    Parallelized computational 3D video microscopy of freely moving organisms at multiple gigapixels per second

    To study the behavior of freely moving model organisms such as zebrafish (Danio rerio) and fruit flies (Drosophila) across multiple spatial scales, it would be ideal to use a light microscope that can resolve 3D information over a wide field of view (FOV) at high speed and high spatial resolution. However, it is challenging to design an optical instrument to achieve all of these properties simultaneously. Existing techniques for large-FOV microscopic imaging and for 3D image measurement typically require many sequential image snapshots, thus compromising speed and throughput. Here, we present 3D-RAPID, a computational microscope based on a synchronized array of 54 cameras that can capture high-speed 3D topographic videos over a 135-cm^2 area, achieving up to 230 frames per second at throughputs exceeding 5 gigapixels (GPs) per second. 3D-RAPID features a 3D reconstruction algorithm that, for each synchronized temporal snapshot, simultaneously fuses all 54 images seamlessly into a globally-consistent composite that includes a coregistered 3D height map. The self-supervised 3D reconstruction algorithm itself trains a spatiotemporally-compressed convolutional neural network (CNN) that maps raw photometric images to 3D topography, using stereo overlap redundancy and ray-propagation physics as the only supervision mechanism. As a result, our end-to-end 3D reconstruction algorithm is robust to generalization errors and scales to arbitrarily long videos from arbitrarily sized camera arrays. The scalable hardware and software design of 3D-RAPID addresses a longstanding problem in the field of behavioral imaging, enabling parallelized 3D observation of large collections of freely moving organisms at high spatiotemporal throughputs, which we demonstrate in ants (Pogonomyrmex barbatus), fruit flies, and zebrafish larvae

    This paper presents a computer controlled stereoscopic vision system that will profile the contour and shape of the bottom of the human foot in its natural weight bearing position. The design will have no moving parts and will be restricted to an error of no more than {dollar}\pm{dollar}1 mm; Stereoscopic vision was chosen as the solution to profiling mainly because its use for this purpose is well documented. It is used in many fields that require range resolution. Mechanical engineers have used it in the development of robot vision for years and their applications range from control systems for robot welding to the determination of metal fatigue in aircraft (2). Radar engineers have used it since the early 1940\u27s to determine the range of incoming flying objects (1). Many other fields have used it for even more far reaching purposes.     Crack detection in "as-cast" steel using laser triangulation and machine learning

    We describe a high-accuracy inspection system designed to automatically detect cracks in "as-cast" steel slabs. Real-time slab inspection requires instrumentation capable of withstanding high temperatures above the steel surface as well as coping with the dirty and dusty environment present in a steel mill. Crack detection is also challenging due to the presence of oxidation scale on the slab surface. A bespoke laser triangulation system has been developed, providing images at 250 fps with a calibrated surface resolution of 97 μm from a 1m standoff distance. Cracks are detected using a combination of morphological detection and SVM classifier. Results are reported from laboratory testing and from extended trials at a production steel mill

    A fiber-based beam profiler for high-power laser beams in confined spaces and ultra-high vacuum

    Laser beam profilometry is an important scientific task with well-established solutions for beams propagating in air. It has, however, remained an open challenge to measure beam profiles of high-power lasers in ultra-high vacuum and in tightly confined spaces. Here we present a novel scheme that uses a single multi-mode fiber to scatter light and guide it to a detector. The method competes well with commercial systems in position resolution, can reach through apertures smaller than 500×500500\times 500~μ\mum2^2 and is compatible with ultra-high vacuum conditions. The scheme is simple, compact, reliable and can withstand laser intensities beyond 2~MW/cm2^2

