目的 :探讨二色桌片参糖蛋白GPMI Ⅰ和GPMI Ⅱ对小鼠移植性肿瘤S1 80 的抑制作用以及对荷瘤小鼠肝脾等器官重量的影响。方法 :通过对二色桌片参体壁进行水煮提取、分离、过SephadexG 2 0 0柱层析 ,获得二色桌片参糖蛋白Ⅰ (GPMI Ⅰ ) ,GPMI Ⅰ里的蛋白部分经酶解后得二色桌片参糖蛋白Ⅱ (GPMI Ⅱ )。结果 :GPMI Ⅰ在 30mg kg及 12 0mg kg剂量下可显著降低荷瘤小鼠的肝指数 (P <0 0 1,P <0 0 2 5 )。同时随着其剂量增加 ,抑瘤作用也明显增强 (P <0 0 0 1)。而经蛋白酶水解后的糖蛋白GPMI Ⅱ在剂量 2 0mg kg时对小鼠肉瘤S1 80 有较显著的抑制作用 (P <0 0 5 ) ,抑瘤率达 4 8 8%。同时发现 ,GP MI Ⅱ在 4 0mg kg剂量下能够显著地降低荷瘤小鼠肝指数 (P <0 0 5 )。结论 :GPMI Ⅰ和GPMI Ⅱ对小鼠S1 80 肿瘤均有较显著的抑制作用Objective:To study the effects of two kinds of glycoproteins GPMI Ⅰand GPMI Ⅱ at inhibiting the growth of mice implanted tumor and the effects on the weight of spleen and liver of mice.Methods:GPMI Ⅰ and GPMI Ⅱ were extracted from sea cucumber mensamaria intercedens by boiling,separating and chromatography.Their anti tumor activity to mice implanted tumor Sarcoma 180 was evaluated in vitro by anti tumor assay.Results:GPMI Ⅱcould significantly inhibit the growth of Sarcoma 180 cells implanted subcutaneously in mice( P <0 05)at the dose of 20mg/kg per day×10 with no sign of toxicity.The ratio of tumor inhibition was 48 8% in Sarcoma 180 mice.GPMI Ⅱ decreased the weight of liver of mice with Sarcoma 180 cells at the dose of 40mg/kg per day×10.On the other hand,GPMI Ⅰdecreased the weight of liver of mice with Sarcoma 180 cells at the dose of 30mg/kg and 120mg/kg per day×10.GPMI Ⅰ could significantly inhibit the growth of Sarcoma 180 cells implanted subcutaneously in mice( P <0 001)at the dose of 120mg/kg per day×10 with no sign of toxicity.The effects appeared to be dose related.Conclusion:GPMI Ⅰ and GPMI Ⅱ can have significant effects on inhibition in Sarcoma 180 mice
