Biogratings: Diffractive Transducers for Biosensing in Photonic Platforms
Tesis por compendio[ES] El desarrollo científico y tecnológico de las últimas décadas ha dado lugar a sistemas sensores capaces de obtener, procesar y transmitir información sobre multitud de aspectos físicos y químicos, y utilizarla para mejorar aspectos clave de multitud de áreas de nuestra sociedad. Los sensores químicos son dispositivos compactos y miniaturizados capaces de ofrecer soluciones alternativas a las técnicas de análisis instrumental convencionales. En especial, los biosensores han adquirido gran relevancia por los avances que han supuesto para sectores estratégicos como el diagnóstico clínico, la industria alimentaria y el medio ambiente.
Los biosensores ópticos se basan en interacciones entre la luz y la materia para transducir eventos de bioreconocimiento y presentan prestaciones importantes como la estabilidad, inmunidad a estímulos externos y versatilidad en el desarrollo de aproximaciones sin marcaje (label-free). Este último aspecto suele aprovechar fenómenos nanoscópicos y su desarrollo se encuentra muy ligado al progreso de la nanociencia y nanotecnología.
Un aspecto clave en el biosensado sin marcaje consiste en descubrir y desarrollar nuevas estrategias de transducción. En este sentido, aunque se encuentren aun en una etapa temprana de desarrollo, los biosensores difractivos presentan un gran potencial en términos de simplicidad, miniaturización, y capacidad para minimizar señales no deseadas fruto de interacciones no específicas, entre otros aspectos.[CA] El desenvolupament científic i tecnològic de les últimes dècades ha donat lloc a sistemes sensors capaços d'obtindre, processar i transmetre informació sobre multitud d'aspectes físics i químics, i utilizar-la per a millorar aspectes clau de multitud d'arees de la nostra societat. Els sensors químics són dispositius compactes i miniaturitzats capaços d'oferir solucions alternatives a les tècniques d'analisi instrumental convencionals. Especialment, els biosensors han adquirit gran rellevància pels avanços que han suposat per als sectors estratègics com el diagnòstic clínic, la industria alimentària i el medi ambient.
Els biosensors òptics es basen en interaccions entre la llum i la matèria per a transduir esdeveniments de bioreconèixement i presenten prestacions importants com estabilitat, immunitat a estímuls externs i versatilitat en el desenvolupament d'aproximacions sense marcatge (label-free). Aquest últim aspecte sol aprofitat fenòmens nanoscòpics i el seu desenvolupament es troba molt lligat al progrés de la nanociència i nanotecnologia.
Un aspecte clau en el biosensat sense marcatge consisteix a descobrir i desenvolupar noves estratègies de transducció. En aquest sentit, encara que es troben fins i tot en una etapa primerenca de desenvolupament, els biosensors difractius presenten un gran potencial en termes de simplicitat, miniaturització, i capacitat per a minimitzar senyals no desitjats fruit d'interaccions no específiques, entre altres aspectes.[EN] The scientific and technological progress in recent decades has given rise to sensor systems capable of obtaining, processing, and transmitting information on a multitude of physical and chemical aspects and using it to improve key aspects of many areas of our society. Chemical sensors are compact, miniaturized devices capable of offering alternative solutions to conventional instrumental analysis techniques. In particular, biosensors have become highly relevant due to the progress they have brought to strategic sectors such as clinical diagnostics, the food industry, and the environment.
Optical biosensors rely on interactions between light and matter to transduce biosensing events and provide important features such as stability, immunity to external stimuli, and versatility in the development of label-free approaches. This last aspect usually exploits nanoscopic phenomena and its development in closely linked to the progress in nanoscience and nanotechnology.
