    A Systemic comparison of different chimeric antigen receptor (car) designs for retargeting of NK-92 cells against tumor antigens

    Cancer immunotherapies focus on the power of the immune system to attack tumor cells. Recently, Chimeric Antigen Receptors expressing T cells (CAR-T cells) have received clinical approval for antigen-specific adoptive immunotherapy against CD19 in B cell malignancies. CAR vector designs have dramatically developed since their initial discovery and now include first-generation CARs (CD3ζ-based CAR), second-generation CARs with additional costimulatory domains such as CD28 or CD137 and third generation CARs (CD3ζ with two costimulatory domains) and recently fourth generation CAR with a transgene for cytokine stimulation. Natural Killer (NK) cells have ability recognize the tumor cells by their native receptors and have grown to be promising candidates for adoptive immunotherapy of cancer. CAR expression in NK cells is also clinically tested and carries the potential to translate into clinical application but the majority of literature on CAR vector design relies on observations from T cells. This thesis aims to use NK-92 cells for evaluation of different designs in order to optimize a CAR vector that could be efficiently used to retarget NK cells against tumor antigens. CAR transgenes comprising identical antigen binding domains that target CD19, combined with different intracellular signaling domains (CD3ζ , CD28 and CD137) are transferred to NK-92 cells via the use of lentiviral vectors. Cytotoxic activity and antigen-specificity of CAR-NK-92 cells are evaluated against the CD19- classical NK cell target K562 cell line and the CD19+ cell line Daudi and Namalwa by analysis of degranulation and cytokine secretion. Our results provide valuable data for optimal CAR vector design in NK cell

The most important criteria for yogurt production is the selection of starter cultures since each culture affects the end-product quality differently. Our natural flora and traditional flavor have been continously changing due to the introduction of imported commercial starter cultures. Because of the necessity to preserve our natural starter cultures and to increase the availability of them for industrial use, these cultures must be isolated from artisanal yogurts, genetically characterized and investigated regarding their technological properties. Aims of our study were to determine the technological and organoleptic properties of starter cultures previously isolated and genetically characterized and to assess the feasibility of near-infrared (NIR) spectroscopy for determining the yogurt quality.Total of 20 different yogurt samples were produced using combinations of 5 Streptococcus thermophilus and 4 Lactobacillus delbrueckii ssp. bulgaricus strains. Yogurt samples were stored at 4 °C for 21 days. Chemical, physical, microbial and organoleptic analyses for samples were conducted at predetermined days.As a conclusion, it was found that 4 Streptococcus thermophilus and 4 Lactobacillus delbrueckii ssp. bulgaricus yogurt isolates have potential to be used in dairy industry regarding their high technological and organoleptic characteristics. In addition, NIR spectroscopy technique could be used successfully on yogurt samples to predict total solids, fat, pH, syneresis, and microbial counts of yogurts based on the calibration models (Genetic Algoritm)

    An Unconventional Example of Big Data: BIST-100 Banking Sub-Index of Turkey

    Resumen de la comunicación[EN] This paper applies Big Data concept to an emerging economy stock exchange market by examining the relationship between price and volume of the Banking index in BIST-100. Stock markets have been commonly analyzed in big data studies as they are one of the main sources of rich data with recordings of hourly and minutely transactions. In this sense, nowcasting the economic outlook has been related to the fluctuations in the stock exchange market as news from companies open to public became important sources of changes in expectations for economic agents. However, most of the previous studies concentrated on the main stock market indices rather than the major sub-indices. This study covers the period 13 December 2017 – 12 March 2018, with minute data and approximately 31000 observations for each of the 11 bank stocks. The effects of stock market movements on exchange rates and interest rates are also examined. The methodologies used are frequency domain Granger causality of Breitung and Candelon (2006) and wavelet coherence of Grinsted et al. (2004). The main finding is the supremacy of the banking index as it seems to have great influence on economic fluctuations in Turkish economy through other high frequency variables and the households’ expectations.Çelik, S.; İşbilen, E.     The Role of Supply Chain Transparency among Supply Chain Analytics Capabilities

