Very strong foehn winds in the Tatra Mountains (Polish Carpathian Mountains) : causes, course and consequences

The foehn wind in the Tatra Mountains is referred to as "halny". It blows from the south and often causes significant temperature rises and drops in air humidity on the leeward side of the mountain range. It is accompanied by strong and abrupt gusts which may cause damage to the forest stand and infrastructure. This paper looks at two cases of very strong foehn winds which occurred in the Tatra Mountains and the Podhale Region in May 1968 and December 2013. It compares the synoptic and weather conditions which accompanied the phenomenon, including wind speed and direction, as well as relative air temperature and humidity. The foehn wind recorded in May 1968 is sometimes referred to as the "wind of the century". Wind gusts at the Tatra peaks reached a speed of 288 km-h^{-1}. In December 2013 the wind had less force at Kasprowy Wierch than in Zakopane, but it caused similar amount of damage to the forest stand as the wind recorded in May 1968

Structural differences between C-terminal regions of tropomyosin isoforms

Tropomyosins are actin-binding regulatory proteins which overlap end-to-end along the filament. High resolution structures of the overlap regions were determined for muscle and non-muscle tropomyosins in the absence of actin. Conformations of the junction regions bound to actin are unknown. In this work, orientation of the overlap on actin alone and on actin–myosin complex was evaluated by measuring FRET distances between a donor (AEDANS) attached to tropomyosin and an acceptor (DABMI) bound to actin’s Cys374. Donor was attached to the Cys residue introduced by site-directed mutagenesis near the C-terminal half of the overlap. The recombinant alpha-tropomyosin isoforms used in this study – skeletal muscle skTM, non-muscle TM2 and TM5a, and chimeric TM1b9a had various amino acid sequences of the N- and C-termini involved in the end-to-end overlap. The donor-acceptor distances calculated for each isoform varied between 36.4 Å and 48.1 Å. Rigor binding of myosin S1 increased the apparent FRET distances of skTM and TM2, but decreased the distances separating TM5a and TM1b9a from actin. The results show that isoform-specific sequences of the end-to-end overlaps determine orientations and dynamics of tropomyosin isoforms on actin. This can be important for specificity of tropomyosin in the regulation of actin filament diverse functions

Dzieła zebrane Zygmunta Krasińskiego – elektroniczna edycja naukowa

The new edition of Zygmunt Krasiński’s Dzieła literackie – apart from its printed version – will be enriched with a digital edition containing the author’s works, critical apparatus and other materials such as the dictionary of language, pictures, links to texts in digital libraries. Why is such a novelty needed? While working on different editions of scholarly writing or scholarly and critical works, it is crucial to be aware of the coexistence of electronic and paper media in our culture and their influence on our lives. Therefore, in order to meet the expectations of prospective readers, we intend to create a new portal allowing to access Krasiński’s works and including additional tools which will enable to take full advantage of the new edition. However, in order to create such an edition it is necessary not only to combine, plan and coordinate the efforts of many individuals, but also to manage numerous problems. The article only indicates some issues requiring attention during working on a digital scholarly edition and introduces basic rules applied to the discussed edition.Powstająca edycja Dzieł literackich Zygmunta Krasińskiego poza papierową, tradycyjną wersją, będzie wzbogacona w wydanie cyfrowe, zawierające teksty romantyka, aparat krytyczny oraz pozostałe przygotowane materiały: słownik języka, fotografie czy odnośniki do tekstów w bibliotekach cyfrowych. Skąd potrzeba takiego rozwiązania? Otóż opracowując współcześnie kolejne wydania prac naukowych lub naukowej edycji krytycznej, należy mieć świadomość koegzystowania w naszej kulturze mediów elektronicznych i papierowych i naszego współdziałania z nimi. Stąd, aby wyjść naprzeciw przyszłemu czytelnikowi, zostanie stworzony portal zawierający teksty w nowym opracowaniu oraz dodatkowe narzędzia, będące dopełnieniem i kompleksowym przedstawieniem dzieł literackich Krasińskiego, dające możliwości wykorzystania nowej edycji. Jednak, aby takie wydanie powstało, trzeba połączyć, zaplanować i skoordynować wysiłki wielu osób oraz uporać się z wieloma problemami. Artykuł jedynie zasygnalizuje pewne kwestie, z którymi należy się zmierzyć, myśląc o elektronicznej edycji naukowej oraz przedstawia podstawowe założenia wydania

Synthesis of novel isothiazolopyridines and their in vitro evaluation against Mycobacterium and Propionibacterium acnes

AbstractIn this paper we describe synthesis, structures and some physicochemical properties of 20 isothiazolopyridines 8–13 substituted differently into an isothiazole ring as well as their in vitro antibacterial assays against Mycobacterium tuberculosis H37Rv, Mycobacterium fortuitum PCM 672 and Propionibacterium acnes PCM 2400. Compound 13a was found to be the most active derivative against M. tuberculosis H37Rv, demonstrating 100% growth inhibition of microorganisms in the primary screen (minimum inhibitory concentration [MIC] 6.25μg/mL). Nineteen of the prepared compounds were evaluated against M. fortuitum PCM 672 and P. acnes PCM 2400 and only compounds 9 and 12d exhibited excellent activity against individual strains of microorganisms with MIC90 <1μg/mL. The inhibitory action of the remaining isothiazolopyridines towards the tested strains of the microorganism was low, absent, or a non-linear correlation prohibited accurate determination of MIC values. Unexpectedly, seven of the remaining isothiazolopyridines tested against M. fortuitum and P. acnes stimulated growth of the microorganisms in the range 10–50% or even more (10b) under experimental conditions

Morphology of Mitochondria in Syncytial Annelid Female Germ-Line Cyst Visualized by Serial Block-Face SEM

Mitochondria change their morphology and distribution depending on the metabolism and functional state of a cell. Here, we analyzed the mitochondria and selected structures in female germ-line cysts in a representative of clitellate annelids – the white worm Enchytraeus albidus in which each germ cell has one cytoplasmic bridge that connects it to a common cytoplasmic mass. Using serial block-face scanning electron microscopy (SBEM), we prepared three-dimensional ultrastructural reconstructions of the entire selected compartments of a cyst at the advanced stage of oogenesis, i.e. the nurse cell, cytophore, and cytoplasmic bridges of all 16 cells (15 nurse cells and oocyte). We revealed extensive mitochondrial networks in the nurse cells, cytophore and mitochondria that pass through the cytoplasmic bridges, which indicates that a mitochondrial network can extend throughout the entire cyst. e dynamic hyperfusion state was suggested for such mitochondrial aggregations. We measured the mitochondria distribution and revealed their polarized distribution in the nurse cells and more abundant accumulation within the cytophore compared to the nurse cell. A close association of mitochondrial networks with dispersed nuage material, which seems to be the structural equivalent of a Balbiani body, not described in clitellate annelids so far, was also revealed
