    Abstract This thesis was titled "the performance of the teacher in the learning process in a sociology class XI IPS 1 SMA Negeri 1 Sambas". As for the problems in this study i.e. how the performance of the teacher sociology study in planning, implementing and evaluating learning outcomes learning in class XI IPS 1 SMA Negeri 1 Sambas. This research uses qualitative, descriptive methods in the form. Data collection tools using the guidelines of observation, interview and documentation study guidelines. The research results indicate (1) the teacher of Sociology have made the planning of learning in class XI IPS 1 SMA Negeri 1 Sambas.this is proven teacher already mention the learning objectives in the RPP; compile learning materials in a sequence, contextual and logical; planning effective learning activities; and select a learning resource or learning media in accordance with the materials and methods of learning. (2) Teacher of Sociology had done the learning process in class XI IPS 1 SMA Negeri 1 Sambas. This is proven teachers already start learning with effective, mastering subject matter, applying an approach or an effective learning methods, utilizing learning resources or media in learning, motivating and nurturing engagement students in learning, using language that is right and proper in learning, ending with effective learning. (3) Teacher of Sociology has been conducting student assessment in class XI IPS 1 SMA Negeri 1 Sambas.     Improving Students\u27 Reading Comprehension on Narrative Text by Using Story Map Technique

    : This thesis is a study on improving students\u27 reading comprehension in narrative text by using Story Map Technique to the tenth grade students of SMA Negeri 10 Pontianak in academic year 2014/2015. The writer took a classroom action research and the teaching learning process took place in two cycles. The subjects of this research were the students of grade X IPA 3 which consist of 39 students. From the observation checklist and field notes, it was found that all the students participated to the activity and did their task well. The students gave positive response to all the explanation and could use the story map technique effectively. For these results, it was concluded that story map technique improved students\u27 reading comprehension skill in narrative text in their Story Map Technique enabled the students to store information in their personal schema more efficiently and facilitated to recall the story elements more completely and accurately

    Розробка та валідація методики визначення супровідних домішок у таблетках рибоксин

    Riboxin (inosine) is used in medicine for treating cardiovascular diseases, so the number of dosage forms containing this substance as an active ingredient is constantly increasing at the Ukrainian pharmaceutical market. When developing quality control methods for a dosage form containing one active substance it is advisable to carry out tests on the presence of related impurities besides identification and assay tests. The article presents the data concerning development of the method for determination of related impurities (hypoxanthine, guanosine) in riboxine tablets using HPLC. The analysis was carried out using a HPLC column (125×4 mm i.d., 5 μm particles) filled with “Lichrospher 60 RP select B”sorbent. The mobile phase adjusted with the phosphate buffer to pH 3.5 was used. The UV detection was performed at 250 nm. The validation characteristics have been studied using the acceptance criteria for the tolerances of the content not more than 0.5% for each impurity, and they confirm specificity (the absence of the effect of excipients), linearity, precision (convergence), accuracy (Δz = 0.79≤max Δz = 5.0, δ = 0.21≤max δ = 0.26, a = 0.48, r = 0.99997>0.9976 for hypoxanthine, and Δz = 0.83≤max Δz = 5.0, δ = 0.17≤max δ = 0.28, a = 0.21, r = 0.99997>0.9976 for guanosine), and the application range for the method proposed.Рибоксин (инозин) применяют в медицинской практике для лечения сердечно-сосудистых заболеваний, поэтому количество готовых лекарственных форм, содержащих в качестве активного фармацевтического ингредиента именно это вещество, на фармацевтическом рынке Украины постоянно растет. При разработке методик контроля качества на готовую лекарственную форму, содержащую одно действующее вещество, целесообразно кроме испытаний на идентификацию и количественное определение также проводить испытания на содержание сопутствующих примесей. В предлагаемой работе приводятся данные по разработке методики определения сопутствующих примесей (гипоксантин, гуанозин) рибоксина в таблетках с использованием метода высокоэффективной жидкостной хроматографии. Определение проводили на колонке размером 125×4 мм, заполненной сорбентом «Lichrospher 60 RP select B» с размером частиц 5 мкм, использовали подвижную фазу на основе фосфатного буферного раствора с рН 3,5 и УФ-детектирование при длине волны 250 нм. Изученные валидационные характеристики с использованием критериев приемлемости для допусков содержания не более 0,5% для каждой из примесей подтверждают специфичность (отсутствие влияния вспомогательных веществ), линейность, прецизионность (сходимость), правильность (Δz = 0.79≤max Δz = 5.0, δ = 0.21≤max δ = 0.26, a = 0.48, r = 0.99997 гипоксантина и Δz = 0.83≤max Δz = 5.0, δ = 0.17≤max δ = 0.28, a = 0.21, r = 0.99997 гуанозина) и диапазон применения предлагаемой методики.Рибоксин (інозин) застосовують у медичній практиці для лікування серцево-судинних захворювань, тому кількість готових лікарських форм, що містять як активний фармацевтичний інгредієнт саме цю речовину, на фармацевтичному ринку України постійно зростає. При розробці методик контролю якості на готову лікарську форму, що містить одну діючу речовину, доцільно окрім випробувань на ідентифікацію та кількісне визначення також проводити випробування на вміст супутніх домішок. У запропонованій роботі наводяться дані по розробці методики визначення супровідних домішок (гіпоксантин, гуанозин) рибоксину в таблетках з використанням методу високоефективної рідинної хроматографії. Визначення проводили на колонці розміром 125×4 мм, заповненій сорбентом «Lichrospher 60 RP select B» з розміром часток 5 мкм, використовували рухому фазу на основі фосфатного буферного розчину з рН 3,5 та УФ-детектування за довжини хвилі 250 нм. Вивчені валідаційні характеристики з використанням критеріїв прийнятності для допусків вмісту не більше 0,5% для кожної з домішок підтверджують специфічність (відсутність впливу допоміжних речовин), лінійність, прецизійність (збіжність), правильність (Δz = 0.79≤max Δz = 5.0, δ = 0.21≤max δ = 0.26, a = 0.48, r = 0.99997>0.9976 гіпоксантину та Δz = 0.83≤max Δz = 5.0, δ = 0.17≤max δ = 0.28, a = 0.21, r = 0.99997>0.9976 гуанозину) та діапазон застосування запропонованої методики