    Nowadays, to take advantage the huge amounts of data collected, companies have started to adapt analytics tools to useful insight their operations and their service level. Such companies attempt to learn from collected data about current and historical processes to define future operational decisions. Even though the importance of supply chain analytics and data-driven decision-making capabilities is recognized by companies, there is a lack of empirical studies. This study demonstrates the role of supply chain transparency (SCT) in the relation between analytics capabilities. The data were gathered from the online survey and were analyzed with partial least squares structural equation modeling (PLS-SEM) using smart-PLS. The results suggest that the existence of significant relation between analytical capabilities in plan, source and make and the mediating effect of supply chain transparency

    The effect of organizational trust on motivation: An application on public hospitals in Erzurum

    The purpose of this study is to measure the effect of organizational trust on motivation. This study was applied to 178 nurses working in Erzurum Palandöken State Hospital, Erzurum Regional Training and Research Hospital and Erzurum Yakutiye Research Hospitals. It is seen that there is a positive relationship between organizational trust and motivation in the result of the study and positively affecting the sub dimensions of motivation and motivation of sub dimensions of this variable. Confidence in managerial trust in sub-dimensions of organizational trust was found to be higher in the degree of motivation of individuals than in other variables. In this context, any work to be done to increase organizational trust seems to trigger internal motivation and increase it.Bu çalışmanın amacı örgütsel güvenin motivasyon üzerindeki etkisini ölçmektir. Bu doğrultuda oluşturulan bu çalışma Erzurum Palandöken Devlet Hastanesi, Erzurum Bölge Eğitim ve Araştırma Hastanesi ve Erzurum Yakutiye Araştırma Hastaneleri’nde çalışan 178 hemşire üzerinde uygulanmıştır. Çalışma sonucunda örgütsel güvenin ve bu değişkenin alt boyutlarının motivasyonu ve motivasyonun alt boyutlarını pozitif yönlü etkilediği ve örgütsel güvenle motivasyon arasında anlamlı bir ilişki olduğu görülmektedir. Örgütsel güvenin alt boyutlarından yöneticiye duyulan güvenin, bireylerin motivasyonlarını etkileme derecesinin diğer değişkenlere göre daha yüksek olduğu tespit edilmiştir. Bu bağlamda örgütsel güveni artırmaya yönelik yapılacak herhangi bir çalışmanın iç motivasyonu tetikleyerek artışına yol açtığı görülmektedir

    Antalya Serik Koşullarında Organik Star Ruby Altıntop Yetiştiriciliğinde Yeşil Gübreleme ve Zeytin Keki Kullanım İmkânlarının Araştırılması

    Bu çalışma ile yeşil gübreleme ve zeytin keki kompostunun organik altıntop yetiştiriciliğinde kullanım olanaklarını araştırmak amaçlanmıştır. Deneme 2006- 2009 yılları arasında Batı Akdeniz Tarımsal Araştırma Enstitüsü Meyvecilik Bölümünün Serik/Kayaburnu arazisinde bulunan Star Ruby Altıntop parselinde yürütülmüştür. Denemede 4 farklı uygulama karşılaştırılmıştır: 1-Konvensiyone1 uygulama (kimyasal NPK gübre uygulaması), 2-Organik-1 : Fiğ+Arpa (8+3)kg/da Zeytin Keki 25kg/ağaç+4kg S/Ağ 3-Organik-2: Fiğ+Arpa (8+3)kg/da Zeytin Keki 50kg/ağaç+4kg S/Ağ, 4-Organik-3 : Fiğ+Arpa (8+3)kg/da Zeytin Keki 75kg/ağaç+4kg S/Ağ Uygulamaların; verim ve bazı meyve kalite özellikleri ile toprak özellikleri ve bitkinin beslenme durumu üzerine olan etkileri incelenmiştir. Meyve verimi değerleri, konvensiyonel uygulamada organik gübrelemeye göre daha yüksek bulunmuştur. Ayrıca inorganik gübre uygulaması sonucu meyve suyu miktarı organik gübre uygulamalarına göre artmıştır. Meyvede tat ve aroma ile yakından ilgili olan meyve suyu asitliği ve suda çözünebilir kuru madde (SÇKM) miktarlarının, organik gübre uygulamalarının etkisiyle inorganik gübrelemeye göre yükseldiği belirlenmiştir. Organik gübre uygulamaları; toprağın tuz ve organik madde içeriğinin yükselmesine neden olmuştur. Bitkilerin azotla beslenme durumu organik uygulamalarda yetersiz düzeyde iken, inorganik gübre uygulamalarında yeterli düzeye ulaşmıştır. Ancak bitkilerin potasyum içeriği, zeytin keki kompostu ve yeşil gübreleme uygulamalarının etkisiyle, inorganik gübre uygulamalarına göre artış göstermiştir. Deneme süresince yapılan incelemeler sonucu, özellikle meyve döneminde unlu bit ve harnup güvesi zararlılarına rastlanmıştır. Unlu bit populasyonu; predatör ve parazitoit salımları ile harnup güvesi populasyonunun da MVP preparatı uygulamaları ile kontrol edilebildiği gözlenmiştir