A key aspect of label-free biosensing is the discovery and development of new transduction strategies. Підвищення метрологічних характеристик сенсора на основі явища поверхневого плазмонного резонансу
Дисертація присвячена підвищенню метрологічних характеристик
оптичного сенсора на основі явища поверхневого плазмонного резонансу (ППР)
при використанні комплексного підходу, що включає вдосконалення
конструкції сенсора та методу визначення резонансного кута. Аналітичний
метод на основі явища ППР використовується для дослідження фізико-хімічних
та біохімічних реакцій у рідких та газоподібних середовищах. Сенсори на
основі явища поверхневого плазмонного резонансу (ППР-сенсори) складаються
з чутливого елемента та фізичного перетворювача. Чутливий елемент реагує на
зміну показника заломлення досліджуваної речовини, внаслідок зміни
концентрації певної її складової або товщини адсорбованого на його поверхню
нанорозмірного шару. Фізичний перетворювач формує на виході ППР-сенсора
різницю потенціалів, пропорційну зміні показника заломлення. Чутливий
елемент таких сенсорів є високопровідним металевим шаром, товщиною від 10
до 100 нанометрів, який розташований на діелектричній підкладинці, що має
показник заломлення більший за показник заломлення досліджуваного
середовища. При опроміненні металевого шару зі сторони підкладинки pполяризованим
монохроматичним світлом під кутом більшим за критичний
виникає резонанс частоти падаючого світла з плазмонною частотою вільних
електронів у металі, внаслідок чого на межі метал-діелектрик виникає
поверхнева оптична хвиля – поверхневий плазмон. Більшість існуючих ППР-сенсорів побудовані за оптичною схемою з
використанням призми для забезпечення умов повного внутрішнього відбиття
(схема Кречмана). На призму через імерсійну рідину для забезпечення
оптичного контакту встановлюють діелектричну підкладинку з металевим
шаром. Для досягнення резонансних умов виконується зміна кута падіння
світла зі сторони призми на межу діелектрик-метал. У цьому випадку резонанс
частот виникає при певному куті падіння світла – резонансному куті, а відгуком
ППР-сенсора є зсув кутового положення мінімуму резонансної характеристики
відбиття, яка представляє собою графік залежності інтенсивності відбитого
світла від кута його падіння. При цьому визначення точного кутового
положення мінімуму характеристики відбиття, а саме резонансного кута, як до
зміни показника заломлення досліджуваного середовища, так і після є
важливою метрологічною задачею, яка потребує вирішення, оскільки
збільшення похибки вимірювання цього кута підвищує похибку величини
відгуку ППР-сенсора та суттєво знижує достовірність результатів дослідження.
Метою дисертаційної роботи є підвищення метрологічних характеристик
сенсора на основі явища поверхневого плазмонного резонансу шляхом
вдосконалення конструкції та методу визначення резонансного кута.
Доповнено наукові дані щодо важливості врахування впливу шорсткості
поверхні металевої плівки та додаткового діелектричного шару на вимірювання
резонансного кута ППР-сенсорами та визначено функції перетворення для
таких сенсорів. Розроблено алгоритм розрахунку невизначеності вимірювання
резонансного кута. Проаналізовано існуючі методи визначення резонансного
кута та визначено їх недоліки та переваги. Особлива увага приділена методу
середньої лінії та найпоширенішими методами апроксимації поліномами 2-го,
3-го степенів. Оцінено абсолютну похибку вимірювання резонансного кута з
використанням теоретичних характеристик відбиття цими методами при зміні
таких параметрів досліджуваного середовища як: показник заломлення,
температура, а також довжина хвилі випромінювання. Також в роботі
досліджено точність визначення резонансного кута різними методами при зміні товщини додаткового діелектричного шару, зміні шорсткості металевої
поверхні та скляної поверхні призми ППР-сенсора.
