    Fine Root Biomass and Dynamics of Casuarina equisetifolia Plantations on Coastal Sandy Soil

    2005年1月到11月对福建省惠安县赤湖林场不同林龄木麻黄人工林细根的生物量及其动态特征进行了研究.结果表明,24a生木麻黄林细根生物量分别占其地下部分生物量和林分总生物量的53.1%和3.8%;活细根的生物量随林龄的增大而逐渐增加,至30a林龄时达到最大值12.373thm-2,而后逐渐下降,死细根的生物量则呈现一直增大的趋势,木麻黄人工林细根的生物量与林分地上部分的生长具有显著的相关关系;细根生物量具有明显的季节动态,各林龄无论活细根还是死细根都表现为双峰型,3a生和18a生的活细根出现在1月和7月,而12a生出现在3月和7月,对于死细根,12a生和18a生的两个峰值出现在3月和7月,5a生则出现在7月和11月.各林龄木麻黄防护林活、死细根密度垂直分布呈单峰型,最大值出现在表层的0~10cm土层中,后随土层厚度增加逐渐减少,其中5a林龄细根生物量随土层深度增加而减少表现最为明显.在0~10cm土层中的活、死细根生物量分别占全部活细根生物量的51.9%和死细根生物量的53.3%,活细根生物量的84.6%和死细根生物量的82.8%分布在0~30cm的土层中.以后随着林龄的增加,表层土壤中细根生物量的比重降低而深层的比重增加.图3表2参31Fine root biomass and its dynamic characteristics of different aged Casuarina equisetifolia plantations on coastal sandy soil were studied in the Chifu Forestry Farm of Huian County, Fujian. The results showed that the fine roots of 24-year-old plantation accounted for 53.1% of the total underground biomass and 3.8% of the total amount of plantation, respectively; the standing crop of living fine roots increased as the forest aging until its maximum reached 12.373 t hm-2 at 30-year-old, then decreased gradually, while the biomass of dead fine roots increased all along as the forest aging. The fine roots biomass had remarkable correlation with the growth of the above-ground. The fine roots biomass was found with remarkable seasonal dynamics that the standing crop of living and dead fine roots of different aged plantations both were shown with double peaks, while the maximum occurred in different seasons for different aged plantations. The maximum of living fine roots of 5-and 18-year-old plantations occurred separately in January and July, while those of 12-year in March and July. As to the dead fine roots, those of 12-and 18-year occurred in March and July, and 5-year in July and November. The vertical distribution of living and dead fine roots at soil depth was shown with single peak for C. equisetifolia plantations, being concentrated at 0~10 cm, and then decreased with depth gradually. This trend was obvious for 5-year-old plantation. The biomasses of living and dead fine roots at 0~10 cm accounted for 51.9% and 53.3% of the totals, respectively. The 84.6% of living and 82.8% of dead fine roots concentrated at the depth of 0~30 cm. The proportion of fine roots decreased in topsoil and increased in deep soil along with aging of the forest.     STUDY ON THE CONSTRUCTION AND IMMUNOLOGY CHARACTERISITCS of SIX-VA-LENT FUSION TOXIN of FOOD POISONING-BORNE BACTERIA

    [目的]构建4种食源性致病菌融合毒素基因及重组表达载体,制备六联融合毒素的血清抗体。[方法]采用柔性lInkEr序列(g-g-g-g-S)对目的基因进行串联(HblA-VT1b-SEA-VT2b-bOnTAHC-SEb),构建重组表达质粒PET-22b(+)-f6并在E.COlI bl21中进行表达,将表达蛋白纯化后免疫豚鼠制备血清抗体,利用ElISA和琼脂扩散试验验证抗体的特异性与敏感性。[结果]成功构建了重组表达质粒PET-22b(+)-f6并在E.COlI bl21中成功表达,37℃表达蛋白主要以包涵体形式存在(表达量10.2%),基因序列全长3384bP,编码1127个氨基酸,蛋白分子量为127205,测序结果与设计序列一致性为100%。ElISA和琼脂扩散试验表明,融合毒素f6与4种食源性致病菌有良好的反应原性,与多种非目标菌不反应。[结论]成功构建了多联融合毒素基因的表达质粒及制备了抗血清,为利用融合毒素的方法检测食源性致病菌,进而建立食源性致病菌广谱、快速的检测方法奠定基础。[Objective] To construct toxin including fusion toxin gene from food poisoning bacteria and its recombinant expression vector,and then prepare serum antibody of Six-valent fusion toxin.[Methods] Six gene(HblA—VT1B—SEA— VT2B—BoNTaHc—SEB)fragments were connected by SOE-PCR via linker sequence encoding five amino acids(G-G-G-G-S),the recombination expression plasmid pET-22b(+)-F6 was constructed and expressed in E.coli BL21.The expression proteinum was purified,and the blood serum was prepared by the immune cobaya,The specificity and sensitivity of antibody were verified by the ELISA and agar diffusion reaction.[Results] The fusion gene F6(HblA—VT1B—SEA—VT2B—BoNTaHc—SEB)and re-combinant plasmid pET-22b(+)-F6 was successfully constructed.The most of the 37℃ expression proteinum showed to be cyt-orrhyctes(expreesion amounted to 10.2%).The sequence encoding the mature fusion protein of the F6 toxin gene was 3384bp,encoding 1127 amino acids.The molecular weight of recombinant fusion toxin protein was 127.205 ku.The result of sequencing was consistent with predicted gene sequences.The results of ELISA and agar diffusion reaction demonstrated that fusion gene F6 and other four kinds of food poisoning-born bacteria had favorable reactionogenicity and did not response to many other non-tar-get bacteria.[Conclusion] The expression plasmid of multi-valent fusion toxin was successful constructed and antiserum was prepared,which laid the foundation for establishing broad spectrum and rapid detection method for food poisoning bacteria by utilization of fusion toxin detection methods.国家自然基金资助项目(30671762);厦门市科技计划项目(3502Z20055009