    A fast high-precision six-degree-of-freedom relative position sensor

    Lasers are commonly used in high-precision measurement and profiling systems. Some laser measurement systems are based on interferometry principles, and others are based on active triangulation, depending on requirements of the application. This paper describes an active triangulation laser measurement system for a specific application wherein the relative position of two fixed, rigid mechanical components is to be measured dynamically with high precision in six degrees of freedom (DOF). Potential applications include optical systems with feedback to control for mechanical vibration, such as target acquisition devices with multiple focal planes. The method uses an array of several laser emitters mounted on one component. The lasers are directed at a reflective surface on the second component. The reflective surface consists of a piecewise-planar pattern such as a pyramid, or more generally a curved reflective surface such as a hyperbolic paraboloid. The reflected spots are sensed at 2-dimensional photodiode arrays on the emitter component. Changes in the relative position of the emitter component and reflective surface will shift the location of the reflected spots within photodiode arrays. Relative motion in any degree of freedom produces independent shifts in the reflected spot locations, allowing full six-DOF relative position determination between the two component positions. Response time of the sensor is limited by the read-out rate of the photodiode arrays. Algorithms are given for position determination with limits on uncertainty and sensitivity, based on laser and spot-sensor characteristics, and assuming regular surfaces. Additional uncertainty analysis is achievable for surface irregularities based on calibration data

    Předložená dizertační práce se zabývá kombinací optických mikromanipulací s mikrospektroskopickými metodami. Využili jsme laserovou pinzetu pro transport a třídění živých mikroorganismů, například jednobuněčných řas, či kvasinek. Ramanovskou spektroskopií jsme analyzovali chemické složení jednotlivých buněk a tyto informace jsme využili k automatické selekci buněk s vybranými vlastnostmi. Zkombinovali jsme pulsní amplitudově modulovanou fluorescenční mikrospektroskopii, optické mikromanipulace a jiné techniky ke zmapování stresové odpovědi opticky zachycených buněk při různých časech působení, vlnových délkách a intenzitách chytacího laseru. Vyrobili jsme různé typy mikrofluidních čipů a zkonstruovali jsme Ramanovu pinzetu pro třídění mikro-objektů, především živých buněk, v mikrofluidním prostředí.The subject of the presented Ph.D. thesis is a combination of optical micromanipulation and microspectroscopic methods. We used laser tweezers to transport and sort various living microorganisms, such as microalgal or yeast cells. We employed Raman microspectroscopy to analyze chemical composition of individual cells and we used the information about chemical composition to automatically select the cells of interest. We combined pulsed amplitude modulation fluorescence microspectroscopy, optical micromanipulation and other techniques to map the stress response of cells to various laser wavelengths, intensities and durations of optical trapping. We fabricated microfluidic chips of various designs and we constructed Raman-tweezers sorter of micro-objects such as living cells on a microfluidic platform.

    High repetition rate temperature and velocity Imaging in turbulent flows using thermographic phosphors

    Turbulent flows involving heat transfer and chemical reactions are prevalent in a huge range of applications such as combustors and engines, boilers, and heating and cooling devices. Directly measuring important variables using laser-based techniques has significantly contributed to our understanding of the underlying flow physics. However, many flows of interest exhibit infrequent or oscillatory behaviour, such as flame extinction or instabilities in thermal boundary layers. Capturing the flow dynamics requires simultaneous, two-dimensional temperature and velocity measurements at sampling rates commensurate with turbulent timescales. Typically this means measuring many thousands of temperature and velocity fields per second, yet there are no high repetition rate diagnostics for temperature imaging in practical, oxygen-containing systems, with the essential capability of simultaneous velocity measurements. This thesis presents a novel laser-based imaging technique based on thermographic phosphor particles. There are a huge variety of thermographic phosphors, which are solid materials with luminescence properties that can be exploited for remote thermometry. Here, phosphor particles are seeded into the flow as a tracer. An appropriate phosphor must be selected, and the particle size chosen so that the particle temperature and velocity rapidly assume that of the surrounding fluid. The particles are probed using high-speed lasers and their luminescence and scattering signals are detected using high-speed cameras to measure the flow temperature and velocity at kHz repetition rates. The development of this method is described in detail. Using the thermographic phosphor BAM:Eu, examples of simultaneous time-resolved measurements are presented in turbulent air flows between 300 and 500 K, consisting of a heated jet (Re = 10,000) and also a flow behind a heated cylinder (Re = 700). The technique permits kHz-rate temperature imaging in oxygen-containing environments. These combined diagnostics currently provide a unique capability for the investigation of transient, coupled heat and mass transfer phenomena in turbulent flows of practical engineering importance. A second objective of this work is to improve the precision of the temperature measurement. The characterisation of a different thermographic phosphor with a high temperature sensitivity, zinc oxide (ZnO), is also reported. Temperature imaging using these tracer particles is demonstrated in a jet (Re = 2,000) heated to 363 K, with a temperature precision of 1%. This extends the capabilities of this versatile technique toward the study of flows with small temperature variations. Also, unlike the majority of phosphors previously investigated for thermometry, this phosphor is a semiconductor. Exploiting the temperature-dependent luminescence of this class of materials presents interesting new opportunities for remote temperature sensing.Open Acces

