    DAPT attenuates ox-LDL-induced human umbilical vein endothelial cell injury

    目的:; 探究gamma-分泌酶抑制剂N-[N-(3,5-difluorophenacetyl)-L-alanyl]-S-phenylglycine; t-butyl ester (DAPT)在氧化低密度脂蛋白(oxidized low-density lipoprotein,; ox-LDL)损伤人脐静脉内皮细胞(human umbilical vein endothelial; cells,HUVECs)模型中的细胞保护作用及其对Notch信号通路的调控。方法:体外培养HUVECs,用ox-; LDL处理HUVECs构建细胞损伤模型。实验分为对照组、ox-LDL处理组、DAPT处理组和DAPT +; ox-LDL处理组。用倒置相差显微镜观察不同处理方法下细胞的形态变化; CCK-8法检测细胞存活率; Western; blot法检测蛋白Notch1、Notch4和Jagged1的表达情况。结果:体外培养HUVECs,倒置相差显微镜下发现ox-LDL处理组细胞死; 亡和碎片增多,经DAPT预处理后,; ox-LDL作用造成的细胞损伤死亡较少,细胞碎片较少。通过CCK-8法检测发现ox-LDL处理组细胞存活率降低,DAPT处理组细胞存活率升高,D; APT预处理后ox-LDL造成存活率降低的幅度变小。在ox-LDL作用下,Notch1和Jagged1蛋白表达量降低,Notch4表达量升高;; 而DAPT作用下Notch1和Jagged1表达量升高,Notch4表达量降低; ox-LDL与DAPT共同作用时蛋白接近正常水平。结论:; ox-LDL对HUVECs具有损伤作用; DAPT减轻ox-LDL对HUVECs造成的损伤;; DAPT保护HUVECs免受ox-LDL损伤的作用与Notch信号通路有关。AIM: To investigate the effect of N-[N-(3,5 -difluorophenacetyl); -L-alanyl]-S-phenylglycine t-butyl ester (DAPT) on the Notch signaling; pathway in a model of oxidized low-density lipoprotein (ox-LDL) -induced; human umbilical vein endothelial cell (HUVEC) damage. METHODS: HUVECs; were divided into control group,ox-LDL group,DAPT group and ox-LDL +; DAPT group. The morphological changes of the HUVECs with different; treatments were observed under light microscope. The viability of the; HUVECs was measured by CCK-8 assay. The protein expression levels of; Notch1,Notch4 and Jagged1 were determined by Western blot. RESULTS:; ox-LDL induced great damage to the HUVECs,evidenced by increased cell; death and debris in the culture. However,the cell damage was abolished; by adding DAPT into the culture. The viability of the HUVECs was; increased by co-treatment with DAPT and ox-LDL. ox-LDL treatment; significantly decreased the protein expression levels of Notch1 and; Jagged1,and elevated Notch4. However,these changes were totally reversed; by DAPT. None of these proteins showed significant change in the HUVECs; co-treated with DAPT and ox-LDL as compared with control group.; CONCLUSION: ox-LDL is able to induce HUVEC damage in vitro. DAPT; attenuates ox-LDL-induced damage in the HUVECs by regulating the Notch; signaling pathway.厦门大学校长基金本科生项

    合成了外围由小分子2-芴醛修饰的树状大分子PAMAM(1G)-FCD,用IR,1H NMR,MALDI-TOF-MS等手段表征了其结构,并对其荧光性能及Sn2+对该性能的影响进行了研究,结果表明,Sn2+能使化合物荧光显著增强.紫外光谱表明,随着PAMAM(1G)-FCD溶液中Sn2+浓度的增加,体系在360 nm处出现了新的吸收峰,表明二者之间存在化学反应.故该树状分子有望作为难得的蓝光区荧光材料及金属-树状大分子杂化材料

    粗细粉混用法制备堇青石膜支撑体

    将粒径分别为1.5和25μm的堇青石粉体按一定比例球磨混匀,添加适当的粘结剂和造孔剂,经捏合、陈腐、挤出成型及烘坯处理后,程序升温至一定温度烧结2h制备膜支撑体。结果表明,细粉含量20%(质量分数)、1400℃烧结所得的支撑体综合性能较好:纯水通量为10.3m3/(m2·h);爆破压力为2.21MPa。其浸渍于5%(质量分数)NaOH中、置于100℃烘箱36h后,爆破压力仍为1.85MPa,表明其还具有良好的耐碱性能,适合碱性条件下的工业应用

