<Original Article> Sister Chromatid Exchange (SCE) Frequency in Human and Simian Lymphoid Cell Lines Infected with Simian Retroviruses Closely Related to Human T-cell Leukemia Virus Type-1
Sister chromatid exchange (SCE) frequencies were examined on 4 simian lymphoid cell lines producing simian retroviruses closely related to human T-cellleukemia virus type-1 and 6 human lymphoid cells immortalized by these simian viruses. The frequencies of SCE in both simian and human lymphoid cell lines were higher than those in normal cultured simian and/or human lymphocytes. We also observed the change of SCE frequencies in human cell lines immortalized by STLV at several passage levels. At an early passage level the frequency of SCE in these human cell lines was already higher than that in normal lymphocytes culture. This rise in SCE frequency did not change significantly during the observation period for 180 days. From these findings we concluded that human lymphocytes were infected with STLV as simian lymphocytes, and that STLV caused same DNA damage in human lymphocytes as in simian lymphocytes. These phenomena induced by the retrovirus infections may correlate with leukemogenesis .journal articl

Unveiling the principle of microRNA-mediated redundancy in cellular pathway regulation
<p>Understanding the multifaceted nature of microRNA (miRNA) function in mammalian cells is still a challenge. Commonly accepted principles of cooperativity and multiplicity of miRNA function imply that individual mRNAs can be targeted by several miRNAs whereas a single miRNA may concomitantly regulate a subset of different genes. However, there is a paucity of information whether multiple miRNAs regulate critical cellular events and thereby acting redundantly. To gain insight into this notion, we conducted an unbiased high-content miRNA screen by individually introducing 1139 miRNA mimics into Chinese hamster ovary (CHO) cells. We discovered that 66% of all miRNAs significantly impacted on proliferation, protein expression, apoptosis and necrosis. In summary, we provide evidence for a substantial degree of redundancy among miRNAs to maintain cellular homeostasis.</p
