improves blood antiradical activity and alters cecal microbial ecology of broiler chickens

    Phytobiotics are usually tested in feed and throughout the production cycle. However, it could be beneficial to evaluate their effects when administered only during critical moments, such as changes in feeding phases. The aim of the trial was to investigate the effect of a commercial plant extract (PE; IQV-10-P01, InQpharm Animal Health, Kuala Lumpur, Malaysia) on growth performance, blood antiradical activity and cecal microbiome when administered in drinking water to broiler chickens during the post-hatching phase and at each change of diet. In the experiment, 480 1-day-old male broiler chicks were assigned to two groups in a 50-day trial. Broilers received drinking water (C) or drinking water plus PE (T) at a rate of 2 mL/L on days 0 to 4, 10\u201311 and 20\u2013 21. PE did not affect performance and water intake, while total antiradical activity was improved (p &lt; 0.05). A greater abundance of lactic acid bacteria (false discovery rate (FDR) &lt; 0.05) was found in the T group and the result was confirmed at a lower taxonomic level with higher Lactobacillaceae abundance (FDR &lt; 0.05). Our findings suggest that PE administration during critical moments of the production cycle of broiler chickens may exert beneficial effects at a systemic level and on gut microbial ecology

    Feed particle size evaluation: conventional approach versus digital holography based image analysis

    The aim of this study was to evaluate the application of image analysis approach based on digital holography in defining particle size in comparison with the sieve shaker method (sieving method) as reference method. For this purpose ground corn meal was analyzed by a sieve shaker Retsch VS 1000 and by image analysis approach based on digital holography. Particle size from digital holography were compared with results obtained by screen (sieving) analysis for each of size classes by a cumulative distribution plot. Comparison between particle size values obtained by sieving method and image analysis indicated that values were comparable in term of particle size information, introducing a potential application for digital holography and image analysis in feed industry

    Effects of nucleotides administration on growth performance and immune response of post-weaning piglets

    The aim of this study was to assess the effect of nucleotides administration on growth perform- ance and immune response in post-weaning piglets. Twenty-eight male weaned piglets, homo- geneous for age and weight were randomly allocated to two experimental treatments. Treated group (T) was daily orally administered 0.8g/head of a mixture of nucleotides suspended in 2.1 mL water solution; while control group (C) received 2.1 mL saline solution. Body weight (BW) and average daily gain (ADG) were individually recorded weekly, while feed intake (FI), and gain:feed (G:F) were recorded and calculated on pen basis. Faecal score was evaluated every seven days. On day 0, 9, 18 and 27 blood samples were collected to determine IgA, IgG and haptoglobin concentration. At day 28 all piglets were sacrificed, and tissue samples of ileal Peyer\u2019s patches were collected for the evaluation of IL1a, IL1b, IL6, IL10, TNFa, TLR2, TLR4 and PPARc gene expression. Nucleotides supplementation significantly increased BW (17.37 vs. 19.00kg/pig; p 1\u20444 &lt;.01), ADG (.351 vs. .400kg/d; p &lt; .01), and FI (3.96 vs. 4.39kg/d; p &lt; .01), but not G:F (.61 vs. .64; p 1\u20444 .29). Faecal consistency was not different between the experimental groups and no occurrence of diarrhoea was reported. IgA and IgG content in blood was not influenced by the treatment, as well as gene expression of inflammatory cytokines in Peyer\u2019s patches. The present trial shows that nucleotide administration is able to improve growth per- formance of post-weaning piglets, with no effects on inflammatory response and the expression of immune-related genes

    Microstructural characterization of alumina-zirconia layered ceramics using positron annihilation spectroscopy

    Positron annihilation spectroscopy (PAS), indentation, nanoindentation experiments and scanning electron microscopy (SEM) observations were performed on Al₂O₃-ZrO₂ laminates samples to assess the effect of residual stresses on their mechanical and microstructural properties. Layered samples were implemented by slip-casting, constituted by two thin Al₂O₃ external layers and an intermediate thick one, consisting of a mixture of Al₂O₃ and monoclinic ZrO₂ in the range 0-30 vol.%. In these systems residual tensile stresses fields were generated inside the external layers during cooling from the sintering temperature, by the expansion of the adjacent ZrO₂-containing layer. SEM observations showed the microstructural effects due to the level of tension related to the zirconia content. A correlation between the PAS parameters and the microstructural changes caused by the presence of residual stresses was found. Nanoindentation measurements were used to trace the sign and magnitude of the residual stress gradient across the interface between the layers.This work was supported by Spanish Government under Contract MAT2006-01038.     Watching the birth of a charge density wave order: diffraction study on nanometer-and picosecond-scales

    Femtosecond time-resolved X-ray diffraction is used to study a photo-induced phase transition between two charge density wave (CDW) states in 1T-TaS2_2, namely the nearly commensurate (NC) and the incommensurate (I) CDW states. Structural modulations associated with the NC-CDW order are found to disappear within 400 fs. The photo-induced I-CDW phase then develops through a nucleation/growth process which ends 100 ps after laser excitation. We demonstrate that the newly formed I-CDW phase is fragmented into several nanometric domains that are growing through a coarsening process. The coarsening dynamics is found to follow the universal Lifshitz-Allen-Cahn growth law, which describes the ordering kinetics in systems exhibiting a non-conservative order parameter.Comment: 6 pages, 5 figure

    SANS evidence for the dispersion of nanoparticles in W-1Y₂O₃ and W-1La₂O₃ processed by hot isostatic pressing

    The development of a dispersion of nanoparticles in the W–1%Y₂O₃ and W–1%La₂O₃ (wt%) alloys processed by hot isostatic pressing have been investigated using small-angle neutron scattering. The analyses of the scattering data using the Beaucage unified approach reveal the presence of a bi-modal distribution of spherical scattering centers with sizes of less of 180 nm in these alloys. The mode values of these centers are found at ~ 10 and 40 nm in W–1%Y₂O₃, and at ~ 15 and 80 nm in W–1%La₂O₃ . The scanning electron microscopy analyses showed the presence of small second phase particles. The contribution of the pore space to the scattering curves has been analyzed using the results obtained for pure W processed following the same procedure used for the alloys, and the porosity measurements of the samples.The present work was supported by the Consejería de Educación de la Comunidad de Madrid, through the program ESTRUMAT CM S2009MAT 1585 and by the Ministerio de Innovación y Ciencia (project ENE2008 06403 C06 04).
