    Cognitive radio adaptive rendezvous protocols to establish network services for a disaster response

    Disasters are catastrophic events that cause great damage or loss of life. In disasters, communication services might be disrupted due to damage to the existing network infrastructure. Temporary systems are required for victims and first responders, but installing them requires information about the radio environment and available spectrum. A cognitive radio (CR) can be used to provide a flexible and rapidly deployable temporary system due to its sensing, learning and decision-making capabilities. This thesis initially examines the potential of CR technology for disaster response networks (DRN) and shows that they are ideally suited to fulfill the requirements of a DRN. A software defined radio based prototype for multiple base transceiver stations based cellular network is proposed and developed. It is demonstrated that system can support a large number of simultaneous calls with sufficient call quality, but only when the background interference is low. It is concluded that to provide call quality with acceptable latency and packet losses, the spectrum should be used dynamically for backhaul connectivity. The deployment challenges for such a system in a disaster include the discovery of the available spectrum, existing networks, and neighbours. Furthermore, to set up a network and to establish network services, initially CR nodes are required to establish a rendezvous. However, this can be challenging due to unknown spectrum information, primary radio (PR) activity, nodes, and topology. The existing rendezvous strategies do not fulfill the DRN requirements and their time to rendezvous (TTR) is long. Therefore, we propose an extended modular clock algorithm (EMCA) which is a multiuser blind rendezvous protocol, considers the DRN requirements and has short TTR. For unknown nodes and topologies, a general framework for self-organizing multihop cooperative fully blind rendezvous protocol is also proposed, which works in different phases, can terminate when sufficient nodes are discovered, and is capable of disseminating the information of nodes which enter or leave a network. A synchronization mechanism is presented for periodic update of rendezvous information. An information exchange mechanism is also proposed which expedites the rendezvous process. In both single and multihop networks, EMCA provides up to 80% improvement in terms of TTR over the existing blind rendezvous strategies while considering the PR activity. A simple Random strategy, while being poorer than EMCA, is also shown to outperform existing strategies on average. To achieve adaptability in the presence of unknown PR activity, different CR operating policies are proposed which avoid the channels detected with PR activity to reduce the harmful interference, provide free channels to reduce the TTR, and can work with any rendezvous strategy. These policies are evaluated over different PR activities and shown to reduce the TTR and harmful interference significantly over the basic Listen before Talk approach. A proactive policy, which prefers to return to channels with recent lower PR activity, is shown to be best, and to improve the performance of all studied rendezvous strategies

    The Selective Attention for Action Model (SAAM). An exploration of affordances in a computational and experimental study

    In this thesis a connectionist model for affordance-guided selective attention for action is presented. The selective attention for action model (SAAM) is aimed at modelling the direct route from vision to action using Gibson's affordances to describe actions. In the model complex affordances are encoded in grasp postures. It is shown that this is a viable method to realise aordances. By deriving complex affordances from grasp postures which in turn are derived from grasp affordances and invariants in the visual information, the model implements a hierarchical structure of affordances as postulated in Gibson's affordance concept. This is a novel way of implementing affordances in a computational model. Three studies were conducted to explore the model. The grasp postures generated by SAAM are verified in Study 1 by comparing them with human grasp postures. These grasps were collected in an experiment which used the same stimulus shapes as the model. In Study 2 the attentional behaviour of SAAM is investigated. It is shown that the model is able to select only one of the objects in visual inputs showing multiple objects as well as selecting the appropriate action category (described by a grasp type). Furthermore, it is shown that the bottom-up selection of the model can be influenced by top-down feedback of action intentions. In Study 3 the fndings of Study 2 are applied to stimuli showing two hand-tools (pliers and hammer) to clarify how the findings of Study 2 link to complex affordances and action intentions. The results demonstrate that SAAM offers a novel and powerful way to implement complex affordances.     Cortical Dynamics of Contextually-Cued Attentive Visual Learning and Search: Spatial and Object Evidence Accumulation

    How do humans use predictive contextual information to facilitate visual search? How are consistently paired scenic objects and positions learned and used to more efficiently guide search in familiar scenes? For example, a certain combination of objects can define a context for a kitchen and trigger a more efficient search for a typical object, such as a sink, in that context. A neural model, ARTSCENE Search, is developed to illustrate the neural mechanisms of such memory-based contextual learning and guidance, and to explain challenging behavioral data on positive/negative, spatial/object, and local/distant global cueing effects during visual search. The model proposes how global scene layout at a first glance rapidly forms a hypothesis about the target location. This hypothesis is then incrementally refined by enhancing target-like objects in space as a scene is scanned with saccadic eye movements. The model clarifies the functional roles of neuroanatomical, neurophysiological, and neuroimaging data in visual search for a desired goal object. In particular, the model simulates the interactive dynamics of spatial and object contextual cueing in the cortical What and Where streams starting from early visual areas through medial temporal lobe to prefrontal cortex. After learning, model dorsolateral prefrontal cortical cells (area 46) prime possible target locations in posterior parietal cortex based on goalmodulated percepts of spatial scene gist represented in parahippocampal cortex, whereas model ventral prefrontal cortical cells (area 47/12) prime possible target object representations in inferior temporal cortex based on the history of viewed objects represented in perirhinal cortex.     Relative Protein Lifetime Measurement in Plants Using Tandem Fluorescent Protein Timers

