    本文探讨二色桌片参糖蛋白 GPMI- 和 GPMI- 对小鼠移植性肿瘤 S180 的抑制作用以及对荷瘤小鼠肝脾等器官重量的影响。实验结果显示 ,经蛋白酶水解后的糖蛋白 GPMI- 在剂量 2 0μg· g-1时小鼠肉瘤S180 有较显著的抑制作用 (P<0 .0 5 ) ,抑瘤率达 4 8.8%。同时发现 ,GPMI- 在 30μg· g-1剂量下可显著增加荷瘤小鼠的脾指数 (P<0 .0 5 ) ,GPMI- 在 4 0 μg· g-1剂量下能够显著降低荷瘤小鼠肝指数 (P<0 .0 5 )。In this study,two kinds of glycoproteins GPMI Ⅰ and GPMI Ⅱ were extractyed from sea cucuber Mensamaria intercedens.Their anti tumor activity to mice implanted tumor S 180 were evaluated in vivo by anti tumor assay.The results indicated that GPMI Ⅱ could significantly inhibit the growth of S 180 cells implanted subcutaneously in mice ( P <0.05) at the dose of 20μg·g -1 per day×10 with no sign of toxicity.The ratio of tumor inhibition is 48.8% in S 180 mice.The effects appear to be dose related.On the other hand, GPMI Ⅰ increases the weight of spleen of mice with S 180 cells at the dose of 30μg·g -1 per day×10 . GPMI Ⅱ decreases the weight of liver of mice with S 180 cells at the dose of 40μg·g -1 per day×10

    目的 研究全反式视黄酸(ATRA)对胃癌细胞生长的影响并从酶水平探索可能的作用机制。 方法 以MTT法测定ATRA对胃癌细胞的生长抑制作用;用酶标仪测定乳酸脱氢酸(LDH)、β葡萄糖醛酸酶(βG)和碱性磷酸酶(ALP)的活性;以细胞化学法测定琥珀酸脱氢酶(SDH)的活性。 结果 ATRA能抑制MGc803细胞的生长并降低LDH、ALP和βG的活性,同时提高SDH的活性;但不能抑制MKN45细胞的生长;除降低其细胞外的βG活性外,也不影响LDH、ALP、SDH和细胞内βG活性。 结论 胃癌细胞对ATRA的敏感性不同,这与细胞酶活性的变化密切相

    目的 :探讨二色桌片参糖蛋白GPMI Ⅰ和GPMI Ⅱ对小鼠移植性肿瘤S1 80 的抑制作用以及对荷瘤小鼠肝脾等器官重量的影响。方法 :通过对二色桌片参体壁进行水煮提取、分离、过SephadexG 2 0 0柱层析 ,获得二色桌片参糖蛋白Ⅰ (GPMI Ⅰ ) ,GPMI Ⅰ里的蛋白部分经酶解后得二色桌片参糖蛋白Ⅱ (GPMI Ⅱ )。结果 :GPMI Ⅰ在 30mg kg及 12 0mg kg剂量下可显著降低荷瘤小鼠的肝指数 (P <0 0 1,P <0 0 2 5 )。同时随着其剂量增加 ,抑瘤作用也明显增强 (P <0 0 0 1)。而经蛋白酶水解后的糖蛋白GPMI Ⅱ在剂量 2 0mg kg时对小鼠肉瘤S1 80 有较显著的抑制作用 (P <0 0 5 ) ,抑瘤率达 4 8 8%。同时发现 ,GP MI Ⅱ在 4 0mg kg剂量下能够显著地降低荷瘤小鼠肝指数 (P <0 0 5 )。结论 :GPMI Ⅰ和GPMI Ⅱ对小鼠S1 80 肿瘤均有较显著的抑制作用Objective:To study the effects of two kinds of glycoproteins GPMI Ⅰand GPMI Ⅱ at inhibiting the growth of mice implanted tumor and the effects on the weight of spleen and liver of mice.Methods:GPMI Ⅰ and GPMI Ⅱ were extracted from sea cucumber mensamaria intercedens by boiling,separating and chromatography.Their anti tumor activity to mice implanted tumor Sarcoma 180 was evaluated in vitro by anti tumor assay.Results:GPMI Ⅱcould significantly inhibit the growth of Sarcoma 180 cells implanted subcutaneously in mice( P <0 05)at the dose of 20mg/kg per day×10 with no sign of toxicity.The ratio of tumor inhibition was 48 8% in Sarcoma 180 mice.GPMI Ⅱ decreased the weight of liver of mice with Sarcoma 180 cells at the dose of 40mg/kg per day×10.On the other hand,GPMI Ⅰdecreased the weight of liver of mice with Sarcoma 180 cells at the dose of 30mg/kg and 120mg/kg per day×10.GPMI Ⅰ could significantly inhibit the growth of Sarcoma 180 cells implanted subcutaneously in mice( P <0 001)at the dose of 120mg/kg per day×10 with no sign of toxicity.The effects appeared to be dose related.Conclusion:GPMI Ⅰ and GPMI Ⅱ can have significant effects on inhibition in Sarcoma 180 mice