Вдосконалено метод визначення резонансного кута. Було встановлено, що
розроблений метод є менш залежним від зміни форми резонансної
характеристики відбиття, на відміну від більшості існуючих методів. Зміна
форми характристики відбиття проявляється у її розширенні та зростанні
інтенсивності відбитого світла в її мінімумі, що є наслідком зміни величини
показника заломлення та температури досліджуваного середовища при
проходженні фізико-хімічних та біохімічних реакцій. Розроблений метод було
порівняно з методом середньої лінії та методами апроксимації поліномами 2-го,
3-го степенів. Для цього було проведене чисельне моделювання характеристик
відбиття, використовуючи формули Френеля і математичний формалізм
матриць розсіювання Джонса для перевірки функціонування методу за умов
зміни довжини хвилі випромінювання, зміни показника заломлення,
температури досліджуваного середовища, зміни товщини додаткового шару та
шорсткості поверхні. За результатами моделювання для чутливого елемента з
додатковим діелектричним шаром було встановлено, що абсолютна похибка
вимірювання резонансного кута для вдосконаленого методу була у 9 разів
менша, ніж для методу середньої лінії, який в свою чергу дозволяє зменшити
абсолютну похибку щонайменше в 2,5 рази у порівнянні з методами
апроксимації поліномом.
Особливістю явища ППР також є те, що частота поверхневого плазмону
та швидкість його загасання, при розповсюдженні вздовж межі металдосліджуване
середовище, залежать не тільки від зміни показника заломлення
на поверхні чутливого елементу, а й від шорсткості поверхні металу, яка
контактує з досліджуваним середовищем. Стан металевої поверхні в свою чергу
залежить від технології формування металевого шару на діелектрику, а також
від стану поверхні самої діелектричної скляної підкладинки, який визначається
технологією її обробки. Скляна підкладинка оптично узгоджена з призмою
ППР-сенсора та призначена для багаторазового використання призми. Для встановлення впливу технології обробки поверхні підкладинки
чутливого елементу було порівняно найпоширеніший метод обробки оптичних
скляних поверхонь (механічне фінішне полірування) із вперше застосованим
для обробки підкладинок ППР-сенсорів – методом електронно-променевої
обробки. Даний метод, на відміну від існуючих, таких як механічне
полірування, йонна обробка та хімічне полірування, не змінюють поверхневого
складу матеріалу підкладинки та не створюють дефектного порушеного
приповерхневого шару. На скляну поверхню з середньоквадратичною
шорсткістю поверхні від 5 нм і більше методом термічного випаровування у
вакуумі було нанесено металевий шар. Внаслідок електронно-променевої
обробки скляних підкладинок чутливих елементів ППР-сенсорів, які пройшли
попередньо механічну обробку, було зменшено шорсткість поверхні металевого
шару в 3 рази з 4,67 нм до 1,64 нм, що зменшило півширину характеристики
відбиття майже в 2 рази з 0,453 град. до 0,867 град., і як наслідок зменшило
абсолютну похибку визначення резонансного кута в 3 рази з 1,8 кут.сек. до 0,58
кут.сек. вдосконаленим методом. При цьому характеристика відбиття зсунулась
у сторону менших кутів на 0,34 градуси, що розширило діапазон вимірювання
ППР-сенсора. Збільшено чутливість по інтенсивності в 1,5 рази через
збільшення глибини проникнення поля поверхневого плазмону у досліджуване
середовище.
Одним з відомих напрямків підвищення чутливості ППР-сенсорів є
підвищення адсорбційної здатності поверхні металевого шару. Для цього
чутливі елементи покривають зі сторони досліджуваного середовища
додатковими діелектричними шарами органічного чи неорганічного
походження з розвиненою поверхнею. Це особливо актуально у сенсорах
газоподібних середовищ, абсорбція яких у порах суттєво підвищує величину
відгуку. До найпоширеніших недоліків ППР-сенсорів з такими покриттями,
слід віднести низьку відновлювальну здатність після абсорбції, зниження з
часом рівня відгуку та деградацію покриття через його руйнування, особливо
після взаємодії з агресивними середовищами. Для підвищення чутливості ППР-сенсора до газоподібних середовищ у
якості додаткового діелектричного покриття було застосовано нанометровий
шар політетрафторетилену, осаджений на металевий шар. Було проведено
експериментальне дослідження вдосконаленого сенсора з шаром
політетрафторетилену різної товщини від 5 до 50 нм при взаємодії з
насиченими парами розчинників: метанолу, етанолу, ацетону та ізопропанолу.