    Uncertainty and error in laser triangulation measurements for pipe profiling

    Underground pipeline infrastructure often receives insufficient attention and maintenance. Those responsible for ensuring the continuing functionality of this infrastructure primarily use subjective information in their decision making, and standards defining the level of damage acceptable before repair or replacement are difficult to implement. Laser pipe profiling is a relatively new technology that has emerged to take a step toward the objective assessment of buried assets. A laser profiler is a device that traverses a section of pipe, taking measurements of radius around the circumference of the inner pipe wall at multiple locations along the length of the pipe. The accuracy of the measurements obtained by a profiler is a critical piece of knowledge for the evaluation of its usefulness. Analytical measurement and uncertainty models were developed for three laser profiling configurations. These configurations involved a digital camera and a laser whose relative position and orientation were fixed relative to one another. The three configurations included (1) a conically projected laser aligned with the pipe axis, (2) a planar laser placed perpendicular to the pipe axis, and (3) a side-facing laser that projected a line onto the pipe wall parallel to the axis of the pipe. The models utilized normalized system parameters to compute pipe geometry from digital images that reveal the intersection of the laser light and the pipe wall; error propagation techniques were applied to compute the variation in measurement uncertainty as a function of position in the measurement space. Analytical evaluation of the conical projection configuration revealed infinite measurement error for a region of the measurement space; the unbounded error was eliminated by utilizing two conical lasers. The accuracy and uncertainty of the perpendicular plane and side facing configurations were much better than for the conical configuration. Physical models of these two configurations were constructed, and measurements were collected for a pipe section to validate the measurement and uncertainty predictions of the analytical models. The difference between observed worst-case laser measurement error and predicted uncertainty was on the order of 0.1% of nominal pipe radius. This work provides pipe profiler designers the analytical detail required to understand the relationship between system geometry, camera parameters and measurement accuracy. The work provides asset managers with a reference against which to evaluate laser profiling for their infrastructure condition monitoring needs

    Imaging through obscurants using time-correlated single-photon counting in the short-wave infrared

    Single-photon time-of-flight (ToF) light detection and ranging (LiDAR) systems have emerged in recent years as a candidate technology for high-resolution depth imaging in challenging environments, such as long-range imaging and imaging in scattering media. This Thesis investigates the potential of two ToF single-photon depth imaging systems based on the time-correlated single-photon (TCSPC) technique for imaging targets in highly scattering environments. The high sensitivity and picosecond timing resolution afforded by the TCSPC technique offers high-resolution depth profiling of remote targets while maintaining low optical power levels. Both systems comprised a pulsed picosecond laser source with an operating wavelength of 1550 nm, and employed InGaAs/InP SPAD detectors. The main benefits of operating in the shortwave infrared (SWIR) band include improved atmospheric transmission, reduced solar background, as well as increased laser eye-safety thresholds over visible band sensors. Firstly, a monostatic scanning transceiver unit was used in conjunction with a single-element Peltier-cooled InGaAs/InP SPAD detector to attain sub-centimetre resolution three-dimensional images of long-range targets obscured by camouflage netting or in high levels of scattering media. Secondly, a bistatic system, which employed a 32 × 32 pixel format InGaAs/InP SPAD array was used to obtain rapid depth profiles of targets which were flood-illuminated by a higher power pulsed laser source. The performance of this system was assessed in indoor and outdoor scenarios in the presence of obscurants and high ambient background levels. Bespoke image processing algorithms were developed to reconstruct both the depth and intensity images for data with very low signal returns and short data acquisition times, illustrating the practicality of TCSPC-based LiDAR systems for real-time image acquisition in the SWIR wavelength region - even in the photon-starved regime.The Defence Science and Technology Laboratory ( Dstl) National PhD Schem