    Bleeding risk in patients with acute coronary syndrome in a Turkish population results from the Turkish acute coronary syndrome registry (TACSER) study

    Objective Bleeding is one of the most important causes of mortality in patients with acute coronary syndrome (ACS). This study therefore aimed to investigate bleeding risk in patients with ACS who were scheduled to receive dual antiplatelet therapy (DAPT) in Turkey. Methods This was a multicentre, observational, cross-sectional cohort study. The study population included 963 patients with ACS from 12 centres in Turkey. We used the Predicting Bleeding Complication in Patients Undergoing Stent Implantation and Subsequent Dual Antiplatelet Therapy (PRECISE-DAPT) score to predict the bleeding risk for all the patients. The patients were divided into high (>= 25) or low (= 25). Compared with the male patients, the female patients had higher PRECISE-DAPT scores (28.2 +/- 15.7 vs 18.4 +/- 13.6,P = 25 was 53%, while among the male patients, the score occurred at a rate of 22%. The female patients had lower haemoglobin (Hb) levels than the male patients (12.1 +/- 1.7 vs 13.8 +/- 1.9,P < .001) and lower creatinine clearance (70.7 +/- 27.5 vs 88.7 +/- 26.3,P < .001). The in-hospital bleeding rates were higher among the patients with high PRECISE-DAPT scores than among those who did not have high scores. Furthermore, the patients with high PRECISE-DAPT scores had a higher in-hospital mortality rate compared with those with low PRECISE-DAPT scores (1% vs 0%,P = .11). Conclusions The mean PRECISE-DAPT score was high among the patients with ACS in this study, indicating that the bleeding tendency was high. This study showed that the PRECISE-DAPT score may help physicians determine the type and duration of DAPT, especially in patients with ACS in Turkey

    Antibacterial activity of bryophyte species against Paenibacillus larvae isolates

    This study was performed to determine the antibacterial activity of methanol extracts of 23 bryophyte species against Paenibacillus larvae isolates that cause American foulbrood diseases in honeybee larvae. The honey and larva samples were collected from nine different locations of Rize in Turkey. A total of 22 gram-positive spore-forming bacteria were isolated from the larva and honey samples. According to the results of morphological, biochemical, and molecular (16S rRNA gene sequencing) tests, 10 isolates of the 22 gram-positive spore-forming bacteria were identified as P. larvae. A total of 10 bryophyte species (Polytrichum formasum,Polytrichum commune, Calliergonella cuspitada, Calliergonella lindbergi, Metzgeria conjugata, Isothecium alopecuroides, Syntrichia calcicola, Syntrichia intermedia, Tortella densa,and Grimmia alpestris) among 23 bryophytes showed good antimicrobial activity against P. larvae isolates according the results of agar-well diffusion method and minimal inhibition concentration experiments
