    Penelitian dilakukan dengan tujuan mengetahui pengaruh berbegai konsentrasi DMSO (0%, 5%, 7%, 10%, 13%, 15%, dan 17%) sebagai krioprotektan terhadap kualitas spermatozoa ikan gurami, Osphronemus goramy Lacepede, 1801, dua puluh empat jam pascakriopreservasi. Penelitian dilakukan di Balai Besar Pengembangan Budidaya Air Tawar (BBPBAT), Sukabumi. Semen dikoleksi dengan cara pengurutan (stripping) dan dievaluasi secara makroskopis (warna, pH, dan volume) dan mikroskopis (persentase motilitas, viabilitas, abnormalitas, dan konsentrasi spermatozoa) baik sebelum maupun sesudah kriopreservasi. Semen diencerkan dengan pengencer yang mengandung ekstender 189M dan krioprotektan DMSO (0%, 5%, 7%, 10%, 13%, 15%, dan 17%), disimpan pada cryotube 2 ml dan dikriopreservasi selama 24 jam. Peningkatan konsentrasi DMSO memengaruhi kualitas spermatozoa ikan gurami pascakriopresevasi. Konsentrasi DMSO yang terbaik berdasarkan hasil pengamatan adalah DMSO konsentrasi 13% (P > 0,05) dengan persentase motilitas sebesar 68,58%; abnormalitas sebesar 29%; dan viabilitas sebesar 63,5%

    Comparative study of estrogen receptor Ī±, Ī² mRNA expressions of endometriosis and normal endometrium in women and analysis of potential synthetic anti-estrogens in silico

    Endometriosis is a multifactorial disease in which genetic and environmental factors interact causing its pathogenesis. The aim of this study was to investigate the expression pattern of estrogen receptor Ī± (ERĪ±) and Ī² (ERĪ²) in endometriosis patients compared to normal endometrioum (n=18) as a control by using Quantitative Real Time PCR method. Moreover, we also measured serum estradiol levels of endometriosis patients in the proliferation phase of the menstrual cycle using the enzyme-linked immunosorbent assay method. The mRNA expression of ERĪ² was significantly higher in the endometriosis group compared to control, and the result of t-test showed that were significantly different (P<0.05). Overexpression of ERĪ² in endometriosis was likely to have other significant important impacts in the pathology of endometriosis that allowed ERĪ² to stimulate prostaglandin production in endometriosis tissue and cells. Estradiol content did not correlate with the ERĪ± expression, and it is weakly correlated with ERĪ² mRNA expression. Molecular docking analysis showed that ERĪ± and ERĪ² have different binding interactions with synthetic antiestrogens, whereas the best inhibitor was Ral2 to ERĪ± and Aco1 to ERĪ². Thus, both inhibitors could be used as leads in further investigation of ERĪ±, ERĪ² inhibitory activities in vitro and in vivo

    Intestinal Morphology, Energy Availability, and Growth Performance of Broilers Treated with the Combination of Probiotic and Inulin

    Probiotic and prebiotic or their combinations can potentially function as an alternative to antibiotics growth promoters (AGPs) for broiler. This study was designed to investigate the growth performance, intestinal microstructure, and nutrients digestibility of broilers administered with probiotics of Lactobacillus plantarum AKK30 and Saccharomyces cerevisiae B18 in combination with inulin. A total of 275 male chickens (initial bodyweight of 47Ā±0.05 g) were reared for growth performance evaluation. At the 32-d-old, 25 male chickens were necropsied for intestinal microstructural analysis, while the other 25 male chickens were selected for evaluation of digestibility (body weight= 1525Ā±0.08 g). Treatments of probiotics in combination with different levels of inulin consisted of control with probiotics without inulin (S0), probiotics with 0.5% of inulin (S1), probiotics with 1.0% of inulin (S2), probiotics with 1.5% of inulin (S3), and commercial probiotics without inulin (Sc), which were arranged in a completely randomized design with five replications. Results showed that body weight gain and performance index in broilers treated prebiotics in combination with 0.5% inulin (S1), 1% inulin (S2), and commercial probiotic without inulin (Sc) were significantly higher (p&lt;0.05) than those in control broiler chickens that were treated with probiotics without inulin. Feed intake showed no differences among treatments, whereas feed conversion ratios in broiler chickens treated with the commercial probiotics (Sc) or probiotics in combination with inulin at 0.5% (S1) and 1.0% (S2) were lower than control chickens. Metabolizable energy, nitrogen retention, and villi height in chickens treated with probiotics in combination with 0.5% inulin (S1) and 1.0% inulin (S2) were higher than those in the control group. In conclusion, the administration of probiotic combined with inulin at the level of either 0.5 or 1.0% improves broiler performance, intestinal microstructure, and nutrients digestibility

