    The purpose of this investigation was to understand a teacher\u27s response to boys and girls in the classroom and determine whether this has an effect on students\u27 perceptions of gender. Using a qualitative study and analyzing interviews, observations, and a gender activity through a data analysis spiral (Creswell 1998) in a first grade classroom with a female teacher, I discovered a teacher\u27s perception of gender influences her interactions with students. Through the gender activity, I found this interaction can help shape a student\u27s perception of gender

    Can Archival Tissue Reveal Answers to Modern Research Questions?: Computer-Aided Histological Assessment of Neuroblastoma Tumours Collected over 60 Years.

    Despite neuroblastoma being the most common extracranial solid cancer in childhood, it is still a rare disease. Consequently, the unavailability of tissue for research limits the statistical power of studies. Pathology archives are possible sources of rare tissue, which, if proven to remain consistent over time, could prove useful to research of rare disease types. We applied immunohistochemistry to investigate whether long term storage caused any changes to antigens used diagnostically for neuroblastoma. We constructed and quantitatively assessed a tissue microarray containing neuroblastoma archival material dating between 1950 and 2007. A total of 119 neuroblastoma tissue cores were included spanning 6 decades. Fourteen antibodies were screened across the tissue microarray (TMA). These included seven positive neuroblastoma diagnosis markers (NB84, Chromogranin A, NSE, Ki-67, INI1, Neurofilament Protein, Synaptophysin), two anticipated to be negative (S100A, CD99), and five research antibodies (IL-7, IL-7R, JAK1, JAK3, STAT5). The staining of these antibodies was evaluated using Aperio ImageScope software along with novel pattern recognition and quantification algorithms. This analysis demonstrated that marker signal intensity did not decrease over time and that storage for 60 years had little effect on antigenicity.     Distributions of Sharks across a Continental Shelf in the Northern Gulf of Mexico

    Declines in shark populations have sparked researchers and fishery managers to investigate more prudent approaches to the conservation of these fish. As managers strive to improve data collection for stock assessment, fisheries-independent surveys have expanded to include data-deficient areas such as coastal regions. To that end, a catch series from a nearshore survey off Alabama was combined with data from a concurrent offshore survey with identical methodology to examine the depth use of sharks across the continental shelf (2ā€“366 m). The combined data set contained 22 species of sharks collected from 1995 to 2008: 21 species in the offshore data set (1995ā€“2008) and 12 species in the nearshore data set (2006ā€“2008). Depth was a significant factor determining speciesā€™ distributions, primarily for Atlantic sharpnose Rhizoprionodon terraenovae, blacknose Carcharhinus acronotus, and blacktip C. limbatus sharks. Blacknose sharks had the highest catch per unit effort (CPUE) in the middepth stratum (10ā€“30 m), blacktip sharks had consistently higher CPUE in the shallow depth stratum (,10 m), and Atlantic sharpnose sharks showed high abundance throughout both the shallow and mid-depth strata. Length frequency and sex ratio analyses suggest that Atlantic sharpnose and blacknose sharks are using waters greater than 30 m deep for parturition, whereas adult blacktip sharks are probably using shallow waters for parturition. Our abundance patterns illustrate a continuum of depth use across the inner continental shelf. Surveys that do not encompass the entirety of this ecosystem fail to accurately characterize the distributions of these important predators

    Rapid desensitization through immunoadsorption during cardiopulmonary bypass. A novel method to facilitate human leukocyte antigen incompatible heart transplantation

    Background: Anti-human leukocyte antigen (HLA)-antibody production represents a major barrier to heart transplantation, limiting recipient compatibility with potential donors and increasing the risk of complications with poor waiting-list outcomes. Currently there is no consensus to when desensitization should take place, and through what mechanism, meaning that sensitized patients must wait for a compatible donor for many months, if not years. We aimed to determine if intraoperative immunoadsorption could provide a potential desensitization methodology. Methods: Anti-HLA antibody-containing whole blood was added to a Cardiopulmonary bypass (CPB) circuit set up to mimic a 20Ā kg patient undergoing heart transplantation. Plasma was separated and diverted to a standalone, secondary immunoadsorption system, with antibody-depleted plasma returned to the CPB circuit. Samples for anti-HLA antibody definition were taken at baseline, when combined with the CPB prime (on bypass), and then every 20Ā min for the duration of treatment (total 180Ā min). Results: A reduction in individual allele median fluorescence intensity (MFI) to below clinically relevant levels (4000 was demonstrated. Reduction occurred in all cases within 120Ā min, demonstrating efficacy in a time period usual for heart transplantation. Flowcytometric crossmatching of suitable pseudo-donor lymphocytes demonstrated a change from T cell and B cell positive channel shifts to negative, demonstrating a reduction in binding capacity. Conclusions: Intraoperative immunoadsorption in an ex vivo setting demonstrates clinically relevant reductions in anti-HLA antibodies within the normal timeframe for heart transplantation. This method represents a potential desensitization technique that could enable sensitized children to accept a donor organ earlier, even in the presence of donor-specific anti-HLA antibodies

    Questioning policy, youth participation and lifestyle sports

    Young people have been identified as a key target group for whom participation in sport and physical activity could have important benefits to health and wellbeing and consequently have been the focus of several government policies to increase participation in the UK. Lifestyle sports represent one such strategy for encouraging and sustaining new engagements in sport and physical activity in youth groups, however, there is at present a lack of understanding of the use of these activities within policy contexts. This paper presents findings from a government initiative which sought to increase participation in sport for young people through provision of facilities for mountain biking in a forest in south-east England. Findings from qualitative research with 40 young people who participated in mountain biking at the case study location highlight the importance of non-traditional sports as a means to experience the natural environments through forms of consumption which are healthy, active and appeal to their identities. In addition, however, the paper raises questions over the accessibility of schemes for some individuals and social groups, and the ability to incorporate sports which are inherently participant-led into state-managed schemes. Lifestyle sports such as mountain biking involve distinct forms of participation which present a challenge for policy-makers who seek to create and maintain sustainable communities of youth participants

    The association of sitting time with sarcopenia status and physical performance at baseline and 18-month follow up in the residential aged care setting.

    This study investigated the association of sitting time with sarcopenia and physical performance in residential aged care residents at baseline and 18-month follow-up. Measures included the International Physical Activity Questionnaire (sitting time), European Working Group definition of sarcopenia, and the short physical performance battery (physical performance). Logistic regression and linear regression analyses were used to investigate associations. For each hour of sitting, the unadjusted odds ratio of sarcopenia was 1.16 (95% confidence interval [0.98, 1.37]). Linear regression showed that each hour of sitting was significantly associated with a 0.2-unit lower score for performance. Associations of baseline sitting with follow-up sarcopenia status and performance were nonsignificant. Cross-sectionally, increased sitting time in residential aged care may be detrimentally associated with sarcopenia and physical performance. Based on current reablement models of care, future studies should investigate if reducing sedentary time improves performance among adults in end of life care
