    Cloning and expression analysis of lipopolysaccharide-induced TNF-α(LITAF) of Japanese scallop (Mizuhopecten yessoensis)

    脂多糖诱导的肿瘤坏死因子(lIPOPOlySACCHArIdE-InduCEd Tnf-AlPHA fACTOr,lITAf)是一类重要的炎症细胞因子,在先天性免疫系统中发挥重要的介质作用。文章根据虾夷扇贝lITAf基因EST序列,应用rACE技术克隆了虾夷扇贝lITAf全长CdnA,对序列及编码的氨基酸进行生物信息学分析。结果显示,该基因CdnA全长1 551 bP,其5′非编码区包含76 bP,3′非编码区包含1 001 bP;开放阅读框(Orf)为474 bP,编码157个氨基酸,氨基酸序列中存在一个保守的lITAf结构域;理论分子量16.99 kdA,等电点为6.24。lITAf基因序列为3 698 bP,由3个外显子和两个内含子组成。利用实时荧光定量PCr技术分析lITAf在虾夷扇贝不同组织、不同胚胎发育阶段以及鳗弧菌(VIbrIO AnguIllAruM)刺激后各时间段的表达情况。结果表明:lITAf基因在所检测的6个成体组织中均有表达,其中肾脏的表达量最高;胚胎发育的7个时期中,担轮幼体时期表达量最高;菌刺激36 H实验组与对照组的表达量差异大。lITAf基因是lITAf家族的一员,推测lITAf基因参与虾夷扇贝的先天性免疫反应。The lipopolysaccharide-induced TNF-alpha factor(LITAF) is an inflammatory cytokine,which plays an important role in innate immunity system.Based on the expressed sequence tag(EST) of Japanese scallop(Mizuhopecten yessoensis),the cDNA of LITAF gene was amplified using rapid amplification of cDNA ends(RACE) approach.Results showed that the full-length cDNA of LITAF is 1 551 bp consisting of a 5′ untranslated region(UTR) of 76 bp,a 3' UTR of 1 001 bp,and an open reading frame(ORF) of 474 bp encoding a polypeptide of 157 amino acids,and there is a conserved LITAF domain in amino acid sequences.The estimated molecular mass is 16.99 kDa and the theoretical isoelectric point is 6.24.The total length of LITAF is 3 698 bp,which includes three exons and two introns.Real-time quantitative PCR was carried out to measure LITAF mRNA expression in adult tissues and monitor mRNA expression patterns during embryonic development after bacteria(Vibrio anguillarum) challenged.The expression level of LITAF mRNA was detected in all the adult tissues with the highest in the kidneys.The trochophore owns the highest expression level of LITAF in embryonic development.LITAF expression showed significant difference(P<0.01)between the control and bacteria challenged specimens at 36 h.These results suggest that the LITAF should be a member of the LITAF family that perhaps involved in the innate immune response of Japanese scallop.国家海洋公益性行业科研专项(编号:200805037);国家自然科学基金项目(编号:31140073)资

    分子筛修饰电极中内电子传输机理的研究

    【中文摘要】用电化学方法聚合分子筛孔道内的苯胺 , 以聚苯胺分子筛修饰电极为模型研究分子筛修饰电极的内电子传输机理 . NaY分子筛的离子交换点位被苯胺修饰后 , 通过电聚合制得聚苯胺分子筛修饰电极 ( Pan+Y-ZM E ),该电极通过聚苯胺链自身的电子跳跃来实现电子传输 , 且只有通过阴极富集后对溶液中Cd 2+才有响应 ,并能用于测定抗坏血酸。【Abstract】This paper is concerned with the mechanism of intrazeolite electron transfer in zeolite-modified electrodes (ZMEs) where the zeolite have been modified with polyaniline, First, data were obtained from ZMEs prepared with zeolite-Y loading aniline and the electropolymerizing of anilines was performed by cyclic voltammery, Polyaniline might not leak out from zeolite due to the bulk size of polyaniline being bigger than the channel of zeolite-Y. At the absence of electron transfer mediators, the Pan(+)Y-ZME showed the expected presence of electron hopping of adjacent polyaniline cations, Second, the data were obtained for the determining Cd2+ and ascorbic acid using Pan+Y-ZME. The Cd2+ might not be preconcentrated at open circuit and could be done at constant potential. At the absence of ion exchange between zeolite and surrounding electrolyte the electron transfer was performed via the electron hopping, i. e. by an intrazeolite mechanism; rather than by an extrazeolite mechanism. In addition, the ascorbic acid molecule and its anion could not cross zeolite film to base electrode therefore, its current was conducted also by the electron hopping between the chains of polyaniline, Experimental results indicate that the electron transfer of Pan(+)Y-ZME was dominated by intrazeolite processes