За результатами експерименту було встановлено, що максимальне збільшення
чутливості у 2,8 рази забезпечила товщина політетрафторетилену 30 нм. Аналіз
поверхні такого сенсора за допомогою атомно-силової мікроскопії дозволив
встановити, що причиною зростання чутливості стало збільшення площі
поверхні взаємодії з досліджуваним середовищем.
Наукова новизна одержаних результатів полягає в наступному:
1. Удосконалено метод визначення резонансного кута, який на відміну від
відомих, враховує вплив зміни інтенсивності в мінімумі характеристики
відбиття сенсора на основі поверхневого плазмонного резонансу, що дало змогу
підвищити точність його визначення у 9 разів при збільшення товщини
додаткового діелектричного верхнього шару в нанометровому діапазоні.
2. Набула подальшого розвитку фізико-математична модель сенсора, яка
містить додатковий діелектричний верхній шар з врахуванням шорсткості
перехідних шарів в нанометровому діапазоні, що дозволило розробити метод
точнішого визначення резонансного кута.
3. Вперше встановлено вплив зміни в нанометровому діапазоні шорсткості
скляних підкладинок сенсора при електронно-променевій обробці їх поверхні
на зміну чутливості внаслідок збільшення крутизни схилів резонансної
характеристики відбиття, що дозволило підвищити чутливість по інтенсивності
в 1,5 рази.
4. Набуло подальшого розвитку дослідження впливу на чутливість сенсора
товщини та топології додаткового діелектричного верхнього шару, що
дозволило встановити залежність між площею поверхні взаємодії з досліджуваним середовищем та чутливістю сенсора та обрати оптимальну
товщину шару.
Практичне значення одержаних результатів полягає у наступному:
1. Вперше запропоновано метод визначення резонансного кута, який
полягає в визначенні цього кута як точки перетину осі кутів падіння світла та
лінії, що проходить через середини відрізків між еквірівневими точками на
схилах характеристики відбиття, та зміщення цієї характеристики на величину
значення інтенсивності в її мінімумі, що дало можливість зменшити похибку
вимірювання резонансного кута щонайменше у 5,5 рази.
2. Вдосконалено спосіб виготовлення сенсорів, який включає електроннопроменеву
обробку скляних, попередньо механічно полірованих, підкладинок,
що дало можливість збільшити чутливість та зменшити похибку вимірювання
резонансного кута щонайменше у 3 рази.
3. Розроблено сенсор для аналізу газоподібних середовищ з додатковим
шаром політетрафторетилену з оптимальною товщиною (30 нм), що забезпечує
підсилення відгуку до насичених парів розчинників щонайменше у 2,8 рази, у
порівнянні з аналогічним сенсором без додаткового шару.
Результати досліджень, проведених в роботі, є перспективними для
застосування у промисловості, в науковій діяльностях та в навчальному
процесі. Розглянуто практичне застосування вдосконаленого високочутливого
ППР-сенсора з додатковим шаром політетрафторетилену у газовій сенсориці.
Вдосконалений метод визначення резонансного кута дозволяє зменшити
похибку вимірювання при дослідженні біологічних взаємодій, оскільки цей
метод менш чутливий до змін коефіцієнта поглинання світла чи адсорбції
тонких поглинаючих шарів. Результати досліджень підтверджені відповідними
актами використання.The dissertation is devoted to increase of metrological characteristics of the
optical sensor based on surface plasmon resonance phenomenon (SPR) at use of the
complex approach including improvement of a design of the sensor and a method of
definition of a resonant angle. Analytical method based on SPR phenomenon is used
to study physicochemical and biochemical reactions in liquid and gaseous media.
