Genomic Diversity among Beijing and non-Beijing Mycobacterium tuberculosis Isolates from Myanmar
Background: The Beijing family of Mycobacterium tuberculosis is dominant in countries in East Asia. Genomic
polymorphisms are a source of diversity within the M. tuberculosis genome and may account for the variation of virulence among M. tuberculosis isolates. Till date there are no studies that have examined the genomic composition of M. tuberculosis isolates from the high TB-burden country, Myanmar.
Methodology/Principle Findings: Twenty-two M. tuberculosis isolates from Myanmar were screened on whole-genome
arrays containing genes from M. tuberculosis H37Rv, M. tuberculosis CDC1551 and M. bovis AF22197. Screening identified 198 deletions or extra regions in the clinical isolates compared to H37Rv. Twenty-two regions differentiated between Beijing and non-Beijing isolates and were verified by PCR on an additional 40 isolates. Six regions (Rv0071-0074 [RD105], Rv1572-1576c [RD149], Rv1585c-1587c [RD149], MT1798-Rv1755c [RD152], Rv1761c [RD152] and Rv0279c) were deleted in Beijing isolates, of
which 4 (Rv1572-1576c, Rv1585c-1587c, MT1798-Rv1755c and Rv1761c) were variably deleted among ST42 isolates, indicating a closer relationship between the Beijing and ST42 lineages. The TbD1 region, Mb1582-Mb1583 was deleted in Beijing and ST42 isolates. One M. bovis gene of unknown function, Mb3184c was present in all isolates, except 11 of 13 ST42 isolates. The CDC1551 gene, MT1360 coding for a putative adenylate cyclase, was present in all Beijing and ST42 isolates (except 1). The pks15/1 gene, coding for a putative virulence factor, was intact in all Beijing and non-Beijing isolates, except in ST42 and ST53 isolates.
Conclusion: This study describes previously unreported deletions/extra regions in Beijing and non-Beijing M. tuberculosis isolates. The modern and highly frequent ST42 lineage showed a closer relationship to the hypervirulent Beijing lineage than to the ancient non-Beijing lineages. The pks15/1 gene was disrupted only in modern non-Beijing isolates. This is the first report of an in-depth analysis on the genomic diversity of M. tuberculosis isolates from Myanmar

Structure of multiphoton quantum optics. II. Bipartite systems, physical processes, and heterodyne squeezed states
Extending the scheme developed for a single mode of the electromagnetic field
in the preceding paper ``Structure of multiphoton quantum optics. I. Canonical
formalism and homodyne squeezed states'', we introduce two-mode nonlinear
canonical transformations depending on two heterodyne mixing angles. They are
defined in terms of hermitian nonlinear functions that realize heterodyne
superpositions of conjugate quadratures of bipartite systems. The canonical
transformations diagonalize a class of Hamiltonians describing non degenerate
and degenerate multiphoton processes. We determine the coherent states
associated to the canonical transformations, which generalize the non
degenerate two--photon squeezed states. Such heterodyne multiphoton squeezed
are defined as the simultaneous eigenstates of the transformed, coupled
annihilation operators. They are generated by nonlinear unitary evolutions
acting on two-mode squeezed states. They are non Gaussian, highly non
classical, entangled states. For a quadratic nonlinearity the heterodyne
multiphoton squeezed states define two--mode cubic phase states. The
statistical properties of these states can be widely adjusted by tuning the
heterodyne mixing angles, the phases of the nonlinear couplings, as well as the
strength of the nonlinearity. For quadratic nonlinearity, we study the
higher-order contributions to the susceptibility in nonlinear media and we
suggest possible experimental realizations of multiphoton conversion processes
generating the cubic-phase heterodyne squeezed states.Comment: 16 pages, 23 figure
On the Non-invasive Measurement of the Intrinsic Quantum Hall Effect
With a model calculation, we demonstrate that a non-invasive measurement of
intrinsic quantum Hall effect defined by the local chemical potential in a
ballistic quantum wire can be achieved with the aid of a pair of voltage leads
which are separated by potential barriers from the wire. B\"uttiker's formula
is used to determine the chemical potential being measured and is shown to
reduce exactly to the local chemical potential in the limit of strong potential
confinement in the voltage leads. Conditions for quantisation of Hall

Molecular Mechanisms of Lymphocyte Homing to Peripheral Lymph Nodes
To characterize the adhesion cascade that directs lymphocyte homing to peripheral lymph nodes (PLNs), we investigated the molecular mechanisms of lymphocyte interactions with the microvasculature of subiliac lymph nodes. We found that endogenous white blood cells and adoptively transferred lymph node lymphocytes (LNCs) tethered and rolled in postcapillary high endothelial venules (HEVs) and to a lesser extent in collecting venules. Similarly, firm arrest occurred nearly exclusively in the paracortical HEVs. Endogenous polymorphonuclear (PMNs) and mononuclear leukocytes (MNLs) attached and rolled in HEVs at similar frequencies, but only MNLs arrested suggesting that the events downstream of primary rolling interactions critically determine the specificity of lymphocyte recruitment. Antibody inhibition studies revealed that L-selectin was responsible for attachment and rolling of LNCs, and that LFA-1 was essential for sticking. LFA-1–dependent arrest was also abolished by pertussis toxin, implicating a requirement for Gαi−-protein–linked signaling. α4 integrins, which play a critical role in lymphocyte homing to Peyer's Patches, made no significant contribution to attachment, rolling, or sticking in resting PLNs. Velocity analysis of interacting LNCs revealed no detectable contribution by LFA-1 to rolling. Taken together, our results suggest that lymphocyte– HEV interactions within PLNs are almost exclusively initiated by L-selectin followed by a G protein–coupled lymphocyte-specific activation event and activation-induced engagement of LFA-1. These events constitute a unique adhesion cascade that dictates the specificity of lymphocyte homing to PLNs
Mass drug administration of azithromycin for trachoma reduces the prevalence of genital Chlamydia trachomatis infection in the Solomon Islands.
OBJECTIVES: Chlamydia trachomatis is the most common bacterial sexually transmitted infection and is frequently asymptomatic; ocular C. trachomatis strains cause trachoma. Mass drug administration (MDA) of azithromycin for trachoma might also reduce the prevalence of genital C. trachomatis. In a survey conducted in the Solomon Islands in 2014, prior to MDA, the prevalence of genital C. trachomatis was 20.3% (95% CI 15.9% to 25.4%). We conducted a survey to establish the impact of MDA with azithromycin on genital C. trachomatis. METHODS: Women attending three community outpatient clinics, predominantly for antenatal care, 10 months after MDA with azithromycin given for trachoma elimination, were enrolled in this survey. Self-taken high vaginal swabs were for C. trachomatis and Neisseria gonorrhoeae using the BD Probetec strand displacement assay. RESULTS: 298 women were enrolled. C. trachomatis infection was diagnosed in 43 women (14.4%, 95% CI 10.6% to 18.9%) and N. gonorrhoeae in 9 (3%, 95% CI 1.4% to 5.7%). The age-adjusted OR for C. trachomatis infection was consistent with a significant decrease in the prevalence of C. trachomatis following MDA (OR 0.58, 95% CI 0.37 to 0.94, p=0.027). There was no change in the prevalence of N. gonorrhoeae between following MDA (OR 0.51, 95% CI 0.22 to 1.22, p=0.13). CONCLUSIONS: This study demonstrated a 40% reduction in the age-adjusted prevalence of genital C. trachomatis infection following azithromycin MDA for trachoma elimination
