Import 29/09/2010Bakalářská práce se zabývá návrhem diagnostického a údržbařského systému. V úvodu práce jsem popsal jednotlivé metody technické diagnostiky aplikovatelné na kolesová rypadla. Popsal jsem zde jednotlivé metody zkoušek využívaných k zjištování technického stavu. Další část práce je věnovaná rozpracování na jednotlivé konstrukční uzly velkostroje. Jsou zde uvedeny měřící přístroje a software, které jsem navrhl k aplikaci. Dále jsem zde uvedl základní informace pro organizační schéma návrhovaného systému údržby.The bachelor thesis deals with the diagnostic and maintenance system. At the prelimiary thesis I described individual methods of technical diagnostic applicable on the wheel excavator. I described the individual test methods here applicable to solving technical condition. Next part is devoted work out on the individual structural units of giant machine. I said there aparaturs and software, which I proposed to the application. Proposal of the manufacturing process of construction of a dwelling house

Import 29/09/2010Náplní této práce bylo řešit projekt obytného domu v kvalitě požadované pro stavební povolení. Práce se skládá z částí technologické a pozemní stavby. V části pozemní stavby je řešena technická zpráva a výkresová část s vybranými detaily. V technologické části je řešen rozpočet stavby s harmonogramem výstavby a technologický postup dílčí etapy, zaměřený na montáž krovu. Stavba je podsklepená se třemi nadzemními podlažími a zastřešená krove. Stavba je založena na železobetonových pasech, vyzděná z cihel typu POROTHERM. Projekt stavby je zasazen do zastavěného území v Ostravě – Porubě na ulici Polská.The aim of this work is an appartment house project solution on the level required for the building permit. The work consists of two parts – a technological part and a structural engineering. The second mentioned part – the structural engineering – solves the problem of a technical report and also specific drawings with some detailed parts. The technological part deals with a building budget along with a building harmonogram and a technology of the particular building phases, focused on the roof truss assemblage. The building includes a basement, three living floors and a roofeed truss. The building is founded on the steel concrete stripfoundation and bricked up as a Porotherm system. Import 03/08/2012Práce se zabývá technickou diagnostikou, konkrétně sledování technického stavu s využitím dvou systémů pro vibrační diagnostiku. Jedná se o on-line systém ifm octavis efektor a pochůzkový systém SKF Microlog/Prism. V teoretické části práce popisuji vibrační diagnostiku, její nástroje, vyhodnocování a normy. Teoretická část je zaměřena na popis jednotlivých systémů, jejich ovládáním s využitím při praktickém měření na papírenském stroji.The work deals with the technical diagnostics, concretely monitoring the technical condition of using two systems for vibration diagnostics. Its the on-line system ifm octavis effector and a foot system SKF Microlog/Prism. The theoretical part describes the vibration diagnostics, its tools, evaluation and standards. Design of digital processing of financial statements for a logistics company

Diplomová práce se zabývá procesy digitalizace účetní závěrky logistické společnosti a snaží se odpovědět na otázku, zda a případně za jakých podmínek je možné implementovat principy digitalizace závěrek v této společnosti. Hlavním požadavkem na cílové řešení je nulový zásah do hlavního systému společnosti. Práce je rozdělena do tří hlavních částí. V první části jsou čtenáři nastíněny nynější procesy účetní závěrky společnosti. Poté je představeno technické řešení a závěrem práce je popis jednotlivých možností řešení a finanční odhad jejich implementace.The diploma thesis deals with the processes of digitalization of financial statements of a logistics company and tries to answer the question if and under what conditions it is possible to implement the principles of digitalization of financial statements in this company. The main requirement for the target solution is zero interference in the company's main system. The thesis is divided into three main parts. In the first part, the reader is given an outline of the company's current financial statement processes. Then the technical solution is presented, and the thesis concludes with a description of the different solution options and a financial estimate of their implementation