    建设世界一流大学需要高度重视本科教育。\"推进一流本科教育,提高人才培养质量\"的前提是要理解本科教育的本意,从不同维度反思\"我们需要什么样的本科教育\"\"什么是好的本科教育\"\"如何推进一流本科教育\"等问题。从宏观层面看,本科教育的使命是为了促进每一个大学生\"成才\"和\"成人\"。从现实需求看,一流本科教育不可忽视跨学科人才培养,需要按照学科标准、职业标准和学生标准对专业进行重新分类,为跨学科专业设置、新工科建设以及高校专业设置与调整开辟理论空间。目前,我国本科教育的最大问题是培养目标不明确,面临学术型与应用型的抉择。好的本科教育需要基于本科生\"关键能力\"的培养,彻底消除他们对于专业的陌生感,满足他们对于理智的好奇心,撤除横亘在大学与社会之间的藩篱并激发他们的创新创业精神。具体来说,一流本科教育是一流\"教\"与\"学\"的统一,需要一流的投入和一流的教育管理,需要在教学改革中侧重技术层面的突破。一流本科教育不是一流大学的\"专利\",不是\"一类本科\",没有固定模式,更不能盲目追求学术GDP。对于应用型本科高校而言,在\"一流本科教育\"中要结合学校特点和办学实际,做出异于\"双一流\"建设高校的现实抉择。作为一类特殊性质的高校,中外合作大学在建设一流本科教育的过程中,要在引入国外优质高等教育资源的基础上,强调对相关资源的消化、吸收、融合、创新,从而培养出符合社会主义现代化建设要求的高素质国际化人才。国家社会科学基金教育学一般项目“新时代中国特色高等教育话语体系构建研究”(BIA180200);;教育部国别和区域研究专项资金资助课题“高等学校专业设置规划与质量保障体系研究

    【中文文摘】传统上调试PLC通常采用手持式PLC编程器,且只能适用于相应型号的PLC,上位计算机上的编程软件无法适用于现场实时调试、编辑程序.本系统在分析PLC编程器和编程软件的功能上,通过系统分析,设计出适合PDA的模块.它采用PDA作上位机,嵌入式VB为开发工具,开发出相应的微型GUI和上层应用软件.该系统可用PDA代替原有的手持式PLC编程器,具有初始化内存、编辑调试程序、监视PLC运行等功能.其既具有PLC编程器的灵活方便的优点,又具有上位计算机易于编程、方便调试且应用范围广的优点. 【英文文摘】Traditionally,the Programming Console is used to debug PLC,but it can only be used for the corresponding CPU Unit.Similarly,it is not adapt to debug and edit the program for real time operation using PLC program system on host computer.For the above reason,a new system was designed and invented based on the function in the Programming Console and PLC program system.In the system,PDA was used as host computer,Embedded Visual Basic as a developing tool,on the side,the relevant interface drivers,micro GUI and app liance software were also developed. The system can substitute PDA for the Programming Console. It could test and debug PLC, including initializing memory, checking and modifying the user programs, adn monitoring programs in different disp laymodes. In conclusion, the system has the advantages of flexibility of the Programming Console, and conveniences for debugging and editing the program of host computer together with wide application

    变构效应是生命体实现复杂生理效应的精妙调控和准确表达的重要途径.本研究利用双硫键的光致翻转特性,以双硫键为变构“枢纽“单元,设计、合成了一种具有非线性响应的光驱动变构荧光分子探针.合成的分子探针dSSb经过光照后,因双硫键翻转而形成刚性的分子内氢键网络,探针溶液的荧光显著增强;同时,由于阴离子的氢键活性,f-、ACO-和H2PO4-等阴离子的存在将促使氢键网络重组而呈现出不同的荧光响应.因此,该探针可实现对3种阴离子的分辨性荧光传感,并具有非线性响应的特性.Allosterism is an important strategy for accurate regulation and expression of complex physiological effects.Taking advantage of the light-driven rotation activity of disulfide bond,a novel allosteric fluorescent probe is developed based on a disulfide receptor.After irradiation,the probe(DSSB)displays remarkable fluorescence enhancement in solution due to the formation of intramolecular hydrogen-bonding networks.Furthermore,the addition of different anions,such as F-,CH3COO-and H2PO4-,will result in different fluorescence responses due to their characteristic disturbances on the hydrogen-bonding format.All the three anions can be recognized by the probe with nonlinear fluorescent responses.国家自然科学基金(21175113

    Application of improved development dominance ratio model of cultivated land resources in sub-humid areas——A case study of Changwu County

    针对以往耕地资源开发优势度模型在计算耕地生产潜力时存在的不足,结合长武县农业耕作的具体特点对该模型在半湿润区适用的科学依据及机理进行了分析,提出了改进耕地资源开发优势度模型,计算了该县耕地资源优势度,并做了进一步分析验证。结果表明:1990、2008年该县耕地资源优势度分别为0.886和0.836;耕地面积从1990年的2.234万hm2减少至2008年的1.146万hm2,18年间共减少了1.088万hm2,减少量达到1990年总面积的49%;粮食播种面积比例从1990的81%上升到了2008年的84%;耕地面积的减少和粮食播种面积比率的提高导致了该县耕地资源优势度的下降。改进后的耕地资源开发优势度模型较好地拟合了实际耕地面积和粮食作物播种面积比重的变化情况