    与南海西边界流有关的区域海洋学进展

    从动力学角度,回顾了与南海西部主流系及其涡旋研究有关的南海物理海洋学的进展.主要内容涉及南海西边界流漂流浮标观测、西边界流源区涡旋遥感观测、西边界流源区跨陆架交换、涡流相互作用、西边界流年际变化、西边界流区海气相互作用、南海贯穿流等方面的进展.西边界流是一个强流系,其与季节和年际变化相关的结构特征、变率及跟某些动力过程的关联有待研究.在西边界流变率、涡旋环流相互作用、海气过程以及南海贯穿流取得了以下成果:(1)利用漂流浮标观测样本对南海西边界环流进行分析,指出南海西边界表层环流在到达越南中部沿岸后伴随着流幅变窄的同时流速加强;探讨了南海北部环流变化机理,定量诊断南海西边界流北支冬季逆风流产生的动力机制;利用航次数据对18°n断面的经向地转流进行诊断,表明南海西边界流的经向输送年际变化明显;(2)结合航次观测数据,对2003/2004年冬季南海北部2个反气旋涡旋和2007年夏季18°n附近的3个反气旋涡旋进行研究,指出冬季2个涡旋产生后以罗斯贝(rOSSby)波速度(约0.1M/S)沿北部陆坡向西南方向传播,并初步揭示了南海西边界环流与夏季3个涡旋的相互作用;南海北部陆架区涡旋西南向传播最大(最大为0.09M/S),而越南以东海域涡动能(EkE)最大,这都说明涡旋活动与南海西边界流存在强的相互作用;(3)发现南海西边界流附近海表面温度(SST)强的季节内振荡特征,进一步研究表明此区域冬季SST季节内变化会使潜热季节内信号减弱20%;(4)探讨了南海贯穿流的长期变化特征以及与整个太平洋环流系统的相互关联.国家自然科学基金重点项目(40830851); 国家重点基础研究发展计划(2011CB403504); 中国科学院近海海洋观测研究网络——西沙南沙海洋观测研究站建设项目(KZCX2-EW-Y040)资

    泛素家族包括泛素及类泛素蛋白,约20种成员蛋白.近年来,泛素家族领域取得了迅猛发展,并已与生物学及医学研究的各个领域相互交叉.泛素家族介导的蛋白质降解和细胞自噬机制的发现分别于2004和2016年获得诺贝尔奖.但是,类泛素蛋白并没有统一规范的中文译名. 2018年4月9日在苏州召开的《泛素家族介导的蛋白质降解和细胞自噬》专著的编委会上,部分作者讨论了类泛素蛋白的中文命名问题,并在随后的\"泛素家族、自噬与疾病\"(Ubiquitinfamily,autophagy anddiseases)苏州会议上提出了类泛素蛋白中文翻译草案,此草案在参加该会议的国内学者及海外华人学者间取得了高度共识.冷泉港亚洲\"泛素家族、自噬与疾病\"苏州会议是由美国冷泉港实验室主办、两年一度、面向全球的英文会议.该会议在海内外华人学者中具有广泛影响,因此,参会华人学者的意见具有一定的代表性.本文介绍了10个类别的类泛素蛋白的中文命名,系统总结了它们的结构特点,并比较了参与各种类泛素化修饰的酶和它们的生物学功能.文章由45名从事该领域研究的专家合作撰写,其中包括中国工程院院士1名,相关学者4名,长江学者3名,国家杰出青年科学基金获得者18名和美国知名高校华人教授4名.他们绝大多数是参加编写即将由科学出版社出版的专著《泛素家族介导的蛋白质降解和细胞自噬》的专家