    Cognitive Function and Activity of Daily Living of the Old Elderly of Zhuang Nationality in Bama County

    目的:了解广西巴马县壮族长寿老人认知功能及日常生活自理能力. 方法:采用简易智力状态检查(MMSE)、日常生活能力量表(ADL)入户调查. 结果:85~89、90~94、95~104岁组MMSE总分分别为24.39±2.36;23.5±2.7;22.3±3.1,符合认知下降诊断者分别占3%;9.3%和13.7%.长寿老人随增龄主要表现为短时记忆、注意力和计算力下降明显(P<0.05).ADL方面,各组自理能力功能丧失率分别为6%;12.5%和31%,其中85~89岁与95~104岁组比较差异显著(P<0.05),而在ADL所有项目中,仅85~89岁与95~104岁组在处理自已钱财功能丧失有显著差异(P<0.05). 结论:巴马长寿老人总体认知功能、自理能力较其他老年人群保持好;增龄仍然是长寿老人认知及日常活动能力下降的重要因素;长寿老人ADL和MMSE相关明显

    水电站跨省跨区月度合同电量曲线分解模型

    水电站跨省跨区送电涉及多省份、多品种、多时间尺度等多维度电量曲线分解,且需要考虑复杂的电网调峰等需求,是电站每月发电运行中亟待解决的重要难题。依托实际工程的生产需求,构建了适合跨省、跨区送电的水电站月度合同电量曲线分解模型,考虑保量保价、保量竞价、市场化三类品种电价差异,以及电网调峰、市场和电站运行复杂约束,提出了多省多品种发电收益总和最大的二次规划目标,并采用混合整数线性规划方法实现了模型高效求解。以溪洛渡右岸水电站为依托工程,通过枯期、汛期两个不同的应用场景进行了模型验证,结果表明在满足电网要求的调峰边界条件下,通过优化两省多尺度多品种电量和出力分配,市场化电量优先分配至负荷高峰和平段,可以有效提高电站发电收益

    Probing the edge-related properties of atomically thin MoS2 at nanoscale

    层状二维材料具有独特的物理化学性质,使其在光电器件、传感、能源和催化等领域得到了高度关注和广泛应用。二维材料在制备过程中不可避免引入结构缺陷,虽然这些缺陷尺度仅为数纳米甚至单原子,但是会极大地改变材料的结构和电子性质,从而影响其应用。化学化工学院任斌教授课题组在层状二维材料缺陷表征方面取得进展。该工作表明了TERS在原位、高空间分辨表征缺陷位的结构和电子性质方面具有独特的优势,可以进一步推广到其他二维材料,从而有效地指导缺陷设计和材料应用。 该工作通过校内外课题组紧密合作,在任斌教授、谭平恒研究员(中科院半导体研究所)和王翔博士共同指导下完成。实验部分主要由黄腾翔博士(第一作者,已毕业化学系博士生)完成,电子能带结构与光谱理论计算由谭平恒研究员课题组从鑫博士生(共同第一作者)完成,吴思思、林楷强、姚旭、何玉韩、吴江滨、包一凡、黄声超等参与了实验与讨论。【Abstract】Defects can induce drastic changes of the electronic properties of two-dimensional transition metal dichalcogenides and influence their applications. It is still a great challenge to characterize small defects and correlate their structures with properties. Here, we show that tipenhanced Raman spectroscopy (TERS) can obtain distinctly different Raman features of edge defects in atomically thin MoS2, which allows us to probe their unique electronic properties and identify defect types (e.g., armchair and zigzag edges) in ambient. We observed an edgeinduced Raman peak (396 cm−1) activated by the double resonance Raman scattering (DRRS) process and revealed electron–phonon interaction in edges. We further visualize the edge-induced band bending region by using this DRRS peak and electronic transition region using the electron density-sensitive Raman peak at 406 cm−1.