    Lactobacillus plantarum AKK30 inoculum in de Maan Rogosa Sharpe Broth (MRSB) grown on media containing oligosaccharides (inulin or mannan oligosaccharides/MOS) was studied in vitro and in vivo. In vitro assay was conducted to characterize metabolite profiles and its effect on pathogenic bacteria, while in vivo assay was conducted to study the metabolic energy, nitrogen retention, and intestinal morphology of broilers. A total of 24 male broilers-40-day-old (average body weight= 1.725Ā±0.05 kg) strains Cobb-500 were used, where 20 birds were added 1% inoculum L plantarum AKK30 of daily intake and randomly distributed in five treatments and four replications consisted of: A) MRSB + L. plantarum AKK-30 (10ā· cfu g-1); B) MRSB + inulin 0.5% (w v-1); C) MRSB + MOS 0.5%(w v-1); D) L. plantarum AKK-30 (10ā· cfu g-1) + MRSB + inulin 0.5% (w v-1).; and E) L. plantarum AKK-30 (10ā· cfu g-1) + MRSB + MOS 0.5% (w v-1), while 4 birds were fasted as endogenous chicken. The results showed that the highest isoleucine and threonine were found in group E. The highest oleic acid and the lowest conjugated linoleic acid (CLA) were observed in all treatments and inoculum did not affect antibacterial activity. The metabolizable energy values and nitrogen retention in group D and E were higher than other treatments (p&lt;0.05). The lowest height of villi was observed in group A (p&lt;0.05). In conclusion, the addition of oligosaccharides inulin or mannan oligosaccharide (MOS) affects the metabolite profiles of Lactobacillus plantarum AKK30 and their synbiotic effects improve intestinal morphology, metabolizable energy, and nitrogen retention in broilers

    The Honey Solution Effect as A Natural Cryoprotectant on Sperm Quality of Botia, Chromobotia Macracanthus Bleeker 1852

    The purpose of study was to obtain the best combination of natural cryoprotectant (honey solution) and synthetic cryoprotectant (methanol) on the quality of spermatozoa Chromobotia macracanthus Bleeker 1852, after freezing at -80 ā„ƒ for 48 hours. The combination of 10% methanol with various concentrations of honey solution (0%, 0.1%, 0.3%, 0.5%, 0.7% and 0.9%) was tested in this study. Ringer's solution was used as an extender. The diluted sperm was then equilibrated for 25 minutes at 4 ā„ƒ, then frozen at -80 ā„ƒ for 48 hours. The sperm was then thawed at 40 ā„ƒ for 13 seconds. Viability, motility and percentage of fertilization were evaluated. The results of the analysis of variance showed that the combination of 10% methanol with several concentrations of honey solution had a significant effect (P &lt;0.05) on the viability of spermatozoa and the percentage of fertilization, but had no effect on sperm motility (P&gt; 0.05). The combination of 0.1% honey solution and 10% methanol showed the highest percentage of both motility (89.4 Ā± 5.45%), viability (85.75 Ā± 4.79%), and the percentage of fertilization (98.55 Ā± 1.69%). %). The conclusion of this study is that 0.1% honey solution combined with 10% methanol in Ringer's solution is the best cryoprotectant for C. macracanthus spermatozoa stored at -80 ā„ƒ for 48 hour

    Molecular analysis of Lactobacillus fermentum isolates is essential to understand their genetic variation in relations to their roles in sayur asin fermentation process. Combination of three molecular techniques which is restriction fragment length polymorphism (RFLP) of 16S23S rDNA intergenic spacer region (ISR), random amplified polymorphic DNA (RAPD-PCR) and an enterobacterial repetitive intergenic consensus (ERIC-PCR) analysis were performed to discriminate 19 representative isolates of L. fermentum isolated from sayur asin. The result showed that L. fermentum strain D11 is distantly related to other isolates based on RFLP using HhaI restriction enzyme and RAPDPCR analyses. In addition, both of RAPD-PCR and ERIC-PCR successfully determined the genetic variation among L. fermentum strains by exhibiting distinct 4-8 bands (800-2080 bp) and 4-10 bands (280-3050 bp), respectively. A dendogram generated from UPGMA cluster analysis of both RAPD-PCR and ERIC-PCR data showed two distinct genotypic groups exist among L. fermentum isolated from sayur asin in Indonesia

    The Impact of Late Follicular Progesterone Level on in Vitro Fertilization-intracytoplasmic Sperm Injection Outcome: Case-control Study

    Background: Studies have been conducted to improve the pregnancy rate through the in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) program. In recent years, researchers have been focusing on finding impact of high progesterone level on endometrial receptivity. However, data on whether progesterone level also affects the quality of the embryo is still limited. Objective: The aim is to assess the effect of late follicular progesterone level on the outcome of in vitro fertilization-intracytoplasmic sperm injection (IVF-ICSI). Materials and Methods: This was a case-control of 245 women who underwent in vitro fertilization cycle at Halim Fertility Center, Indonesia. The outcomes assessed were number of oocytes retrieved (OR), maturation rate (MR), fertilization rate (FR), number of good embryos (GE), number of fair embryos (FE), and number of poor embryos (PE). The progesterone (P4) and estradiol (E2) levels were analyzed on the day of human chorionic gonadotropin injection. Serum progesterone level was divided into three groups: 1. low progesterone (ā‰¤ 0.50 ng/ml), 2. normal progesterone (0.51-1.50 ng/ml), and 3. high progesterone (&gt; 1.50 ng/ml). All outcomes were compared amongst the groups. Results: Significant differences occurred between progesterone level on the day of human chorionic gonadotropin administration. The number of OR in group 1, 2, and 3 were 8.41 Ā± 5.88 vs. 12.99 Ā± 8.51 vs. 17.58 Ā± 9.52, respectively. Conclusion: Progesterone level on the day of human chorionic gonadotropin injection may have an impact on the outcome of IVF-ICSI.