Sensors based on surface plasmon resonance (SPR-sensors) consist of a sensitive
element and a physical transducer. The sensitive element responds to changes in the
refractive index of the test substance, due to changes in the concentration of a certain
component, or the thickness of the nano-layer adsorbed on its surface. The physical
transducer generates a difference of electrical potential at the output of the SPR
sensor, proportional to the change in the refractive index. The sensitive element of
such sensors is a high-conductivity metal layer, 10 to 100 nanometers thick, which is
located on a dielectric substrate having a refractive index greater than the refractive
index of the test medium. When the metal layer is irradiated from the substrate side
with p-polarized monochromatic light at an angle greater than critical, the frequency
of incident light resonates with the plasmon frequency of free electrons in the metal,
as a result of which a surface optical wave - surface plasmon - is formed at the metaldielectric
interface. Most of the existing SPR sensors are built on an optical scheme
using a prism to provide conditions of total internal reflection (Kretschman's scheme)
on which through an immersion fluid to ensure optical contact set a dielectric
substrate with a metal layer. To achieve resonant conditions, the angle of incidence of
light from the prism to the dielectric-metal boundary is changed. In this case, the resonance of frequencies occurs at a certain angle of incidence of light - resonant
angle, and the response of the SPR sensor is a shift of the angular position of the
minimum resonant reflection characteristic, which is a graph of the intensity of
reflected light from its angle of incidence. In this case, determining the exact angular
position of the minimum reflection characteristics, namely the resonant angle, both
before and after the refractive index of the studied medium is an important
metrological problem that needs to be solved, because increasing the measurement
error increases the response error and reduces the reliability of the study results.
The aim of the dissertation is to increase the metrological characteristics of the
sensor based on the phenomenon of surface plasmon resonance by improving the
design and method of determining the resonance angle.
The scientific data on the importance of taking into account the influence of the
surface roughness of the metal film and the additional dielectric layer on the
measurement of the resonance angle by SPR sensors are supplemented and the
transformation functions for such sensors are determined. An algorithm for
calculating the uncertainty of resonance angle measurement has been developed. The
existing methods of determining the resonance angle are analyzed and their
disadvantages and advantages are determined. Particular attention is paid to the most
common methods, namely: the method of approximation by polynomials of 2nd and
3rd degrees, as well as the method of the middle line. The absolute error of resonance
angle measurement is estimated using the theoretical characteristics of reflection by
these methods when changing such parameters of the studied medium as: refractive
index, temperature, as well as the wavelength of radiation. The paper also
investigates the accuracy of determining the resonance angle by different methods
when changing the thickness of the additional dielectric layer, and changing the
roughness of the metal surface and the glass surface of the prism of SPR sensor.
The method of determining the resonant angle has been improved. It was found
that the developed method is less dependent on the change in the shape of the
resonant reflection characteristic, in contrast to most existing methods. The change in
the shape of the reflection characteristics is manifested in its expansion and increase in the intensity of reflected light in its minimum, which is a consequence of changes
in the refractive index and temperature of the studied medium during
physicochemical and biochemical reactions. The developed method was compared
with the middle line method and the most common methods of approximation by
polynomials of 2nd and 3rd degrees. To do this, numerical modeling of reflection
characteristics was performed using Fresnel formulas and mathematical formalism of
Jones scattering matrices to test the method under the conditions of changing the
wavelength of radiation, changes in refractive index, temperature of the medium,
changes in additional layer thickness and surface roughness. According to the
simulation results for the sensitive element with an additional dielectric layer, it was
found that the absolute error of resonance angle measurement for the advanced
method was 9 times less than for the midline method, which in turn reduces the
absolute error by at least 2.5 times compared with polynomial approximation
methods.
The peculiarity of the SPR phenomenon is also that the frequency of the
surface plasmon and its rate of attenuation, when propagating along the metalmedium
medium,
depend
not
only
on
the
change
in
refractive
index
on
the
surface
of
the
sensing element, but also on the surface roughness of the metal contacting the
medium. The condition of the metal surface in turn depends on the technology of
forming a metal layer on the dielectric, as well as on the condition of the surface of
the dielectric glass substrate, which is determined by the technology of its processing.
The glass substrate is in optical contact with the prism of the SPR sensor and is
designed for multiple use of the prism.