    Tissue distribution of a plasmid DNA encoding Hsp65 gene is dependent on the dose administered through intramuscular delivery

    In order to assess a new strategy of DNA vaccine for a more complete understanding of its action in immune response, it is important to determine the in vivo biodistribution fate and antigen expression. In previous studies, our group focused on the prophylactic and therapeutic use of a plasmid DNA encoding the Mycobacterium leprae 65-kDa heat shock protein (Hsp65) and achieved an efficient immune response induction as well as protection against virulent M. tuberculosis challenge. In the present study, we examined in vivo tissue distribution of naked DNA-Hsp65 vaccine, the Hsp65 message, genome integration and methylation status of plasmid DNA. The DNA-Hsp65 was detectable in several tissue types, indicating that DNA-Hsp65 disseminates widely throughout the body. The biodistribution was dose-dependent. In contrast, RT-PCR detected the Hsp65 message for at least 15 days in muscle or liver tissue from immunized mice. We also analyzed the methylation status and integration of the injected plasmid DNA into the host cellular genome. The bacterial methylation pattern persisted for at least 6 months, indicating that the plasmid DNA-Hsp65 does not replicate in mammalian tissue, and Southern blot analysis showed that plasmid DNA was not integrated. These results have important implications for the use of DNA-Hsp65 vaccine in a clinical setting and open new perspectives for DNA vaccines and new considerations about the inoculation site and delivery system

    Validity of the Empatica E4 wristband to measure heart rate variability (HRV) parameters:A comparison to electrocardiography (ECG)

    Wearable monitoring devices are an innovative way to measure heart rate (HR) and heart rate variability (HRV), however, there is still debate about the validity of these wearables. This study aimed to validate the accuracy and predictive value of the Empatica E4 wristband against the VU University Ambulatory Monitoring System (VU-AMS) in a clinical population of traumatized adolescents in residential care. A sample of 345 recordings of both the Empatica E4 wristband and the VU-AMS was derived from a feasibility study that included fifteen participants. They wore both devices during two experimental testing and twelve intervention sessions. We used correlations, cross-correlations, Mann-Whitney tests, difference factors, Bland-Altman plots, and Limits of Agreement to evaluate differences in outcomes between devices. Significant correlations were found between Empatica E4 and VU-AMS recordings for HR, SDNN, RMSSD, and HF recordings. There was a significant difference between the devices for all parameters but HR, although effect sizes were small for SDNN, LF, and HF. For all parameters but RMSSD, testing outcomes of the two devices led to the same conclusions regarding significance. The Empatica E4 wristband provides a new opportunity to measure HRV in an unobtrusive way. Results of this study indicate the potential of the Empatica E4 as a practical and valid tool for research on HR and HRV under non-movement conditions. While more research needs to be conducted, this study could be considered as a first step to support the use of HRV recordings provided by wearables

    The paper presents the results of the ongoing research coordinated by the Chair of Ancient Topography at the University of Padova. The work concerns the medieval site of “Mura della Bastia” in Onigo di Pederobba (Treviso), in the North-Eastern part of Italy. Historical information from written documents and results obtained from the archaeological investigation at the Onigo castle (1994-2007) are discussed. The study provided the historical reconstruction of recent collapses of the Onigo tower and allowed the virtual reconstruction of still-cohesive blocks, which had fallen down in 1989, and which were recovered during the summer of 2007. At this time, five of the ten building blocks have been positioned; the procedure was carried out by comparing the panoply of each agglomerate with the prospect of the tower, which was obtained from the photogrammetric rectification of the historical images (2D). The 3D survey of the tower and of nine blocks was performed with a Leica HDS 2500 laser scanner. The resulting point clouds were aligned with Cyclone software, also using the coordinates of some laser scanning targets, which were measured with the Leica TC 2003 total station in a local reference system. The final 3D model obtained by the co-registration of the scans was textured with the digital images acquired with a Canon EOS 1 ds Mark II camera. Subsequently, three of the five recognized blocks were positioned on the 3D model using Reconstructor software