    二色桌片参〔Mensamaria intercedens( L ampert)〕的体壁经水煮提、分离、最后经 Sephadex G- 2 0 0柱层析纯化得二色桌片参糖蛋白 ( GPMI- )。GPMI- 经酶解后得二色桌片参糖蛋白 ( GPMI- )。分别研究了GPMI- 和 GPMI- 的性状、化学组成和抗肿瘤活性 ,结果表明 :GPMI- 和 GPMI- 都是糖蛋白 ,但蛋白组成和糖含量明显不同。GPMI- 对小鼠 S180 肉瘤有明显抑制作用 ,而无直接毒性 ,抑瘤率为 4 8.8%。而 GPMI- 却能显著提高 S180 肉瘤荷瘤小鼠的脾指数 ,呈免疫增强作用。Glycoprotein of Mensamaria intercedens(GPMI Ⅰ) was extracted from the body of Mensamaria intercedens with hot water,and purification was finished with column chromatography on Sephadex G 200. GPMI Ⅰ was hydrolyzed with enzymes to obtain glycoprotein of Mensamaria intercedens Ⅱ(GMI Ⅱ).The chemical property,composition and antitumor activity of GPMI Ⅰ and GPMI Ⅱ were studied. The results showed that GPMI Ⅰ and GPMI Ⅱ were two kinds of glycoproteins in which the content of protein and sugar were different. GPMI Ⅱ could significantly inhibit the growth of Sarcoma 180 cells implanted subcutaneously in mice (P<0.05) at the dose of 20 mg·kg -1 per day ×10 with no sign of toxicity. The ratio of tumor inhibition is 48.8%. On the other hand, GPMI Ⅰ increased the weight of spleen of mice with S 180 cells at the dose of 30 mg·kg -1 per day×10.福建省重中之重项目资

    对海参动物——二色桌片参 [Mensamaria intercedens] (Lampert)的主要化学成分的分析结果表明 ,这种海参干品蛋白含量高达 85.98% ,维生素 A、E含量也较高 ,有较高的营养价值。更重要的是这种海参还含有丰富的活性蛋白多糖、多糖和皂甙类物质 ,是开发海洋药物的丰富资源。The main chemical components from Sea cucumber Mensamaria intercedens (Lampert) are detected. The data indicate that it contains fairly rich protein which is about 85.98% in the dry powder. The reducing carbohydrates and free amino acids reach 535.0 mg/L and 6934.0 mg/L respectively in the enzyme digested solution (100 mL contain 5.5g dry powder). So it can be seen that this Sea cucumber has fairly high nutritional value. The important is it also contains fairly active glycoprotein, polysaccharide, as well as saponin which including triterpene-oligoglycoside and steroid-oligoglycoside.     Determination of Astaxanthin by High Performance Liquid Chromatography with Photodiode Array Detector

    建立虾青素含量测定的高效液相色谱–光电二极管阵列法。采用PurOSPHEr STAr rP 18(4.6 MMx250MM,5μM)色谱柱,以甲醇–水(体积比为95∶5)为流动相,流速1.0 Ml/MIn,检测波长为482 nM,柱温为30℃,进样量为20μl。在所选定的液相色谱条件下,虾青素主峰与其它杂质峰分离良好,虾青素在0.2~16μg/Ml范围内线性良好,线性相关系数r=0.999 9,检出限为0.01μg/Ml,测定结果的相对标准偏差为0.42%(n=6),平均回收率为100.4%。该法分析快速准确、灵敏度高、重现性好。A method using high-performance liquid chromatography(HPLC) coupled with photodiode array detector(PDA) was established for determination of astaxanthin.The separation was performed on Purospher STAR RP 18 column(4.6 mm×250 mm,5μm) with the mobile phase of methanol–water(Volume ratio was 95∶5).The detector was photodiode array detector and the variable wavelength detector(VWD) was set at 482 nm.The column temperature was 30℃ and the injection volume was 20 μL.The calibration curve was linear in the range of 0.2–16 μg/mL,and the correlation coefficient was 0.999 9.The relative standard deviation of determination results was 0.42%(n=6).The average recovery was 100.4%, and the detection limit was 0.01 μg/mL.This method is characteristic of rapidity,accuracy,high sensitivity and good reproducibility.海洋生物产业化中试技术研发公共服务平台项目(厦海渔合[2013]19号

    高岭土中铁的赋存状态和含量影响其白度和增白效果,因此快速准确地测定高岭土中不同种类的铁含量具有实际应用意义。高岭土中自由铁和结构铁定量分析困难,为此本文将高岭土中的铁分为可溶铁和非可溶铁两类。高岭土经盐酸酸溶处理后溶液中的铁含量为可溶铁含量,经氢氧化钠碱熔处理后溶液中的铁含量为总铁含量,非可溶铁含量由总铁含量减去可溶铁含量计算得出。采用邻菲啰啉光度法测定溶液中的铁含量,该方法的相对标准偏差小于5%,加样回收率范围在95%~105%,最低检测含量为1.06μg/g。高岭土中的其他元素对铁的测定均没有干扰。所测得的可溶铁为大部分自由铁和少数结构铁。通过对高岭土中的可溶铁进行定量分析,可得知漂白工艺中可除去的最大铁含量
