    高效液相色谱法同时测定水产品中6种喹诺酮药物的残留

    建立用固相萃取-反相高效液相色谱法同时检测水产品中6种喹诺酮药物残留量的方法。通过对提取方法和C18固相萃取柱净化条件、色谱条件选择与优化、洗脱液浓度和用量的选择方面的研究,确定采用高效液相色谱分离、荧光检测器检测、外标法定量的分析方法。本方法对环丙沙星、恩诺沙星、氧氟沙星、诺氟沙星、噁喹酸、氟甲喹标准曲线的线性回归系数均在0.99以上,线性范围为0.02~2.0μg/mL。以3倍信噪比RSN计算,环丙沙星、恩诺沙星、诺氟沙星、氧氟沙星检出限为2μg/kg,噁喹酸、氟甲喹检出限为5μg/kg;以10倍信噪比RSN计算,环丙沙星、恩诺沙星、氧氟沙星、诺氟沙星的定量下限为5μg/kg;噁喹酸、氟甲喹的定量下限为10μg/kg。在南美白对虾、罗非鱼、鳗鱼、斑点叉尾鮰中进行这6种喹诺酮类药物加标回收率的验证实验,实验结果满意,回收率在78.8%~92.9%之间;相对标准偏差为2.97%~7.10%。说明本方法简单、灵敏,结果可靠,可用于水产品中环丙沙星、恩诺沙星、氧氟沙星、诺氟沙星、噁喹酸、氟甲喹的同时检测

    紫菜降血压肽大鼠体内降压效果研究

    目的观察紫菜降血压肽对高血压大鼠(SHR)和京都Wistar大鼠(WKY)的降压效果,并与降压药物卡托普利的作用效果比较。方法分别以500、1 000和1 500 mg.kg-1剂量的紫菜降血压肽一次性给药后,每隔2 h测量大鼠收缩压(SBP),连续测量8 h;长期实验持续4 w,每天1 500 mg.kg-1灌胃一次,每隔1周测量一次SBP、心率及体重。结果 SHR给予紫菜降血压肽后,血压均显著下降;而正常血压大鼠WKY服用紫菜降血压肽后血压没有显著变化,服用卡托普利血压仍保持显著下降的趋势。结论紫菜降血压肽对SHR具有显著的降压效果,长期服用可稳定血压,对正常血压大鼠无明显影响;卡托普利对SHR的降压效果虽高于紫菜降血压肽,但会导致正常大鼠血压过低

    Accumulation and elimination of avermectin in perch (Lateolabrax japonicus) muscle

    为了解实际养殖过程中使用阿维菌素药物后,鲈(lATEOlAbrAX JAPOnICuS)对阿维菌素的蓄积及消除规律,以指导阿维菌素药物在实际生产中的应用。以毒性试验为基础,在确定96 H半致死质量浓度(lC50)后,以0.25倍lC50,即指导用药量,质量浓度为4 ng·Ml-1,结合实际养殖用药情况,药浴72 H后换水,观察阿维菌素在鲈鱼肌肉组织中的残留与消除规律。文章建立高效液相色谱-串联质谱法测定鲈鱼中阿维菌素残留量的方法,并以此法测定鲈鱼肌肉组织中阿维菌素的质量浓度。结果表明,随着药浴时间的延长,鲈鱼肌肉组织中的阿维菌素质量分数逐步增加,在药浴结束时达到峰值8.767μg·kg-1,随后阿维菌素在体内的残留量随消除试验的进行逐渐下降,至第528小时降至检测限以下。To investigate the accumulation and elimination rules of avermectin in perch(Lateolabrax japonicus) muscle during actual breeding process,we determined the median lethal concentration in 96 h as 4 ng·mL-1 based on a toxicity test,and exposed the perch to 4 ng·mL-1 of avermectin for 72 h,and then transfered the solution into seawater.Thus,we develop a method by high performance liquid chromatography-tandem mass spectrometry to determine avermectin in perch muscle.The results show that the avermectin concentration in perch muscle gradually increases with passing medicated bath time,and reaches the peak of 8.767·kg-1 at the end of the medicated bath.Later,the avermectin in perch muscle gradually decreases and reduces below the detection limit at 528th hour.福建省海洋与渔业厅重点项目(KJXH-2010-007); 国家星火计划项目(2011GA72001