    2005年2月中下旬,高岚河库湾发生拟多甲藻水华,本文研究了水华爆发期间藻类的生消过程及其基本组成,并分析了环境因子在水华形成和消亡过程中的作用机制。研究结果表明:超高的营养盐,如TN、TP等是此次水华发生的主要原因之一,其他物理条件,如气温、水温的回升等是重要的诱因。而水华消亡的原因是营养盐被不断繁殖的甲藻消耗,以至于不能满足甲藻的营养需求,最终部分甲藻下沉到底层

    大幅度提高石油采收率对于稳定我国东部油田产量具有十分重要的现实意义。依靠地层能量开采的一次采油和以注水为主的二次采油技术所能达到的最终采收率一般不超过 40%,这意味着水驱后仍有一半以上石油储量未能采出。为了提高原油的最终采收率,我国石油工业界发展了以化学驱和热力采油为主要代表的三次采油技术。化学驱采油方法在我国油田实践应用中取得了较好的效果,但也出现了大量需要解决的理论和工程问题,这些问题的解决对化学驱方法在油田的进一步推广和应用有着重要意义

    Research on Impact of Trade Openness on Wages of China's Manufacturing Workers: Evidence from Sampling Survey Data

    本文使用高度细化的关税数据和大样本人口抽样调查数据,实证研究了贸易开放对我国制造业工人的工资水平和技能溢价的影响。研究发现:在控制住个体微观特征、产业特征及地区特征后,贸易开放对我国制造业工人的工资水平有显著的促进作用;我国制造业的贸易开放对工人技能溢价有显著的降低效应;教育对我国制造业工人工资有显著的促进作用,国有单位工人获得更高工资,女性受到显著的性别歧视,同工不同酬现象明显存在。Against the background of Chin's great tariff reduction after its entry into WTO, this paper utilizes both micro-level tariff data and the sampling survey data in 2005 to empirically investigate the impact of trade openness on the wage level and skill premium of China's manufacturing workers.The result shows that:(1) trade openness significantly promotes wage level of China's manufacturing workers when characteristic factors of workers, industries and regions are controlled;(2) trade openness significantly reduces the skill premium among China's manufacturing workers;(3) workers of higher education level and workers in state-owned enterprises earn more than others, and female workers are suffering from significant gender discrimination in China.To narrow the income distribution gap in China, more attention should be given to the reduction of various monopoly barriers and gender discrimination than trade openness.教育部哲学社会科学研究重大课题攻关项目(项目批准号13JZD010); 中央高校基本科研业务费专项资金资助(项目编号:T2013221017

    Studies on the Hydrogen Production of immobilized Klebsiella oxytoca HP1

    为了保持菌体固有的生物活性,使其重复使用,将产酸克雷伯氏菌(HP1,Klebsiella oxytoca)包埋在凝胶中,采用注滴法制备海藻酸钙凝胶球来固定产酸克雷伯氏菌HP1,达到截留菌体产氢的目的.结果表明,选择培养6~8 h的菌体作为海藻酸钙凝胶球包埋的菌体,Tris做培养基中的缓冲盐,当体积质量为5 g/L时,最佳产氢初始pH值为9.0,温度为37℃,采用柱反应器连续产氢7 d,平均产氢速率为3.38×10-2 moL(/L.h)-1.Biological H2-production is considered as an important and economic way to produce hydrogen in the future for energy supply.Ca-alginate gel beads with Klebsiella oxytoca HP1 were prepared by dripping method,so it is easy to keep bacterium and continue to produce hydrogen.The result of experiment showed that choosing the germ body of 6~8 h as the germ fixed by Ca-alginate gel beads and Tris as the buffer the density of which is 5 g/L.The optimal conditions for hydrogen production were: initial pH=9.0,ferment temperature 37 ℃.The average rate of H2-production was achieved as 33.8 mmol/L·h,during a 7days continuous hydrogen production