To establish the influence of the technology of surface treatment of the
substrate of the sensitive element, the most common method of treatment of optical
glass surfaces (mechanical finishing polishing) was compared with the first used for
the treatment of SPR sensor substrates - the method of electron beam treatment. This
method, in contrast to existing ones, such as mechanical polishing, ion processing and
chemical polishing, does not change the surface composition of the substrate material
and does not create a defective disturbed near-surface layer. A metal layer was applied to a glass surface with a root mean square surface roughness of 5 nm or more
by thermal evaporation in vacuum. As a result of electron beam treatment of glass
substrates of sensitive elements of SPR sensors, which have undergone premachining,
the surface roughness of the metal layer was reduced by 3 times from
4.67 nm to 1.64 nm, which reduced the half-width of the reflection characteristic by
almost 2 times from 0.453 deg. to 0.867 deg., and as a consequence reduced the
absolute error of determining the resonant angle in 3 times from 1.8 arc.sec. up to
0.58 arc.sec. for the advanced method. At the same time, the reflection characteristic
shifted towards smaller angles by 0.34 degrees, which expanded the measurement
range of the SPR sensor. The sensitivity in intensity is increased by 1.5 times due to
the increase in the depth of penetration of the surface plasmon field into the studied
medium.
One of the known ways to enhance the sensitivity of SPR sensors is to increase
the adsorption capacity of the surface of the metal layer. To do this, the sensitive
elements are covered on the side of the test medium with additional dielectric layers
of organic or inorganic origin with a developed surface. This is especially true in
gaseous media sensors, the absorption of which in the pores significantly increases
the magnitude of the response. The most common disadvantages of SPR sensors with
such coatings include low reproducibility after absorption, reduced over time, the
level of response and degradation of the coating due to its destruction, especially after
interaction with aggressive media.
To increase the sensitivity of the SPR sensor to gaseous media, a nanometer
layer of polytetrafluoroethylene deposited on a metal layer was used as an additional
dielectric coating. An experimental study of an advanced sensor with a layer of
polytetrafluoroethylene of different thickness from 5 to 50 nm when interacting with
saturated pairs of solvents: methanol, ethanol, acetone and isopropanol. According to
the results of the experiment, it was found that the maximum increase in sensitivity
by 2.8 times was provided by the thickness of polytetrafluoroethylene at 30 nm.
Analysis of the surface of such a sensor using atomic force microscopy revealed that the reason for the increase in sensitivity was an increase in the surface area of
interaction with the studied medium.
The scientific novelty of the obtained results is as follows:
1. Improved the method of determining the resonance angle, which, in contrast
to the known methods, takes into account the effect of changing the intensity in the
minimum reflection characteristics of the sensor based on surface plasmon resonance,
which increased the accuracy of its determination by 9 times increasing the thickness
of the additional dielectric upper layer in the nanometer range.
2. The physico-mathematical model of the sensor, which contains an additional
dielectric upper layer taking into account the roughness of the transition layers in the
nanometer range, was further developed, which allowed developing a method for
more accurate determination of the resonance angle.