    建立用高效液相色谱-串联质谱法测定水产品中19喹诺酮类药物——氟甲喹、噁喹酸、诺氟沙星、依诺沙星、环丙沙星、培氟沙星、洛美沙星、丹诺沙星、恩诺沙星、氧氟沙星、沙拉沙星、司帕沙星、双氟沙星、西诺沙星、奥比沙星、麻保沙星、萘啶酸、氟罗沙星和吡哌酸残留量的方法。用酸化乙腈提取水产品中喹诺酮类药物,正己烷脱脂。采用电喷雾电离源(ESI),正离子扫描模式,选择反应监测模式(SRM)监测,外标法定量。该方法对19种喹诺酮类药物标准曲线的线性回归系数均在0.99以上,线性范围为2~200ng/mL,氟罗沙星和吡哌酸检出限为1.0μg/kg、定量限为2.0μg/kg,其余17种喹诺酮的检出限为0.5μg/kg、定量限为1.0μg/kg。19种喹诺酮类药物回收率在76.3%~108%之间,相对标准偏差为0.743%~13.1%。该方法简单、灵敏,结果可靠,可满足实验室批量样品分析的需求

    本文研究了不同规格黑脊倒刺鲃的耗氧率和排氨率。实验在室内水族箱内进行,实验鱼按体重分成三种规格(187~245g,10.58~14.32g,1.96~2.95g),每个规格设三个平行组。实验结果表明,水温30℃时,黑脊倒刺鲃的平均耗氧率0.389mg/(g.h);氨氮排泄率1.608&mu;mol/(g.h);O:N=15.49。不同规格对耗氧率和排氨率有显著影响(P&lt;0.01)。耗氧率OR=0.6458W~(-0.1766)(r=0.9666),耗氧量M_0=0.6458W~(0.8233)(r=0.9984),排氨率NR=2.185W~(-0.0971)(r=0.7831),排氨量M_N=50.328W~(0.9038)(r=0.9959)。耗氧率和排氨率随规格增大而减小,耗氧量和排氨量随规格增大而增大

    Detection of inorganic arsenic in algae by electrospray ionization mass spectrometry

    建立了基于分子量测定海藻中无机砷含量的新方法——电喷雾质谱法。探讨了提取方法、溶液PH、还原剂、螯合剂、提取剂、质谱仪参数等实验条件对实验准确度、灵敏度的影响。在优化条件下,AS(Ⅲ)的线性范围为0~151.4 ng/Ml,AS(V)的线性范围为0~175.0 ng/Ml,相关线性回归系数均在0.99以上,方法检出限(S/n>3)0.024 Mg/kg,方法定量限(S/n>10)0.048 Mg/kg。实验在不同浓度下加标回收率为81.0%~95.6%,相对标准偏差为2.52%~7.54%。该方法简单,灵敏度高,重现性好,避免了有机砷引起的假阳性现象,适用于海藻中无机砷的测定,可为海藻中无机砷监督检验提供有力技术支撑。A new method based on molecular mass for rapid determination of inorganic arsenic in algae by electrospray ionization mass spectrometry was developed.Several conditions affecting accuracy and sensitivity such as extraction method,pH of solvent,reductant,chelating agent,extractant and instrument parameters were studied.Under the optimum conditions,the calibration curve of As( Ⅲ) was linear in the range of 0-151.4 ng /mL,the calibration curve of As( Ⅴ) was linear in the range of 0- 175.0 ng /mL,and the correlation coefficients were all above 0.99.The limit of detection( S /N > 3) was 0.024 mg /kg and the limit of quantitation( S /N > 10) was 0.048 mg /kg.The recoveries at different spiked concentration levels ranged from 81.0% to 95.6% with relative standard deviations( 2.52%- 7.54%).This method is simple,of high sensitivity and good reproducibility,without interference of organic arsenic.It can be applied to analyze inorganic arsenic in algae and can provide technological support for food safety management.福建省自然科学基金面上项目(2010J01020); 厦门市科技局项目(3502z2011201); 福建省海洋与渔业厅项目(KJXH-2010-007); 石狮市重点项目(2010SN04