Import 27/01/2012Náplní této práce je řešení projektu pro realizaci stavby Administrativní budovy v požadované kvalitě. členění práce je provedeno do šesti částí. V první části je řešena výkresová dokumentace stavby v rozsahu dle zadání spolu s projektovou dokumentací stavby. V druhé části se provádí porovnání variantního řešení výkopových prací (zajištění stěny pomocí svahování, pažení) a jejich porovnání z hlediska finanční náročnosti. V třetí části je řešen technologický postup s návrhem pracovní sestavy pro obě varianty. Ve čtvrté části je řešen rozpočet stavby, v páté části je řešen harmonogram výstavby a v poslední šesté části jsou řešeny Zásady organizace výstavby. Objekt je řešen jako monolitický železobetonový sloupový skelet s výplňovým zdivem typu Porotherm a zastřešen pomocí ploché střechy. Objekt má jedno podzemní podlaží, ve kterém je fitness centrum a čtyři nadzemní, ve kterých jsou kancelářské prostory.The scope of this work is the project for the Construction Office buildings on the level required quality. Breakdown of work is done in six parts. The first part dealt with construction drawings to the extent to specification together with the project documentation of the building. In the second part of the comparison is made variant solution excavation work (securing walls with sloping, sheeting) and their comparison in terms of financial cost. In the third part is solved technological process proposed by the Task reports for both variants. In the fourth part is solved by the construction budget, in the fifth part is solved by the construction schedule and the last sixth part are dealt with principles of construction organizations. The building is designed as a monolithic reinforced concrete column frame with masonry infill type Porotherm and roofed a flat roof. Specifics of financial management of non-profit organizations: case study Cesta domu

The aim of this bachelor thesis is to define the specifics of financial management of non-profit organizations and their differences from the commercial sector with a closer focus on the non-profit organization called Cesta domů. The bachelor thesis consists of two main parts – theoretical and practical. The first methodological part describes the individual financial indicators of non-profit organizations. In the analytical part, these indicators are analyzed on a case study of the non-profit organization Cesta domů. The quantitative research approach in the form of a case study was used as a research method. The result of the work is a comparison of these two sectors and a subsequent recommendation for new non-profit organizations in this sector.Cílem této bakalářské práce je definovat specifika finančního řízení neziskových organizací a jejich odlišností od komerčního sektoru s užším zaměřením na neziskovou organizaci Cesta domů. Bakalářská práce se skládá ze dvou hlavních částí – teoretické a praktické. Úvodní metodická část popisuje jednotlivé finanční ukazatele neziskových organizací. V analytické části jsou následně rozebrány tyto ukazatele na případové studii neziskové organizace Cesta domů. Jako výzkumná metoda byl použit přístup kvantitativního výzkumu formou případové studie. Výsledkem práce je porovnání těchto dvou sektorů a následné doporučení pro nově vznikající neziskové organizace v tomto odvětví

Myeloid - derived suppressor cells in Type 1 diabetes are an expanded population exhibiting diverse T-cell suppressor mechanisms.

Myeloid-derived suppressor cells (MDSC) represent a heterogeneous group of immature myeloid cells with immunoregulatory function in cancer and autoimmune diseases. In humans, two subsets of MDSC were determined based on the characteristic surface markers, monocytic MDSC (M-MDSC) and granulocytic MDSC (G-MDSC). Expansion of MDSC has been reported in some murine models and patients with autoimmune diseases and their immune-suppressive properties were characterized. However, the exact role of MDSC in the pathogenesis of autoimmune diseases is more complex and/or controversial. In type 1 diabetes mellitus (T1D), the increased frequency of MDSC was found in the blood of T1D patients but their suppressor capacity was diminished. In our study, we assessed the role of M-MDSC in the pathogenesis of T1D and showed for the first time the increased frequency of M-MDSC not only in the blood of T1D patients but also in their at-risk relatives compared to healthy donors. T1D patients with inadequate long term metabolic control showed an elevation of M-MDSC compared to patients with better disease control. Furthermore, we described the positive correlation between the percentage of M-MDSC and Th17 cells and IFN-γ producing T cells in T1D patients and their at-risk relatives. Finally, we found that the ability of M-MDSC to suppress autologous T cells is efficient only at the high MDSC: T cells ratio and dependent on cell-cell-contact and TGF-β production. Our data show that the engagement of MDSC in the pathogenesis of T1D is evident, yet not entirely explored and more experiments are required to clarify whether MDSC are beneficial or harmful in T1D