    Serum Amyloid P Therapeutically Attenuates Murine Bleomycin-Induced Pulmonary Fibrosis via Its Effects on Macrophages

    Macrophages promote tissue remodeling but few mechanisms exist to modulate their activity during tissue fibrosis. Serum amyloid P (SAP), a member of the pentraxin family of proteins, signals through Fcγ receptors which are known to affect macrophage activation. We determined that IPF/UIP patients have increased protein levels of several alternatively activated pro-fibrotic (M2) macrophage-associated proteins in the lung and monocytes from these patients show skewing towards an M2 macrophage phenotype. SAP therapeutically inhibits established bleomycin-induced pulmonary fibrosis, when administered systemically or locally to the lungs. The reduction in aberrant collagen deposition was associated with a reduction in M2 macrophages in the lung and increased IP10/CXCL10. These data highlight the role of macrophages in fibrotic lung disease, and demonstrate a therapeutic action of SAP on macrophages which may extend to many fibrotic indications caused by over-exuberant pro-fibrotic macrophage responses

Així mateix, durant l’elaboració d’aquest treball, des de l’ASPCAT (ACSA i Sub-direcció General de Seguretat Alimentària i Protecció de la Salut) hem rebut propostes de noves vies de difusió del nostre model de PNT, tant als OE com als SVOE (serveis veterinaris oficials d’escorxador) perquè en tinguin coneixement, i també la possibilitat que se’n derivi un document de format més breu i pedagògic (Codi de bones pràctiques).En el anterior trabajo de la CoP de bienestar animal (Modelo de PNT de bienestar animal para pequeños mataderos de ungulados y solípedos domésticos, 2017) elaboramos un modelo de procedimiento normalizado de trabajo (PNT) para pequeños mataderos de ungulados y solípedos domésticos. Este procedimiento estaba dirigido al operador económico (OE); así, este año, hemos querido comprobar su encaje. Asimismo, durante la elaboración de este trabajo, desde la ASPCAT (ACSA y Subdirección General de Seguridad Alimentaria y Protección de la Salud) hemos recibido propuestas de nuevas vías de difusión de nuestro modelo de PNT, tanto en los OE como a los SVOE (servicios veterinarios oficiales de matadero) para su conocimiento, así como la posibilidad de que se derive un documento de formato más breve y pedagógico (Código de buenas prácticas).In the previous work of the Animal Welfare CoP (Animal Welfare SOP Model for Small Slaughterhouses of Domestic Ungulates and Solipeds, 2017) we developed a standard operating procedure (SOP) model for small abattoirs of domestic ungulates and solipeds.     Targeting of human interleukin-12B by small hairpin RNAs in xenografted psoriatic skin

    <p>Abstract</p> <p>Background</p> <p>Psoriasis is a chronic inflammatory skin disorder that shows as erythematous and scaly lesions. The pathogenesis of psoriasis is driven by a dysregulation of the immune system which leads to an altered cytokine production. Proinflammatory cytokines that are up-regulated in psoriasis include tumor necrosis factor alpha (TNFα), interleukin-12 (IL-12), and IL-23 for which monoclonal antibodies have already been approved for clinical use. We have previously documented the therapeutic applicability of targeting TNFα mRNA for RNA interference-mediated down-regulation by anti-TNFα small hairpin RNAs (shRNAs) delivered by lentiviral vectors to xenografted psoriatic skin. The present report aims at targeting mRNA encoding the shared p40 subunit (IL-12B) of IL-12 and IL-23 by cellular transduction with lentiviral vectors encoding anti-IL12B shRNAs.</p> <p>Methods</p> <p>Effective anti-IL12B shRNAs are identified among a panel of shRNAs by potency measurements in cultured cells. The efficiency and persistency of lentiviral gene delivery to xenografted human skin are investigated by bioluminescence analysis of skin treated with lentiviral vectors encoding the luciferase gene. shRNA-expressing lentiviral vectors are intradermally injected in xenografted psoriatic skin and the effects of the treatment evaluated by clinical psoriasis scoring, by measurements of epidermal thickness, and IL-12B mRNA levels.</p> <p>Results</p> <p>Potent and persistent transgene expression following a single intradermal injection of lentiviral vectors in xenografted human skin is reported. Stable IL-12B mRNA knockdown and reduced epidermal thickness are achieved three weeks after treatment of xenografted psoriatic skin with lentivirus-encoded anti-IL12B shRNAs. These findings mimick the results obtained with anti-TNFα shRNAs but, in contrast to anti-TNFα treatment, anti-IL12B shRNAs do not ameliorate the psoriatic phenotype as evaluated by semi-quantitative clinical scoring and by immunohistological examination.</p> <p>Conclusions</p> <p>Our studies consolidate the properties of lentiviral vectors as a tool for potent gene delivery and for evaluation of mRNA targets for anti-inflammatory therapy. However, in contrast to local anti-TNFα treatment, the therapeutic potential of targeting IL-12B at the RNA level in psoriasis is questioned.</p
