    本文报道了Hg、Cd及二者共同作用明显不同程度地影响烟草叶绿素含量及抗氧化酶系统。受Hg、Cd胁迫后,随着土壤中Hg、Cd浓度的增加,叶绿素含量、叶绿素a/b值、CAT活性逐渐减小,SOD活性先升后降,POD活性则逐渐增加。同时也表现出单一Hg、Cd对烟草叶绿素含量及抗氧化酶系统的影响明显大于Hg、Cd二者的共同作用

    The Acetylation of Thymosin α1 is Independent on RimL in Escherichia coli

    目的:考察大肠杆菌乙酰转移酶rIMl对胸腺素α1(Tα1)乙酰化修饰的影响。方法:构建含500bP同源臂的卡那抗性基因打靶片段,利用rEd同源重组系统,使大肠杆菌bl21(dE3)的rIMl基因插入失活,随后导入质粒PCP20去除抗性基因,构建突变菌株rIMl-bl21(dE3);将重组质粒PET-Tα1-l12分别转入出发菌株和突变菌株中进行表达,经固定金属离子亲和层析和反向高效液相层析后,将所得纯品进行质谱分析,精确测定相对分子质量。结果:PCr鉴定结果证明成功敲除rIMl基因;质谱结果表明,rIMl基因敲除菌中所表达的Tα1-l12融合蛋白与出发菌株一样,均有部分乙酰化修饰。结论:Tα1的乙酰化修饰并不依赖于rIMl。Objective:To investigate the effect of RimL,a N-terminal acetyltransferase of E.coli,on thymosin α1(Tα1) acetylation.Methods:A kanamycin cassette with two 500 bp long arms homologous to the regions around rimL as the replacement fragment was electroporated into cells,in which the Red recombinant functions was induced.Subsequently,the plasmid pCP20 was transformed to eliminate the kanamycin resistant gene.The plasmid pET-Tα1-L12 was transformed into the original and mutant strains respectively.The expressed fusion protein Tα1-L12 was purified by IMAC and RP-HPLC.The accurate molecular weight of the fusion protein was measured by Q-Tof-MS.Results:The gene rimL was inactivated by insertion of the replacement fragment successfully.According to the MS results,the fusion protein Tα1-L12 was still partly acetylated when it was expressed in the mutant strains as in the original strains.Conclusion:Tα1 acetylation was independent on RimL

    利用重叠PCR技术将PTH(parathyroidhormone,甲状旁腺激素)基因与TFN(transferrinN_terminalhalf_molecule,转铁蛋白N端半分子)基因在体外融合,融合基因克隆至真核表达载体pPIC9中,转化毕赤酵母GS115。转化子经甲醇诱导后,融合蛋白得到了表达并分泌到发酵上清液中。经SPSepharoseFF阳离子交换层析、PhenylSepharoseFastFlow疏水层析纯化获得了纯度大于95%的PTH_TFN样品。Westernblot分析及腺苷酸环化酶实验证明融合蛋白中的PTH具有与抗PTH抗体结合能力及刺激腺苷酸环化酶的活性,铁饱和实验证明融合蛋白中的TFN和单独的TFN具有相同铁结合能力。因而TFN可望作为PTH的天然运输载体。The fused gene (PTH_TFN) of parathyroid hormone (PTH) gene and transferring N_terminal half_molecule (TFN) gene was amplified by multiple PCR and inserted into pPIC9 vector. The recombinant plasmid pPIC9_PTH_TFN was transformed into Pichia pastoris GS115 by PEG. After methanol induction, the target protein was expressed in fermentation supernatant at high level.The fused protein PTH_TFN with purity being higher than 95% was finally obtained after purification through two_step chromatography : SP Sepharose Fast Flow and Phenyl Sepharose Fast Flow.Western blot analysis and adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis and the ability to bind Fe ~3+ in the Fe ~3+ saturation study as the recombinant TFN did indicating that TFN could be used as the transcellar carrier of PTH.国家高技术研究与发展项目基金资助(No.2004AA215172)。~

    Green Emission from a Strain-Modulated InGaN Active Layer

    Strain-induced quantum dots (QDs) like island formations are demonstrated to effectively suppress pits/dislocation generation in high indium content (26.8%) InGaN active layers. In addition to the strain redistribution in the QD-like islands, strain modulation on the InGaN active layers by using the GaN island capping is employed to form an increased surface potential barrier around the dislocation cores, which inhibits the carrier transport to the surrounding dislocations. Cathodoluminescence shows distinct double-peak emissions at 503 nm and 444 nm, corresponding to the In-rich QD-like emission and the normal quantum well emission, respectively. The QD-like emission becomes dominated in photoluminescence due to the carrier localization effect of In-rich InGaN QDs at relatively low "carrier injection current". Accordingly, green emission may be enhanced by the following origins: (1) reduction in pits/dislocations density, (2) carrier localization and strain reduction in QDs, (3) strain modulation by GaN island capping, (4) enhanced light extraction with faceted GaN islands on the surface.National Natural Science Foundation of China [60876008, 61076091]; Program for New Century Excellent Talents in Fujian Province Universit