    Targeted protein degradation plays a wide range of important roles in plant growth and development, but analyzing protein turnover in vivo is technically challenging. Until recently, there has been no straightforward methodology for quantifying protein dynamics at subcellular resolution during cellular transitions in plants. A tandem fluorescent protein timer (tFT) is a fusion of two different fluorescent proteins with distinct fluorophore maturation kinetics, which allows estimation of relative protein age from the ratio of fluorescence intensities of the two fluorescent proteins. Here, we describe approaches to use this technology to report relative protein lifetime in both transient and stable plant transformation systems. tFTs enable in vivo, real-time protein lifetime assessment within subcellular compartments and across tissues, permitting the analysis of protein degradation dynamics in response to stresses or developmental cues and in different genetic backgrounds

    Mesenchymal Stem Cell-Derived Extracellular Vesicles in Tendon and Ligament Repair-A Systematic Review of In Vivo Studies.

    Tendon and ligament injury poses an increasingly large burden to society. This systematic review explores whether mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) can facilitate tendon/ligament repair in vivo. On 26 May 2021, a systematic search was performed on PubMed, Web of Science, Cochrane Library, Embase, to identify all studies that utilised MSC-EVs for tendon/ligament healing. Studies administering EVs isolated from human or animal-derived MSCs into in vivo models of tendon/ligament injury were included. In vitro, ex vivo, and in silico studies were excluded, and studies without a control group were excluded. Out of 383 studies identified, 11 met the inclusion criteria. Data on isolation, the characterisation of MSCs and EVs, and the in vivo findings in in vivo models were extracted. All included studies reported better tendon/ligament repair following MSC-EV treatment, but not all found improvements in every parameter measured. Biomechanics, an important index for tendon/ligament repair, was reported by only eight studies, from which evidence linking biomechanical alterations to functional improvement was weak. Nevertheless, the studies in this review showcased the safety and efficacy of MSC-EV therapy for tendon/ligament healing, by attenuating the initial inflammatory response and accelerating tendon matrix regeneration, providing a basis for potential clinical use in tendon/ligament repair

    A BIASED COMPETITION COMPUTATIONAL MODEL OF SPATIAL AND OBJECT-BASED ATTENTION MEDIATING ACTIVE VISUAL SEARCH

    A computational cognitive neuroscience approach was used to examine processes of visual attention in the human and monkey brain. The aim of the work was to produce a biologically plausible neurodynamical model of both spatial and object-based attention that accounted for observations in monkey visual areas V4, inferior temporal cortex (IT) and the lateral intraparietal area (LIP), and was able to produce search scan path behaviour similar to that observed in humans and monkeys. Of particular interest currently in the visual attention literature is the biased competition hypothesis (Desimone & Duncan. 1995). The model presented here is the first active vision implementation of biased competition, where attcntional shifts are overt. Therefore, retinal inputs change during the scan path and this approach raised issues, such as memory for searched locations across saccades, not addressed bv previous models with static retinas. This is the first model to examine the different time courses associated with spatial and object-based effects at the cellular level. Single cell recordings in areas V4 (Luck et al., 1997; Chelazzi et al., 2001) and IT (Chelazzi ct al., 1993, 1998) were replicated such that attentional effects occurred at the appropriate time after onset of the stimulus. Object-based effects at the cellular level of the model led to systems level behaviour that replicated that observed during active visual search for orientation and colour feature conjunction targets in psychophysical investigations. This provides a valuable insight into the link between cellular and system level behaviour in natural systems. At the systems level, the simulated search process showed selectivity in its scan path that was similar to that observed in humans (Scialfa & Joffe, 1998; Williams & Reingold, 2001) and monkeys (Motter & Belky. 1998b), being guided to target coloured locations in preference to locations containing the target orientation or blank areas. A connection between the ventral and dorsal visual processing streams (Ungerleider & Mishkin. 1982) is suggested to contribute to this selectivity and priority in the featural guidance of search. Such selectivity and avoidance of blank areas has potential application in computer vision applications. Simulation of lesions within the model and comparison with patient data provided further verification of the model. Simulation of visual neglect due to parietal cortical lesion suggests that the model has the capability to provide insights into the neural correlates of the conscious perception of stimuli The biased competition approach described here provides an extendable framework within which further "bottom-up" stimulus and "top-down" mnemonic and cognitive biases can be added, in order to further examine exogenous versus endogenous factors in the capture of attention
