Modification and applications of Taq DNA polymerase

本论文对TaqDNA聚合酶展开了研究，从TaqDNA聚合酶表达载体出发，对其进行定向改造，分别用于探针检测PCR体系和焦磷酸水解激活的聚合酶反应（PAP）技术，最后制备其抗体达到热启动PCR的目的。 第一章，概述了TaqDNA聚合酶在PCR中的应用，普通TaqDNA聚合酶虽然得到广泛使用，但是仍然存在不足。各个实验室针对TaqDNA聚合酶的不足进行了不同形式的改造，以降低或消除酶的5'→3'核酸外切酶活性，提高酶的续进性、忠实性、特异性和耐热性等。 第二章，我们也通过定点突变和缺失突变的方法在基因水平对TaqDNA聚合酶进行改造，以消除酶的5'→3'核酸外切酶活性和降低TaqDNA聚合酶对...This dissertation consists of three parts. It focuses on modification and applications of Taq DNA polymerase. Starting from complete sequence of Taq DNA polymerase, we made a variety of modified Taq DNA polymerases and applied them to real-time PCR, pyrophosphorolysis-activated polymerization (PAP) and hot-start PCR. Study on educational system for assembly language by using effective visualization method

In this paper, I propose an educational system for assembly language and computer architecture by using effective visualization method. The System allows students to study assembly program technique and internal structure and behavior of computer visually. Current educational system has a problem that visualization of control structures and data structures. The system supports it and necessary function such as editor, debugger, tracer, and profiler which reference current educational system