3. For the first time the influence of the change in the nanometer roughness
range of the sensor glass substrates during electron beam treatment of their surface on
the change in sensitivity due to the increase in the slope of the resonant reflection
characteristics, which increased the sensitivity by 1.5 times

Holographic Point-of-Care Diagnostic Devices
Developing non-invasive and accurate diagnostics that are easily manufactured, robust and reusable will provide monitoring of high-risk individuals in any clinical or point-of-care environment, particularly in the developing world. There is currently no rapid, low-cost and generic sensor fabrication technique capable of producing narrow-band, uniform, reversible colorimetric readouts with a high-tuneability range. This thesis aims to present a theoretical and experimental basis for the rapid fabrication, optimisation and testing of holographic sensors for the quantification of pH, organic solvents, metal cations, and glucose in solutions. The sensing mechanism was computationally modelled to optimise its optical characteristics and predict the readouts. A single pulse of a laser (6 ns, 532 nm, 350 mJ) in holographic “Denisyuk” reflection mode allowed rapid production of sensors through silver-halide chemistry, in situ particle size reduction and photopolymerisation. The fabricated sensors consisted of off-axis Bragg diffraction gratings of ordered silver nanoparticles and localised refractive index changes in poly(2-hydroxyethyl methacrylate) and polyacrylamide films. The sensors exhibited reversible Bragg peak shifts, and diffracted the spectrum of narrow-band light over the wavelength range λpeak ≈ 500-1100 nm. The application of the holographic sensors was demonstrated by sensing pH in artificial urine over the physiological range (4.5-9.0), with a sensitivity of 48 nm/pH unit between pH 5.0 and 6.0. For sensing metal cations, a porphyrin derivative was synthesised to act as the crosslinker, the light absorbing material, the component of a diffraction grating, as well as the cation chelating agent. The sensor allowed reversible quantification of Cu2+ and Fe2+ ions (50 mM - 1 M) with a response time within 50 s. Clinical trials of a glucose sensor in the urine samples of diabetic patients demonstrated that the glucose sensor has an improved performance compared to a commercial high-throughput urinalysis device. The experimental sensitivity of the glucose sensor exhibited a limit of detection of 90 µM, and permitted diagnosis of glucosuria up to 350 mM. The sensor response was achieved within 5 min and the sensor could be reused about 400 times without compromising its accuracy. Holographic sensors were also tested in flake form, and integrated with paper-iron oxide composites, dyed filter and chromatography papers, and nitrocellulose-based test strips. Finally, a generic smartphone application was developed and tested to quantify colorimetric tests for both Android and iOS operating systems. The developed sensing platform and the smartphone application have implications for the development of low-cost, reusable and equipment-free point-of-care diagnostic devices
Biosensors for Diagnosis and Monitoring
Biosensor technologies have received a great amount of interest in recent decades, and this has especially been the case in recent years due to the health alert caused by the COVID-19 pandemic. The sensor platform market has grown in recent decades, and the COVID-19 outbreak has led to an increase in the demand for home diagnostics and point-of-care systems. With the evolution of biosensor technology towards portable platforms with a lower cost on-site analysis and a rapid selective and sensitive response, a larger market has opened up for this technology. The evolution of biosensor systems has the opportunity to change classic analysis towards real-time and in situ detection systems, with platforms such as point-of-care and wearables as well as implantable sensors to decentralize chemical and biological analysis, thus reducing industrial and medical costs. This book is dedicated to all the research related to biosensor technologies. welcome to the 18th edition of the Merck & Elsevier Young Chemists Symposium, formerly SAYCS and MYCS.
This conference is an international scientific event organized by the Young Group of the Italian Chemical Society (SCI Giovani) with the financial support of Merck and Elsevier.
This symposium is fully devoted to young researchers, such as MSc and PhD students, post-doc fellows and young researchers in companies. All the disciplines of Chemistry are covered: analytical, physical, industrial, organic, inorganic, theoretical, pharmaceutical, biological, environmental, macromolecular and electrochemistry. This year, a special emphasis will be given to chemistry from knowledge to innovation: how chemistry is increasingly present in all of the fields that are essential for human life, and how chemical fundamentals are pushing novel technologies?

Mycobacterium tuberculosis DNA Detection Using Surface Plasmon Resonance Modulated by Telecommunication Wavelength
A surface plasmon resonance sensor for Mycobacterium tuberculosis (MTB) deoxyribonucleic acid (DNA) is developed using repeatable telecommunication wavelength modulation based on optical fiber communications laser wavelength and stability. MTB DNA concentrations of 1 μg/mL and 10 μg/mL were successfully demonstrated to have the same spectral half-width in the dip for optimum coupling. The sensitivity was shown to be −0.087 dB/(μg/mL) at all applied telecommunication wavelengths and the highest sensitivity achieved was 115 ng/mL without thiolated DNA immobilization onto a gold plate, which is better than the sensor limit of 400 ng/mL possible with commercial biosensor equipment
